Multi-omics pan-cancer study of cuproptosis core gene FDX1 and its role in kidney renal clear cell carcinoma.

BACKGROUND: The mechanism of copper-induced cellular death was newly discovered and termed cuproptosis. Inducing cuproptosis in cancer cells is well anticipated for its curative potential in treating tumor diseases. However, ferredoxin 1 (FDX1), the core regulatory gene in cuproptosis, is rarely studied, and the regulation of FDX1 in tumor biology remains obscure. A comprehensive pan-cancer analysis of FDX1 is needed. METHODS: Thirty-three types of tumors were included with paired normal tissues in The Cancer Genome Atlas (TCGA) and the Genotype-Tissue Expression (GTEx) datasets. The interact

Restraint of Fumarate Accrual by HIF-1α Preserves miR-27a-Mediated Limitation of Interleukin 10 during Infection of Macrophages by Histoplasma capsulatum.

Hypoxia-inducible factor 1α (HIF-1α) regulates the immunometabolic phenotype of macrophages, including the orchestration of inflammatory and antimicrobial processes. Macrophages deficient in HIF-1α produce excessive quantities of the anti-inflammatory cytokine interleukin 10 (IL-10) during infection with the intracellular fungal pathogen Histoplasma capsulatum (R. A. Fecher, M. C. Horwath, D. Friedrich, J. Rupp, G. S. Deepe, J Immunol 197:565-579, 2016, https://doi.org/10.4049/jimmunol.1600342). Thus, the macrophage fails to become activated in response to proinflammatory cytokines and remains

Flexible Metabolism and Suppression of Latent Enzymes Are Important for Escherichia coli Adaptation to Diverse Environments within the Host.

Bacterial metabolism is necessary for adaptation to the host microenvironment. Flexible metabolic pathways allow uropathogenic Escherichia coli (UPEC) to harmlessly reside in the human intestinal tract and cause disease upon extraintestinal colonization. E. coli intestinal colonization requires carbohydrates as a carbon source, while UPEC extraintestinal colonization requires gluconeogenesis and the tricarboxylic acid cycle. UPEC containing disruptions in two irreversible glycolytic steps involving 6-carbon (6-phosphofructokinase; pfkA) and 3-carbon (pyruvate kinase; pykA) substrates have no f

Muscle biopsies from human muscle diseases with myopathic pathology reveal common alterations in mitochondrial function.

Muscle diseases are clinically and genetically heterogeneous and manifest as dystrophic, inflammatory and myopathic pathologies, among others. Our previous study on the cardiotoxin mouse model of myodegeneration and inflammation linked muscle pathology with mitochondrial damage and oxidative stress. In this study, we investigated whether human muscle diseases display mitochondrial changes. Muscle biopsies from muscle disease patients, represented by dysferlinopathy (dysfy) (dystrophic pathology; n = 43), polymyositis (PM) (inflammatory pathology; n = 24), and distal myopathy with rimmed vacuol

Antagonism of vWF inhibits both injury induced arterial and venous thrombosis in the hamster.

von Willebrand factor (vWF) is instrumental in arterial but has also been implicated in venous thrombogenesis. To address its role in venous thrombosis, experimental thrombosis was induced in the carotid artery and the femoral vein of hamsters, following which thrombus prevention by two different antagonists of vWF was studied. The first antagonist was the anti-human vWF monoclonal antibody AJvW-2, which inhibits the botrocetin and ristocetin induced aggregation of human blood platelets. AJvW-2 reacts with an epitope present in the A1 domain of vWF in very different species (human, pig, rabbit

Alterations in nicotinamide and adenine nucleotide systems during mixed-function oxidation of p-nitroanisole in perfused livers from normal and phenobarbital-treated rats.

The contents of adenine nucleotides as well as steady-state concentrations of a number of glycolytic, pentose phosphate-pathway and tricarboxylic acid-cycle intermediates were measured in extracts of livers from normal and phenobarbital-treated rats that were perfused with p-nitroanisole. Metabolites were measured in livers that were freeze-clamped during periods of maximal rates of drug metabolism. Treatment of rats with phenobarbital increased rates of p-nitroanisole O-demethylation approx. fivefold. The concentrations of lactate, xylulose 5-phosphate and ribulose 5-phosphate were increased

α-Ketoglutarate enhances milk protein yield in dairy cows accompanied by increased rumen microbial protein synthesis and improved dietary crude protein digestibility.

