Advancing vector biology research: a community survey for future directions, research applications and infrastructure requirements.

Vector-borne pathogens impact public health, animal production, and animal welfare. Research on arthropod vectors such as mosquitoes, ticks, sandflies, and midges which transmit pathogens to humans and economically important animals is crucial for development of new control measures that target transmission by the vector. While insecticides are an important part of this arsenal, appearance of resistance mechanisms is increasingly common. Novel tools for genetic manipulation of vectors, use of Wolbachia endosymbiotic bacteria, and other biological control mechanisms to prevent pathogen transmis

Benefits of genetically modified crops for the poor: household income, nutrition, and health.

The potential impacts of genetically modified (GM) crops on income, poverty and nutrition in developing countries continue to be the subject of public controversy. Here, a review of the evidence is given. As an example of a first-generation GM technology, the effects of insect-resistant Bt cotton are analysed. Bt cotton has already been adopted by millions of small-scale farmers, in India, China, and South Africa among others. On average, farmers benefit from insecticide savings, higher effective yields and sizeable income gains. Insights from India suggest that Bt cotton is employment generat

Increased control of the sheep biting louse Bovicola (Damalinia) ovis with deltamethrin formulated in a fractionated wool grease carrier.

The synthetic pyrethroid deltamethrin (DM) containing a trace of [(14)C]-DM was formulated with non-oxidised sterol and wax ester fractions (F1) of wool grease and as the commercial preparation 'Clout-S'. These were applied as a 'backline' strip to sheep immediately after shearing and the concentration of [(14)C]-DM at meridians adjacent to the application strip and at 1/4 and 3/4 of the dorsal-ventral distance was determined. The F1 formulation resulted in significantly greater lateral spread of DM with less remaining at the application site (66+/-8% of dose) 98 days after treatment compared

Ecological origin and natural Trypanosoma cruzi infection are associated with reconfiguration of the Rhodnius pallescens midgut microbiome.

The intrusion of the secondary vector Rhodnius pallescens into urban and peri-urban environments in Colombia poses an emerging risk for Chagas disease transmission, compounded by rising insecticide resistance. Developing novel control strategies requires the identification of stable gut symbionts; however, high-resolution microbial profiles for urban-intruding vector populations remain poorly defined. To characterize the intestinal microbiome and identify candidate taxa for paratransgenesis, we generated high-resolution taxonomic composition and predicted functional profiles using 16S rRNA amp…

The XylC-mediated aromatic metabolic network modulates the metabolic response of Pseudomonas aeruginosa to deltamethrin.

The widespread emergence of pyrethroid resistance represents a major challenge in pest and vector management. This study examined the function of the xylC-encoded benzaldehyde dehydrogenase in Pseudomonas aeruginosa under deltamethrin stress. The metabolic response of a ΔxylC mutant cultured with 50 mg/L deltamethrin was compared to the wild-type strain, and functional complementation was achieved by adding purified XylC protein. Integrated metabolomic and proteomic analyses were used to characterize the associated metabolic adaptations. Deltamethrin exposure inhibited the production of 3-(2-c

Host Plant Trichome Architecture Mediates the Predatory Efficacies of Nesidiocoris tenuis and Chrysoperla carnea Against Tuta absoluta.

The tomato leafminer, Tuta absoluta, is a globally important pest that causes severe losses in tomato production, and effective biological control strategies are increasingly needed due to widespread insecticide resistance. The effectiveness of natural enemies is influenced by plant structural traits, such as trichomes, which create complex microhabitats that shape predator-prey interactions. Here, we compared the predatory performances, plant-level control efficacies, and predator-plant interactions of two widely used biological control agents, Nesidiocoris tenuis and Chrysoperla carnea, agai…

Mechanism of locust egg susceptibility to diafenthiuron: micropyle permeation and serosal cuticle disruption lead to embryonic lethality.

Chemical pesticides constitute a core cornerstone of modern agricultural pest control practices. Contemporary management paradigms remain narrowly focused on post-embryonic pest stages, creating critical knowledge gaps in ovicidal intervention strategies that are pivotal for effective source-specific pest population suppression. Locusta migratoria is one of the key pests that pose a serious threat to agricultural production. However, current control measures for this pest are mainly focused on the nymph or adult stages. As a widely deployed thiourea insecticide, diafenthiuron exhibits excellen

Ecological and engineered modulation of the mosquito microbiome: mechanisms, vector competence, and translational prospects for disease control.

Malaria, dengue fever, Zika, chikungunya, yellow fever, and West Nile fever are mosquito-borne diseases that collectively impose an enormous global health burden, disproportionately affecting low- and middle-income countries where vector-control tools remain limited or compromised by insecticide resistance. Over the past two decades, the characterization of mosquito-associated microbiomes has transformed our understanding of vector biology, revealing complex, ecologically contingent assemblages of bacteria, fungi, viruses, and protists that profoundly influence mosquito physiology, immunity, a…

Developing methods for chilling, compacting, and sterilizing adult Aedes aegypti (Diptera: Culicidae) and comparing mating competitiveness between males sterilized as adults versus pupae for sterile male release.

