Mosquito E-20-Monooxygenase Gene Knockout Increases Dengue Virus Replication in Aedes aegypti Cells.

E-20-monooxygenase (E20MO) is an enzymatic product of the shade (shd) locus (cytochrome p450, E20MO). Initially discovered in Drosophila, E20MO facilitates the conversion of ecdysone (E) into 20-hydroxyecdysone (20E) and is crucial for oogenesis. Prior research has implicated 20E in growth, development, and insecticide resistance. However, little attention has been given to the association between the E20MO gene and DENV2 infection. The transcriptome of Ae. aegypti cells (Aag2 cells) infected with DENV2 revealed the presence of the E20MO gene. The subsequent quantification of E20MO gene expres…

Bridging the Gap from Molecular Surveillance to Programmatic Decisions for Malaria Control and Elimination.

An increasing number of molecular and genomic assays are available to study malaria parasite populations. However, so far they have played a marginal role in informing policy and programmatic decision-making. Currently, molecular data are mainly used for monitoring drug efficacy against Plasmodium falciparum; assessing molecular markers of drug and insecticide resistance; and assessing P. falciparum histidine-rich protein 2 and 3 genes (Pfhrp2/3) deletion. We argue that additional use cases for molecular routine surveillance could be implemented in the near future, especially in transmission s…

The influence of feeding behaviour and temperature on the capacity of mosquitoes to transmit malaria.

Insecticide-treated bed nets reduce malaria transmission by limiting contact between mosquito vectors and human hosts when mosquitoes feed during the night. However, malaria vectors can also feed in the early evening and in the morning when people are not protected. Here, we explored how the timing of blood feeding interacts with environmental temperature to influence the capacity of Anopheles mosquitoes to transmit the human malaria parasite Plasmodium falciparum. In laboratory experiments, we found no effect of biting time itself on the proportion of mosquitoes that became infectious (vector

Knockdown of vATPase subunit A impairs larval-pupal development in Plutella xylostella.

Plutella xylostella is a destructive pest damaging the cruciferous plants worldwide. Transgenic plants expressing dsRNA can be widely deployed for pest control. However, highly effective lethal target genes are currently lacking. Here, we assessed the efficacy of RNA interference (RNAi) against P. xylostella through microinjection of in vitro synthesized dsRNA targeting PxvATPaseA. The full-length ORF of PxvATPaseA was 1848 bp, encoding 615 amino acids. PxvATPaseA was widely transcribed during different developmental stages from egg to adult. It was abundantly expressed in the foregut, hindgut

Repellency of Eucalyptol and DEET Against Triatoma infestans, a Chagas Disease Vector: A Proof-of-Concept Study Under a Human Odor Background.

Triatomines are vectors of Chagas disease. Control of Triatoma infestans, one of its main vectors, relies largely on the use of pyrethroid insecticides. However, due to resistance to them as well as environmental concerns, alternative control strategies are required. Here, we evaluated the volatile repellent activity of eucalyptol, carvacrol, carvone, and DEET (N, N-diethyl-3-methylbenzamide) under conditions that prevented the contact of the insects with the compounds, and under a human odor background, providing a realistic assessment of repellency for human protection. Importantly, eucalypt

Multi-Plant Screening and Controlled Evaluation of Two Selected Plant Species Against Tuta absoluta (Meyrick).

Tuta absoluta is an invasive species that has managed to expand beyond its native geographical area and causes severe yield losses in tomato. Despite the application of synthetic insecticides in getting rid of the pest, farmers have reported reduced efficacy over the course of time due to resistance. With regard to ecological pest management approaches, studies regarding the use of companion plants in the management of T. absoluta are scarce. Therefore, we screened fourteen potential companion plants for repellency using a Y-tube olfactometer followed by cage and screenhouse experiments. Data

Genome-Guided Dissection of ser and shlB Uncovers Multifactorial Virulence Mechanisms of Serratia marcescens PXG6 in Plutella xylostella.

The rapid evolution of pesticide resistance in Plutella xylostella has reduced the effectiveness of conventional pest control methods in cruciferous crops. This study investigated the virulence of Serratia marcescens PXG6 against P. xylostella and identified two putative virulence factors, ser (serralysin) and shlB (hemolysin transporter). Whole-genome sequencing revealed a GC content of 59.65%, 4650 predicted genes, multiple secretion systems (types I, V, and VI), and 715 putative virulence-related genes. Bioinformatic analyses predicted type IV pili, serralysin, hemolysin, and flagella as pa

RNAi-Mediated Knockdown of Tektins Does Not Affect Male Fertility in Lygus hesperus.

Mirid plant bugs (Hemiptera: Miridae), including Lygus hesperus (western tarnished plant bug), are key pests of numerous agricultural crops. While management of this pest relies heavily on chemical insecticides, the evolution of resistance and environmental concerns underscore the need for new and more effective approaches. Genetic-based strategies that target male fertiliy are currently being evaluated for population suppression. However, a lack of candidate gene targets with appropriate function, specifically in non-model species like L. hesperus, has hindered progress in the development and

In vitro larvicidal activity of selected azabenzimidazole and diarylquinoline derivatives against Anopheles gambiae and in silico mechanistic analysis.

