Sweet Corn Sentinel Monitoring for Lepidopteran Field-Evolved Resistance to Bt Toxins.

As part of an insect resistance management plan to preserve Bt transgenic technology, annual monitoring of target pests is mandated to detect susceptibility changes to Bt toxins. Currently Helicoverpa zea (Boddie) monitoring involves investigating unexpected injury in Bt crop fields and collecting larvae from non-Bt host plants for laboratory diet bioassays to determine mortality responses to diagnostic concentrations of Bt toxins. To date, this monitoring approach has not detected any significant change from the known range of baseline susceptibility to Bt toxins, yet practical field-evolved

Phenotypic plasticity, canalisation and developmental stability of Triatoma infestans wings: effects of a sublethal application of a pyrethroid insecticide.

BACKGROUND: Triatomine control campaigns have traditionally consisted of spraying the inside of houses with pyrethroid insecticides. However, exposure to sublethal insecticide doses after the initial application is a common occurrence and may have phenotypic consequences for survivors. Here, using Triatoma infestans (the main vector of Chagas disease in the Southern Cone of South America) as a model species, we quantified the effects of exposure to a sublethal dose of pyrethroid insecticide on wing morphology. We tested if the treatment (i) induced a plastic effect (change in the character mea

Dual immunological and oxidative responses in Oreochromis niloticus fish exposed to lambda cyhalothrin and concurrently fed with Thyme powder (Thymus vulgaris L.): Stress and immune encoding gene expression.

The present study was performed to explore the immunotoxicological effects of the lambda cyhalothrin (LCH) insecticide and evaluate the efficiency of Thyme powder (TP) as a fish supplement in attenuation of LCH impact on Oreochromis niloticus (O. niloticus) fish. Fish was sampled following 30-days exposure to LCH (1/6 LC50: 0.48 μg/L) and TP (2%) supplementation, individually or in combination. The growth performance, immune status, biochemical indices, and mRNA expression pattern changes of stress and immune-encoding genes in the liver and spleen tissues, respectively, through real-time polym

Genetic analysis of the orthologous crt and mdr1 genes in Plasmodium malariae from Thailand and Myanmar.

BACKGROUND: Plasmodium malariae is a widely spread but neglected human malaria parasite, which causes chronic infections. Studies on genetic polymorphisms of anti-malarial drug target genes in P. malariae are limited. Previous reports have shown polymorphisms in the P. malariae dihydrofolate reductase gene associated with pyrimethamine resistance and linked to pyrimethamine drug pressure. This study investigated polymorphisms of the P. malariae homologous genes, chloroquine resistant transporter and multidrug resistant 1, associated with chloroquine and mefloquine resistance in Plasmodium falc

Spinosad resistance of melon thrips, Thrips palmi, is conferred by G275E mutation in α6 subunit of nicotinic acetylcholine receptor and cytochrome P450 detoxification.

To examine the resistance mechanisms of Thrips palmi against spinosad, we cloned partial nucleotide sequences of the nicotinic acetylcholine receptor α6 subunit (TPα6) gene from susceptible (OK) and resistant (TS1 and TS5) strains and compared the deduced amino acid sequences among the three strains. The OK, TS1, and TS5 strains respectively showed LC50 values of 3.4mg/L, 2838.5mg/L, and 6655.5mg/L. The deduced amino acid sequence of TPα6 gene showed 96% identity with that of Frankliniella occidentalis. Comparison of the deduced amino acid sequences of TPα6 gene among the three strains showed

Effect of fluralaner administration to domestic dogs on the bloodmeal profile of Triatoma infestans in a cluster-randomized trial in the Argentine Chaco.

BACKGROUND: Residual spraying with pyrethroid insecticides remains the main tool in the control of vector-borne transmission of Chagas disease. However, its efficacy has been compromised in regions with pyrethroid-resistant populations of triatomines, as in the Gran Chaco region, where Triatoma infestans has developed high levels of resistance. To cope with it, xenointoxication has emerged as a promising strategy. As part of a cluster-randomized trial aimed at evaluating the effects of treating dogs with fluralaner on (peri)domestic infestation and T. cruzi infection, we evaluated the effect o

Sequencing of five poultry strains elucidates phylogenetic relationships and divergence in virulence genes in Morganella morganii.

BACKGROUND: M. morganii is a bacterium frequently associated with urinary infections in humans. While many human strains are sequenced, only the genomes of few poultry strains are available. Here, we performed a detailed characterization of five highly resistant Morganella morganii strains isolated in association with Escherichia coli from diseased domestic Austrian poultry flocks, namely geese, turkeys and chicken layers. Additionally, we sequenced the genomes of these strains by NGS and analyzed phylogenetic clustering, resistance and virulence genes in the context of host-specificity. RESU

Transcriptome analysis of Barbarea vulgaris infested with diamondback moth (Plutella xylostella) larvae.

BACKGROUND: The diamondback moth (DBM, Plutella xylostella) is a crucifer-specific pest that causes significant crop losses worldwide. Barbarea vulgaris (Brassicaceae) can resist DBM and other herbivorous insects by producing feeding-deterrent triterpenoid saponins. Plant breeders have long aimed to transfer this insect resistance to other crops. However, a lack of knowledge on the biosynthetic pathways and regulatory networks of these insecticidal saponins has hindered their practical application. A pyrosequencing-based transcriptome analysis of B. vulgaris during DBM larval feeding was perfo

Community knowledge, perceptions, and practices regarding malaria and its control in Jabi Tehnan district, Amhara Region, Northwest Ethiopia.

