A knockout screen of genes expressed specifically in Ae. aegypti pupae reveals a critical role for stretchin in mosquito flight.

Aedes aegypti is a critical vector for transmitting Zika, dengue, chikungunya, and yellow fever viruses to humans. Genetic strategies to limit mosquito survival based upon sex distortion or disruption of development may be valuable new tools to control Ae. aegypti populations. We identified six genes with expression limited to pupal development; osi8 and osi11 (Osiris protein family), CPRs and CPF (cuticle protein family), and stretchin (a muscle protein). Heritable CRISPR/Cas9-mediated gene knockout of these genes did not reveal any defects in pupal development. However, stretchin-null mutati

Control of Phlebotomus argentipes (Diptera: Psychodidae) sand fly in Bangladesh: A cluster randomized controlled trial.

BACKGROUND: A number of studies on visceral leishmaniasis (VL) vector control have been conducted during the past decade, sometimes came to very different conclusion. The present study on a large sample investigated different options which are partially unexplored including: (1) indoor residual spraying (IRS) with alpha cypermethrin 5WP; (2) long lasting insecticide impregnated bed-net (LLIN); (3) impregnation of local bed-nets with slow release insecticide K-O TAB 1-2-3 (KOTAB); (4) insecticide spraying in potential breeding sites outside of house using chlorpyrifos 20EC (OUT) and different c

Evaluation of efficacy of 18 strains of entomopathogenic nematodes (Rhabditida) against Planococcus citri (Risso, 1813) (Hemiptera: Pseudococcidae) under laboratory conditions.

Planococcus citri (Risso, 1813) (Hemiptera: Pseudococcidae) is an important plant virus vector in grapevine crops in Brazil and other countries. The mealybug grows in roots and leaves of the grapes. Entomopathogenic nematodes (EPNs) are efficient control agents against insects associated to the soil and could be applied with the same equipment used for chemical insecticides. The aim of this study was to select effective EPNs for controlling P. citri females in laboratory conditions (25±1°C, UR 60±10%). We tested 17 native [Steinernema rarum (6 strains), Steinernema glaseri, Steinernema feltiae

Improved chemical control of Chagas disease vectors in the dry Chaco region.

The effectiveness of two doses of suspension concentrate (SC) pyrethroid insecticides in suppressing peridomestic populations of Triatoma infestans (Klug) was evaluated in 28 rural communities located in Santiago del Estero province, northwestern Argentina, including 388 houses and 1,516 identified sites. Four treatments were randomly assigned to peridomiciles within each community: 5% SC beta-cypermethrin at standard (S, 50 mg active ingredient [AI]/m2) and double dose (2S), and 2.5% SC deltamethrin at standard (D, 25 mg [AI]/m2) and double dose (2D). Simultaneously, we assessed the effects o

[Twenty-year surveillance of insects relevant to public health during the construction of hydroelectric facilities in Antioquia, Colombia].

INTRODUCTION: Entomological studies conducted in large hydroelectric infrastructure projects are a tool for the prevention and control of vector-borne diseases. These diseases emerge as a consequence of changes made to the terrain that often increase the natural and artificial mosquito larval habitats. Many of these insects are of public health importance and population increases result in an increased risk of disease transmission. OBJECTIVE: The culicine (mosquito) and phlebotomine (sand fly) populations were characterized in the area of the Porce II and Porce III hydroelectric projects of A

Serological markers of sand fly exposure to evaluate insecticidal nets against visceral leishmaniasis in India and Nepal: a cluster-randomized trial.

BACKGROUND: Visceral leishmaniasis is the world' second largest vector-borne parasitic killer and a neglected tropical disease, prevalent in poor communities. Long-lasting insecticidal nets (LNs) are a low cost proven vector intervention method for malaria control; however, their effectiveness against visceral leishmaniasis (VL) is unknown. This study quantified the effect of LNs on exposure to the sand fly vector of VL in India and Nepal during a two year community intervention trial. METHODS: As part of a paired-cluster randomized controlled clinical trial in VL-endemic regions of India and

Preliminary evidence that synanthropic flies contribute to the transmission of trachoma- causing Chlamydia trachomatis in Latin America.

