Primate adult brain cell autotransplantation produces behavioral and biological recovery in 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine-induced parkinsonian St. Kitts monkeys.

The potential for "replacement cells" to restore function in Parkinson's disease has been widely reported over the past 3 decades, rejuvenating the central nervous system rather than just relieving symptoms. Most such experiments have used fetal or embryonic sources that may induce immunological rejection and generate ethical concerns. Autologous sources, in which the cells to be implanted are derived from recipients' own cells after reprogramming to stem cells, direct genetic modifications, or epigenetic modifications in culture, could eliminate many of these problems. In a previous study on

Onchocerciasis: the pre-control association between prevalence of palpable nodules and skin microfilariae.

BACKGROUND: The prospect of eliminating onchocerciasis from Africa by mass treatment with ivermectin has been rejuvenated following recent successes in foci in Mali, Nigeria and Senegal. Elimination prospects depend strongly on local transmission conditions and therefore on pre-control infection levels. Pre-control infection levels in Africa have been mapped largely by means of nodule palpation of adult males, a relatively crude method for detecting infection. We investigated how informative pre-control nodule prevalence data are for estimating the pre-control prevalence of microfilariae (mf)

Regeneration of the infarcted heart with stem cells derived by nuclear transplantation.

Nuclear transfer techniques have been proposed as a strategy for generating an unlimited supply of rejuvenated and histocompatible stem cells for the treatment of cardiac diseases. For this purpose, c-kit-positive fetal liver stem cells obtained from cloned embryos were injected in the border zone of infarcted mice to induce tissue reconstitution. Cloned embryos were derived from somatic cell fusion between nuclei of cultured LacZ-positive fibroblasts and enucleated oocytes of a different mouse strain. We report that regenerating myocardium replaced 38% of the scar at 1 month. The rebuilt tiss

Cytomegalovirus retinitis in the era of highly active antiretroviral therapy.

A number of striking changes have occurred recently in the presentation and course of cytomegalovirus (CMV) retinitis in patients with acquired immunodeficiency syndrome (AIDS) who are receiving highly active antiretroviral therapy (HAART). Before the use of HAART, CMV retinitis was the most common intraocular infection in patients with AIDS, occurring in up to 40% of patients, typically when CD4+ cell counts have decreased to less than 0.10 x 10(9)/L. By studying CMV retinitis, clinicians can investigate whether the rejuvenated immune system that results from HAART can effectively control opp

Oxidative DNA damage and senescence of human diploid fibroblast cells.

Human diploid fibroblast cells cease growth in culture after a finite number of population doublings. To address the cause of growth cessation in senescent IMR-90 human fibroblast cells, we determined the level of oxidative DNA damage by using 8-oxoguanine excised from DNA and 8-oxo-2'-deoxyguanosine in DNA as markers. Senescent cells excise from DNA four times more 8-oxoguanine per day than do early-passage young cells. The steady-state level of 8-oxo-2'-deoxyguanosine in DNA is approximately 35% higher in senescent cells than in young cells. Measurement of protein carbonyls shows that senesc

CRISPR screens identify targets to rescue age-related T cell dysfunction in cancer.

Immune aging is being increasingly recognized as a critical barrier to effective cancer immunotherapy, as the aged tumor microenvironment (TME) drives T cell dysfunction and impairs immune control of cancer. However, the key molecular drivers of this process as well as potential targets to rescue T cell dysfunction in aged tumors remain incompletely understood. Therefore, we performed in vivo single-cell CRISPR screens in CD8 + T cells within aged tumors and tumor-draining lymph nodes (tdLNs). We identified Dusp5 and Zfp219 as key regulators of T cell persistence and effector differentiation i

Mortality associated biological age improves independently of weight loss after bariatric surgery.

Obesity increases the risk of common diseases and mortality, placing a significant burden on our aging society. Bariatric surgery results in significant weight loss; however, the amount of associated health gain is currently less studied, particularly in the first two years. We modelled mortality-associated biological age according to established blood markers in a prospective cohort of 505 patients that underwent bariatric surgery. The difference between biological age and chronological age (age acceleration) as a molecular marker of health gain was evaluated at different time points with mix

"I'm not a volunteer; I'm an equal player": the social factor of quality of life in mixed ability basketball players.

