Neuronal nitric oxide synthase is permanently decreased in the cerebellum of rats subjected to chronic neonatal blockade of N-methyl-D-aspartate receptors.

Pharmacological blockade of the (NMDA) receptor at critical stages of brain development may have long-lasting effects on brain chemistry and on animal behavior. We report here experiments in which the competitive NMDA receptor antagonist CGP 39551 was administered to rat pups from postnatal day 7 (P7) to P18. The stage of treatment was selected to primarily target the cerebellum, whose granule cells undergo post-mitotic migration and establishment of synaptic connections during this period. We focused our study on the long-term consequences of CGP 39551 treatment on the neuronal isoform of nit

Evidence that enhanced nasal reactivity to bradykinin in patients with symptomatic allergy is mediated by neural reflexes.

OBJECTIVE: The aim of this study was to determine whether allergic inflammation induces nasal hyperreactivity to bradykinin by enhancing neuronal responsiveness. METHODS: We compared the response to localized, unilateral nasal challenge with bradykinin in patients with perennial allergic rhinitis and nonallergic subjects, and in patients with seasonal allergic rhinitis challenged in and out of season. Weights of secretions from each nostril were recorded, and levels of albumin and lactoferrin in secretions recovered from each nostril were assayed. Contralateral administration of atropine (0.3

Evidence for thymopoietin and thymopoietin/alpha-bungarotoxin/nicotinic receptors within the brain.

Источник пометил публикацию как отозванную

Thymopoietin, a polypeptide hormone of the thymus that has pleiotropic actions on the immune, endocrine, and nervous systems, potently interacts with the neuromuscular nicotinic acetylcholine receptor. Thymopoietin binds to the nicotinic alpha-bungarotoxin (alpha-BGT) receptor in muscle and, like alpha-BGT, inhibits cholinergic transmission at this site. Evidence is given that radiolabeled thymopoietin similarly binds to a nicotinic alpha-BGT-binding site within the brain and does so with the characteristics of a specific receptor ligand. Thus specific binding to neuronal membranes was saturab

Selective distribution of the 57 kDa neural intermediate filament protein in the rat CNS.

In order to determine the CNS distribution of the 57 kDa neural intermediate filament protein (NIFP), a specific antiserum was used in immunofluorescence studies on serial sections taken from each spinal cord level and at 300-microns intervals through the rat brain. The labeling pattern was recorded onto camera lucida tracings of adjacent sections stained with hematoxylin and eosin/luxol fast blue. Three major immunolocalization patterns were revealed. (1) Both large-caliber and fine-caliber axons of optic and all brain stem cranial nerves and their tracts except for the auditory portion of th

Secretion and biosynthesis of COOH-terminal glycine extended progastrin (gastrin-G) in rat gastric antrum.

The effects of serotonin (5-HT) on gastrin and COOH-terminal glycine extended progastrin (gastrin-G) secretion, biosynthesis of gastrin and gastrin-G, and the effects of gastrin-G on gastric acid secretion were examined in rats. 5-HT 10(-5) M significantly stimulated gastrin and gastrin-G secretion. Atropine 10(-6) M, which abolished the effects of cholinergic agent to stimulate gastrin secretion, blocked the 5-HT-stimulated gastrin and gastrin-G secretion. In biosynthesis of gastrin and gastrin-G the peak of 35S radioactivity appeared in gastrin-G immunoreactive peak at 30 minutes incubation,

Inhibition of neuronal apoptosis and axonal regression ameliorates sympathetic atrophy and hemodynamic alterations in portal hypertensive rats.

BACKGROUND AND AIM: A neuronal pathway participates in the development of portal hypertension: blockade of afferent sensory nerves in portal vein ligated (PVL) rats simultaneously prevents brain cardiovascular regularory nuclei activation, neuromodulator overexpression in superior mesenteric ganglia, sympathetic atrophy of mesenteric innervation and hemodynamic alterations. Here we investigated in PVL rats alterations in neuromodulators and signaling pathways leading to axonal regression or apoptosis in the superior mesenteric ganglia and tested the effects of the stimulation of neuronal proli

Combating Cadmium-Induced Neurotoxicity, Oxidative Stress, and Inflammatory Pathways Using DOPA-31, a Dioxopiperidinamide Derivative in an In Vivo Zebrafish Model.

