Impaired liver regeneration of steatotic rats after portal vein ligation: a particular emphasis on (99m)Tc-DISIDA scintigraphy and adiponectin signaling.

BACKGROUND & AIMS: Portal vein ligation (PVL) is increasingly employed prior to major hepatectomy in order to enhance the volume of the future liver remnant (FLR) and to avoid post-hepatectomy liver failure. The efficacy of PVL on subjects with non-alcohol fatty liver disease (NAFLD) is largely unknown. METHODS: Sprague-Dawley rats fed with normal diet (control) and methionine-choline deficient diet (MCD) were used. The animals underwent PVL and were sacrificed at indicated time points. RESULTS: Livers from MCD rats exhibited a decreased BrdU and Ki-67 labelling index, and an increased apopt

Sustained IL-4 exposure leads to a novel pathway for hemophagocytosis, inflammation, and tissue macrophage accumulation.

Erythrophagocytosis and inflammation from activated macrophages occur in distinct clinical scenarios. The presence of CD8(+) T cells and interferon-γ (IFN-γ) production is required to induce disease in mouse models of hemophagocytic lymphohistiocytosis. We investigated the roles of a different class of proinflammatory cytokines, interleukin-4 (IL-4) and IL-13, in the induction of inflammatory tissue macrophage accumulation and/or hemophagocytosis. We found that large amounts of IL-4, but not IL-13, delivered via an implanted mini-pump or IL-4/anti-IL-4 complexes, lead to substantial YM1(+) tis

Cellular organization and phylogeny of Cryptonaiada gen. n. (Cryptonaiadidae fam. n.) shed light on the evolution of intracellular vertebrate parasitism in Apicomplexa.

Apicomplexan parasites have coevolved with animal hosts for hundreds of millions of years, yet only a handful of their lineages have made the transition to infecting vertebrates. Among the least understood vertebrate-infecting apicomplexans is Nematopsis temporariae, recently rediscovered after nearly a century of neglect. Here, we isolated this parasite from tadpole tissues and examined its host relationship, ultrastructure, and phylogenetic placement. We show that N. temporariae's monozoic oocysts are persistently associated with hepatic Kupffer cells in tadpoles of several frog species and …

Therapeutic potential and mechanisms of flavonoids from Citrus grandis 'Tomentosa' in metabolic dysfunction-associated steatotic liver disease: a focus on immune-inflammatory signaling pathways.

INTRODUCTION: Metabolic dysfunction-associated steatotic liver disease (MASLD) constitutes a major global health burden, with limited therap … e rigorous studies validating CGT-specific regulatory effects on hepatic immune components, such as Kupffer cells and intrahepatic T-cell subsets under MASLD pathological conditions. DISCUSSION: The present scoping review confirms the preclinical potential of CGT flavonoids as novel botanical candidates for MASLD treatment, primarily through the suppression of …

Dysregulation of antioxidant responses in patients diagnosed with concomitant Primary Sclerosing Cholangitis/Inflammatory Bowel Disease.

OBJECTIVE: Primary Sclerosing Cholangitis (PSC) is a chronic cholestatic liver disease that is characterized by severe peri-biliary tract inflammation and fibrosis, elevated oxidative stress and hepatocellular injury. A hallmark of PSC patients is the concurrent diagnosis of Inflammatory Bowel Disease occurring in approximately 70%-80% of PSC patients (PSC/IBD). The objective of this study was to determine the impact of end stage PSC/IBD on cellular antioxidant responses and the formation of protein carbonylation. METHODS: Using hepatic tissue and whole cell extracts isolated from age-matched

ER stress-mediated impairment of hepatic lipid export is associated with steatosis in AKI-induced remote liver injury.

Acute kidney injury (AKI) frequently causes remote organ injury including hepatic steatosis, yet whether lipid accumulation reflects increased synthesis or impaired clearance has not been resolved. We used a murine ischemia-reperfusion AKI model. Unbiased liver proteomics was performed at 24 h after reperfusion, and dysregulated pathways were identified by Gene Set Enrichment Analysis. Results were validated by Western blotting, qPCR, and immunohistochemistry. These findings were complemented by retrospective analysis of two intensive care unit (ICU) databases (the Medical Information Mart for

Protein tyrosine phosphatase 1b deficiency protects against hepatic fibrosis by modulating nadph oxidases.