Alpha-ketoglutarate (AKG), a key intermediate in the tricarboxylic acid cycle, has been demonstrated to exert multiple physiological benefits, including promoting growth, enhancing nitrogen utilization, improving immunity, and optimizing intestinal health in monogastric livestock and aquatic animals. However, studies investigating its effects in ruminants remain limited. This study systematically evaluated the effects of AKG on rumen fermentation characteristics, production performance, nutrient digestibility, and health parameters in mid-lactation dairy cows through a combination of in vitro

Ion transport and membrane potential in CNS myelinated axons. II. Effects of metabolic inhibition.

Compound resting membrane potential was recorded by the grease gap technique (37 degrees C) during glycolytic inhibition and chemical anoxia in myelinated axons of rat optic nerve. The average potential recorded under control conditions (no inhibitors) was -47 +/- 3 (SD) mV and was stable for 2-3 h. Zero glucose (replacement with sucrose) depolarized the nerve in a monotonic fashion to 55 +/- 10% of control after 60 min. In contrast, glycolytic inhibition with deoxyglucose (10 mM, glucose omitted) or iodoacetate (1 mM) evoked a characteristic voltage trajectory consisting of four distinct phas

OGDH primes macrophage for M1-like polarization and ferroptosis in sepsis associated acute lung injury.

BACKGROUND: Sepsis-associated acute lung injury (S-ALI) is a clinical syndrome characterized by dysregulated inflammation and overwhelming oxidative stress, and is associated with a poor prognosis. Alpha-ketoglutarate dehydrogenase (OGDH) is a key enzyme in the tricarboxylic acid cycle that catalyzes the oxidative decarboxylation of α-ketoglutarate (α-KG) to succinyl-CoA, a critical step linking mitochondrial energy metabolism to immune response. However, the mechanism by which OGDH participates in S-ALI remains unclear. METHODS: The mouse model of sepsis was established via intraperitoneal i

Machine learning identifies autophagy biomarkers driving microvascular injury in cardiac ischemia/reperfusion.

BACKGROUND: Ischemic heart disease (IHD) remains a leading global cause of mortality. While reperfusion is essential for treating ischemic cardiomyopathy, it paradoxically induces myocardial ischemia/reperfusion (I/R) injury targeting the microvascular endothelium. The link between autophagy and microvascular damage in myocardial I/R requires clarification. OBJECTIVES: This study aimed to identify autophagy-related biomarkers of myocardial I/R injury using integrated bioinformatics and experimental validation, characterize the associated molecular and immune features, and evaluate the role of

Nucleo-mitochondrial asymmetry profiles the proliferative engine and spatial niche reconstruction in clear cell renal cell carcinoma.

BACKGROUND: Tumor heterogeneity is the key driver of disease progression and therapeutic resistance in clear cell renal cell carcinoma (ccRCC). Within this landscape, mitochondrial (MT) heterogeneity has emerged as a critical but poorly understood feature. This study identified a specific manifestation of MT heterogeneity termed "nucleo-mitochondrial expression asymmetry (NMA)". It is characterized by a dysregulated burst of mitochondrial DNA (mtDNA)-encoded genes compared to the nuclear genome, marking a pivotal tipping point in tumor proliferation and spatial reconstruction. METHODS: We emp

Influence of a 12 week at-home resistance exercise program on 13C-glucose metabolism in patients with metabolic myopathies.

BACKGROUND: Metabolic myopathies (MM) are a diverse group of genetic disorders that impair cellular energy metabolism, resulting in exercise intolerance and fatigue. Previous studies have reported benefits of physiotherapy in patients with MM, however, objective assessments of their efficacy remain limited. The 13C glucose breath test (13C-GBT) is a non-invasive measure of glucose metabolism through the detection of 13CO2 expelled from the tricarboxylic acid cycle. This study utilizes the in vivo13C-GBT assessment pre- and post-therapeutic intervention to evaluate the impact of a supervised 12

Itaconate alleviates anesthesia/surgery-induced cognitive impairment by activating a Nrf2-dependent anti-neuroinflammation and neurogenesis via gut-brain axis.

