Ocular absorption behavior of palmitoyl tilisolol, an amphiphilic prodrug of tilisolol, for ocular drug delivery.

The objective of this study was to examine the ocular absorption behavior of an amphiphilic prodrug after instillation onto the cornea of rabbits. A micellar solution of O-palmitoyl tilisolol (PalTL), an amphiphilic prodrug, was prepared. After instillation of tilisolol (TL) and PalTL, the drug concentrations in the tear fluid, cornea, aqueous humor, iris-ciliary body, vitreous body, and blood were measured. In addition, in situ ocular absorption behavior was also evaluated. After instillation of TL, the concentration of TL in the tear fluid quickly decreased. After instillation of PalTL, prol

In Vitro Investigations of Acetohexamide Binding to Glycated Serum Albumin in the Presence of Fatty Acid.

The interaction of drugs with human serum albumin (HSA) is an important element of therapy. Albumin affects the distribution of the drug substance in the body, as well as its pharmacokinetic and pharmacodynamic properties. On the one hand, inflammation and protein glycation, directly associated with many pathological conditions and old age, can cause structural and functional modification of HSA, causing binding disorders. On the other hand, the widespread availability of various dietary supplements that affect the content of fatty acids in the body means that knowledge of the binding activity

Fatty acid 2-hydroxylation restrains gastric cancer by blocking CD36 palmitoylation and fatty acid uptake.

Fatty acid (FA)-induced internalization of the FA transporter CD36 normally functions as an intrinsic brake on FA uptake and lipid overload, but cancer cells can bypass this checkpoint to sustain lipid-fueled growth. The mechanism underlying this failure remains unclear. Here, we identify a FA 2-hydroxylase (FA2H)-CD36 signaling axis that operates as an endogenous brake on lipid fueling in gastric cancer cells. FA2H-derived (R)-2-hydroxy palmitic acid ((R)-2-OHPA) disrupts CD36 interaction with its palmitoyltransferase DHHC4, thereby limiting CD36 palmitoylation and trafficking to the plasma m

Lipase from Stenotrophomonas maltophilia strain HO5 for efficient biodiesel synthesis using non-edible plant oils.

The increasing global demand for sustainable and renewable energy has intensified interest in microbial lipases as efficient biocatalysts for biodiesel production. A lipase-producing bacterium isolated from waste-oil-contaminated environments was identified as Stenotrophomonas maltophilia strain HO5 through 16 S ribosomal RNA gene sequencing. The strain exhibited a maximum lipase production of 28 U mL⁻¹. The purified monomeric enzyme (approximately 55 kDa) showed a Michaelis constant (Km) of 1.728 mM and a maximum reaction velocity (Vmax) of 80 U mL⁻¹, indicating high catalytic efficiency. The

Nobiletin Alleviates Npy1r-Mediated Insulin Secretion Deficiency of Islet β-Cells via the Clock-Modulatory Signaling.

Current research indicates that insulin secretion deficiency in β-cells contributes to Type 2 diabetes mellitus (T2DM), which is associated with neuropeptide Y1 receptor (Npy1r) overexpression from neuropeptide Y (NPY) system dysregulation. To date, limited literature has explored nobiletin (NOB) as a circadian modulator for restoring β-cell function through Npy1r regulation. This study investigates NOB's stimulatory effects on insulin secretion via Npy1r and clock-modulatory signaling to elucidate its underlying mechanism. The findings demonstrated that NOB ameliorated hyperglycemia and promo

Broad-Spectrum Antibacterial Activity of Postbiotic From Lacticaseibacillus paracasei BGP1 Against Multidrug-Resistant Skin Wound Pathogens.

The increasing problem of antibiotic resistance indicates the need for alternative therapeutic strategies for skin wound infections. While probiotics exhibit potential for developing such alternatives, the majority of antimicrobial studies focus on live cells or lysates and oral delivery. Notably, in dermatology, formulating products with live strains poses technical challenges due to stability issues in water-based systems. Postbiotics, substances made from probiotics, offer a promising, stable, and safe alternative. This study addresses the gap by evaluating the antibacterial potential of ce

MitoSNO inhibits mitochondrial hydrogen peroxide generation by α-ketoglutarate dehydrogenase.

