Antifertility effect of ethanolic extract of Juniperus phoenica (L.) in male albino rats.

The plant Juniperus phoenica (L.) (Cupressaceae) is widely growing on the rocky soils of the Mediterranean regions. In Jordan, the plant is distributed in different locations and is used as a folk medicine to treat rheumatism, edema, and urinary tract diseases. This study aimed to investigate the antifertility effect of J. phoenica in male albino rats. Animals were administered single daily intraperitoneal injections of 400 or 800 mg/kg of J. phoenica cones ethanol extract or the vehicle (dimethyl sulfoxide) for 21 consecutive days. A marked dose-dependent decrease in the counts and motility o

Paternal High-Fat Diet Altered Sperm 5'tsRNA-Gly-GCC Is Associated With Enhanced Gluconeogenesis in the Offspring.

Background: Paternal lifestyle, stress and environmental exposures play a crucial role in the health of offspring and are associated with non-genetic inheritance of acquired traits, however the underlying mechanisms are unclear. In this study, we aimed to find out how the sperm tsRNA involved in paternal high-fat diet induced abnormal gluconeogenesis of F1 offspring, and explore the underlying molecular mechanism of its regulation. Method: We generated a paternal high fat diet (42% kcal fat) model to investigate the mechanism by which paternal diet affects offspring metabolism. Four-week-old C

Variation in post-thaw sperm quality of white-tailed deer bucks (Odocoileus virginianus) during rut.

White-tailed deer farming is a growing industry in the United States, with breeding operations contributing significantly to the industry's economic impact. Artificial insemination with frozen semen allows for selection and dissemination of valuable genetics, yet surprisingly little is known regarding the best time throughout rut to perform semen cryopreservation. The objective of this study was to compare semen quality following cryopreservation of white-tailed deer bucks collected early in the breeding season (September, n = 6), at peak rut (December, n = 8), and late season (March, n = 7).

Functional characterization of MrOrb in Macrobrachium rosenbergii reveals its crucial role in spermatogenesis.

Orb proteins, members of the cytoplasmic polyadenylation element-binding (CPEB) family, play critical roles in germline development in various organisms. Studies in Drosophila melanogaster have indicated that Orb genes are essential for oocyte development, and are crucial for spermatogenesis, including meiotic completion and spermatid differentiation. However, their role in crustacean spermatogenesis remains largely unexplored. This study investigates the function of an Orb homolog in the crustacean freshwater prawn Macrobrachium rosenbergii. We identified and characterized MrOrb, revealing it

Histological intersex (ovotestis) in the European lobster Homarus gammarus and a commentary on its potential mechanistic basis.

This paper provides the first report of the intersex (ovotestis) condition in the European lobster Homarus gammarus. A single specimen (10% of males sampled) presenting the condition was discovered as part of routine sampling, from the Weymouth Bay region of the English Channel, UK. The lobster presented externally as a male, but upon histological examination was seen to contain an ovotestis, containing elements of both male and female gonadal tissue. Previtellogenic oocytes were present in several otherwise normal seminiferous tubules throughout the testis. The seminiferous tubules were also

Oocyte quality and treatment outcome in intracytoplasmic sperm injection cycles of polycystic ovarian syndrome patients.

Patients with polycystic ovarian syndrome (PCOS) have higher miscarriage rates. It is postulated that this is caused by a lower rate of mature oocytes, and a lower quality of embryos. Retrospectively we analysed 51 intracytoplasmic sperm injection (ICSI) cycles of 31 PCOS patients. These data were compared to age-matched controls (105 cycles) during the same period. All patients of both groups received gonadotrophin-releasing hormone (GnRH) agonists prior to gonadotrophin treatment. The rate of metaphase II oocytes (MII) was not different. However, the mean absolute number of normally fertiliz

Varicocoele-associated male infertility: Cellular and molecular perspectives of pathophysiology.

BACKGROUND: Varicocoele is a common risk factor associated with reduced male fertility potential. The current understanding of varicocoele pathophysiology does not completely explain the clinical manifestation of infertility. The present treatment options such as antioxidant supplementation and varicocoelectomy only help ≈35% of men to achieve spontaneous pregnancy. OBJECTIVE: This review aims to summarize the available knowledge on cellular and molecular alterations implicated to varicocoele-associated male infertility and also highlights the new knowledge generated by "omics" technologies.

Study on the mechanism of different doses of lycium barbarum polysaccharides affecting sperm motility in Drosophila melanogaster.

Lycium barbarum, a Solanaceae plant, yields bioactive polysaccharides (LBP) in its dried mature fruits. LBP has demonstrated protective effects on the reproductive system. This study took Drosophila melanogaster as the experimental subject to investigate the dose-dependence of LBP on male reproductive capacity. The flies were orally administered LBP (0-2000 mg/L) by the CAFE method. Twenty-four hours later, sperm motility, ovulation rate and testicular gene expression were evaluated. Key results: LBP modulates sperm motility and female ovulation rate in a dose-dependent manner. The dose-respon

KIF2C Deletion Causes Meiotic Abnormalities and Nonobstructive Azoospermia in Mice.

