From marine predator to pharmacology: Conotoxin diversity, discovery, and therapeutic potential.

Cone snails ( Conus spp.) biosynthesize a highly specialized venom comprising a vast arsenal of neurotoxic peptides, termed conotoxins. These peptides exhibit exceptional structural and functional diversity, characterized by unique disulfide-bond architectures, highly variable amino acid sequences, and precise interactions with defined molecular targets. This review presents a comprehensive synthesis of current knowledge on conotoxin biology, beginning with evolutionary origin and proceeding through the classification systems used to define their gene superfamilies and pharmacological families

Artificial intelligence in central nervous system autoimmune diseases: from clinical decision support to mechanistic discovery and translation.

Central nervous system (CNS) autoimmune diseases, including multiple sclerosis (MS), neuromyelitis optica spectrum disorder (NMOSD), autoimmune encephalitis (AE), and myelin oligodendrocyte glycoprotein antibody-associated disease (MOGAD), remain difficult to diagnose, stratify, and treat because clinical phenotypes overlap, biomarkers are imperfect, and the disease course requires longitudinal observation. Artificial intelligence (AI) is increasingly being evaluated to integrate clinical, imaging, immunological, and molecular data for decision-making in neuroimmunology. Clinical applications

A Two-Stage Ensemble Machine Learning Pipeline for Breast Cancer Diagnosis from Digital Mammograms.

Breast cancer is the most common cancer among women, and early detection through mammography is essential for reducing mortality. Artificial intelligence can support radiologists by improving diagnostic accuracy. To develop and evaluate a two-stage ensemble machine learning pipeline for breast cancer diagnosis from digital mammograms. The proposed framework combines image preprocessing, multiple convolutional neural networks trained under different conditions, and a second-stage classifier that integrates the CNN outputs. Several machine learning models and feature selection techniques were ev

A comprehensive framework for glioma surgery by the PIONEER Consortium and RANO resect group, part 2: perioperative recommendations for neurological, language, functional, and quality-of-life assessment.

Historically, the assessment of surgical outcomes in patients with glioma has been focused on technical outcomes, such as volumetric analysis of the residual tumour, progression-free survival, and overall survival. Other outcomes, such as neurological deficits, can be challenging to assess in an objective, quantifiable, and comprehensive manner. As a result, no consensus is available on methods to systematically evaluate perioperative neurological, language, cognitive, and functional outcomes in patients with glioma. This variability contributes to suboptimal outcomes, hinders uniformity acros

Artificial intelligence standardizes CT-based body composition analysis in breast cacer to address methodological heterogeneity.

Computed tomography (CT)-derived body composition parameters-including skeletal muscle area, muscle density, and visceral and intermuscular adipose tissue-are robust prognostic and predictive biomarkers in breast cancer. However, their clinical translation is hampered by substantial methodological heterogeneity across studies. Key sources of variability include image acquisition parameters (contrast phase, tube voltage, slice thickness), inconsistent vertebral level selection (L3 versus thoracic levels), the absence of standardized normalization methods, and fragmented diagnostic thresholds fo

Quantitative Volumetric Analysis of the Brain Using Magnetic Resonance Imaging in Sickle Cell Anaemia.

BACKGROUND: Sickle cell disease (SCD) is a group of inherited hemoglobinopathies caused by a mutation in the β-globin gene, with sickle cell anaemia (SCA) representing the homozygous and most severe form. The disease burden is highest in sub-Saharan Africa, India, and the Mediterranean region. Neurological complications, including overt stroke and silent cerebral infarcts (SCI), contribute significantly to morbidity, with a markedly increased risk observed among affected individuals. The objective of the study is to assess and compare brain gray matter and white matter volumes in patients with

Clinical proteomics in inborn errors of metabolism: from biomarker discovery to implementation.

INTRODUCTION: Inborn errors of metabolism (IEMs) are rare, heterogeneous disorders traditionally diagnosed through genetic testing, enzyme assays, and metabolite measurements. However, these tools often do not fully explain phenotypic variability, organ involvement, disease progression, or treatment response. Clinical proteomics provides a complementary functional layer by capturing changes in protein abundance, proteoforms, post-translational modifications (PTM), and biological pathways, offering insights beyond genotype- and metabolite-based approaches. AREAS COVERED: This review examines t

A novel extracellular flux assay workflow uncovers impaired sciatic nerve mitochondrial respiration in diabetic db/db mice.

Abnormal mitochondrial function contributes to the development of diabetic neuropathy by inducing oxidative stress and altering energy metabolism, ultimately leading to neuronal damage. However, direct and reproducible assessment of mitochondrial bioenergetics in peripheral nerves remains technically challenging. Here, we describe an optimized ex vivo approach for measuring mitochondrial respiration in isolated murine sciatic nerve segments using extracellular flux analysis. Although extracellular flux-based assays have previously been applied to nerve tissue, we introduce key methodological r

Quantum-driven frequency stability in Indian prospect smart grid with electric vehicle charging station integration and real-time hardware validation.

