NLRP3 inflammasome blockade reduces liver inflammation and fibrosis in experimental NASH in mice.

BACKGROUND & AIMS: NOD-like receptor protein 3 (NLRP3) inflammasome activation occurs in Non-alcoholic fatty liver disease (NAFLD). We used … cluding designation as NASH pathology, and liver fibrosis. In vitro, cholesterol crystals activated Kupffer cells and macrophages to release IL-1β; MCC950 abolished this, and the associated neutrophil migration. MCD diet-fed mice developed fibrotic steatohepatitis; MCC950 suppressed the increase in hepatic caspase 1 and IL-1β, lowered numbers of macrophages and …

Strain-Dependent Effects of Dietary Cholic Acid on Liver Fibrogenesis and Gut Microbiota in TSNO and TSOD Mice.

Background: Metabolic dysfunction-associated steatohepatitis (MASH) is defined by hepatocellular damage accompanied by inflammation and fibrotic changes. Bile acids (BAs) and gut microbiota play pivotal roles in disease progression. However, the contribution of dietary cholic acid (CA), a primary BA, remains unclear. Methods: We investigated the effect of dietary CA supplementation in Tsumura-Suzuki obese diabetic (TSOD) and Tsumura-Suzuki non-obese (TSNO) mouse strains with distinct metabolic phenotypes. The mice were fed normal diet (ND) or 0.5% CA-supplemented ND. Liver injury, fibrosis, an

Virulence attenuation in ST11-K64 Klebsiella pneumoniae explains its divergent clinical manifestation from ST23-K1.

The clinical threat posed by Klebsiella pneumoniae is dual-faceted, encompassing both hypervirulence and carbapenem resistance. The emergence of hypervirulent carbapenem-resistant K. pneumoniae (hv-CRKP) merges these threats, with the ST11-K64 clone being a dominant and concerning lineage. However, its clinical presentation diverges from the classic hypervirulent Klebsiella pneumoniae (hvKp); hv-CRKP is isolated from respiratory sites but is notably absent from pyogenic liver abscesses. This distinct clinical niche prompted us to investigate the underlying pathogenicity differences. Among 847

Impact of Allostatic Load on Chronic Hepatitis B and C: A Narrative Review.

The clinical progression of chronic hepatitis B (HBV) and C (HCV) is highly variable and not fully explained by virological factors alone. This narrative review explores the role of allostatic load, defined as the cumulative physiological burden of chronic stress, as a host factor that may contribute to this heterogeneity by influencing immune dysregulation, hepatic inflammation, and clinical outcomes. We conducted a narrative synthesis of English-language studies identified through PubMed, Web of Science, Scopus, PsycINFO, and gray literature. We included studies (cross-sectional, case-contro

Role of host immunity and HBx among inactive chronic hepatitis B patients in a highly endemic region.

The hepatitis B virus (HBV) infection has a wide range, from fulminant hepatitis to inactive chronic hepatitis B (ICB) infection. The present study evaluated critical factors in the outcomes of HBV infection in a highly endemic region of Iran (approximately 12% HBV positive). The expression of seven genes involved in host immunity (Foxp3, T-bet, ROR-γt, AKT, CREB, IL-28/or IFN-λ2, and IL-28R) and HBx for viral activities were evaluated using real-time PCR, TaqMan method. A total of 58 subjects were randomly chosen, including 28 ICB and 30 healthy controls (HCs) from the Esfandiar district, Sou

Novel Targets and Therapeutic Strategies to Protect Against Hepatic Ischemia Reperfusion Injury.

Hepatic ischemia reperfusion injury (IRI), a fascinating topic that has drawn a lot of interest in the last few years, is a major complication caused by a variety of clinical situations, such as liver transplantation, severe trauma, vascular surgery, and hemorrhagic shock. The IRI process involves a series of complex events, including mitochondrial deenergization, metabolic acidosis, adenosine-5'-triphosphate depletion, Kupffer cell activation, calcium overload, oxidative stress, and the upregulation of pro-inflammatory cytokine signal transduction. A number of protective strategies have been

Hepatic NAD(+) deficiency as a therapeutic target for non-alcoholic fatty liver disease in ageing.

BACKGROUND AND PURPOSE: Ageing is an important risk factor of non-alcoholic fatty liver disease (NAFLD). Here, we investigated whether the deficiency of nicotinamide adenine dinucleotide (NAD(+) ), a ubiquitous coenzyme, links ageing with NAFLD. EXPERIMENTAL APPROACH: Hepatic concentrations of NAD(+) , protein levels of nicotinamide phosphoribosyltransferase (NAMPT) and several other critical enzymes regulating NAD(+) biosynthesis, were compared in middle-aged and aged mice or patients. The influences of NAD(+) decline on the steatosis and steatohepatitis were evaluated in wild-type and H247A

Environmental toxin-linked nonalcoholic steatohepatitis and hepatic metabolic reprogramming in obese mice.

Obesity is associated with strong risks of development of chronic inflammatory liver disease and metabolic syndrome following a second hit. This study tests the hypothesis that free radical metabolism of low chronic exposure to bromodichloromethane (BDCM), a disinfection byproduct of drinking water, causes nonalcoholic steatohepatitis (NASH), mediated by cytochrome P450 isoform CYP2E1 and adipokine leptin. Using diet-induced obese mice (DIO), mice deficient in CYP2E1, and mice with spontaneous knockout of the leptin gene, we show that BDCM caused increased lipid peroxidation and increased tyro

Intragastric ethanol infusion model for cellular and molecular studies of alcoholic liver disease.

