Antimicrobial resistance (AMR) is recognised as a significant global health threat, yet research, surveillance and intervention strategies remain largely focused on clinical and veterinary settings, with limited attention to environmental reservoirs and transmission pathways. Growing global evidence, including from Aotearoa New Zealand (NZ), points to a clear link between dominant sources of AMR entering the environment (such as wastewater) and its dissemination in receiving environments. Once in these environments, a wide range of microorganisms, both environmental and introduced, interact in

Climate change and biological invasions pose synergistic threats; however, organisms may rapidly adapt through microbial symbiosis. We investigated how defensive symbionts in invasive pea aphids, Acyrthosiphon pisum, respond to emerging threats. Previously rare strains of the protective symbiont Hamiltonella defensa increased from <0.5% to 58% in aphid populations over just a few years. Bioassays revealed that these strains confer reciprocal, enemy-specific defences. One strain (C11) protected against Praon pequodorum, a native parasitoid that only began attacking pea aphids post-introduction,

The enrichment of antibiotic resistance genes (ARGs) during composting poses a substantial risk to the safe utilization of chicken manure. However, the role of multidrug-resistant plasmid (MRP)-harboring bacteria in ARG dissemination and microbial succession during composting remains poorly understood. In this study, we investigated the regulatory effects of thermophilic Parageobacillus toebii G12 (G12) on ARG dynamics, as well as its performance under interference from MRP-harboring bacteria. Our results showed that G12 inoculation elevated compost temperature, extended the thermophilic phase

Carbapenemase-producing Escherichia coli (E. coli) has emerged as a critical contributor to antimicrobial resistance (AMR), significantly compromising the efficacy of last-resort carbapenem antibiotics. Carbapenemase-producing E. coli has significantly reduced the effectiveness of carbapenems, which were previously considered last-resort antibiotics for treating severe infections caused by extended-spectrum β-lactamase (ESBL)-producing organisms. Numerous β-lactam antibiotics, including carbapenems, are hydrolyzed by these enzymes, which results in fewer therapy choices, greater rates of treat

A diverse microbial community exists within the human oral cavity that plays an essential role in maintaining health or inducing diseases such as dental caries, periodontal disease, and halitosis. Probiotics, live microorganisms that provide health benefits when consumed in adequate amounts, have been found to be promising as a means of modulating the oral microbiome and combating these diseases. This review incorporates present knowledge about the mechanism of probiotic action, including competitive exclusion of pathogens, antimicrobial metabolite production, biofilm disruption, and immune mo

Natural hallucinogenic compounds have arisen independently across plants, fungi, and animals, evolving into a diverse chemical arsenal that includes phenethylamines, indolealkylamines, and terpenoid scaffolds. Beyond clinical and cultural frameworks, their ecological origins and evolutionary trajectories may help explain why such potent modulators of perception, emotion, and cognition persist in nature. Here, integrating chemical ecology, comparative genomics, biosynthetic logic, and evolutionary biology, we propose that these molecules may function as defensive agents or symbiosis-associated

The global spread of antimicrobial resistance (AMR) poses a major challenge to food safety and public health. To evaluate foodborne transmission as a critical pathway, we analyzed 713,343 Salmonella enterica genomes from the NCBI Pathogen Detection database across 164 countries. A total of 363,409 ARGs conferring resistance to aminoglycosides, tetracyclines, and β-lactams were identified. Source-specific resistome signatures were evident, with primary sources (human feces, animal waste) exhibiting the highest ARG abundance and diversity, followed by secondary sources (food products) and tertia

This narrative review explores triazole resistance and pathogenesis of Aspergillus fumigatus (A. fumigatus) under a unified One Health framework, integrating evidence across host immune spectrums, agricultural environmental selection pressures, veterinary aspergillosis and clinical antifungal management. We illustrate layered host-fungus immune interactions underlying distinct clinical aspergillosis phenotypes, and delineate core resistance mechanisms including cyp51-related target alterations, enhanced efflux pump activity, biofilm-mediated tolerance and adaptive stress response rewiring. Urinary tract infections (UTIs) caused by Klebsiella pneumoniae are an increasing challenge in pediatric practice due to the combined effects of biofilm formation, multidrug resistance (MDR), and limited therapeutic options for children. Biofilm development promotes bacterial persistence by impairing antibiotic penetration, enabling metabolic adaptation, promoting persister-cell formation, facilitating horizontal gene transfer (HGT), and inducing stress-induced mutagenesis, thereby reducing the effectiveness of conventional antimicrobial therapy. These mechanisms are further compounded by pedi