Different species of mosquitoes are responsible for transmitting infectious diseases such as chikungunya, dengue, Japanese encephalitis, lymphatic filariasis, rift valley fever, west nile fever, yellow fever, zika virus, and malaria. Particularly, malaria infection is endemic in sub-Saharan Africa region, and female anopheles mosquitoes is responsible for the transmission of the parasite causing the infection. The growing resistance of mosquitoes to conventional insecticides and the need to complement existing strategies for the elimination of malaria transmission necessitate the exploration o

Development of transgenic corn planthopper Peregrinus maidis that express the tetracycline transactivator.

The corn planthopper, Peregrinus maidis, is a vector of several maize viruses and is consequently a significant agricultural pest in many tropical and subtropical regions. As P. maidis has developed resistance to insecticides, the aim of this study was to develop transgenic P. maidis strains that could be used for future genetic biocontrol programs. To facilitate the identification of transgenic P. maidis, we isolated and characterized the promoters for the P. maidis ubiquitin-like and profilin genes. Transient expression assays with P. maidis embryos showed that both promoters were active. Tr

Cu2+-dependent stereoselective hydrolysis of a chiral organophosphonothioate insecticide for domestic mammals' sera and its albumins.

Acute toxicity of organophosphate (OPs) pesticides is a public health problem. The adverse effects are associated with the inhibition and aging of nervous system B-esterases such as acetyl cholinesterase (AChE) and neuropathic target esterase (NTE). Treatment based on A-esterases such as mammal serum paraoxonase-1 has been suggested. This ex vivo study shows the Cu2+-dependent hydrolysis of trichloronate (TCN), a racemic organophosphonothioate insecticide, in human and domestic mammal serum (dog, goat, pig, sheep and cow). Ca2+-dependent (2.5 mM) or EDTA-resistant (5 mM) activity (1-6%) was no

Compelling Evidence for the Activity of Antiviral Peptides against SARS-CoV-2.

Multiple outbreaks of epidemic and pandemic viral diseases have occurred in the last 20 years, including those caused by Ebola virus, Zika virus, and severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). The emergence or re-emergence of such diseases has revealed the deficiency in our pipeline for the discovery and development of antiviral drugs. One promising solution is the extensive library of antimicrobial peptides (AMPs) produced by all eukaryotic organisms. AMPs are widely known for their activity against bacteria, but many possess additional antifungal, antiparasitic, insecticid

Role of Endocrine-Disrupting Chemicals in the Pathogenesis of Non-Alcoholic Fatty Liver Disease: A Comprehensive Review.

Non-alcoholic fatty liver disease (NAFLD) is considered the most common liver disorder, affecting around 25% of the population worldwide. It is a complex disease spectrum, closely linked with other conditions such as obesity, insulin resistance, type 2 diabetes mellitus, and metabolic syndrome, which may increase liver-related mortality. In light of this, numerous efforts have been carried out in recent years in order to clarify its pathogenesis and create new prevention strategies. Currently, the essential role of environmental pollutants in NAFLD development is recognized. Particularly, endo

Can Generalist Predators Control Bemisia tabaci?

The whitefly, Bemisia tabaci, has developed resistance to many insecticides, renewing interest in the biological control of this global pest. Generalist predators might contribute to whitefly suppression if they commonly occur in infested fields and generally complement rather than interfere with specialized natural enemies. Here, we review literature from the last 20 years, across US cropping systems, which considers the impacts of generalist predators on B. tabaci. Laboratory feeding trials and molecular gut content analysis suggest that at least 30 different generalist predator species will

Effect of Wheat Cultivars and Blends on the Oviposition and Larval Mortality of Cephus cinctus (Hymenoptera: Cephidae) and Parasitism by Bracon cephi (Hymenoptera: Braconidae).

The wheat stem sawfly (Cephus cinctus Norton) is a major historical pest of wheat in the northern Great Plains of North America. The insect spends most of its life as a larva protected inside grass stems so that its management has relied on strategies other than insecticides. We conducted a study in southern Alberta from 2006-2009 to assess the effects of wheat species, cultivar, seeding rate, and blending a resistant and a vulnerable cultivar, on oviposition, larval infestation, and cutting damage. The mortality caused by its primary parasitoid, Bracon cephi (Gahan), was also assessed to inve

Genome sequence of Yersinia pestis, the causative agent of plague.

The Gram-negative bacterium Yersinia pestis is the causative agent of the systemic invasive infectious disease classically referred to as plague, and has been responsible for three human pandemics: the Justinian plague (sixth to eighth centuries), the Black Death (fourteenth to nineteenth centuries) and modern plague (nineteenth century to the present day). The recent identification of strains resistant to multiple drugs and the potential use of Y. pestis as an agent of biological warfare mean that plague still poses a threat to human health. Here we report the complete genome sequence of Y. p

An outbreak of plague in Hlegu, Burma in 1977.

An outbreak of plague occurred in Hlegu Town, Burma in February and March 1977. Altogether 14 bubonic plague cases, with 2 deaths were reported, mostly children under 15 years of age. Twice as many males as females were affected. A retrospective investigation showed that a rat fall took place in the last week of January. Only 9.5% of the rodents which died in the epizootic were found to be infected with Y. pestis; this apparent low infection rate probably resulted from the inclusion in the survey of some dead rats which died of other causes. 14.5% out of 48 trapped rodents were found to be ser