Synanthropic flies have been shown to be important mechanical vectors of Chlamydia trachomatis, which causes trachoma. However entomological studies have not been forthcoming in Latin America. This study assesses the relationship between household dipteran fly densities and active childhood trachoma in a village on Marajó Island, Pará state, Brazil. For 78 households, members were examined for signs of trachoma, relative abundance of potential trachoma vectors (Diptera, Chloropidae and Diptera, Muscidae) was quantified by trap counts, and additional measures of household hygiene associated wit

Effect of anti-mosquito hemolymph antibodies on fecundity and on the infectivity of malarial parasite Plasmodium vivax to Anopheles stephensi (Diptera:Insecta).

Rabbit antibodies to hemolymph antigens (102.5, 101, 100, 96, 88, 80, 64, 55, 43, 29, and 23 kDa) of Anopheles stephensi reduced fecundity as well as viability in An. stephensi. However, ingestion of these antibodies was not associated with a marked effect on the engorgement of mosquitoes but egg laying was significantly delayed. Antisera raised against hemolymph proteins were also used to identify cross reactive antigens/epitopes present in other tissues by Western blotting, as well as by in vivo ELISA. In addition, a significant reduction in oocyst development was also observed in An. stephe

The evolution of ocular onchocerciasis in the Volta River Basin Area over a period of five years of vector control.

The results of an ophthalmological evaluation conducted in seven West African savannah villages before and after 5 years of vector control, were analysed to determine the effect of an interrupted or greatly reduced transmission of Onchocerca volvulus on the evolution of ocular onchocerciasis. Cross-sectional data showed a significant reduction of the prevalence of ocular onchocerciasis in five of the villages, and the rates of irreversible ocular lesions and blindness were generally lower after 5 years of vector control. A longitudinal study of a defined population showed that the ocular statu

Broadening the application of evolutionarily based genetic pest management.

Insect- and tick-vectored diseases such as malaria, dengue fever, and Lyme disease cause human suffering, and current approaches for prevention are not adequate. Invasive plants and animals such as Scotch broom, zebra mussels, and gypsy moths continue to cause environmental damage and economic losses in agriculture and forestry. Rodents transmit diseases and cause major pre- and postharvest losses, especially in less affluent countries. Each of these problems might benefit from the developing field of Genetic Pest Management that is conceptually based on principles of evolutionary biology. Thi

An experience in the control of plague in Botswana.

In October 1989, cases of human plague started to occur in Boteti district of Botswana. One hundred and seventy cases were recorded in total from 15 October 1989 to 6 April 1990. There were 12 deaths, giving a case fatality rate of 7%. All cases were of the bubonic type. A concerted control strategy based on sentinel surveillance, involving public education, flea and rodent control, chemotherapy and chemoprophylaxis controlled the epidemic.

Ectoparasites of medical and veterinary importance: drug resistance and the need for alternative control methods.

OBJECTIVES: Despite multiple attempts at eradication, many ectoparasites of humans and domestic livestock remain a persistent problem in the modern world. For many years, a range of pesticide drugs including organophosphates, organochlorides and synthetic pyrethroids provided effective control of these parasites; but intensive use of these drugs has led to the evolution of resistance in many target species. This paper aims to review the effectiveness of current control methods and discuss potential alternatives for the long term sustainable control of ectoparasites. KEY FINDINGS: Important me

Pathogen survival in the external environment and the evolution of virulence.

Recent studies have provided evolutionary explanations for much of the variation in mortality among human infectious diseases. One gap in this knowledge concerns respiratory tract pathogens transmitted from person to person by direct contact or through environmental contamination. The sit-and-wait hypothesis predicts that virulence should be positively correlated with durability in the external environment because high durability reduces the dependence of transmission on host mobility. Reviewing the epidemiological and medical literature, we confirm this prediction for respiratory tract pathog

Fresh, dried or smoked? Repellent properties of volatiles emitted from ethnomedicinal plant leaves against malaria and yellow fever vectors in Ethiopia.

