Projection neurons and interneurons in the lateral geniculate nucleus undergo distinct forms of degeneration ranging from retrograde and transsynaptic apoptosis to transient atrophy after cortical ablation in rat.

The cytological responses of thalamic interneurons to selective degeneration of thalamocortical projection neurons after cortical damage in the adult brain are poorly understood. We used a unilateral neocortical lesion model (occipital cortex ablation) in the adult rat to test the hypothesis that interneurons and projection neurons in the lateral geniculate nucleus undergo distinct forms of degeneration. In situ nuclear DNA fragmentation in neurons in the lateral geniculate occurs maximally at 7 days postlesion. Geniculocortical projection neurons that are identified by the retrograde tracer F

Projections from primary somatosensory cortex to the neostriatum: the role of somatotopic continuity in corticostriatal convergence.

We characterized the organization of corticostriatal projections from rodent primary somatosensory cortex (SI), testing the hypothesis that projections from SI areas representing subcomponents of the forelimb exhibit greater neostriatal overlap than projections from areas representing separate body parts. The anterograde tracers Fluoro-Ruby (FR), Alexa Fluor (AF), and biotinylated dextran amine (BDA) were injected into physiologically identified regions of rat SI. Injection locations were confirmed by examining the SI barrel fields and limb representations in tangential sections processed for

Neuronal changes during forebrain evolution in amniotes: an evolutionary developmental perspective.

Embryology is the interface of genetic inheritance and phenotypic expression in adult forms, and as such is uniquely positioned to illuminate both. Embryonic cell migration pattern, transient connectivity, axonal growth kinetics and fasciculation patterns can clearly be substantially impacted at the striatocortical junction, which appears to be critical for telencephalic development. Similarly, the big questions concerning pallial evolution in amniotes all involve the pivotal region at the pallial-subpallial boundary, an area where complex developmental cross-currents may be involved in the sp

Intrinsic connectivity of human auditory areas: a tracing study with DiI.

The human supratemporal plane contains the primary as well as several other auditory areas. We have investigated the intrinsic connectivity of these areas by means of antero- and retrograde labelling with the carbocyanin dye DiI (1,1'-dioctadecyl-3,3,3',3'-tetramethylindocarbocyanine perchlorate). A total of 30 injections was placed in both hemispheres of four freshly fixed postmortem brains. Labelled neurons and axons were found in cortex around the injection. The retrograde labelling varied from faint to Golgi-like; most of the retrograde labelled neurons were layers II-III pyramids and only

Posteroventral pallidotomy for midbrain tremor after a pontine hemorrhage. Case report.

This 49-year-old man gradually developed a disabling action tremor in the proximal right upper extremity 8 months after suffering a pontine tegmental hemorrhage. The intraoperative microrecording in the nucleus ventralis intermedius (VIM) of the left thalamus revealed tremor-synchronous grouped discharges with a vigorous (2.7 Hz) action tremor predominantly in the shoulder and upper arm. High frequency electrical stimulation in the VIM did not affect the tremor. A posteroventral pallidotomy (PVP) was performed and resulted in the successful alleviation of all tremor activity. Posteroventral pa

A neurophysiological approach to brainstem reflexes. Blink reflex.

The blink reflex (BR) is a generalised phenomenon in mammals. Its teleological protective eye function is perhaps the reason why the BR can be provoked by a multitude of stimuli. As corneal and glabellar reflexes, BR has an inveterate use in the neurological exploration. Some of its physiopathological aspects were discussed more than 100 years ago, and soon half a century will have passed since the first electrophysiological study was published. This review focuses on the BR elicited by the electrical stimulation of the trigeminal supraorbital nerve, a controlled and reliable model in clinical

The irritable larynx syndrome.

Muscular tension dysphonia, episodic laryngospasm, globus, and cough may be considered to be hyperfunctional laryngeal symptoms. Suggested etiological factors for these symptoms include gastroesophageal reflux, psychological problems, and/or dystonia. We propose a unifying hypothesis that involves neural plastic change to brainstem laryngeal control networks through which each of the above etiologies, plus central nervous system viral illness, can play a role. We suggest that controlling neurons are held in a "spasm-ready" state and that symptoms may be triggered by various stimuli. Inclusion

Phrenic nerve inputs to upper cervical (C1-C3) spinothalamic tract neurons in monkeys.

Afferent input from the phrenic nerve enters mid-cervical spinal segments, and previous results in monkeys show that mid-cervical spinothalamic (STT) neurons are activated by groups II and III phrenic afferent input arising from the diaphragm. In rats, dorsal horn neurons in C1-C2 segments receive phrenic input stimulated above the heart, but are not activated by phrenic input arising from diaphragm or abdomen. It is not known if this differential organization occurs in primate STT neurons. We hypothesized that high cervical STT neurons in monkeys also would be preferentially activated by phre

Interocular interactions in esotropia.

In cats the unilateral convergent strabismus surgically induced in the early postnatal life causes monocular spatial vision deficits and loss of binocular activation of the primary visual cortex. It is assumed that the asynchrony of inputs from the two eyes, by disrupting binocular convergence, leads to unbalanced binocular interactions that favour the non-deviated eye. The competitive advantage of this eye reduces both the excitatory drive to the cortex and the behavioural visual capabilities of the strabismic eye that exhibits the loss of the nasal portion of the visual field. Our experiment

The role of the thalamus in schizophrenia.