Cadmium (Cd), a prevalent environmental toxin and pollutant capable of causing neurodegenerative diseases (NDs) like Alzheimer's and Parkinson's, primarily through oxidative stress, calcium imbalance, and neuroinflammation-induced mechanisms. Cd exposure increases the level of reactive oxygen species (ROS) and disrupts neurotransmitters by lowering antioxidants, leading to neuron death. Cd exposure in zebrafish results in neurodegeneration, with motor, mental, and behavioral impairments. The efficacy of the DOPA-31 intervention at varying concentrations was evaluated through the behavioral tes

Revolutionizing Therapeutic Approaches Against Pathophysiology of Alzheimer's Disease: A Therapeutic Review.

Alzheimer's disease (AD) is a multifactorial neuron-degenerative old age illness, which deteriorates the neuronal cells of the brain ultimately leading to dementia, decreased thinking ability and intricacy in performing daily routine activities. In most of the cases, AD is suspected to be caused by a combination of numerous factors, like environmental, genetic and lifestyle affecting the brain functioning. The present permitted treatments include N-methyl-Daspartate (NMDA) receptor antagonists, cholinesterase inhibitors, and their combinations, which provide only momentary and symptomatic reli

Nitrergic innervation of the cardiac small intensely fluorescent cells in rats.

BACKGROUND: This study aims to analyse the innervation of the small intensely fluorescent (SIF) cells within the rat heart, seeking to examine the supposable neuroanatomical basis for the paradoxical increase of the sympathetic activity of the heart following electrical stimulation of the vagal nerves. METHODS: Neuronal markers and confocal laser scanning microscopy were used to analyse cardiac SIF cells and their innervation in 12 rats. RESULTS: The findings of the present study demonstrate the limited number of nerve fibres (NFs) adjacent to SIF cells. These NFs were of nitrergic phenotype

Morphological substrates of dementia in parkinsonism. A critical update.

Dementia in parkinsonism is caused by a variety of central nervous system (CNS) lesions, of which the molecular and pathogenic causes are poorly understood but probably include: 1. Degeneration of subcortical ascending systems with neuronal losses in dopaminergic, noradrenergic, serotonergic, cholinergic or multiple systems including the amygdyloid nucleus; 2. limbic and/or cortical Alzheimer and/or Lewy body pathologies, with loss of synapses and neurons, and 3. a combination of these lesions or additional CNS pathologies. In general, degeneration of subcortical neuronal networks appears insu

Evaluation of early events in the creation of amyenteric opossum model of achalasia.

Benzyldimethyltetradecylammonium chloride (BAC) has previously been used to create amyenteric rat jejunal models. Fifteen opossums (D. virginiana) were injected with 10-15 mL 4 mM BAC or saline in the distal oesophagus and along with controls underwent oesophagoscopy, manometry and barium oesophagrams. Atropine and sodium nitroprusside were studied in six of the BAC-treated and five controls using oesophageal manometry. Histologically several neuronal markers, B-NADPH-diaphorase and acetylcholine esterase histochemical staining were used. NADPH-diaphorase activity was assayed at the lower oeso

The extracellular matrix glycoprotein tenascin-X regulates peripheral sensory and motor neurones.

KEY POINTS: Tenascin-X (TNX) is an extracellular matrix glycoprotein with anti-adhesive properties in skin and joints. Here we report the novel finding that TNX is expressed in human and mouse gut tissue where it is exclusive to specific subpopulations of neurones. Our studies with TNX-deficient mice show impaired defecation and neural control of distal colonic motility that can be rescued with a 5-HT4 receptor agonist. However, colonic secretion is unchanged. They are also susceptible to internal rectal intussusception. Colonic afferent sensitivity is increased in TNX-deficient mice. Correspo

Neuroimmune interaction in the gut: from bench to bedside.

The enteric nervous system plays a crucial role in the control of gastrointestinal motility and coordinates relaxation and contraction of the smooth muscle cells. Relaxation of the gut is mainly mediated by inhibitory non-adrenergic non-cholinergic (NANC) neurons, a subpopulation of enteric neurons which release a neurotransmitter different from acetylcholine and noradrenaline. Via in vitro work performed in Antwerp, we demonstrated in 1990 that these neurons release nitric oxide. Thereafter, my research has focused on the role of the inhibitory NANC neurons in the pathophysiology of postopera

Central cholinergic system mediates working memory deficit induced by anesthesia/surgery in adult mice.