PURPOSE: Kinesin Family Member 2C (KIF2C) is a key regulator of microtubule dynamics and chromosome segregation in mitosis. However, its role in spermatogenesis remains unclear. Recent transcriptomic analyses suggest a potential link between KIF2C and male infertility. This study aimed to clarify KIF2C's roles in spermatogenesis using Kif2c knockout (KO) mice. METHODS: To overcome the preweaning lethality associated with Kif2c deletion, we generated Kif2c KO mice with a mixed genetic background of 129X1/SvJ and B6D2. We assessed male fertility, epididymal sperm counts, and testicular sections

Cryopreservation effect on DNA methylation profile in rainbow trout spermatozoa.

Spermatozoa are the cells that are most commonly used for cryopreservation of valuable genetic resources in aquaculture. It is known that fish spermatozoa transmit to the embryo not only their genetic but also their epigenetic profile, especially DNA methylation. Therefore, any alteration of the DNA methylation profile in spermatozoa induces the risk of transmitting epigenetic alterations to the offspring. The aim of this study was to assess the effect of cryopreservation on DNA methylation in rainbow trout spermatozoa. To trigger variable cellular response after freezing-thawing, spermatozoa

The Cannabinoid Receptor CB1 Stabilizes Sperm Chromatin Condensation Status During Epididymal Transit by Promoting Disulphide Bond Formation.

The cannabinoid receptor CB1 regulates differentiation of spermatids. We recently characterized spermatozoa from caput epididymis of CB1-knock-out mice and identified a considerable number of sperm cells with chromatin abnormality such as elevated histone content and poorly condensed chromatin. In this paper, we extended our findings and studied the role of CB1 in the epididymal phase of chromatin condensation of spermatozoa by analysis of spermatozoa from caput and cauda epididymis of wild-type and CB1-knock-out mouse in both a homozygous or heterozygous condition. Furthermore, we studied the

Regulation of apical blebbing in the porcine epididymis.

Apical blebbing, a non-classical secretion mechanism, occurs in the mature porcine epididymis as part of its normal function. Proteins secreted by this mechanism contribute to the modification of the sperm plasma membrane during epididymal transit and are thought to contribute to acquisition of fertilizing ability. However, little is known about the regulation of this secretion mechanism in an in vivo model. Previous work demonstrated apical blebbing in the epididymis developed pubertally, suggesting androgens, sperm or other luminal factors regulated this process. Hence, the objective was to

SLC9A3 Protein Is Critical for Acrosomal Formation in Postmeiotic Male Germ Cells.

Solute carrier family 9 isoform 3 (SLC9A3), a Na⁺/H⁺ exchanger, regulates the transepithelial absorption of Na⁺ and water and is primarily expressed on the apical membranes of the intestinal epithelium, renal proximal tubule, epididymis, and vas deferens. Loss of the Slc9a3 allele in mice enhances intestinal fluid and causes diarrhoea as a consequence of diminished Na⁺ and HCO₃- absorption. Hence, the loss also causes male infertility and reveals the abnormal dilated lumen of the rete testis and calcification in efferent ductules. However, whether loss of Slc9a3 alleles also disrupts mammalian

Unilateral orchidectomy in donkey (Equus asinus): Evaluation of different surgical techniques, histological and morphological changes on remaining testis.

Unilateral orchidectomy (UO) is required when further breeding potential is important. It is sometimes necessary to remove a single testis in a matured stallion for therapeutic reasons. In this study, twelve donkeys were used to evaluate three techniques of unilateral castration, histological and morphological changes on the remaining testis. Results of the study showed that each of the surgical techniques used had its advantages and disadvantages in comparison with the other two techniques. Therefore the selection among the three techniques depends on the surgeon preferences and the environme

Antifertility effect of Tinospora cordifolia (Willd.) stem extract in male rats.

Oral administration of 70% methanolic extract of T. cordifolia stem to male rats at the dose level of 100 mg/rat/day for 60 days did not cause body weight loss but decreased the weight of testes, epididymis, seminal vesicle and ventral prostate in a significant manner. Sperm motility as well as sperm density were reduced significantly which resulted in reduction of male fertility by 100%. The stem extract brought about an interference with spermatogenesis. The round spermatids were decreased by 73.12%. However, the population of preleptotene and pachytene spermatocytes were decreased by 47.60%

Mechanism of Shu-Mu Brain-Kidney Acupoint in Treating Oligoasthenospermia in Rats via the Hypothalamic-Pituitary-Testicular Axis.

Oligoasthenospermia is a common cause of male infertility. The hypothalamic-pituitary-testicular (HPT) axis regulates gonadal differentiation and maturation through reproductive hormone synthesis and release, playing a vital role in male fertility. Disrupting HPT axis stability impairs sperm production, reducing semen quality. Investigating electroacupuncture's effect on HPT axis regulation may provide insights into treating oligoasthenospermia. Fifty 8-week-old male SD rats were randomly divided into blank, model, Shu-Mu Brain-Kidney, non-acupoint, and L-carnitine groups (n = 10 per group). E

Cracking the code: how piRNA pathway shapes spermatogenesis and combats male infertility.

Male infertility, accounting for approximately 50% of global infertility cases, is a growing concern in reproductive medicine. A fundamental cause lies in disrupted spermatogenesis-a complex, highly regulated process involving mitotic proliferation, meiotic division, and spermiogenic remodeling. Among the key regulatory pathways, PIWI-interacting RNAs (piRNAs) and their associated PIWI proteins have emerged as essential players in maintaining germline genome integrity and ensuring successful sperm development. However, their clinical relevance remain underexplored. This review provides a compr