BACKGROUND: Explaining the diversity of symptoms that occur in schizophrenia is a major conceptual challenge. Perhaps the most powerful strategy is to identify a fundamental cognitive process and/or a fundamental neural circuit. METHODS: Convergent data from our research group in Iowa and from investigators in other centres are summarized. RESULTS: The thalamus plays a key role in information processing. A defect in circuitry connecting the thalamus, frontal cortex, and cerebellum could explain a wide range of symptoms. Neuropathology and imaging studies suggest that patients with schizophre

Reversible changes of motor cortical outputs following immobilization of the upper limb.

We mapped the cortical representations of the abductor pollicis brevis, flexor carpi radialis, biceps and deltoid muscles in six subjects with unilateral wrist fractures, immediately after the removal of the splint. This was repeated 1 month later in three out of the six subjects. Duration of immobilization was 1 month. Muscle maps were obtained by delivering four focal magnetic pulses for each scalp position (1 cm apart with reference to Cz) over the contralateral hemisphere. Motor evoked potentials (MEPs) were averaged off-line and expressed as a percentage of the motor action potential evok

Cutaneous representations of the hand and other body parts in the cuneate nucleus of a primate, and some relationships to previously described cortical representations.

Dynamic properties of primate somatosensory maps are dependent on normal central adjacencies of cutaneous representations. The cuneate nucleus is an important brainstem processing center of cutaneous information. Surprisingly, there are no descriptions of functional representations of the skin in the primate cuneate nucleus; as a result, the relationships of functional representations at the brainstem level and other levels of the somatosensory neuraxis remain obscure. The present neurophysiological study indicates that the main cuneate nucleus of marmoset monkeys (Callithrix jacchus) contains

Neural projections from the frontal cortex to the oculomotor nucleus: an anatomical study using retrograde axonal, anterograde axonal and transneuronal transport of wheat germ agglutinin-conjugated horseradish peroxidase in cats.

Neural projections from the frontal cerebral cortex to the oculomotor nucleus (3N) were investigated in 1- to 2-year-old cats by retrograde and anterograde axonal and transneuronal transport of wheat germ agglutinin-conjugated horseradish peroxidase (WGA-HRP). Following injection of WGA-HRP into the 3N area and its surrounding tissues, retrogradely labeled cells were observed in the anterior sigmoid gyrus, ventral bank of the cruciate sulcus, medial and lateral walls and base of the presylvian sulcus, gyrus rectus and gyrus proreus. Following injection of WGA-HRP into these frontal cortical ar

Maternal alcohol consumption before pregnancy and ultrastructure of neurons and interneuronal connections in rat offspring.

The ultrastructure of neurons and interneuronal connections in sensorimotor cortex have been studied in the offspring at 14- and 21-day-old female rats damaged by alcohol intoxication before pregnancy. We found three categories of ultrastructural changes in cortical neurons and interneuronal connections: delayed maturation of these structures, dystrophic and destructive changes and a signs of compensatory-adaptive processes. The first two categories of neuronal changes more marked at 14 days after birth. Later compensatory-adaptive changes become more significant, but dystrophic and destructiv

Cell interactions control the direction of outgrowth, branching and fasciculation of the HSN axons of Caenorhabditis elegans.

The two serotonergic HSN motor neurons of the nematode Caenorhabditis elegans innervate the vulval muscles and stimulate egg laying by hermaphrodites. By analyzing mutant and laser-operated animals, we find that both epithelial cells of the developing vulva and axons of the ventral nerve cord are required for HSN axonal guidance. Vulval precursor cells help guide the growth cone of the emerging HSN axon to the ventral nerve cord. Vulval cells also cause the two HSN axons to join the ventral nerve cord in two separate fascicles and to defasciculate from the ventral nerve cord and branch at the

Location and connectivity of abdominal motoneurons in the embryo and larva of Drosophila melanogaster.

The soma location and peripheral connectivity of motoneurons in abdominal segments of the embryo and larva of the fruitfly, Drosophila melanogaster are described as an initial step in determining the mechanisms by which motoneurons make connections with their target muscles in a genetically accessible organism. Embryonic motoneuron somata were retrogradely labelled by application of the fluorescent dye, DiI, to the whole peripheral nerve or to its separate anterior or posterior fascicles in segments A5-A7 of late stage 15/early stage 16 embryos. This technique reveals a stereotyped, segmentall

Magnetic stimulation study in mirror movements.

A young man with congenital mirror movements was studied by non-invasive magnetic stimulation. Radiological examination showed no craniocervical or pituitary abnormality except for mild atrophy of the right hippocampus. Magnetic stimulation of the motor cortex caused large amplitudes in the ipsilateral hand muscles, indicating the possibility of functional disorder in the motor pathways from the motor cortex to muscles in the upper extremities.

Postnatal construction of neural circuitry in the mouse olfactory bulb.

We have undertaken a quantitative analysis of the mouse olfactory bulb to address several major questions concerning the development of neural circuitry in the postnatal mammalian brain. These are: (1) To what degree are new elements and circuits added during maturation? (2) How long do such processes go on? and (3) Does postnatal development involve a net addition of circuits and their constituent elements, or is there elimination of some portion of an initial surfeit? Using male mice of known age, weight, and length, we measured the overall size of the bulb, the numbers of processing units (

Selective labeling by propidium iodide injected into the lateral cerebral ventricle of the rat.

Injections of propidium iodide (PI) into the lateral cerebral ventricle of the rat resulted in a bilateral labeling in the septohippocampal nuclei, substantia nigra (SN), ventral tegmental area (VTA), retrorubral nuclei (rr), dorsal and median raphe nuclei, regions within and dorsal to the medial lemniscus of the caudal midbrain, and Purkinje cells in the cerebellum. No labeled neurons were seen in other areas of the brain. The data suggest that PI appears to exhibit selective labeling, and the mechanism underlying the selective labeling is discussed. Combined with Faglu histofluorescence, it