BACKGROUND: Postoperative cognitive dysfunction (POCD) is consistently associated with increased morbidity and mortality, which has become a major concern of patients and caregivers. Although POCD occurs mainly in aged patients, it happens at any age. Previous studies demonstrated that anesthesia/surgery had no effects on reference memory of adult mice. However, whether it impairs working memory remains unclear. Working memory deficit would result in many deficits of executive function. We hypothesized that anesthesia/surgery impaired the working memory of adult mice and the central cholinergi

Increase in cochlear microphonic potential after toluene administration.

Human and animal studies have shown that toluene can cause hearing loss. In the rat, the outer hair cells are first disrupted by the ototoxicant. Because of their particular sensitivity to toluene, the cochlear microphonic potential (CMP) was used for monitoring the cochlea activity of anesthetized rats exposed to both noise (band noise centered at 4 kHz) and toluene. In the present experiment, the conditions were specifically designed to study the toluene effects on CMP and not those of its metabolites. To this end, 100-microL injections of a vehicle containing different concentrations of sol

Altered neuronal physiology, development, and function associated with a common chromosome 15 duplication involving CHRNA7.

BACKGROUND: Copy number variants (CNVs) linked to genes involved in nervous system development or function are often associated with neuropsychiatric disease. While CNVs involving deletions generally cause severe and highly penetrant patient phenotypes, CNVs leading to duplications tend instead to exhibit widely variable and less penetrant phenotypic expressivity among affected individuals. CNVs located on chromosome 15q13.3 affecting the alpha-7 nicotinic acetylcholine receptor subunit (CHRNA7) gene contribute to multiple neuropsychiatric disorders with highly variable penetrance. However, th

RGS Proteins in Sympathetic Nervous System Regulation: Focus on Adrenal RGS4.

The sympathetic nervous system (SNS) consists largely of two different types of components: neurons that release the neurotransmitter norepinephrine (NE, noradrenaline) to modulate homeostasis of the innevrvated effector organ or tissue and adrenal chromaffin cells, which synthesize and secrete the hormone epinephrine (Epi, adrenaline) and some NE into the blood circulation to act at distant organs and tissues that are not directly innervated by the SNS. Like almost every physiological process in the human body, G protein-coupled receptors (GPCRs) tightly modulate both NE release from sympathe

Neuroprotective effects of dexpanthenol on streptozotocin-induced neuronal damage in rats.

AIM: Although the most common age-related neurodegenerative disease defined by memory loss is Alzheimer's disease (AD), only symptomatic therapies are present. A complex pathway for the AD pathogenesis that includes an increase in inflammation has recently been suggested. Since in previous animal experiments dexpanthenol has anti-inflammatory and neuroprotective activities, effects and role of dexpanthenol in an intracerebroventricular (ICV)-streptozotocin (STZ) induced sporadic-AD(memory impairment) animal model have been examined. DESIGN AND METHODS: In total, 18 adult sprague-dawley rats w

Kinetic organization of Ca2+ signals that regulate synaptic release efficacy in sympathetic neurons.

Calcium regulation of neurotransmitter release is essential for maintenance of synaptic transmission. However, the temporal and spatial organization of Ca(2+) dynamics that regulate synaptic vesicle (SV) release efficacy in sympathetic neurons is poorly understood. Here, we investigate the N-type Ca(2+) channel-mediated kinetic structure of Ca(2+) regulation of cholinergic transmission of sympathetic neurons. We measured the effect of Ca(2+) chelation with fast 1,2-bis(2-aminophenoxy) ethane-tetraacetic acid (BAPTA) and slow ethyleneglycol-tetraacetic acid (EGTA) buffers on exocytosis, synapti

Activation of the pharmacologically defined M3 muscarinic receptor depolarizes hippocampal pyramidal cells.

Acetylcholine has a variety of actions on hippocampal neurons, possibly through the activation of different muscarinic receptor subtypes. We have used several competitive muscarinic antagonists and the hippocampal slice preparation to test the hypothesis that a particular muscarinic response can be ascribed to a specific subtype. Initially we performed experiments to determine the relative ability of 3 antagonists to inhibit the cholinergic slow EPSP. Dose-response curves were constructed, and the IC50s determined for 4-diphenylacetoxy-N-methylpiperidine methiodide (4-DAMP), pirenzepine (PRZ)