Inflammation is typically associated with the development of fibrosis, cirrhosis and hepatocellular carcinoma. The key role of protein tyrosine phosphatase 1B (PTP1B) in inflammatory responses has focused this study in understanding its implication in liver fibrosis. Here we show that hepatic PTP1B mRNA expression increased after bile duct ligation (BDL), while BDL-induced liver fibrosis was markedly reduced in mice lacking Ptpn1 (PTP1B-/-) as assessed by decreased collagen deposition and α-smooth muscle actin (α-SMA) expression. PTP1B-/- mice also showed a significant increase in mRNA levels

Decrease of platelet-endothelial cell adhesion molecule 1-gene-expression in inflammatory cells and in endothelial cells in the rat liver following CCl(4)-administration and in vitro after treatment with TNFalpha.

UNLABELLED: Platelet-endothelial cell adhesion molecule (PECAM-1), a member of the Ig superfamily is strongly expressed at endothelial cell-cell junctions, on most leukocytes and on monocytes. PECAM-1 has been implicated as a key mediator of the transendothelial migration of leukocytes and monocytes. To further define the physiological role of PECAM-1, we studied the modulation of PECAM-1-expression in a model of liver inflammation in both mononuclear cells (MCs) and sinusoidal endothelial cells (SECs). In normal rat liver sections, PECAM-1 immunohistology indicated a sinusoidal pattern simila

Characterization of fumonisin toxicity in orally and intravenously dosed swine.

Fumonisin B1 (FB1), a recently identified mycotoxin produced by Fusarium moniliforme in corn, has been shown to cause death in swine due to … cyte microvilli; membranous material in hepatic sinusoids; and multilamellar bodies in hepatocytes, Kupffer cells, pancreatic acinar cells and pulmonary macrophages. Pulmonary intravascular macrophages (PIMs) contained large amounts of membranous material. Thus, the target organs of fumonisin in the pig are the lung, liver, and pancreas. At lower doses, slowly …

IL-22 and non-ELR-CXC chemokine expression in chronic hepatitis B virus-infected liver.

Hepatitis B virus infection is still a major global health problem, despite decades of research. Interleukin (IL)-22 induces acute phase reactants and chemokines, favors anti-microbial defence and protects tissues from damage. IL-22 is important in chronic skin inflammation, but its role in chronic hepatitis B (CHB) is unclear. This study explores the association between intra-hepatic IL-22 expression, its relevant associated cytokines and the severity of liver inflammation/fibrosis in CHB patients. IL-22, IL-17, IL-10, IL-6, non-ELR-CXC chemokines (CXCL-9, CXCL-10, CXCL-11), fibroblast growth

Depletion of activated hepatic stellate cell correlates with severe liver damage and abnormal liver regeneration in acetaminophen-induced liver injury.

Hepatic stellate cells (HSCs) are important part of the local 'stem cell niche' for hepatic progenitor cells (HPCs) and hepatocytes. However, it is unclear as to whether the products of activated HSCs are required to attenuate hepatocyte injury, enhance liver regeneration, or both. In this study, we performed 'loss of function' studies by depleting activated HSCs with gliotoxin. It was demonstrated that a significantly severe liver damage and declined survival rate were correlated with depletion of activated HSCs. Furthermore, diminishing HSC activation resulted in a 3-fold increase in hepatoc

Damage-associated molecular patterns in hepatic ischemia-reperfusion injury: spatiotemporal signatures, biomarker potential, and clinical translation.

Hepatic ischemia-reperfusion injury (HIRI) remains a key contributor to early allograft dysfunction (EAD), post-hepatectomy liver failure, biliary complications, and the underutilization of marginal donor grafts. Damage-associated molecular patterns (DAMPs) are central mediators of sterile inflammation in HIRI; however, their translational relevance extends beyond being upstream inflammatory triggers. In this review, we reframe DAMP biology by considering spatiotemporal release, sampling accessibility, biomarker utility, and therapeutic tractability. Rather than providing another broad overvie

Albumin Nanoparticles Increase the Efficacy of Doxorubicin Hydrochloride Liposome Injection Based on Threshold Theory.

One of the most significant reasons hindering the clinical translation of nanomedicines is the rapid clearance of intravenously injected nanoparticles by the mononuclear phagocyte system, particularly by Kupffer cells in the liver, leading to an inefficient delivery of nanomedicines for tumor treatment. The threshold theory suggests that the liver's capacity to clear nanoparticles is limited, and a single high dose of nanoparticles can reduce the hepatic clearance efficiency, allowing more nanomedicines to reach tumor tissues and enhance therapeutic efficacy. Building upon this theory, researc…
