A new method for the experimental production of necrotic enteritis and its use for studies on the relationships between necrotic enteritis, coccidiosis and anticoccidial vaccination of chickens.

A new method for the experimental production of necrotic enteritis in chickens is described. The main features are the use of a diet high in wheat and fish meal content; oral administration of a non-lethal inoculum of the coccidium Eimeria maxima followed 6 days later by the bacterium Clostridium perfringens type A per cloaca, so that the bacterial inoculum is deposited at the time and place when and where the intestinal coccidial lesions are maximal; grading of coccidial and clostridial lesions in individual birds sampled during the 14 days following the coccidial infection. The new method wa

Experimental production and modulation of human cytotoxic dermatitis in human-murine chimeras.

Human dermatitis-involving cytotoxic interaction between effector lymphocytes and epithelial target cells has thus far been documented in vivo only as naturally occurring disease or as an iatrogenic complication of organ engraftment. In this report, we reproduce human cytotoxic dermatitis via local microinjection of heterologous human lymphocytes into human skin xenografted to mice with severe combined immune deficiency syndrome. Injection sites develop progressive T cell epidermotropism culminating in cytotoxic dermatitis resembling human lichen planus within 4 weeks. Effector T cells express

Development of a Babesia bovis live attenuated vaccine.

Babesia bovis is the blood parasite causing the highest morbidity and mortality of cattle in Cuba. Thus a control program based on immunization with a vaccine using an attenuated strain of the parasite was developed. For this purpose, a virulent B. bovis strain was isolated and then attenuated by the traditional method in which biological characteristics, proteolytic activity and vector transmissibility were studied. The protection conferred by the vaccine was measured by vaccinating cattle which then was challenged with a heterologous strain. In order to assess postvaccination accident risks,

Production and immunochemical characterization of Neisseria meningitidis group B antiserum for the diagnosis of purulent meningitis.

Unlike Neisseria meningitidis groups A, C, Y and W135, the group B capsular polysaccharide has been shown to be chemically and immunologically identical to the capsular polysaccharide of Escherichia coli K1. Both components are sialic acid homopolymers and are poorly immunogenic. Nevertheless, due to the high incidence of Neisseria meningitidis group B meningitis in the population of the State of São Paulo, preparing antiserum to this serogroup for diagnostic purposes has become a matter of high priority. Of the many immunization schemes proposed, intravenous inoculation of whole bacteria prev

[Intolerance reactions of the skin].

Intolerance reactions of the skin in which a decision is made on allergic and toxic genesis have increased in significance in recent years. Whereas antibodies blocking bacterial or viral antigens are formed in the normergic immune reaction, nonblocking antibodies to animal, plant or chemical heterologous substances are formed in an allergy as a result of an misdirected immunological reaction. Sensitization is favored by a) large structure and protein affinity of the allergen, b) irritation of the affected part of the skin by fungi or chemicals and c) genetic predisposition. Depending on the cl

Production of interferons during experimental African trypanosomiasis.

African trypanosomiasis is associated with profound changes in the function of the immune system. In this study we find that alpha/beta and gamma interferon (IFN) are released into the serum of mice infected with Trypanosoma brucei. The parasite-induced rise in serum IFNs is associated with a detectable parasitaemia, but the serum IFN peak precedes the peak parasitaemia in some cases. Unlike other protozoan interferon inducers, no parasite-dependent IFN production was observed in the pre-patent period of infection; while the most virulent clone induced very high IFN levels, no clear difference

Efficacy of a Modified-Live Virus Combination Vaccine (CDV, CAV, CPV, CPiV), CanigenTM DHPPi, in Puppies Vaccinated at Six Weeks of Age.

During their early life, puppies are protected against infectious agents with the presence of maternal derived antibodies (MDA). Vaccination is recommended to start as soon as the levels of MDA begin to wane to ensure that the puppy's immune system can respond effectively to the vaccines and develop active immunity against diseases. Two studies were designed to assess the efficacy of the CanigenTM DHPPi vaccine in puppies from 6 weeks of age. The studies comprised two parts: the efficacy assessment of the Canine Parainfluenza Virus (CPiV) vaccine component against a virulent challenge with CPi

Preclinical assessment of an anti-HTLV-1 heterologous DNA/MVA vaccine protocol expressing a multiepitope HBZ protein.

