Time-dependent elastic extensional RBC deformation by micropipette aspiration: redistribution of the spectrin network?

The time dependence of small elastic extensional RBC deformation by micropipette aspiration has been analyzed. This process shows two-phases which are characterized by time constants of the order of some tenths of seconds and about ten seconds, respectively. The equilibrium tongue length is reached after about 30 s. For the first, fast step we assume that the membrane model of immobilized boundaries holds, i.e., the skeleton is tightly associated with the lipid bilayer and no redistribution of the skeleton with respect to the lipid bilayer is allowed. This lipid-spectrin interaction or anchora

Cyanine dye labeling reagents--carboxymethylindocyanine succinimidyl esters.

Ten carboxymethylindocyanine dyes which form the basis of a new series of fluorescent probes have been synthesized and converted into succinimidyl active esters for fluorescent labeling of proteins or other amino-containing substances. Fluorescence emission maxima for members of the series range from 575 to 780 nm. Hydrophilic, water-soluble reagents have been obtained which yield labeled antibodies with little tendency to form precipitates. The fluorescence intensities achieved are higher than those produced by labeling with the cyanine isothiocyanates described previously (Mujumdar et al.: C

Opioid peptides modulate the organization of vimentin filaments, phagocytic activity, and expression of surface molecules in monocytes.

It is theorized that intermediate filaments are important in the modulation of membrane activity and cell motility; however, their functions are unknown. The assembly and organization of these filaments are under hormonal regulation. We investigated in human monocytes the in vitro effects of Met-enkephalin, Leu-enkephalin, and beta-endorphin on the expression of immunoreactive cytoskeletal vimentin filaments. We simultaneously examined their effect on the phagocytosis of Candida albicans and on the membrane display of surface molecules. The three opioid peptides markedly reduced the expression

The ultrastructural organization of the basement membrane of Bowman's capsule in the rat renal corpuscle.

The basement membrane of Bowman's capsule (BCBM) of the rat was studied by means of a modified tissue-preservation technique for transmission electron microscopy, which avoids the usual thorough fixation in OsO4 and applies tannic acid and uranyl acetate for staining (Sakai et al. 1986). At most sites the BCBM is multilayered, consisting of one to seven dense layers separated by electron-lucent layers. The latter, which can be termed laminae rarae, contain fine filaments which connect the dense layers to each other and the innermost dense layer to the basal cell membrane of the parietal epithe

Repair of the wounded guinea pig tympanic membrane: organization of filamentous actin and spatial cellular reorganization.

Since the role of the epithelial cell in the repair of the wounded tympanic membrane is not well understood, the epithelial cell layers were examined using rhodamine phalloidin to localize F-actin in situ following a full-thickness traumatic perforation. The change in shape of the epithelial cells and the morphological changes in F-actin were characterized. The mucosal cells were remarkably resistant to wounding and their morphology and F-actin distribution remained unchanged. Within 24 hours following perforation, basal cells were prominently stained adjacent to the perforation, whereas the r

A flow EPR study of deformation and orientation characteristics of erythrocyte ghosts: a possible effect of an altered state of cytoskeletal network.

Using the flow EPR technique, we investigated the resealed ghost deformability in shear flow and the effects of the altered state of cytoskeletal network induced by hypotonic incubation of ghosts. Isotonically resealed ghosts in the presence of Mg-ATP, in which alteration of cytoskeletal network is not effected, have smooth biconcave discoid shapes, and show a flow orientation and deformation behavior similar to that of erythrocytes, except that higher viscosities are required to induce the same degrees of deformation and orientation as in erythrocytes. The flow behavior of resealed ghosts is

Cytoskeletal immunohistochemistry of Alzheimer's disease.

The first half of this paper is devoted to a review on the cytoskeletal immunocytochemistry of the various morphological changes in Alzheimer's disease (AD), i.e. neurofibrillary tangles (NFT), senile plaques (SP), granulovacuolar degeneration (GV) and Hirano bodies (HB). In the second half it is demonstrated that sera raised against the paired helical filaments (PHF) in NFT, and monoclonal antibodies to phosphorylated epitopes on neurofilament proteins, also stain structures within neuronal perikarya without PHFs in some diseases. These include Pick bodies and swollen cells in Pick's disease,

Ultrastructural observation on the thalamic neuronal inclusions in young mice.

Neuronal intracytoplasmic inclusions were found in the ventrolateral and posterior nuclei of the thalamus of 3-month-old thiamine-deficient mice and quaking mice. An electron-microscopic study revealed that these inclusions were composed of condensation of parallel rectilinear 10-nm filaments with interconnecting cross-bridges as a hallmark of neurofilaments. They were morphologically indistinguishable from those described in aged mice. This observation indicates that neuronal inclusions in the thalamus, which have been constantly reported in aging mice, can be seen even in young mice in the p

Distortions of the nodes of Ranvier from axonal distension by filamentous masses in hexacarbon intoxication.

A study has been made of the structural changes of nodal and paranodal regions of the nodes of Ranvier of peripheral nerves of rats in which marked accumulations of neurofilaments have occurred within axons under the influence of 2,5-hexanediol over 10 weeks. The neurofilamentous masses caused distension of the axon at two points of apparent weakness as they attempted to slide through the axonal constriction at the nodes. Principally, a spiral axonal protrusion pushed into the zone of unattached myelin loops in the proximal paranodal spinous bracelet of Nageotte. This led to a conical widening

The pathogenesis of Dupuytren's contracture: contractile mechanisms of the myofibroblasts.

