Repeat cesarean delivery with hysterectomy: a sheep model to examine uterine scars.

ABSTRACT: Cesarean delivery (CD) is one of the most common surgical procedures worldwide, with rising rates leading to an increasing impact of complications, such as uterine rupture during a trial of labor in a subsequent pregnancy. To date, only little information on uterine wound healing is available. The current challenge is to establish a suitable model and to define quantifiable outcome parameters of uterine scar tissue with the aim of comparing the effect of different surgical techniques on uterine scarring. This study presents a sheep model to examine the effects of different hysterotom…

Establishment of an ELISA for detecting oocyst-derived Toxoplasma gondii infections in sheep.

BACKGROUND: Ovine toxoplasmosis is a zoonotic disease that severely impacts the development of the sheep industry. The primary routes of Toxoplasma gondii infection in sheep are cyst infection and oocyst infection. However, current technologies are unable to distinguish between these two infection pathways. METHODS: In this study, we initially screened eight proteins that are highly specifically expressed during the oocyst stage. Through Western blot (WB) analysis, we identified a protein (TGME49_267410) that could serve as a diagnostic antigen. Subsequently, we optimized the conditions for a…

Integrated 16S rRNA and Metagenomic Analysis of Pulmonary Microbiota in Sheep with Pneumonia.

Sheep are a major livestock species in China, yet pneumonia-related mortality poses a significant obstacle to intensive farming. In this study, 115 sheep lung samples were collected and classified into different pneumonia severity groups based on lung lesion scoring. Subsequently, this study employed 16S rRNA sequencing to systematically investigate the structure and diversity of the pulmonary microbiota in sheep, including alpha diversity, beta diversity, and LEfSe analyses. Metagenomic techniques were also applied to analyze the abundance of metabolic pathways, exploring the associations bet…

A Wolf in Sheep's Clothing: Laryngeal Myxoma Masquerading as a Vocal Cord Polyp.

Myxoma is a rare benign tumor of mesenchymal origin and is very rarely seen in the larynx. Because of its appearance, it is often mistaken for a vocal cord polyp. We report the case of a 19-year-old female who presented with hoarseness of voice for 1 month. Laryngoscopic examination showed a small polypoidal lesion on the vocal cord. The lesion was surgically removed. Histopathological examination showed sparse spindle-shaped cells in abundant myxoid stroma without atypia or mitotic activity, confirming the diagnosis of myxoma. Complete excision was curative. This case highlights the importanc

Engraftment of sheep splenic lymphocytes into NBSGW mice and application in Brucella infection.

BACKGROUND: The development of immune reconstituted xenograft mouse models has addressed the limitations imposed by interspecies immunological differences in disease research. However, mouse models reconstituted with a sheep immune system remain largely unexplored. METHODS: In this study, we successfully established a sheep splenocyte-reconstituted mouse model (Sh-Spl mice) by intravenously transplanting ovine splenocytes into severely immunodeficient NBSGW mice, and evaluated the efficiency of immune reconstitution. RESULTS: Results showed that ovine-derived CD45+, CD4+, and CD8+ cells were…

Comprehensive analysis of the transcriptome-wide m6A Methylome in sheep testicular development.

N6-methyladenosine (m6A) modification of RNA is a critical post-transcriptional modification, that dynamically contributes to testicular development and spermatogenesis. Nevertheless, the investigation into the role of m6A in testicular development of sheep remains insufficient. Herein, we conducted a comprehensive analysis of the m6A transcriptome landscape in the testes of F1 hybrid Southdown × Hu sheep across M0 (0 months old, newborn), M3 (3 months old, sexually immature), M6 (6 months old, sexually mature), and Y1 (1 years old, adult). By profiling the m6A signatures across the transcript…

The Autocrine Impact of Nerve Growth Factor on Sheep Uterine Epithelial Cells.

Nerve growth factor (NGF) plays a critical role in reproduction through paracrine and endocrine mechanisms. However, its autocrine effects on uterine receptivity and inflammatory pathways remain unknown. This study is the first to demonstrate NGF's direct autocrine action on sheep endometrial luminal epithelial cells (SELECs), primary cultures treated with NGF for 12, 24, and 48 h, with or without the NTRK1 antagonist. NGF significantly increased PGE2 (p < 0.0001) and PGF2α (p < 0.0001) levels only at 12 h, with no significant changes at 24 and 48 h. NGF also upregulated the expression of NGF,…

Epidemiological and molecular study of Theileria spp. in sheep and goats in Erbil, Iraq.