BACKGROUND: Human T-lymphotropic virus 1 (HTLV-1) is associated with the development of several pathologies and chronic infection in humans. The inefficiency of the available treatments and the challenge in developing a protective vaccine highlight the need to produce effective immunotherapeutic tools. The HTLV-1 basic leucine zipper (bZIP) factor (HBZ) plays an important role in the HTLV-1 persistence, conferring a survival advantage to infected cells by reducing the HTLV-1 proteins expression, allowing infected cells to evade immune surveillance, and enhancing cell proliferation leading to i

Evaluation of the systemic and mucosal immune response induced by COVID-19 and the BNT162b2 mRNA vaccine for SARS-CoV-2.

BACKGROUND: The currently used SARS-CoV-2 mRNA vaccines have proven to induce a strong and protective immune response. However, functional relevance of vaccine-generated antibodies and their temporal progression are still poorly understood. Thus, the central aim of this study is to gain a better understanding of systemic and mucosal humoral immune response after mRNA vaccination with BNT162b2. METHODS: We compared antibody production against the S1 subunit and the RBD of the SARS-CoV-2 spike protein in sera of BNT162b2 vaccinees, heterologous ChAdOx1-S/BNT162b2 vaccinees and COVID-19 patients

Assessment of immune responses to H5N1 inactivated influenza vaccine among individuals previously primed with H5N2 live attenuated influenza vaccine.

During the past decade, a number of H5 subtype influenza vaccines have been developed and tested in clinical trials, but most of them induced poor serum antibody responses prompting the evaluation of novel vaccination approaches. One of the most promising ones is a "prime-boost" strategy, which could result in the induction of prompt and robust immune responses to a booster influenza vaccine following priming with homologous or heterologous vaccine strains. In our study we evaluated immunogenicity of an adjuvanted A(H5N1) inactivated influenza vaccine (IIV) in healthy adult subjects who receiv

The safety and immunogenicity of a MF59-adjuvanted H5N1 prepandemic influenza vaccine in healthy adults primed with homologous or heterologous H5N1 vaccines: an observational study.

BACKGROUND: World Health Organization (WHO) has recommended individuals with increased risk of contracting influenza A H5N1 infection to be immunized against the virus during the inter-pandemic period. Safety and immunogenicity of H5N1 vaccine among participants primed with homologous or heterologous H5N1 vaccines produced by diverse manufactures have not been reported. METHODS: Healthy individuals aged 20 to 60 years old were recruited and stratified into three groups: participants without priming (control group), participants primed with A/Indonesia/05/2005 vaccine, participants primed with

Live attenuated tetravalent dengue virus host range vaccine is immunogenic in African green monkeys following a single vaccination.

UNLABELLED: The causative agent of dengue fever, dengue virus (DENV), is transmitted by mosquitoes, and as distribution of these insects has expanded, so has dengue-related disease. DENV is a member of the Flaviviridae family and has 4 distinct serotypes (DENV-1, -2, -3, and -4). No lasting cross protection is afforded to heterologous serotypes following infection by any one of the individual serotypes. The presence of nonneutralizing antibodies to one serotype can facilitate the occurrence of more-severe dengue hemorrhagic fever through immune enhancement upon infection with a second serotype

Immune signaling of Litopenaeus vannamei c-type lysozyme and its role during microsporidian Enterocytozoon hepatopenaei (EHP) infection.

