Kojic acid (KA) has been widely used as a quasi-drug ingredient. Possible promotion activity of KA was suggested on livers of mouse and rat by findings obtained in genotoxicity and carcinogenicity studies performed thus far. Therefore, in order to examine safety as a quasi-drug ingredient, we investigated the presence of initiation activity in rat liver and the photo-genotoxicity and carcinogenicity in mouse skin. In medium-term carcinogenesis test in rats, 2.0% KA was orally given to F344/DuCrj rats for 4 weeks of the initiation period, followed by the combination of partial hepatectomy and t

INTRODUCTION: Seed-borne fungal contamination is an important challenge in maize production and may contribute to grain deterioration and mycotoxin contamination. This study evaluated peroxynitric acid (PNA), alone and in combination with nonequilibrium gaseous plasma pretreatment, as a rapid oxidative strategy for disinfecting maize seeds contaminated with Aspergillus flavus and Fusarium fujikuroi.
METHODS: Maize seeds were artificially inoculated with approximately 10⁶ spores of either A. flavus or F. fujikuroi. For plasma pretreatment, seeds were sequentially exposed to hydrogen (0.2 s) an

Studies have shown that levels of fungal contamination and mycotoxin concentrations in oil-producing seeds vary across regions. In addition, extreme exposure to seeds dust particulate matter can lead to respiratory health problems. In Ethiopia oil-seeds processing factories workers exposure to particulate matter and magnitude of its respiratory symptoms have not been widely explored. To determine particulate matter concentration exposure levels and identify factors associated with respiratory symptoms among oil-seed processing factories workers. Using cross-sectional study design, 14 factories

Aspergillus welwitschiae is a widespread fungus with diverse roles as a plant mutualist, opportunistic human pathogen and industrial enzyme producer. The endophytic strain AwOcstreb1, isolated from halophytic rice (Oryza coarctata), promotes growth in commercial rice under normal and saline conditions. Despite its significance, genomic and metabolic resources for A. welwitschiae remain limited, with no complete genome information available for endophytic strains within the species. Moreover, the close relationship of this species to Aspergillus niger complicates its taxonomic resolution. We pe

Deoxynivalenol (DON) contamination of grains and coccidiosis are two major health and economic challenges in layer production, yet their combined effect during pullet rearing phase remains poorly understood. This study evaluated whether dietary DON exposure exacerbates the severity of Eimeria infection and delays recovery in growing pullets. A total of 288 Hy-Line W36 pullets (4-wk-old) were fed either a control diet containing 2.5 mg/kg DON or a naturally contaminated diet containing 14-15 mg/kg DON for 14 d. Birds were then sham‑dosed or orally inoculated with E. acervulina (50 000 oocysts),

Ochratoxin A (OTA) is one of the most abundant mycotoxin contaminants in food stuffs and possesses carcinogenic, nephrotoxic, teratogenic, and immunotoxic properties. Specifically, a major concern is severe nephrotoxicity, which is characterized by degeneration of epithelial cells of the proximal tubules and interstitial fibrosis. However, the mechanism of OTA toxicity, as well as the genetic risk factors contributing to its toxicity in humans has been elusive due to the lack of adequate models that fully recapitulate human kidney function in vitro. The present study attempts to evaluate dose-

If the diagnosis is not made early, dissemination often occurs. Therapy, if it is to be effective, must be started early and requires combinations of antifungal drugs, surgical intervention, and reversal of the underlying risk factors. The Entomoph …

Hypoxia contracts the pulmonary vein, but the underlying cellular effectors remain unclear. Utilizing contractile studies and whole cell patch-clamp electrophysiology, we report for the first time a hypoxia-sensitive K(+) current in porcine pulmonary vein smooth muscle cells (PVSMC). Hypoxia induced a transient contractile response that was 56 ± 7% of the control response (80 mM KCl). This contraction required extracellular Ca(2+) and was sensitive to Ca(2+) channel blockade. Blockade of K(+) channels by tetraethylammonium chloride (TEA) or 4-aminopyridine (4-AP) reversibly inhibited the hypox

The bioactivity-guided fractionation of chloroform extracts of the fruit bodies of Hypsizigus marmoreus led to our isolation of (22E,24R)-ergosta-7,22-diene-3beta,5alpha,6beta-triol (1), ergosterol-3-O-beta-D-glucopyranoside (2), 5alpha,8alpha-epidioxyergosta-6,22-dien-3beta-ol (3), hypsiziprenol A9 (4), hypsiziprenol AA8 (5), hypsiziprenol AA9 (6) and hypsiziprenol BA10 (7). Among these seven isolates, compound 2 was identified for the first time from this plant. All compounds (1-7) exhibited moderate cytotoxicity towards cultured human colon carcinoma (HT-29), human breast carcinoma (MCF-7)

Autophagy is a lysosome-dependent cellular catabolic mechanism mediating the turnover of intracellular organelles and long-lived proteins. Reduction of autophagy activity has been shown to lead to the accumulation of misfolded proteins in neurons and may be involved in chronic neurodegenerative diseases such as Huntington's disease and Alzheimer's disease. To explore the mechanism of autophagy and identify small molecules that can activate it, we developed a series of high-throughput image-based screens for small-molecule regulators of autophagy. This series of screens allowed us to distinguis

Tumor necrosis factor alpha (TNFalpha) is involved in fumonisin-induced hepatotoxic effects in mice. The hepatic response to fumonisin B(1) (FB(1)) was reduced in transgenic animals lacking either of the two TNFalpha receptors. In the present study, we hypothesized that the effect of a similar fumonisin treatment in animals lacking either TNFalpha or both TNFalpha receptors would be considerably less than their wild type (WT) counterparts. The FB(1)-induced increase in circulating liver enzymes was enhanced by deletion of TNFalpha or unchanged in mice lacking both TNFalpha receptors. These fin

Our previous studies have indicated that tumour necrosis factor alpha (TNFalpha) is involved in fumonisin B1 (FB1)-induced toxic responses. To investigate the role of TNFalpha in FB1 toxicity further we employed male transgenic mice expressing human TNFalpha gene (TG) and their wild-type equivalent C57BL/6 (WT). It was hypothesized that TG animals would have enhanced response to FB1. Repeated subcutaneous treatment of animals with 2.25 mg/kg per day of FB1 for 5 days caused minimal changes in body weight, organ weights, blood cell counts, and TNFalpha levels in plasma 1 day after the last inje
