The fascial involvement score retains diagnostic value for complicated appendicitis on CT when specific signs are absent.

OBJECTIVE: To evaluate the diagnostic performance of the fascial involvement score in predicting complicated appendicitis among patients without extraluminal air, extraluminal appendicolith, or abscess on CT. METHODS: This single-center retrospective study initially included 307 patients with surgically and/or pathologically confirmed appendicitis who underwent unenhanced CT and lacked the three specific CT signs of complicated appendicitis. Of these, 287 patients had assessable fascial structures and formed the primary analytic sample for the fascial involvement score. The fascial involvemen

CDKN2A alterations as possible molecular link between environmental pollutants and pathogenesis of liver cancer: Epidemiology, mechanisms, and insights.

Hepatocellular carcinoma (HCC) represents a global public health burden of increasing magnitude, driven by risk factors including viral hepatitis, metabolic dysfunction-associated fatty liver disease and various environmental carcinogens, which may inactivate key tumor suppressor genes or activates oncogenes. Recent studies have linked various environmental exposures with inactivation of tumor suppressor gene CDKN2A (cyclin dependent kinase inhibitor 2A) and its encoded proteins p16INK4a and p14ARF to various molecular pathways leading to hepatocarcinogenesis. Current studies suggest p16INK4a

Rapidly separable bubble microneedle-patch system present superior transdermal mRNA delivery efficiency.

Traditional mRNA vaccine formulation loaded by lipid nanoparticle (mRNA-LNP) has several shortcomings in clinical application, including the need for cryopreservation, discomfort associated with intramuscular injections, and the risk of liver aggregation. Dissolvable microneedles (DMNs), as a novel transdermal drug delivery platform, can overcome the skin barrier to deliver drugs directly into the skin in a minimally invasive manner. However, mRNA-LNP is unstable and easily degraded during the solidification of DMN. In this study, we proposed to establish a rapidly dissolvable bubble microneed

Lifestyle, environment and other major determinants of frailty in older adults: a population-based study from the UK Biobank.

Frailty is a geriatric multidimensional syndrome characterized by a loss of physiologic reserves and disproportionate vulnerability to external stressors and associated with increased risk of multiple negative health outcomes. Since frailty can be prevented, controlled, and even reverted in its early stages, identifying the main factors involved in its development is crucial to implement preventive and/or restorative interventions. The aim of this study was to assess the impact of a broad range of parameters, including host factors, lifestyle, diet, and environmental and occupational condition

[Research advances of exosomal micrornas in regulating the pathogenesis of acute lung injury/acute respiratory distress syndrome].

Acute lung injury/acute respiratory distress syndrome (ALI/ARDS) is a severe critical condition marked by rapid progression and high fatality. It results from direct/indirect lung-related or systemic triggers, leading to widespread injury of lung epithelial and endothelial cells. Its pathogenesis involves uncontrolled inflammation and breakdown of the lung's blood-air barrier due to leaky blood vessels and epithelial damage. Current management of ALI/ARDS remains primarily supportive, offering symptomatic relief but limited improvement in prognosis, necessitating deeper exploration of upstream

Intertidal encapsulated embryos exposed to environmental stressors associated with the tidal cycle: A study based on oxidative stress and antioxidant response in early stages of the muricid snail Acanthina monodon.

Intertidal environments are highly dynamic, exposing organisms to multiple physical stressors simultaneously. This study evaluates the physiological responses of encapsulated Acanthina monodon embryos to stressors associated with tidal cycles, using oxidative damage and antioxidant activity as indicators. Egg capsules collected from the rocky intertidal zone were subjected to a tidal cycle including 3 h of emersion followed by re-immersion. Environmental variables such as temperature, humidity, PAR and UV-B radiation were recorded. Samples were analyzed for lipid peroxidation, protein carbonyl

Zein Monolayers: Characterization and Interaction with (Bio)surfactants.

Zein is the main protein of corn seeds, which is often employed in food packaging and as a model of keratin. In this study, zein monolayers were deposited from nonconventional solvents: aqueous ethanol and acetic acid, on pure water that was later exchanged for 1% (bio)surfactant solutions: SDS, CTAB, Triton X-100, and the saponin-rich plant extracts of soapwort (Saponaria officinalis L.) and cowherb (Saponaria vaccaria [P. Mill.] Rauschert), as well as Quillaja bark saponins (QBS). The monolayers on pure water could be reversibly compressed up to ∼47 mN/m. On the basis of neutron reflectivity

Steam Cooking Methods Promote the Transfer of Viable Antibiotic-Resistant Pathogens from Water into Air.

Steam cooking is an ancient and widely used method for sterilizing water and food globally. However, its effectiveness may be compromised by the ubiquitous presence of antibiotic resistance genes (ARGs) and antibiotic-resistant bacteria (ARB) in these media. Here, we combined metagenomic sequencing, quantitative PCR analysis, plate culture, and Sanger sequencing to examine the effects of steam cooking on the profiles of antibiotic resistance in cooked fish, tap water, and indoor air in real cooking environments (i.e., a canteen and a home kitchen) and a laboratory chamber. We found that while

Optimized microfluidic blow spinning process for single-step manufacturing of crosslinked chitosan nanofilaments using a quality-by-design approach.

