Molecular detection of novel Glutamate-gated chloride channel mutations in field collected human head lice (Phthiraptera: Pediculidae) from Iran.

OBJECTIVE: Recently, insecticides such as ivermectin, which targets glutamate-gated chloride channel (GluCl) channels in the nervous system of invertebrates, have gained attention for the treatment of head lice. However, resistance to this insecticide threatens the effectiveness of head louse control programs. RESULTS: Molecular bioinformatics sequence analysis revealed that the most common mutations were R37K and E50K with a frequency of 85.71%, followed by D93N (64.28%), M101R (35.71%), and R100Q (28.57%). These mutations are reported here for the first time. The identification of these nov…

LaIT6: A Novel Insect-Selective K+-Channel Toxin from Liocheles australasiae Scorpion Venom.

Scorpion venom contains various insecticidal peptides. Previously, through transcriptome analysis of the venom gland of Liocheles australasiae, we identified precursor sequences of several peptides that share sequences similar to those acting on K+ channels. In this study, we chemically synthesized five of the peptides which were found in the venom and evaluated their insecticidal activity against crickets. This revealed that one of the peptides, named LaIT6, exhibited significant insecticidal activity without mammalian toxicity. To identify amino acid residues important for the insecticidal a

Discriminating lethal concentrations for pyrethroid compounds used in susceptibility monitoring of Anopheles epiroticus, a malaria vector in Thailand.

Anopheles epiroticus is a malaria vector in Thailand found primarily along coastal areas with brackish water habitats. Insecticides, particularly pyrethroid class compounds, are commonly used to control malaria vectors in Thailand. The lack of specific discriminating lethal concentrations for An. epiroticus has possibly compromised a more accurate assessment of physiological susceptibility to various chemicals. The routine assessment of vector response to insecticides is a key program management component to prevent or mitigate the development of resistance. The purpose of this study was to de…

Insights into factors sustaining persistence of high malaria transmission in forested areas of sub-Saharan Africa: the case of Mvoua, South Cameroon.

BACKGROUND: In Mvoua, a village situated in a forested area of Cameroon, recent studies have reported high prevalence of Plasmodium falciparum infection among the population. In order to understand factors that can sustain such a high malaria transmission, we investigated the biology of Anopheles vectors and its susceptibility to insecticides, as well as long-lasting insecticidal net (LLIN) coverage, use and bio-efficacy. METHODS: A longitudinal entomological survey was conducted from July 2018 to April 2019. Adult mosquitoes were collected using the human landing catch (HLC) method and ident…

The Effectiveness of Indoor Residual Spraying for Malaria Control in Sub-Saharan Africa: A Systematic Protocol Review and Meta-Analysis.

BACKGROUND: Indoor residual spraying (IRS) is a core insecticide-based vector control tool employed in most malaria-affected settings globally. However, mosquito vectors have developed resistance to nearly all of the insecticides currently used in IRS. This has necessitated a transition to new classes of insecticides, from mostly using dichlorodiphenyltrichloroethane (DDT) and pyrethroids from 1997 to 2010 to carbamates in 2011 and organophosphates in 2013. In addition, other vector control measures, like the use of long-lasting insecticide-treated bed nets (LLINs), have also been employed for…

Computational Modeling Oriented Substructure Splicing Application in the Identification of Thiazolidine Derivatives as Potential Low Honeybee Toxic Neonicotinoids.

With the aim of identifying novel neonicotinoid insecticides with low bee toxicity, a series of compounds bearing thiazolidine moiety, which has been shown to be low bee toxic, were rationally designed through substructure splicing strategy and evaluated insecticidal activities. The optimal compounds A24 and A29 exhibited LC50 values of 30.01 and 17.08 mg/L against Aphis craccivora, respectively. Electrophysiological studies performed on Xenopus oocytes indicated that compound A29 acted on insect nAChR, with EC50 value of 50.11 μM. Docking binding mode analysis demonstrated that A29 bound to L…

Alteration of unfolded protein responses and autophagy signaling represented the molecular basis underlying saccharin toxicity to Drosophila (Diptera: Drosophilidae).

The purpose of this study was to develop a new control method for Drosophila using saccharin sodium dihydrate (saccharin), an artificial sweetener that is safe for humans and the environment, and to elucidate its mode of action. In this study, we confirmed that saccharin can dose-dependently inhibit the development of or kill vinegar flies (VFs) and spotted wing Drosophila (SWDs). In addition, we found that low concentrations of saccharin induced a similar effect as starvation in Drosophila, whereas high concentrations of saccharin induced changes in the unfolded protein response (UPR) and aut

Exposure to spinosad induces histopathological and cytotoxic effects on the salivary complex of the non-target predator Podisus nigrispinus.

