Skeletal dysplasias: 38 prenatal cases.

OBJECTIVE: To assess the prenatal diagnosis of skeletal dysplasias in a single center over a ten-years period. METHODS: All antenatal detected skeletal dysplasias during the period January 1st 1996 until December 31 2005 (10 years) were retrieved from the genetic database. This database includes all skeletal dysplasias where invasive prenatal diagnosis (chorionic villus sampling/amniocentesis) was performed. The final diagnosis was sought on the basis of fetopathological examination, radiographic studies and if possible molecular testing. RESULTS: A total of 46 antenatal skeletal dysplasias

Familial risk of lymphoproliferative tumors in families of patients with chronic lymphocytic leukemia: results from the Swedish Family-Cancer Database.

The importance of genetic factors in etiology of chronic lymphocytic leukemia (CLL) is suggested by family and population studies. However, the spectrum of malignancies sharing common genetic factors with CLL and the effects of sex and age on familial risk are unknown. We used the Swedish Family-Cancer Database to test for increased familial risks of CLL and other lymphoproliferative tumors. Cancer diagnoses from 1958 to 1998 were assessed in 14 336 first-degree relatives of 5918 CLL cases and in 28 876 first-degree relatives of 11 778 controls. Cancer risks in relatives of cases were compared

Genetic and nongenetic factors affecting vancomycin population pharmacokinetics and pharmacodynamics: A systematic review.

Despite frequent vancomycin prescribing, achieving optimal therapeutic outcomes is challenging due to its complex pharmacokinetic (PK) and pharmacodynamic (PD) properties. Those characteristics are influenced by various parameters, including both genetic and non-genetic variables, rendering accurate dosing and monitoring challenging for clinicians. This systematic review was conducted in accordance with the 2020 Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines, ensuring a standardized and comprehensive approach to the literature search, data extraction, an

Myopia as a Global Public Health Challenge a Narrative Review.

(1) Background: The global prevalence of myopia has increased substantially in recent decades. Myopia development is influenced by both environmental factors and a complex genetic architecture involving more than 400 susceptibility loci. The interaction between genetic predisposition and environmental exposures plays a critical role in myopia onset and progression. Unequal access to preventive strategies and eye care services continue to limit effective global myopia control. (2) Methods: This structured narrative review synthesizes evidence identified through systematic database searches, man

Genetic and environmental origins of gambling behaviors from ages 18 to 25: A longitudinal twin family study.

Gambling behaviors tend to increase in prevalence from late adolescence to young adulthood, and the underlying genetic and environmental influences during this period remain largely understudied. We examined the genetic and environmental influences on gambling behaviors contributing to stability and change from ages 18 to 25 in a longitudinal, behavioral genetic mixed-sex twin study design. Participants were enrolled in the Minnesota Twin Family Study. A range of gambling behaviors (maximum frequency, average frequency, money lost, and gambling problems) were assessed at ages 18 and 25. The re

Primary dystonias and genetic disorders with dystonia as clinical feature of the disease.

Dystonia is probably the most common form of movement disorder encountered in the clinical practice. It is characterized by sustained muscle contractions, usually producing twisting and repetitive movements or abnormal postures or positions. Dystonias can be classified in several ways, including primarily by the clinical phenomenology or by the underlining etiology, in particular to understand if the presentation is genetically determined. By advances of genetics, including contemporary genomic technologies, there is a growing understanding of the molecular underpinnings of genetically determi

A region on chromosome 16 is associated with Doberman Pinscher dilated cardiomyopathy.

Doberman Pinschers are known for their increased susceptibility to dilated cardiomyopathy (DCM) relative to other domestic dogs. This makes the Doberman Pinscher a key model for gene-disease investigations. We conducted a genome-wide association study (GWAS) leveraging a database of genetic profiles obtained through collaboration with the Doberman Diversity Project (DDP). We worked in parallel to increase the depth and power of the DDP database. We exchanged direct correspondences with listed breeders and owners to establish health updates for participant dogs. In total, our study included dat

The Taiwan Precision Medicine Initiative provides a cohort for large-scale studies.

Han Chinese people comprise nearly 20% of the global population but remain under-represented in genetic studies1,2, so there is an urgent need for large-scale cohorts to advance precision medicine. Here we present the Taiwan Precision Medicine Initiative (TPMI), established by Academia Sinica in collaboration with 16 major medical centres around Taiwan, which has recruited 565,390 participants who consent to provide DNA samples for genetic profiling and grant access to their electronic medical records (EMRs) for research. EMR access is both retrospective and prospective, allowing longitudinal

Exploring the link between antihypertensive drugs targets and pancreatitis risk through Mendelian randomization.

BACKGROUND: Observational studies indicate an association between antihypertensive drugs and the risk of acute pancreatitis (AP) and chronic pancreatitis (CP). This study aims to utilize Mendelian randomization (MR) analysis to assess the causal impact of antihypertensive drugs on the risk of AP and CP. METHOD: Protein target information for eight commonly used antihypertensive drugs was collected from the DrugBank database. The inverse variance-weighted (IVW) method was the primary approach used to investigate the association between genetic instruments from the discovery cohorts and the ris

A novel frameshift variation of PKD1 in familial autosomal dominant polycystic kidney diseases: expanding the clinical phenotype and genetic spectrum of PKD1 disorders.

BACKGROUND: Autosomal dominant polycystic kidney disease is a common monogenic hereditary kidney disorder. Mutations in the PKD1 gene lead to the development of fluid-filled cysts in the kidney, liver, and other organs, which can eventually result in end-stage renal failure. Affected individuals have a 50% chance of passing the mutation to each of their offspring. Families with ADPKD are advised to undergo genetic testing to causative variants and would benefit from genetic counseling to reduce the inherited risk. METHOD: We performed genetic analysis on a family with ADPKD. DNA samples extrac

Tomato yellow leaf curl virus (TYLCV) in Kuwait and global analysis of the population structure and evolutionary pattern of TYLCV.

Tomato yellow leaf curl virus (Family Geminiviridae, Genus Begomovirus) is a serious menace in the cultivation of tomato causing Tomato leaf curl disease (ToLCD). Recently, we presented the TYLCV isolates having additional genomic features (nucleotides insertions) characterized from the tomato fields of Kuwait adding up to the genetic diversity repertoire of these viruses. The widespread prevalence of disease in tropics across the continents, emergence of genetic variants and ever increasing complete genome sequences of virus isolates in public database warrant a global analysis to infer the g

Folic acid: friend or foe in cancer therapy.

Folic acid plays a crucial role in diverse biological processes, notably cell maturation and proliferation. Here, we performed a literature review using articles listed in electronic databases, such as PubMed, Scopus, MEDLINE, and Google Scholar. In this review article, we describe contradictory data regarding the role of folic acid in cancer development and progression. While some studies have confirmed its beneficial effects in diminishing the risk of various cancers, others have reported a potential carcinogenic effect. The current narrative review elucidates these conflicting data by highl