Antimicrobial resistance is a critical global public health challenge, with drug-resistant infections contributing to more than one million deaths annually. The widespread dissemination of multidrug-resistant bacteria poses a severe threat to the management of infectious diseases. Bacterial evolution via genetic mutation and horizontal gene transfer diminishes antimicrobial efficacy, often leading to therapeutic failure, increased morbidity and mortality. However, the development of novel antibiotics lags far behind the rapid evolution of drug-resistant bacteria. Therefore, scientists worldwid

Multidrug resistance (MDR) in bacterial and fungal pathogens poses a growing global health crisis, rendering many conventional antimicrobial therapies ineffective. The rise of MDR strains complicates treatment, prolongs illness, increases healthcare costs, and contributes to higher mortality rates. Mechanisms driving MDR include enzymatic drug inactivation, target modification, efflux pump activity, decreased permeability, and biofilm formation-often fueled by horizontal gene transfer and selective pressure from antimicrobial overuse. In response to the urgent need for novel therapeutic strate

Antimicrobial multidrug resistance is the ability of microorganisms to withstand the effects of several antimicrobial agents, presenting a major challenge to modern healthcare systems worldwide. Although considerable research has been conducted, the molecular and evolutionary mechanisms underlying resistance are still not completely understood. This review brings together current knowledge to explain how resistance originates, spreads, and persists in different pathogens. Microorganisms may show primary resistance, which arises naturally without prior exposure to drugs, or acquired resistance,

Streptococcus anginosus is a common isolate of the oral cavity and an opportunistic pathogen for systemic infections. Although the pyogenic infections caused by S. anginosus are similar to those caused by Streptococcus pyogenes, S. anginosus lacks most of the well-characterized virulence factors of S. pyogenes. To investigate the pathogenicity of S. anginosus, we analysed the genome of a newly identified S. anginosus strain, KH1, which was associated with toxic shock-like syndrome in an immunocompetent adolescent. The genome of KH1 contains nine genomic islands, two Clustered Regularly Intersp

Few strains of Dehalococcoides mccartyi harbour and express the vinyl chloride reductase (VcrA) that catalyzes the dechlorination of vinyl chloride (VC), a carcinogenic soil and groundwater contaminant. The vcrA operon is found on a Genomic Island (GI) and, therefore, believed to participate in horizontal gene transfer (HGT). To try to induce HGT of the vcrA-GI, we blended two enrichment cultures in medium without ammonium while providing VC. We hypothesized that these conditions would select for a mutant strain of D. mccartyi that could both fix nitrogen and respire VC. However, after more th

The Rcs phosphorelay is a bacterial stress response system that responds to envelope stresses and in turn controls several virulence-associated pathways, including capsule, flagella, and toxin biosynthesis, of numerous bacterial species. The Rcs system also affects antibiotic tolerance, biofilm formation, and horizontal gene transfer. The Rcs system of the ocular bacterial pathogen Serratia marcescens was recently demonstrated to influence ocular pathogenesis in a rabbit model of keratitis, with Rcs-defective mutants causing greater pathology and Rcs-activated strains demonstrating reduced inf

Porphyromonas gingivalis is a keystone pathogen of chronic periodontitis. The virulence of P. gingivalis is reported to be strain related and there are currently a number of strain typing schemes based on variation in capsular polysaccharide, the major and minor fimbriae and adhesin domains of Lys-gingipain (Kgp), amongst other surface proteins. P. gingivalis can exchange chromosomal DNA between strains by natural competence and conjugation. The aim of this study was to determine the genetic variability of P. gingivalis strains sourced from international locations over a 25-year period and to

Mesenchymal stem cells (MSC) are multipotent cells able to differentiate into several cell types, hence providing cell reservoirs for therapeutic applications. The absence of detectable MSC homing at injury sites suggests that paracrine functions could, at least in part, be mediated by extracellular vesicles (EVs); EVs are newly identified players that are studied mainly as predictive or diagnostic biomarkers. Together with their clinical interests, EVs have recently come to the fore for their role in cell-to-cell communication. In this context, we investigated gene-based communication mechani

Natural genetic transformation of Streptococcus pneumoniae, an important human pathogen, mediates horizontal gene transfer for the development of drug resistance, modulation of carriage and virulence traits, and evasion of host immunity. Transformation frequency differs greatly among pneumococcal clinical isolates, but the molecular basis and biological importance of this interstrain variability remain unclear. In this study, we characterized the transformation frequency and other associated phenotypes of 208 S. pneumoniae clinical isolates representing at least 30 serotypes. While the vast ma

Aminoacyl-tRNA synthetases are pivotal in determining how the genetic code is translated in amino acids and in providing the substrate for protein synthesis. As such, they fulfill a key role in a process universally conserved in all cellular organisms from their most complex to their most reduced parasitic forms. In contrast, even complex viruses were not found to encode much translation machinery, with the exception of isolated components such as tRNAs. In this context, the discovery of four aminoacyl-tRNA synthetases encoded in the genome of mimivirus together with a full set of translation