BACKGROUND: Postoperative cognitive dysfunction (POCD) is a common neurological complication of anesthesia and surgery in aging individuals. Neuroinflammation has been identified as a hallmark of POCD. However, safe and effective treatments of POCD are still lacking. Itaconate is an immunoregulatory metabolite derived from the tricarboxylic acid cycle that exerts anti-inflammatory effects by activating the nuclear factor erythroid 2-related factor 2 (Nrf2) pathway. In this study, we investigated the effects and underlying mechanism of 4-octyl itaconate (OI), a cell-permeable itaconate derivati

Enhancement of β-Lactam-Mediated Killing of Gram-Negative Bacteria by Lysine Hydrochloride.

Widespread bacterial resistance among Gram-negative bacteria is rapidly depleting our antimicrobial arsenal. Adjuvants that enhance the bactericidal activity of existing antibiotics provide a way to alleviate the resistance crisis, as new antimicrobials are becoming increasingly difficult to develop. The present work with Escherichia coli revealed that neutralized lysine (lysine hydrochloride) enhances the bactericidal activity of β-lactams in addition to increasing bacteriostatic activity. When combined, lysine hydrochloride and β-lactam increased expression of genes involved in the tricarbox

The reduction of XIAP is associated with inflammasome activation in RPE: implications for AMD pathogenesis.

BACKGROUND: Age-related macular degeneration (AMD) is a multifactorial chronic disease of the eye. Several candidate pathways have been hypothesized to play a role in AMD pathogenesis. Our work and those of others suggests inflammasome activity as a mechanism associated with retinal pigment epithelial (RPE) cell demise. X-linked inhibitor of apoptosis protein (XIAP), an anti-apoptosis factor, has recently been shown to regulate inflammasome activity in non-ocular cells. The purpose of this study is to characterize XIAP's regulatory role in RPE. METHODS: Protein lysates of eye tissues from rat

New insights into the genic and metabolic characteristics of induced pluripotent stem cells from polycystic ovary syndrome women.

BACKGROUND: Polycystic ovary syndrome (PCOS) is a common endocrine and metabolic disorder that affects female fertility. However, with the lack of a corresponding research model, the pathology mechanism of PCOS is poorly understood. Induced pluripotent stem cell (iPSC) technology has been recognized as means to generate patient-specific stem cells for disease modeling. METHODS: The mRNA abundance of iPSCs was analyzed by RNA microarray and real-time polymerase chain reaction (RT-PCR). Karyotyping of iPSCs was performed with cytogenetic analysis. In Vivo-Selected Compensatory Mutations Restore the Fitness Cost of Mosaic penA Alleles That Confer Ceftriaxone Resistance in Neisseria gonorrhoeae.

Resistance to ceftriaxone in Neisseria gonorrhoeae is mainly conferred by mosaic penA alleles that encode penicillin-binding protein 2 (PBP2) variants with markedly lower rates of acylation by ceftriaxone. To assess the impact of these mosaic penA alleles on gonococcal fitness, we introduced the mosaic penA alleles from two ceftriaxone-resistant (Cror) clinical isolates (H041 and F89) into a Cros strain (FA19) by allelic exchange and showed that the resultant Cror mutants were significantly outcompeted by the Cros parent strain in vitro and in a murine infection model. Four Cror compensatory m

The ncRNA-Epigenetic-Cuproptosis Axis in Liver Diseases: From Copper Homeostasis to Precision Hepatology.

Cuproptosis, a recently defined copper-dependent form of regulated cell death driven by excess intracellular copper, is characterized by copper binding to lipoylated tricarboxylic acid (TCA) cycle proteins, causing aberrant protein oligomerization, iron-sulfur cluster loss, and proteotoxic stress. Given the liver's central role in copper homeostasis, hepatocytes are particularly vulnerable to this death modality. This review examines the emerging non-coding RNA (ncRNA)-epigenetic-cuproptosis axis in liver diseases, critically evaluating the evidence across a spectrum from well-established bioc