The microsporidian Enterocytozoon hepatopenaei (EHP) is a fungi-related, spore-forming parasite. EHP infection causes growth retardation and size variation in shrimp, resulting in severe economic losses. Studies on shrimp immune response have shown that several antimicrobial peptides (AMPs) were upregulated upon EHP infection. Among those highly upregulated AMPs is c-type lysozyme (LvLyz-c). However, the immune signaling pathway responsible for LvLyz-c production in shrimp as well as its function against the EHP infection are still poorly understood. Here, we characterized major shrimp immune

Outer membrane vesicles derived from Bordetella pertussis are potent adjuvant that drive Th1-biased response.

For several years, we have been committed to exploring the potential of Bordetella pertussis-derived outer membrane vesicles (OMVBp) as a promising third-generation vaccine against the reemerging pertussis disease. The results of our preclinical trials not only confirm its protective capacity against B. pertussis infection but also set the stage for forthcoming human clinical trials. This study delves into the examination of OMVBp as an adjuvant. To accomplish this objective, we implemented a two-dose murine schedule to evaluate the specific immune response induced by formulations containing O

An attenuated variant of Eastern encephalitis virus: biological properties and protection induced in mice.

Wild type Eastern equine encephalitis virus (E) was compared with a mutant (Em) derived from it. The latter was tested as an attenuated vaccine in mice. They differed in the following properties: Em formed smaller plaques on chick embryo (CE) cell monolayers and, unlike E, did not plaque on mouse embryo (ME) monolayers. Futher, Em had a longer latent period and attained a lower peak titer than E after infection of CE cells, was more senssitive than E to chick interferon, and was less virulent for mice (SC and IP routes) and hamsters (IP route) than E. Both viruses were similar in several other

Evaluating the Immune Response in Rabbits to an Escalating Dose of mRNA-Based HIV-1 Env Immunogens.

BACKGROUND: The development of an effective HIV-1 vaccine remains a major challenge due to HIV-1's extraordinary diversity, high mutation rate, and the rarity of broadly neutralizing antibody (bnAb) precursors. To address these challenges, we have previously immunized rabbits with mRNA-LNPs encoding for HIV-1 envelope glycoproteins (Envs), together with mRNA-LNPs encoding for HIV-1 Gag, which likely mediated the generation of virus-like particles presenting HIV-1 Envs to the immune system in vivo. METHODS: Here, we investigated whether an escalating dose (ED) immunization using mRNA-LNP primi

Broad-Spectrum Vaccines: Challenges and Opportunities (A Systematic Review).

BACKGROUND: Though advances in technologies have created an arena for multiple vaccinology platforms, the world population is still facing health challenges from the deadliest pathogens with a high mutation rate and their tremendous ability of immune evasion. Presently, there is a constant exigence to shift the vaccine development efforts from traditional vaccines to broad-spectrum vaccines for the effective prevention of contagious diseases. MATERIALS AND METHODS: Using several databases, a comprehensive systematic review of the literature pertinent to this article's discussion was carried o

MdLYK7 enhances resistance to Valsa canker by activating salicylic acid signaling and PbeBGLU12-mediated fungal cell wall stress.

Valsa canker, caused by necrotrophic fungi in genus Valsa, is a devastating disease that threatens apple and pear production in China and other regions of East Asia. Receptor-like kinases (RLKs) play crucial roles in pathogen signal perception and the initiation of immune responses. However, key members of RLKs contribute to Valsa canker resistance and potential functions remain largely elusive. In this study, we identified a Malus domestica lysin-motif RLK gene, MdLYK7, whose overexpression substantially enhanced the Valsa canker resistance in apple and pear fruits and in suspension cells of

Influenza vaccination as a modulator of inflammaging: Trained tolerance as a mechanistic lens on heterologous effects in age-related diseases.

Influenza vaccination may confer benefits that extend well beyond seasonal protection against respiratory infection: reduced cardiovascular morbidity and mortality, lower dementia incidence, fewer cardio-renal complications, improved survival in the very old, and emerging signals in oncology and diabetes. Although vulnerable to confounding and evidence in adults aged ≥ 75-80 years and in frail populations remains scarce, their magnitude, cross-organ consistency and apparent dose-response suggest that protection is not explained solely by prevention of infection, but points to a shared upstream