In India, the growth of the Electric Vehicle(EV) is gradually increasing due to the high price of fossil fuel and global warming. But in India, the Electric Vehicle Charging Station(EVCS) infrastructure is inadequate due to the limited number of charging points, insufficient grid support for integration with RES. The integration of Electric Vehicle Charging Station(EVCS) along with RES in the Distributed Generation(DG) system confronts frequency fluctuations, variations in tie-line power, and instability. Thus, the employment of a Load Frequency Controller (LFC) with the DG diminishes the issu

Unraveling the catalytic mechanisms of defective carbons for selective 2e-/4e- oxygen reduction: from intrinsic defects to synergies with heteroatoms and single-metal atoms.

Selective oxygen reduction via either the two-electron pathway for distributed H2O2 electrosynthesis or the four-electron route for high-efficiency energy conversion represents a cornerstone of sustainable electrocatalysis, where defect engineering offers unprecedented opportunities for atomic-level site manipulation. This review comprehensively summarizes defect-centric functional active sites, encompassing intrinsic carbon defects, synergistic structures between heteroatom dopants and defects, and synergistic motifs combining metal single-atom sites with surrounding defect configurations. Diagnosing Anaerobic Digesters' Function and Performance: From Meta-Omics to Integrated Meta-Omics Analyses.

Anaerobic digestion (AD) of organic wastes by microorganisms into biomethane contributes significantly towards bioenergy generation. However, the bioprocess remains challenging due to its complex microbial ecology, dynamic biochemical pathways and sensitivity to perturbations. Over the past decade, using meta-omics like metataxonomics, metagenomics, metatranscriptomics, metaproteomics and metabolomics to diagnose its functioning has become necessary and common. However, a single meta-omics method can only provide limited biological understanding of the bioprocess. This led to the use of multi-

Fast 3D-HEVC Depth Map Coding Method Based on Spatio-Temporal Correlation and a Two-Stage Mode Decision Framework.

Efficient intra-mode decision for depth maps assumes a pivotal role in augmenting the overall performance of 3D-HEVC. Existing research endeavors predominantly rely on fast mode screening strategies grounded in texture characteristics or machine learning techniques. These strategies, to a certain extent, mitigate the complexity of mode search. Nevertheless, these approaches often fall short of fully leveraging the intrinsic spatio-temporal correlations within depth maps. Moreover, strategies relying on deterministic classifiers exhibit insufficient discrimination reliability in regions featuri

Eye-Tracking Based Measurement Framework For Quantifying Visual Attention In Screen-Based Interactive Digital Art Environments.

This article presents a gaze-enabled measurement framework designed to quantify visual attention during interactive digital art experiences and to link attention dynamics to experiential outcomes. Unlike traditional static art viewing, interactive digital art involves dynamic media, multi-source information, and continuous feedback loops, which challenge standard eye-tracking methodologies. The proposed system integrates eye-tracking acquisition, interaction logging, timestamp synchronization, dynamic Area of Interest (AOI) mapping, and metric computation into a unified pipeline. Visualization of text on bowed sheets via High-resolution 3D-Magnetic Resonance Micro-imaging for potential reading of closed books: the proof-of-concept.

Non-invasive Magnetic Resonance Imaging on clinical MR-scanners at spatial resolution ≂1mm3 could in principle be used also for insights in the structure of valuable ancient objects like books if the spatial resolution could be improved. We demonstrate, that Magnetic-Resonance-Microscopy is even able to visualize printed letters at thickness < 30 µm on superposed paper-sheets. The physical-technical methodology is based on prototype hardware installed as insert on a human MR-scanner relying on strong magnetic and gradient-fields with sensitive radiofrequency-sensors. Three-way multi-criteria decision-making with linguistic information using prospect-regret theory and generalised TODIM for heart failure data analysis.

Cardiovascular disease is a leading global cause of mortality, and timely monitoring can help assess its severity. Electronic medical records contain information on patients’ symptoms, physical characteristics, and clinical laboratory test results. These data can be used in biostatistical analysis to identify patterns and relationships that may not be apparent to medical practitioners. However, when experts confront risk in complex settings, their bounded rationality may influence their decision-making behaviour. Moreover, the irrational behaviour of a single expert can substantially affect de

Synthetic Priors for Real-World Detection: A Label-Free Framework for Identifying Ultra-Rare Objects.

Detecting ultra-rare objects (frequency < 10-6) without positive annotations remains a critical challenge in computer vision. We propose a Synthesis-Prior Driven paradigm that eliminates the need for real positive samples. Our core insight is that models trained on statistically and perceptually indistinguishable synthetic data can generalize effectively to real-world environments. To achieve this, we introduce a Differentiable Morphological Modeling framework, which embeds explicit physical and geometric priors into a differentiable synthesis engine. Unlike deep learning generative models, th