The yellow fever mosquito, Aedes aegypti L., can transmit several pathogens responsible for human diseases. With insecticide resistance development becoming a concern, alternative control strategies are needed for Ae. aegypti. Sterile insect technique (SIT) is an increasingly popular option being explored. However, logistical issues in mass production and sterilization make it difficult to maintain a SIT program. Male mosquitoes are typically irradiated as pupae because this is the earliest developmental point at which females can be separated from males, but asynchrony in pupation and high va…

A population genomic unveiling of a new cryptic mosquito taxon within the malaria-transmitting Anopheles gambiae complex.

The Anopheles gambiae complex consists of multiple morphologically indistinguishable mosquito species including the most important vectors o … of Anopheles ecology and evolution in western Africa, including phenotypic and genotypic surveys of insecticide resistance. Reproductive barriers between cryptic species may also complicate novel vector control efforts, for example gene drives, and hinder predictions about evolutionary dynamics of Anopheles and Plasmodium.

Incursion pathways of the Asian tiger mosquito (Aedes albopictus) into Australia contrast sharply with those of the yellow fever mosquito (Aedes aegypti).

BACKGROUND: Understanding pest incursion pathways is critical for preventing new invasions and for stopping the transfer of alleles that reduce the efficacy of local control methods. The mosquitoes Aedes albopictus (Skuse) and Ae. aegypti (Linnaeus) are both highly invasive disease vectors, and through a series of ongoing international incursions are continuing to colonize new regions and spread insecticide resistance alleles among established populations. This study uses high-resolution molecular markers and a set of 241 reference genotypes to trace incursion pathways of Ae. albopictus into m…

Detection of arboviruses and other micro-organisms in experimentally infected mosquitoes using massively parallel sequencing.

Human disease incidence attributed to arbovirus infection is increasing throughout the world, with effective control interventions limited by issues of sustainability, insecticide resistance and the lack of effective vaccines. Several promising control strategies are currently under development, such as the release of mosquitoes trans-infected with virus-blocking Wolbachia bacteria. Implementation of any control program is dependent on effective virus surveillance and a thorough understanding of virus-vector interactions. Massively parallel sequencing has enormous potential for providing compr…

Synthesis of pediculocidal and larvicidal silver nanoparticles by leaf extract from heartleaf moonseed plant, Tinospora cordifolia Miers.

Insecticide resistance and inadequate attention to the application instructions of topical pediculicides are common reasons for treatment failure. Essential oils or plant extracts are good and safe alternatives due to their low toxicity to mammals and easy biodegradability. The present study was carried out to establish the pediculocidal and larvicidal activity of synthesized silver nanoparticles (AgNPs) using leaf aqueous extract of Tinospora cordifolia Miers (Menispermaceae) against the head louse Pediculus humanus capitis De Geer (Phthiraptera: Pediculidae) and fourth instar larvae of malar…

A target product profile for a rapid diagnostic test to monitor mosquito gene drive presence and frequency.

Malaria remains a major global health challenge, with over 263 million cases and nearly 600,000 deaths reported in 2023, the majority in sub-Saharan Africa. While conventional interventions such as insecticide-treated nets, indoor residual spraying and antimalarial drugs have reduced transmission, progress has stalled due to the limitations of these interventions and the emergence of resistance. Gene drive-modified mosquitoes represent a promising, potentially transformative vector control strategy, capable of spreading malaria-refractory traits or suppressing mosquito populations. Molecular characterization of vitellogenin and its receptor with CRISPR-based sgRNA validation in the legume pod borer, Maruca vitrata (Geyer) (Lepidoptera: Crambidae).

Maruca vitrata, the legume pod borer, causes yield losses of up to 80% in grain legumes. Increasing insecticide resistance and environmental concerns necessitate sustainable pest management alternatives. In the present study, the complete vitellogenin (Vg) coding sequence (CDS), a key reproductive gene involved in oogenesis and embryonic development, was cloned and molecularly characterised from M. vitrata. The assembled Vg CDS (∼5.3 kb) shared 99.04% sequence identity with the reported M. vitrata Vg sequence (MG799570.1). Phylogenetic analysis demonstrated close evolutionary association with …

Estimating the dispersal of the malaria vector Anopheles farauti through a natural ecosystem in north Queensland, Australia using mark release and recapture experiments.

Anopheles farauti (Laveran, 1902) is a major malaria vector in the Southwest Pacific region that is showing behavioural resistance to indoor insecticide-based vector control. This study utilised fluorescent dust-based mark release recapture (MRR) experiments to quantify the dispersal characteristics of An. farauti through a non-village natural ecosystem in tropical north Queensland, Australia. Good quality adult mosquitoes were collected from the field using a Fan Box Traps (FBT), with initial parity dissections of this material showing a daily survival rate of 0.68 during the wet season and 0

Discovery of novel skeletons of aphid repellents based on the key fragment of nepetalactone and plant volatiles.

BACKGROUND: Aphids are significant agricultural pests, causing extensive crop damage and transmitting plant viruses. Conventional insecticide-based pest control faces challenges such as human and environmental toxicity and resistance. As alternative solutions, pheromone-based repellents have emerged as a promising approach. However, the instability of key compounds like (E)-β-Farnesene (EBF) has limited their effectiveness in field applications. To address these limitations, this study focuses on developing novel aphid repellent compounds based on plant volatiles and nepetalactone key fragment
