Delta class glutathione S-transferase (TuGSTd01) from the two-spotted spider mite Tetranychus urticae is inhibited by abamectin.

GSTs (Glutathione S-transferases) are known to catalyze the nucleophilic attack of the sulfhydryl group of reduced glutathione (GSH) on electrophilic centers of xenobiotic compounds, including insecticides and acaricides. Genome analyses of the polyphagous spider mite herbivore Tetranychus urticae (two-spotted spider mite) revealed the presence of a set of 32 genes that code for secreted proteins belonging to the GST family of enzymes. To better understand the role of these proteins in T. urticae, we have functionally characterized TuGSTd01. Moreover, we have modeled the structure of the enzym

Self-limiting paratransgenesis.

Presently, the principal tools to combat malaria are restricted to killing the parasite in infected people and killing the mosquito vector to thwart transmission. While successful, these approaches are losing effectiveness in view of parasite resistance to drugs and mosquito resistance to insecticides. Clearly, new approaches to fight this deadly disease need to be developed. Recently, one such approach-engineering mosquito resident bacteria to secrete anti-parasite compounds-has proven in the laboratory to be highly effective. However, implementation of this strategy requires approval from re

Antiplasmodial activity of Indigofera spicata root extract against Plasmodium berghei infection in mice.

BACKGROUND: In addition to pharmacovigilance and pharmaco-economic concerns, resistance to anti-malarial medicines has been documented in all classes of anti-malarials and this is further worsened by resistance to common insecticides by malaria vector, which is a major threat to malaria control. As a means of facing the challenges of searching for new anti-malarial agents, the current study focused on evaluation of anti-malarial activity of root extract of Indigofera spicata. METHODS: Chloroquine-sensitive rodent malaria parasite, Plasmodium berghei (ANKA strain) was used to infect the Swiss

Skin conditions: emerging drug-resistant skin infections and infestations.

Methicillin-resistant Staphylococcus aureus (MRSA) skin infections are increasingly common. Automated microbiology systems are now available to detect MRSA and to determine antibiotic resistance patterns. Abscesses should be drained and antibiotics administered, with systemic antibiotics used to manage more severe infections. Until sensitivities are known and depending on local resistance rates, clindamycin is an option for empiric management of stable patients without bacteremia. For patients who are more ill, linezolid and vancomycin are alternatives, the latter being first-line treatment fo

The symbiotic Wolbachia in Anopheles and its role in reducing the transmission of Plasmodium: updates and prospects.

Reducing malaria transmission and the prospects for vector control include multi-pronged strategies, such as interrupting the parasite cycle in both vectors and mosquitoes. Effective vector control remains essential to prevent malaria transmission. This is all the more important as problems such as resistance to insecticides and the lack of a highly effective malaria vaccine remain. New generation vector control measures and optimised products are essential to address the public health needs for malaria eradication. Strategies to reduce malaria transmission include the use of insecticide-treat

Ecological drivers of malaria vector habitat and transmission over 1 year of long-lasting insecticidal net intervention in Côte d'Ivoire.

BACKGROUND: Malaria is a mosquito-borne parasitic disease that causes significant morbidity and mortality in at-risk populations, especially in children in sub-Saharan Africa. Despite reductions in malaria burden owing to the scale-up of effective interventions, there are concerns that long-lasting insecticidal net (LLIN) effects may not be sustained owing to widespread insecticide resistance and differential impacts of LLIN on vector species. In this study, we aimed to test the effect of different LLIN products and other environmental factors on the ecological niche of three mosquito vector s…

Installation of insecticide-treated durable wall lining: evaluation of attachment materials and product durability under field conditions.

BACKGROUND: Insecticide-treated durable wall lining (DL) is a new method of vector control designed to supplement LLINs and overcome two inherent limitations of LLINs and IRS: nightly behavioural compliance and short residual activity, respectively. DL is a deltamethrin-treated polyethylene material, which when used to cover interior house walls, functions as long-lasting IRS. Because the DL concept anticipates minimal upkeep, a primary challenge is how to guarantee correct household installation and in situ longevity for several years. Field trials were undertaken on various wall surfaces in

Effect of Nanoemulsions of Chrysanthemum cinerariaefolium and Oenothera biennis Flowers Against the West Nile Virus Vector, Culex pipiens, with Phytochemical Profile.