In integrated pest management systems, biological and chemical controls must be compatible. The insecticide spinosad affects some non-target insects and might compromise their fitness. The objective of this study was to evaluate the histopathological and cytotoxic effects of spinosad on the salivary complex of the predatory bug Podisus nigrispinus (Heteroptera: Pentatomidae). Spinosad toxicity and insect survival were determined using six concentrations of insecticide. Ultrastructural changes and cell death of salivary glands were analyzed after P. nigrispinus exposure to spinosad LC50 (3.15 μ

Field evaluation of Cucurbit yellow stunting disorder virus transmission by Bemisia tabaci.

Cucurbit yellow stunting disorder virus (CYSDV) is a whitefly-transmitted Crinivirus (Closteroviridae) that impacts melon production in many parts of the world including the USA. It has been responsible for melon crop loss in the southwestern U.S. since 2006 when it was first identified. Control strategies have revolved mainly around chemical control, but research to identify suitable products and approaches to implementing them have lagged. The current study investigated the performance of four systemic insecticides in the field while concurrently tracking CYSDV disease progression after cont…

Efficacy of resmethrin aerosols applied from the road for suppressing Culex vectors of West Nile virus.

We determined whether aerosol applications of resmethrin, delivered from the road, suppress the reproductive activity of Culex pipiens pipiens and Cx. restuans mosquitoes in suburban sites located near Boston. Oviposition implies a prior blood-feeding event and hence a potential West Nile virus (WNV) transmission-related event. Droplet size, rate of delivery and meteorological conditions were monitored. The target populations proved to be fully susceptible to the insecticide that was used. The roads in the test sites generally gave adequate opportunity for insecticidal coverage. We found that

Culling structured hosts to eradicate vector-borne diseases.

A compartmental model is developed, in the form of a nonautonomous system of delay differential equations subject to impulses at specific times, for mosquito-born disease control involving larvicides and insecticide sprays. Sufficient conditions in terms of the frequencies and rates of larvicides and insecticide sprays are derived, and numerical simulations are provided to illustrate the sharpness of these disease eradication conditions.

The molecular determinants of pesticide sensitivity in bee pollinators.

Bees carry out vital ecosystem services by pollinating both wild and economically important crop plants. However, while performing this function, bee pollinators may encounter potentially harmful xenobiotics in the environment such as pesticides (fungicides, herbicides and insecticides). Understanding the key factors that influence the toxicological outcomes of bee exposure to these chemicals, in isolation or combination, is essential to safeguard their health and the ecosystem services they provide. In this regard, recent work using toxicogenomic and phylogenetic approaches has begun to ident…

"People are afraid of getting injections": feasibility of community-placed atropine auto-injectors in treating organophosphate poisoning in Sri Lanka.

INTRODUCTION: Organophosphorus insecticide poisoning remains a major problem in rural low- and middle-income communities. Atropine auto-injectors are used for organophosphorus nerve agent poisoning, but not organophosphorus insecticide poisoning. We explore the social and practical feasibility of introducing atropine auto-injectors in Sri Lankan villages to initiate first-line treatment for acute organophosphorus insecticide poisoning. We investigate how auto-injectors might be best integrated into the social and spatial context of rural communities with high levels of organophosphorus insecti

Voltage-Gated Sodium Channel (Vgsc) Mutation-Based Pyrethroid Resistance in Aedes aegypti Populations of Three Endemic Dengue Risk Areas of Sri Lanka.

BACKGROUND: Pyrethroid insecticides are widely used in many countries for chemical-based control of Ae. aegypti. Regardless of their efficacy, the constant use of insecticides has induced insecticide resistance mechanisms, such as knockdown resistance (kdr) in mosquitoes. Sri Lankan Vector Controlling Entities (VCE) have been using a variety of pyrethroid insecticides as the primary approach for dengue control. However, development of any resistance among the Aedes mosquitoes has been limitedly studied in the country. Therefore, the current study was conducted to evaluate the prevalence of F15…

Temperature and time of host-seeking activity impact the efficacy of chemical control interventions targeting the West Nile virus vector, Culex tarsalis.

West Nile virus (WNV) is the leading mosquito-borne disease causing-pathogen in the United States. Concerningly, there are no prophylactics or drug treatments for WNV and public health programs rely heavily on vector control efforts to lessen disease incidence. Insecticides can be effective in reducing vector numbers if implemented strategically, but can diminish in efficacy and promote insecticide resistance otherwise. Vector control programs which employ mass-fogging applications of insecticides, often conduct these methods during the late-night hours, when diel temperatures are coldest, and…

Conflict of interest: use of pyrethroids and amidines against tsetse and ticks in zoonotic sleeping sickness endemic areas of Uganda.

BACKGROUND: Caused by trypanosomes and transmitted by tsetse flies, Human African Trypanosomiasis and bovine trypanosomiasis remain endemic across much of rural Uganda where the major reservoir of acute human infection is cattle. Following elimination of trypanosomes by mass trypanocidal treatment, it is crucial that farmers regularly apply pyrethroid-based insecticides to cattle to sustain parasite reductions, which also protect against tick-borne diseases. The private veterinary market is divided between products only effective against ticks (amidines) and those effective against both ticks …