BACKGROUND: In the search for plant-based mosquito repellents, volatile emanations were investigated from five plant species, Corymbia citriodora, Ocimum suave, Ocimum lamiifolium, Olea europaea and Ostostegia integrifolia, traditionally used in Ethiopia as protection against mosquitoes. METHODS: The behaviour of two mosquitoes, the malaria vector Anopheles arabiensis and the arbovirus vector Aedes aegypti, was assessed towards volatiles collected from the headspace of fresh and dried leaves, and the smoke from burning the dried leaves in a two-choice landing bioassay and in the background of

Do anthropogenic transports facilitate stored-product pest moth dispersal? A molecular approach.

Stored-product moths cause large economic damage in food processing industries and storage facilities. Control of indoor pests is currently dealt with locally, and control strategies seldom include different mills or cooperative industries in joint efforts to reduce infestations. In colder climates where conditions hinder flight dispersal of stored-product moths, we hypothesize that human transport between mills will facilitate dispersal. Albeit considered intuitive, this hypothesis has so far never been tested. Male moths from three mills (populations) in southern Sweden and Denmark were coll

RNA interference-based antiviral immune response against the salivary gland hypertrophy virus in Glossina pallidipes.

BACKGROUND: Glossina pallidipes salivary gland hypertrophy virus (GpSGHV; Hytrosaviridae) is a non-occluded dsDNA virus that specifically infects the adult stages of the hematophagous tsetse flies (Glossina species, Diptera: Glossinidae). GpSGHV infections are usually asymptomatic, but unknown factors can result to a switch to acute symptomatic infection, which is characterized by the salivary gland hypertrophy (SGH) syndrome associated with decreased fecundity that can ultimately lead to a colony collapse. It is uncertain how GpSGHV is maintained amongst Glossina spp. populations but RNA inte

High Pyrethroid Resistance in Triatoma infestans Is Strongly Associated with kdr Variants: A Multiplex High-Resolution Melting Tool for Vector Surveillance.

Triatoma infestans is the main vector of Trypanosoma cruzi in the Southern Cone, where vectorial transmission of Chagas is still occurring despite control campaigns with pyrethroid insecticides. High pyrethroid resistance was detected in T. infestans populations from Argentina and Bolivia. This raises the urgent need for including strategies for monitoring and management of insecticide resistance as an essential component of control campaigns. Voltage-gated sodium channels are the target site of pyrethroids; non-silent single-nucleotide polymorphisms in specific sites of the gene encoding this

Worldwide Status of Insecticide Resistance of Aedes aegypti and Ae. albopictus, Vectors of Arboviruses of Chikungunya, Dengue, Zika and Yellow Fever.

BACKGROUND: Controlling of Aedes aegypti and Ae. albopictus, vectors of five important mosquito-borne diseases, is known as the most effective method to prevent the transmission of arboviruses to humans, but the emergence of insecticide resistance is threat for control and prevention of vector borne diseases. A better understanding of mosquito resistance to insecticides will help to develop more effective methods to control insecticide resistance in mosquito vectors. High Triatoma brasiliensis Densities and Trypanosoma cruzi Prevalence in Domestic and Peridomestic Habitats in the State of Rio Grande do Norte, Brazil: The Source for Chagas Disease Outbreaks?

AbstractA total of 2,431 Triatoma brasiliensis were collected from 39 populations of Paraíba (PB) and Rio Grande do Norte (RN) states, Brazil. In PB, Trypanosoma cruzi infection was not detected in either peridomestic or domestic vector populations. In contrast, in RN, T. brasiliensis was detected with high parasite prevalence in these ecotopes (30.7-40.0%). Moreover, peridomicile insect population densities were more than double the average densities of all other settings evaluated (19.17 versus < 8.94 triatomine/man-hour). Genotyped parasites evidenced a mix of T. cruzi lineages circulating

CRISPR/Cas9-Mediated Knockout of White Gene Produces Eye Color Mutants in the Southern Green Stinkbug, Nezara viridula.

CRISPR/Cas9 gene editing is a transformative tool for genetic studies in non-model organisms like the southern green stinkbug, Nezara viridula. However, current protocols depend on embryonic microinjection of CRISPR/Cas9, which remains technically difficult. An alternative method of delivering Cas9 ribonucleoprotein directly into female ovaries has been tested in only a few insect species, such as mosquitoes and whiteflies. Here, we developed a simple technique for gene editing by injecting Cas9 ribonucleoprotein into adult Nezara viridula females using ReMOT control or BAPC delivery methods p