The intragastric alcohol infusion rat model (IAIRM) of alcoholic liver disease (ALD) has been utilized in various laboratories to study various aspects of ALD pathogenesis including oxidative stress, cytokine upregulation, hypoxic damage, apoptosis, ubiquitin-proteasome pathway and CYP2E1 induction. The basic value of the model is that it produces pathologic changes which resemble ALD including microvesicular and macrovesicular fat, megamitochondria, apoptosis, central lobular and pericellular fibrosis, portal fibrosis, bridging fibrosis, central necrosis, and mixed inflammatory infiltrate inc

The mechanism of anti-CD20-mediated B cell depletion revealed by intravital imaging.

Since the first description of paraneoplastic pemphigus, several cases have been described in the literature. However, curative therapy is usually a challenge to the physicians treating this disease. Several publications are available discussing the efficacy of steroids, cyclophosphamide and cyclosporin A. Recently, a report of the successful use of rituximab was also published. However, the use of cyclosporin A is controversial in the case of B cell malignancies, as there are reports showing the cytotoxic effect of this drug on B cells. However, other authors report no effect, or even unwante

Hepatoprotective Potential of Curcumin in the Prevention of Liver Dysfunction in a Porcine Model.

Curcumin, the major polyphenolic constituent of Curcuma longa, has been widely investigated as a hepatoprotective adjunct due to its antioxidant and immunomodulatory properties. This review evaluates the relevance of curcumin for the prevention and management of liver dysfunction and hepatitis in pigs by synthesizing available porcine evidence and integrating mechanistic insights from translational liver injury models where pig-specific data remain limited. Across experimental hepatic injury contexts, curcumin administration is most consistently associated with reduced biochemical and structur

Advanced materials for non-alcoholic fatty liver disease and cardiovascular disease comorbidity therapeutics.

Non-alcoholic fatty liver disease (NAFLD) and cardiovascular disease (CVD) frequently co-occur, driven by inter-organ metabolic crosstalk that current single-target therapies fail to disrupt. This review reframes NAFLD-CVD comorbidity through a materials-first lens and advances a translational design framework that couples pathophysiology with engineering principles. We synthesize three major pathological axes, including lipid overflow and impaired reverse cholesterol transport, inflammation-oxidative stress amplification driven by Kupffer cell-NLRP3 and mitochondrial ROS/MAPK signaling, as we

Pharmacokinetics and Biodistribution of Polymeric Nanoparticles Loaded with a Small Molecule Drug in Mice with Alcoholic Liver Disease.

We recently demonstrated the therapeutic efficacy of a novel phosphodiesterase 4B (PDE4B) inhibitor, KVA-D88, loaded in mPEG-b-P(CB-co-LA) polymeric nanoparticles (NPs) for alcohol-associated liver disease (AALD) treatment. In this study, we investigated the pharmacokinetics (PK) parameters and biodistribution at whole body and organ (liver) levels of this nanoformulation compared to the KVA-D88 free drug as well as their in vivo efficacies. Following administration, KVA-D88 was distributed to all major organs, with the highest accumulation in the liver and kidney and the lowest accumulation i

Thromboxane A2/thromboxane A2 receptor axis facilitates hepatic insulin resistance and steatosis through endoplasmic reticulum stress in non-alcoholic fatty liver disease.

BACKGROUND AND PURPOSE: Defective insulin signalling and dysfunction of the endoplasmic reticulum (ER), driven by excessive lipid accumulation in the liver, is a characteristic feature in the pathogenesis of non-alcoholic fatty liver disease (NAFLD). Thromboxane A2 (TXA2 ), an arachidonic acid metabolite, is significantly elevated in obesity and plays a crucial role in hepatic gluconeogenesis and adipose tissue macrophage polarization. However, the role of liver TXA2 /TP receptors in insulin resistance and lipid metabolism is largely unknown. EXPERIMENTAL APPROACH: TP receptor knockout (TP-/-

Hepatic muscarinic acetylcholine receptors are not critically involved in maintaining glucose homeostasis in mice.

OBJECTIVE: An increase in the rate of hepatic glucose production is the major determinant of fasting hyperglycemia in type 2 diabetes. A better understanding of the signaling pathways and molecules that regulate hepatic glucose metabolism is therefore of great clinical importance. Recent studies suggest that an increase in vagal outflow to the liver leads to decreased hepatic glucose production and reduced blood glucose levels. Since acetylcholine (ACh) is the major neurotransmitter of the vagus nerve and exerts its parasympathetic actions via activation of muscarinic ACh receptors (mAChRs), w

An experimental study on fluorocarbons as blood substitutes.

A fluorocarbon emulsion (Fluosol DA 20%) was experimented as a blood substitute. Fourty-five rats weighing 320 +/- 30 g. underwent different degrees of hemodilution, by isovolemic exanguino-transfusions. A first group (15 rats) had maximal exchange transfusion (Ht = 0,5%) with Fluosol DA 20% and was followed for 12 hours. A second group (15 rats) (control group) underwent the same procedure, but hydrolyzed gelatin 3% (Emagel) was used instead of Fluosol DA 20%. A third group (15 rats) had partial exchange-transfusion (Ht = 23 +/- 2%) with Fluosol DA 20%, and was followed for 2 months. Cerebral