INTRODUCTION: Despite regulatory and conceptual progress in the field of inclusion, people with disabilities continue to face significant barriers which limit their full participation in mainstream sports and affect key social factors of quality of life. Within this context, the Mixed Ability (MA) model represents an alternative where people with disabilities can be included into grassroots sports settings without the need to modify rules or implement classification or identification systems. This study aimed to analyse perceived changes in the social factor of MA basketball players, as well a

IL-12 augments adoptive cell therapy by reshaping CD8+ T dynamics and broadening antitumor immunity.

Adoptive cell therapy (ACT) against solid tumors is constrained by tumor heterogeneity, immunosuppressive microenvironments, and insufficient T cell potency and persistence. Although IL-12 has long been recognized as a potent enhancer of T-cell immunity, its clinical application has been hindered by systemic toxicity and the lack of a strategy to harness its benefits without chronic exposure. Here, we show that ex vivo IL-12 preconditioning programs tumor-specific T cells with increased antigen sensitivity, yielding superior antitumor activity compared with IL-7/IL-15. Systemic delivery of Pro

How bones learn to listen: In vivo models for investigating mechanosensitivity at bony interfaces in the ageing mouse.

Beyond the well-documented loss of bone volume, the aged skeletal environment is marked by structural decay and the degenerated network of osteocytes, the bone cells responsible for orchestrating bone formation and resorption. Recent findings point towards the loss of mechanosensitivity and thus the 'deafened' adaptive response to mechanical loading as the driver of decreased bone formation. This imbalance results in a net loss of bone mass and, over time, increases skeletal fragility and fracture incidence. Although the responsiveness of bones to mechanical loading is typically diminished wit

Integrated multi-tissue transcriptomic analysis identifies hub genes associated with egg production recovery after fasting-induced molting.

Fasting-induced molting is widely employed in the poultry industry to rejuvenate the reproductive system of aging laying hens, thereby improving post-molt laying performance and extending productive lifespan. However, substantial individual variation in post-molt reproductive recovery may compromise the overall effectiveness of molting programs. Elucidating the molecular basis underlying this variation is therefore important for poultry production and genetic improvement. In this study, laying hens were classified into high-recovery (HR) and low-recovery (LR) groups according to their egg prod

Immunosenescence and Bone Homeostasis: From Mechanisms of Homeostasis Disruption to Therapeutic Opportunities in Age-Related Skeletal Disorders.

The progressive decline in immune function during aging, termed immunosenescence, is increasingly recognized as a critical driver of skeletal fragility and impaired bone regeneration. This age-associated phenomenon-driven by thymic involution, inflammaging, and the accumulation of senescent immune cells-disrupts bone homeostasis primarily through the establishment of a pro-inflammatory milieu, wherein senescence-associated secretory phenotype (SASP) factors directly reprogram the function and fate of mesenchymal stem cells, osteoblasts, osteoclasts, and chondrocytes. Clinically, this immune-dr

The mitochondrial logic of inflammaging: how energy imbalance drives fibroblast SASP and tissue-specific aging.

The conversion of metabolic disequilibrium into chronic inflammatory signaling represents a central and actively investigated question in ageing biology. Among stromal cells, fibroblasts are key effectors of tissue remodeling and inflammation, acquiring a senescence-associated secretory phenotype (SASP) that sustains age-related pathology. Here, we delineate a mechanistic framework in which disruption of energy homeostasis drives mitochondrial dysfunction, innate immune activation, and SASP secretion. Mitochondria act as metabolic sentinels that sense energetic stress through altered AMP/ATP a

Biomarkers of aging as it relates osteoarthritis: we can't improve what we can't measure.

Aging is the largest risk factor for the development of osteoarthritis (OA), a major contributor to increased years lived with disability. This review reflects on how age-related changes relevant to OA have been measured at various length scales. Key discoveries include increased chondrocyte DNA damage with age and the disruption of matrix homeostasis by cellular senescence. Epigenetic clocks have yet to show predictive value for OA, while transcriptomic changes and miRNA profiles are linked to aging and senescence. Protein biomarkers have gained traction in the context of post-traumatic OA an