Here, we demonstrate mitochondrial hydrogen peroxide (mtH2O2) production by α-ketoglutarate dehydrogenase (KGDH) can be inhibited by mitochondria-targeted S-nitrosating agent (MitoSNO), alleviating lipotoxicity. MitoSNO in the nanomolar range inhibits mtH2O2 by ∼50% in isolated liver mitochondria without disrupting respiration, whereas the mitochondria-selective derivative used to synthesize MitoSNO, mitochondria-selective N-acetyl-penicillamine, had no effect on either mtH2O2 generation or oxidative phosphorylation. Additionally, mtH2O2 generation in isolated liver mitochondria was almost abo

From byproduct to bioactive: genetic profiling, GC-MS, and nutritional analysis of citrus peel extracts for nutraceutical development.

The phytochemical, nutritional, and therapeutic qualities of citrus species (Rutaceae) make them significant on a global scale. Despite having a wealth of bioactive compounds with significant commercial and nutraceutical potential, citrus by-products, including peels, seeds, and pulp, are routinely discarded in large amounts. In this study, we examine the nutritional composition, elemental diversity, metabolite profile, and genetic variability of citrus species collected from various regions of Uttarakhand, India. Citrus peels are a good source of calcium (up to 104.10mg/100 g), zinc (58.3 mg/

Characterization of intestinal microbiota in patients with psoriasis combined with metabolic syndrome.

The co-morbidity of psoriasis (PsO) and metabolic syndrome (MetS) is prevalent. While the mechanisms causing PsO in MetS remain unknown, it is understood that gut microbiota plays a critical role in maintaining immune and metabolic balance. However, the specific mechanisms and whether biomarkers are involved in the pathophysiological process of PSO in MetS patients have yet to be elucidated. This study involved 29 patients with PSO and MetS (PM) and 29 with MetS (M). It analyzed their clinical parameters, microbial composition, function, diagnostic, and predictive performance. The results unve

PKD2 regulates autophagy and forms a protein complex with BECN1 at the primary cilium of hypothalamic neuronal cells.

The primary cilium, hereafter cilium, is an antenna-like organelle that modulates intracellular responses, including autophagy, a lysosomal degradation process essential for cell homeostasis. Dysfunction of the cilium is associated with impairment of autophagy and diseases known as "ciliopathies". The discovery of autophagy-related proteins at the base of the cilium suggests its potential role in coordinating autophagy initiation in response to physiopathological stimuli. One of these proteins, beclin-1 (BECN1), it which is necessary for autophagosome biogenesis. Additionally, polycystin-2 (PK

Self-assembling modified neuropeptide S enhances nose-to-brain penetration and exerts a prolonged anxiolytic-like effect.

Anxiety disorders are the most common mental diseases and can greatly disrupt everyday life. Although there has been substantial research on anxiety disorders, novel therapeutics are needed. Neuropeptide S (NPS) is a potential therapeutic candidate owing to its strong anxiolytic activity; however, some disadvantages, such as its poor metabolic stability and inability to cross the blood-brain barrier (BBB), limit its use in the clinic. Herein, inspired by nose-to-brain drug delivery strategies, an endogenous 20-amino-acid-long mNPS peptide was modified by incorporating palmitic acid into its fu

Asiatic acid ameliorates obesity-related osteoarthritis by inhibiting myeloid differentiation protein-2.

Obesity is related to osteoarthritis (OA). Aberrant lipid metabolism results in increased levels of free fatty acids, such as palmitate (PA), leading to inflammatory responses and excess catabolism of chondrocytes. Asiatic acid (AA), a plant anti-inflammatory compound, has been reported to exert protective effects for several diseases, but its effect on obesity-related OA is still unclear. The aim of this study is to evaluate the chondro-protective effect of AA on PA-induced human chondrocytes and a high fat diet (HFD)-fed mouse cartilage degeneration model. In vitro, the levels of the inflamm

Early evolutionary loss of the lipid A modifying enzyme PagP resulting in innate immune evasion in Yersinia pestis.

Immune evasion through membrane remodeling is a hallmark of Yersinia pestis pathogenesis. Yersinia remodels its membrane during its life cycle as it alternates between mammalian hosts (37 °C) and ambient (21 °C to 26 °C) temperatures of the arthropod transmission vector or external environment. This shift in growth temperature induces changes in number and length of acyl groups on the lipid A portion of lipopolysaccharide (LPS) for the enteric pathogens Yersinia pseudotuberculosis (Ypt) and Yersinia enterocolitica (Ye), as well as the causative agent of plague, Yersinia pestis (Yp).