Theileria spp. are a group of hemoprotozoal parasites pose a significant threat to small ruminants in tropical and subtropical regions of the world. Because of that and due to the limited data on the genetic diversity of Theileria species in Erbil Governorate, northern Iraq, this study was conducted to assess the prevalence of Theileria spp. in sheep and goats using both Giemsa-stained blood smears and PCR methods as a diagnostic criterion. The overall infection rate detected by blood smears was 26.2% in sheep and 18.3% in goats, while PCR showed higher infection rates, 47.7% in sheep and 38.3…

Single-cell RNA sequencing reveals a cellular atlas of the sheep testis.

Spermatogenesis is a highly regulated biological process involving various cell types, making the accurate identification and classification of different cell populations challenging. Single-cell RNA sequencing (scRNA-seq) has emerged as a powerful tool for resolving cellular heterogeneity at high resolution. In this study, 10× Genomics Chromium™ scRNA-seq was employed to analyze 15,826 testicular cells from three 6-month-old sexually mature Hu sheep, and a high-resolution single-cell transcriptomic atlas was generated. Specifically, six somatic cell subtypes and five germ cell subtypes were i…

Sheep femoral artery occlusion is well tolerated and does not result in ischemia.

OBJECTIVE: Sheep are commonly used as large animal pre-clinical models for investigating cardiovascular therapies, interventions, anatomy and physiology. Further, novel small diameter vascular grafts are frequently tested via implantation into sheep carotid arteries (CAs). This is because, unlike humans, acute occlusion of one or both sheep CAs is not associated with morbidity or mortality and thus provides safer experimental testing, with reduced ethical constraints, animal numbers and costs. However, to date there has been no evidence regarding sheep tolerance of femoral artery (FA) occlusio…

The effects of different silage types on rumen bacteria and metabolites in Tibetan sheep.

BACKGROUND: The objective of this study was to examine the relationship between two different types of silage (saline; SQ and non-saline; NSQ) and rumen metabolism in Tibetan sheep. RESULTS: It was found that sheep in the saline silage (SQ group) exhibited superior apparent quality (e.g., crude protein, crude fat, DM, crude polysaccharide, polyphenols, and flavonoid), secondary metabolite profiles (e.g., Proline, Leucine, Alanine, Pyruvate), and fermentation microbial populations (e.g., Firmicutes, Bacteroidota, and Lactobacillus) compared to the non-saline (NSQ) silage group. However, the hi…

Status of Selenium and Other Essential and Toxic Elements in Oregon Grazing Sheep.

Mineral imbalances in sheep can have significant health and economic consequences, yet regional assessments of trace mineral and toxic element status are limited. Oregon is an endemically selenium (Se)-deficient region, but a statewide evaluation has not been conducted. This study assessed whole blood concentrations of 18 elements in 370 clinically normal ewes from 56 farms across Oregon to determine the prevalence of deficiencies and potential toxic exposures. A deficiency threshold of <120 ng Se/mL whole blood was used. We found that 28% of animals were Se-deficient, and 27% of farms had an …

Comparison of Dexmedetomidine/Morphine and Xylazine/Morphine as Premedication in Isoflurane-Anesthetized Sheep.

In this study we investigated the sedative, anesthetic, and pulmonary histopathologic effects of dexmedetomidine/morphine (DM) and xylazine/morphine (XM) in sheep. We hypothesized that DM would provide profound sedation and better maintain physiologic parameters under anesthesia than XM in sheep undergoing laparoscopic surgery. Nineteen male sheep were premedicated with either DM (dexmedetomidine [0.006 mg/kg] and morphine [0.3 mg/kg]) or XM (xylazine [0.1 mg/kg] and morphine [0.3 mg/kg]). After DM or XM administration, 3 blinded veterinarians evaluated sedation scores (0 [no sedation], 1 [mil…

Seroprevalence of malignant catarrhal fever in cattle and sheep from South Brazil.

Malignant Catarrhal Fever (MCF) is caused by a group of gammaherpesviruses in the genus Macavirus. The virus is transmitted via nasal shedding from adapted hosts, such as sheep, which do not develop the disease, to non-adapted hosts such as cattle, pigs, bison, and deer, which can develop an often-fatal form of the disease. There is currently no specific treatment or vaccine available to control MCF, and the resulting losses significantly affect livestock industries worldwide. To assess the seroprevalence of MCF in southern Brazil, blood samples were collected from sheep and cattle reared simu…

Sexually dimorphic impact of prenatal hyperandrogenism on offspring growth trajectory in sheep.

In a translationally relevant ovine model of gestational hyperandrogenism we have previously reported cardiometabolic disruption in female offspring (males were not extensively studied). We hypothesized that gestational hyperandrogenism would lead to sex-specific disruption in offspring's growth, cytokine and metabolic milieu, potential mediators of cardiometabolic disease (CMD). 100 mg Testosterone propionate (T) or vehicle (C) was administered intramuscularly twice weekly between gestational days (GD) 30-90. Two cohorts of offspring were generated, including both males and females. Fetal wei