The role of myofibroblasts in the pathogenesis of Dupuytren's contracture was investigated by light and electron microscopic histochemical methods. Dupuytren's myofibroblasts contain an intracellular contractile mechanism that is driven by the dephosphorylation of adenosine triphosphate. Our study of calcium adenosinetriphosphatase (ATPase) activities verifies that the site of this energy system is on the myofilaments of the myofibroblasts. The degree of ATPase activity, as determined by cell counts, appeared to correlate with the residual contracture as predicted by the Legge and McFarlane Ou

Microfilament-associated adhering junctions (6 nm F-maculae adherentes) connect bovine pulmonary fibroblasts in vivo.

Fibroblasts in the pulmonary alveolar septa of neonatal and mature cattle form spot-shaped intercellular junctions with each other where 6 nm microfilaments adhere to the plasma membranes. The junctions have variable cleft widths (10-20 nm) and a diffuse periodic intracleft substance. Impinging 6 nm microfilaments and plaque specializations were present in thin sections of junctions ranging from 0.4 to 0.6 micron in length. The microfilaments involved in junctions are parts of either cortical cytoplasmic webs or highly organized bundles. Two or more fibroblasts may form one complex of junction

Clinical and electron microscopic observations of the conjunctiva of adult patients with Bitot's spots.

Three adult patients with Bitot's spots of the conjunctiva were observed in the American Southwest; all were in good health without obvious cause for vitamin A deficiency. Serum vitamin A levels were low normal in two; the third patient was on replacement vitamin A therapy with resultant high serum levels. Serum beta-carotene levels were low in two and normal in one. Schirmer test without anesthesia was low normal in two and with topical anesthesia was abnormal in all. After 8 weeks of oral vitamin A therapy, the Schirmer test and Bitot's spots showed little response. Electron microscopy of Bi

Bilateral juvenile granulosa cell tumors in a 4-month-old dysmorphic infant. A clinical, histologic, and ultrastructural study.

Juvenile granulosa cell tumors were encountered within bilateral cystic ovarian masses in a 4-month-old infant. The child was the product of a consanguinous pregnancy, and manifested poor growth, relative microcephaly, facial asymmetry, and a malformed left ear. There was no history of gestational drug or hormone ingestion, and no evidence of abnormal endocrine activity after birth, however, the serum alpha-fetoprotein level was mildly elevated. The tumors were well defined by sonography and there was no evidence of metastasis. Histologically, a dense proliferation of round to oval tumor cells

Morphological alterations and induction of oxidative stress in glial cells caused by the branched-chain alpha-keto acids accumulating in maple syrup urine disease.

Maple syrup urine disease (MSUD) is an inherited neurometabolic disorder biochemically characterized by the accumulation of the branched-chain alpha-keto acids (BCKA) alpha-ketoisocaproic (KIC), alpha-keto-beta-methylvaleric (KMV) and alpha-ketoisovaleric (KIV) and their respective branched-chain alpha-amino acids in body fluids and tissues. Affected MSUD patients have predominantly neurological features, including cerebral edema and atrophy whose pathophysiology is not well established. In the present study we investigated the effects of KIC, KMV and KIV on cell morphology, cytoskeleton reorg…

[The role of lipids in the pathogenesis of lateral amyotrophic sclerosis].

Amyotrophic lateral sclerosis (ALS) is an incurable neurodegenerative disease characterized by selective degeneration of motor neurons of the motor cortex, brain stem and brain stem. Mutations in genes coding for SOD1, C9ORF72, TDP-43, FUS and others are associated with ALS and result in abnormal processing and transport of RNA as well as changes in the dynamics of cytoskeleton. In addition, a sharp change in the metabolism of various lipid classes, including phospholipids, fatty acids, sphingolipids, etc., was detected. This review describes changes in lipid content and activity of enzymes in…

Pushing myelination - developmental regulation of myosin expression drives oligodendrocyte morphological differentiation.

Oligodendrocytes are the central nervous system myelin-forming cells providing axonal electrical insulation and higher-order neuronal circuitry. The mechanical forces driving the differentiation of oligodendrocyte precursor cells into myelinating oligodendrocytes are largely unknown, but likely require the spatiotemporal regulation of the architecture and dynamics of the actin and actomyosin cytoskeletons. In this study, we analyzed the expression pattern of myosin motors during oligodendrocyte development. We report that oligodendrocyte differentiation is regulated by the synchronized express…

Wiskott-Aldrich Syndrome (WAS) and Dedicator of Cytokinesis 8- (DOCK8) Deficiency.

Both Wiskott-Aldrich syndrome (WAS) and dedicator of cytokinesis 8 (DOCK8) deficiency are primary immunodeficiency diseases caused by mutations in genes that result in defective organization of the cytoskeleton in hematopoietic tissues. They share some overlapping features such as a combined immunodeficiency, eczema and a predisposition to autoimmunity and malignancy, but also have some unique features that make them relatively easy to diagnose by clinical means. Both diseases can be cured by HSCT in a large proportion of patients. In WAS it is sometimes difficult to establish an indication fo…

[Outer dense fiber 2 and sperm function: Progress in studies].

Oligoasthenozoospermia, teratozoospermia or low sperm motility is the main cause of male infertility. Low sperm motility can be induced by abnormalities of the sperm tail structure and sperm function. The outer dense fiber protein 2 (ODF2) is a protein fiber maintaining cytoskeleton, as a major component of the mammalian sperm tail and centrosome, and its abnormality is closely related to asthenospermia. Recent studies indicate that ODF2 includes many proteins of the same name and homologous splices located in the sperm centrosomes and spindles of cleaved-embryos, necessary for animal ciliogen…