Nanofibrous patches composed of chitosan (CS) present promising solutions for wound healing by supporting cell growth and enabling controlled drug release. This study introduces a new single-step microfluidic blow-spinning process that blends CS with polyethylene oxide (PEO) and crosslinks it with tripolyphosphate (TPP) using a series of laminar flow chips integrated into a blow-spinning apparatus. The microfluidic mixing between PEO and CS solutions at different concentrations was simulated with computational fluid dynamics (CFD). This manufacturing process was optimized through a Design of E

Bioinspired Iontronic Gas Sensor for Selective CO2 Sensing via Reversible Lysine Carbamylation.

Carbon dioxide (CO2) is ubiquitous in both natural and indoor atmospheres, playing a pivotal role in global climate regulation, air quality, and biological processes. However, their chemical inertness poses inherent challenges for developing compact, high-sensitivity detection platforms. In living organisms, CO2 often modulates protein function through reversible carbamylation of lysine residues, illustrating a finely tuned molecular recognition mechanism. Inspired by this principle, we developed a bioinspired iontronic sensor by embedding lysine moieties into a cross-linked poly(ethylene glyc

Comparing UV and Diesel Cutaneous Damage and Evaluating the Protective Role of a Topical Antioxidant Mixture Containing Vitamin C, E and Ferulic Acid.

Cutaneous tissue is one of the main targets of outdoor stressors, and nowadays, the effect of pollution on skin conditions and premature skin ageing has been well correlated, although the exact effect that different pollutants have on the skin has not been well defined, especially when compared to other stressors. Among the air pollutants, UV radiation and particulate matter (PM) have been found among the most aggressive in terms of skin damage, inducing oxinflammatory responses, promoting degradation of extracellular matrix (ECM) components, and compromising the cutaneous defensive barrier. Subdural Empyema and Pneumocephalus From Chronic Polypoid Sinusitis in an Elderly Woman With Rapid Neurological Recovery: A Report of a Rare Case.

BACKGROUND Subdural empyema (SDE) is a rare, life-threatening intracranial infection characterized by pus accumulation between the dura and arachnoid mater. It is most commonly associated with sinusitis, typically affecting young males. The presence of pneumocephalus without trauma or surgery is exceedingly rare and should raise concern for intracranial infection. Early diagnosis through neuroimaging and prompt multidisciplinary intervention are crucial for preventing morbidity and mortality. CASE REPORT We present a case of SDE and atraumatic pneumocephalus in a 76-year-old woman who was foun

Evaporation-driven digital ELISA with micro-droplet arrays for ultrafast detection of low-abundance proteins.

Early detection of ultra-low-abundance protein biomarkers is critical for improving clinical outcomes. However, many conventional immunoassays and proteomic platforms exhibit high limits of detection (LOD) and long incubation times, complicating low-to sub-femtomolar measurements. In addition, state-of-the-art digital ELISAs often rely on magnetic beads, oil emulsions, and pump-valve benchtop analyzers-bulky and costly systems that hinder broad deployment. Here, we present a digital ELISA (dELISA) that lowers LOD and shortens time-to-result by harnessing rapid, repeated femtoliter-scale drople

Probiotic acoustic biosensors for noninvasive imaging of gut inflammation.

Inflammatory bowel diseases (IBD) affect millions of people globally, result in severe symptoms, and are difficult to diagnose and monitor - often necessitating the use of invasive and costly methods such as colonoscopies or endoscopies. Engineered gut bacteria offer a promising alternative due to their ability to persist in the gastrointestinal (GI) tract and sense and respond to specific environmental signals. However, probiotics that have previously been engineered to report on inflammatory and other disease biomarkers in the Gl tract rely on fluorescent or bioluminescent reporters, whose s

SCG2 Transcriptionally Activated by SP1 Promotes Nasal Epithelial Cell Proliferation and Epithelial Mesenchymal Transition.

Chronic rhinosinusitis with nasal polyps (CRSwNP) is characterized by a tendency to recur and a poor prognosis. Epithelial mesenchymal transition (EMT) and proliferation of nasal epithelial cells (NECs) play an important role in CRSwNP development. Secretogranin II (SCG2) is reported to be an EMT-related gene, but its role in CRSwNP has not been reported. In this study, human NECs were cultured in an air-liquid interface culture system and stimulated with IL-13 to maintain or promote the CRSwNP state. EMT-associated protein expression levels were examined by real-time quantitative PCR and West

Proposing Bromo-Epi-Androsterone (BEA) for Stiff Person Syndrome (SPS).

SPS is characterized by progressive spasmodic muscular rigidity. SPS is thought to be an autoimmune disease with a prominent feature of antibodies against glutamic acid decarboxylase (GAD). GAD is responsible for the enzymatic conversion of glutamic acid (glutamate) into the inhibitory neurotransmitter gamma-aminobutyric acid (GABA). Reduced GABA activity leads to increased excitability in the central nervous system, resulting in muscle rigidity and spasms characteristic of SPS. While SPS is rare, anti-GAD antibodies seen in SPS are also seen in the much more common autoimmune disease, type 1