(1) Background: Mosquitoes represent major vectors of human pathogens, and their control remains a global public health priority. However, the widespread use of synthetic insecticides has resulted in environmental contamination, adverse effects on non-target organisms, and the rapid emergence of insecticide resistance. These limitations have intensified the search for sustainable botanical alternatives. (2) Methods: The present study investigated the phytochemical composition and larvicidal efficacy of flower extracts from Chrysanthemum cinerariaefolium and Oenothera biennis, alongside their n…

Estimates of the population size and dispersal range of Anopheles arabiensis in Northern KwaZulu-Natal, South Africa: implications for a planned pilot programme to release sterile male mosquitoes.

BACKGROUND: Anopheles arabiensis is a major malaria vector, recently implicated as contributing to ongoing residual malaria transmission in South Africa, which feeds and rests both indoors and outdoors. This species is, therefore, not effectively targeted using core malaria vector control interventions alone. Additionally, increasing resistance to available insecticides necessitates investigations into complementary non-insecticide-based vector control methods for outdoor-resting mosquitoes. The feasibility of the sterile insect technique (SIT) as a complementary vector control intervention is

Phe362Tyr in AChE: A Major Factor Responsible for Azamethiphos Resistance in Lepeophtheirus salmonis in Norway.

Organophosphates (OP) are one of the major treatments used against the salmon louse (Lepeophtherius salmonis) in Norwegian salmonid aquaculture. The use of OP since the late 1970s has resulted in widespread resistant parasites. Recently, we reported a single mutation (Phe362Tyr) in acetylcholinesterase (AChE) as the major mechanism behind resistance in salmon louse towards OP. The present study was carried out to validate this mechanism at the field level. A total of 6658 salmon louse samples were enrolled from 56 different fish farms across the Norwegian coast, from Vest Agder in the south to

Observations on the habits and control of Culex pipiens fatigans in Guyana.

Investigations in Guyana in 1961-63 showed that Wuchereria bancrofti was the only filarial parasite causing elephantiasis and that Culex pipiens fatigans was the primary vector. Mansonia titillans and Anopheles aquasalis were found to be secondary vectors.C. p. fatigans bred primarily in pit latrines and secondarily in the clean, confined water of drums and barrels near latrines. No breeding occurred in the relatively clean open water of trenches, ditches and drains.Of 21016 mosquitos collected from houses, 39.9% were found resting on walls, 34.9% on clothing and other hanging objects, and 23.

Culturable microbiota of reared mosquito fauna in Osogbo, Nigeria and their potential biological roles: a molecular insight.

OBJECTIVE: Mosquitoes are vectors of deadly, life-threatening diseases worldwide. There is limited information on the interactions between microbes and local mosquito fauna in the Nigerian ecotype and Osun State. This study employed molecular techniques to characterize microorganisms isolated from the internal tissues of adult mosquitoes reared from larvae and pupae collected from various breeding sites in the Osogbo metropolis, Nigeria. Bacteria and yeasts were isolated from immobilized, surface-sterilized, and homogenized mosquitoes. Molecular identification of microbes was based on the Poly

Can the leafhopper microbiome unlock new strategies for its control?

Leafhoppers (Hemiptera: Cicadellidae) are significant agricultural pests worldwide, causing direct feeding injury and transmitting plant pathogens. Conventional management still relies heavily on insecticides, but resistance development, non-target effects, and environmental concerns increasingly limit their effectiveness. Recent progress in leveraging insect microbiomes for sustainable pest control, mostly in well-studied groups such as mosquitoes, whiteflies, and aphids, suggests that symbiotic manipulation could offer new tools. Whether such strategies can be developed for leafhoppers remai

Soil microbial responses to multiple global change factors as assessed by metagenomics.

Anthropogenic activities impose multiple concurrent pressures on soils globally, but responses of soil microbes to multiple global change factors are poorly understood. Here, we apply 10 treatments (warming, drought, nitrogen deposition, salinity, heavy metal, microplastics, antibiotics, fungicides, herbicides and insecticides) individually and in combinations of 8 factors to soil samples, and monitor their bacterial and viral composition by metagenomic analysis. We recover 742 mostly unknown bacterial and 1865 viral Metagenome-Assembled Genomes (MAGs), and leverage them to describe microbial

Unstable laboratory Wolbachia strain w-Anga is negatively correlated with Plasmodium falciparum in wild malaria vectors.

Spread of insecticides resistance threatens the control of malaria. In this context, biological control using an endosymbiotic bacterium Wolbachia is being explored as a complementary method for its control. However, for optimal use of this bacterium in biocontrol strategies, it is imperative to characterize it. So, Anopheles gambiae complex mosquitoes were collected, morphologically identified, then blood fed and gravid female mosquitoes oviposited individually. After oviposition, the species of parent was molecularly determined, along with their w-Anga infection status. Additionally, we perf