BACKGROUND: Use of long-lasting insecticidal nets (LLINs), indoor residual spraying (IRS), community-based malaria education, prompt diagnosis and treatment are key programme components of malaria prevention and control in Ethiopia. However, the effectiveness of these interventions is often undermined by various challenges, including insecticide and drug resistance, the plasticity of malaria vectors feeding and biting behaviour, and certain household factors that lead to misuse and poor utilization of LLINs. The primary objective of this study was to document households' perceptions towards ma

Complex intracellular messenger pathways regulate one type of neuronal alpha-bungarotoxin-resistant nicotinic acetylcholine receptors expressed in insect neurosecretory cells (dorsal unpaired median neurons).

Although molecular biology provides new insights into the subunit compositions and the stoichiometries of insect neuronal nicotinic acetylcholine receptors (nAChRs), our knowledge about the phosphorylation/dephosphorylation mechanisms of native neuronal nAChRs is limited. The regulation of alpha-bungarotoxin-resistant nAChRs was studied on dissociated adult dorsal unpaired median neurons isolated from the terminal abdominal ganglion of the cockroach Periplaneta americana, using whole-cell, patch-clamp technique. Under 0.5 microM alpha-bungarotoxin treatment, pressure ejection application of ni

Combining sterile insect releases with refuge areas to delay the evolution of resistance to Bt sugarcane: an agent-based modeling approach.

The strategic use of refuge areas is a well-known method for delaying the development of pest resistance to Bacillus thuringiensis (Bt) crop. A lesser-known method to control against resistance development is sterile insect releases. In this article, an agent-based simulation model is used to test the effectiveness of combining the use of Bt sugarcane, refuge areas, and sterile insect releases as an integrated strategy against Eldana saccharina Walker (Lepidoptera: Pyralidae) infestation and resistance development. Individual insects are modeled with their own genetic traits on a simulated sug

Studies in vitro on the relative efficacy of current acaricides for Sarcoptes scabiei var. hominis.

Resistance of Sarcoptes scabiei to various topical therapies has been described, but clinical assessment of treatment failure is problematic and in-vitro assays are generally not available. We describe a simple in-vitro analysis used to evaluate the relative efficacy of a range of topical, oral, and herbal treatments available in Australia for the treatment of scabies. S. scabiei var. hominis mites were collected from skin scrapings obtained from 7 crusted scabies patients over a period of 2 years (1997 and 1998). Larvae, nymphal instars, and adult mites were tested within 3 h of collection an

[Effect of pesticides on Bacillus thuringiensis strains under controlled conditions].

Little is known about native populations of Bacillus thuringiensis (Bt) isolated from soils of Argentina. We undertook this study to determine the resistance to different pesticides of two commercial and fourteen native strains of Bt under in vitro conditions. An agar plate bioassay test conducted with ten pesticides and sixteen strains of Bt showed that Basagran, Scepter, Fungoxan and Decis were not toxic for the bioinsecticide bacteria at recommended application rates (RAR). In contrast, low concentrations (3.2% RAR) of Agil, Select and Isomero showed a deleterious effect on the bacteria inv

Blood host preferences and competitive inter-species dynamics within an African malaria vector species complex inferred from signs of animal activity around aquatic larval habitats.

BACKGROUND: The feeding behaviours of the malaria vector Anopheles arabiensis, and its competitive relationships with other sibling species … vestock, may confound efforts to eliminate this key malaria vector. However, they might also enable insecticide resistance management strategies that could restore the effectiveness of pyrethroids in particular. This new approach to indirectly identifying commonly utilized blood sources may also be applicable to an unprecedented diversity of zoophagic mosquitoes, enabli …

Community benefits of mass distribution of three types of dual-active-ingredient long-lasting insecticidal nets against malaria prevalence in Tanzania: evidence from a 3-year cluster-randomized controlled trial.

BACKGROUND: Long-lasting insecticidal nets (LLINs) were once fully effective for the prevention of malaria; however, mosquitoes have developed resistance to pyrethroids, the main class of insecticides used on nets. Dual active ingredient LLINs (dual-AI LLINs) have been rolled out as an alternative to pyrethroid (PY)-only LLINs to counteract this. Understanding the minimum community usage at which these LLINs elicit an effect that also benefits non-users against malaria infection is important. METHODS: We conducted a secondary analysis of a 3-year randomized controlled trial (RCT) in 84 cluste

Malaria epidemiology in the Korhogo area, Northern Côte d'Ivoire: baseline assessment prior to a randomized controlled trial.

BACKGROUND: Widespread Insecticide resistance among malaria vector populations poses a significant threat to the efficacy of current vector control strategies. This highlights the urgent need for innovative approaches that integrate existing tools with those under development. The primary objective of this study was to characterize the malaria epidemiological profile in the Korhogo region prior to the implementation of complementary vector control tools alongside long-lasting insecticidal nets (LLINs). Both passive and active case detection methods were employed. METHODS: Passive case detecti…
