Development of machine learning-based models to predict congenital heart disease: A matched case-control study.

BACKGROUND: The current congenital heart disease (CHD) prediction tools lack adequate interpretability and convenience, hindering the development of personalized CHD management strategies. We developed a machine learning-based risk stratification model for CHD prediction. METHODS: This study utilized data from 1,759 participants in a case-control study of CHD conducted across six birth defects surveillance hospitals located in Xi'an, Shaanxi Province, Northwest China, spanning from January 2014 to December 2016. The data was partitioned into training and testing datasets with a ratio of 7:3.

Artificial Intelligence in Predicting Postoperative Nausea and Vomiting Among Surgical Patients Following General Anesthesia: A Systematic Review.

Artificial intelligence (AI) may improve perioperative risk prediction by integrating complex clinical and physiological data, but its performance and readiness for clinical use remain uncertain. We evaluated AI models predicting postoperative nausea and vomiting (PONV), perioperative hypotension, prolonged intensive care unit (ICU) stay, in-hospital mortality, and neurological complications after general anesthesia. We searched bibliographic databases from inception to June 2026 for studies developing or validating AI-based prediction models in surgical patients undergoing general anesthesia.

DBI Mediates the Progression of Ankylosing Spondylitis by Regulating CD56dim NK Cells Cytotoxicity Function.

BACKGROUND: Ankylosing spondylitis (AS) is an autoimmune disease characterized by low back stiffness and pain, with no cure and a dearth of therapeutic targets. METHODS: Identifying novel AS targets from a list of 5884 druggable genes using weighted gene coexpression network analysis (WGCNA), machine learning, and Mendelian randomization analysis. Investigating the biological functions of the targets through comprehensive bio-functional analyses; exploring immune-related functions of the targets based on single-cell analyses; developing a reliable AS risk prediction model based on the identif

Distinguishing benign from malignant pelvic mass utilizing an algorithm with HE4, menopausal status, and ultrasound findings.

OBJECTIVE: The purpose of this study was to develop a risk prediction score for distinguishing benign ovarian mass from malignant tumors using CA-125, human epididymis protein 4 (HE4), ultrasound findings, and menopausal status. The risk prediction score was compared to the risk of malignancy index and risk of ovarian malignancy algorithm (ROMA). METHODS: This was a prospective, multicenter (n=6) study with patients from six Asian countries. Patients had a pelvic mass upon imaging and were scheduled to undergo surgery. Serum CA-125 and HE4 were measured on preoperative samples, and ultrasound

Evaluation and optimization of the syndromic management of female genital tract infections in Nairobi, Kenya.

BACKGROUND: Genital tract infections pose a public health concern. In many low-middle-income countries, symptom-based algorithms guide treatment decisions. Advantages notwithstanding, this strategy has important limitations. We aimed to determine the infections causing lower genital tract symptoms in women, evaluated the Kenyan syndromic treatment algorithm for vaginal discharge, and proposed an improved algorithm. METHODS: This cross-sectional study included symptomatic non-pregnant adult women presenting with lower genital tract symptoms at seven outpatient health facilities in Nairobi. Cli

A Comparative Study of Deep Learning and Classical Modeling Approaches for Protein-Ligand Binding Pose and Affinity Prediction in Coronavirus Main Proteases.

The accurate prediction of protein-ligand binding poses and affinities is central to structure-based drug design. In this study, we first benchmarked three distinct pose generation strategies for data sets from the ASAP Antiviral Challenge 2025: molecular docking (Glide and AutoDock Vina), ligand-based superposition (FlexS), and deep learning-based modeling (AlphaFold3, Boltz-2, DiffDock and Gnina). We evaluated their performance on binding pose prediction for ligands targeting SARS-CoV-2 and MERS-CoV main protease (Mpro). For binding affinity estimation, we implemented a machine learning-base

Advancing healthcare allocation and prevention of disability: the role of disease-based predictive model for disability in aging adults.

BACKGROUND: Disability in the aging population was a major public health challenge for aging nations, imposing a significant burden on healthcare resources. Accurate disability prediction models were essential for efficiently allocating long-term care resources and preventing disability. This study utilized healthcare claims data to construct a disease-based disability risk prediction model that identified high-risk disability groups and diseases with significant impacts on disability. The model informed the formulation of prevention strategies and resource allocation. METHODS: This study ado

Comparative Performance of Machine Learning and Traditional Risk Scores in Predicting Adverse Events After Transcatheter Aortic Valve Replacement in Patients With Atrial Fibrillation.

Patients with atrial fibrillation (AF) following transcatheter aortic valve replacement (TAVR) remain at risk of ischemic stroke (IS) and bleeding. However, traditional risk scores provide modest predictions of IS and bleeding in these patients. We aimed to develop machine learning (ML) models that predict IS, major gastrointestinal bleeding (MGIB), all clinically relevant bleeding (CRB), and net adverse clinical events (NACE) using data from patients in the ENVISAGE-TAVI AF trial. Ten ML algorithms were trained per outcome using nested cross-validation; the best-performing model (highest F1 s

Predicting resistance to fluoroquinolones among patients with rifampicin-resistant tuberculosis: A cross-country validation study.

BACKGROUND: Fluoroquinolones (FQs) are a cornerstone of most all-oral, shorter regimens endorsed by the World Health Organization for the treatment of rifampicin-resistant or multidrug-resistant tuberculosis (RR/MDR-TB). Knowledge of resistance to FQs can help guide regimen selection at the point of care. In settings where rapid testing for FQ resistance is unavailable, prediction models could support treatment decisions by identifying FQ resistance based on patient characteristics observable at the point of care. These prediction models have been typically developed and evaluated within a sin

Reliability of Genetic Alterations in Predicting Ceftriaxone Resistance in Neisseria gonorrhoeae Globally.

Antimicrobial resistance in N. gonorrhoeae is increasing globally, and ceftriaxone is the recommended treatment for empirical therapy in most settings. Developing molecular assays to detect decreased ceftriaxone susceptibility is critical. Using PathogenWatch, a public database of N. gonorrhoeae genomes, antibiotic susceptibility data and DNA sequences of different genes associated with ceftriaxone resistance were extracted. That information was used to determine the sensitivity and specificity of different molecular markers and algorithms to predict decreased susceptibility to ceftriaxone. A

BACKGROUND: Late gadolinium enhancement (LGE) border zone on cardiac magnetic resonance imaging has been proposed as an independent predictor of ventricular arrhythmias. The purpose was to determine whether size and heterogeneity of LGE predict appropriate implantable cardioverter defibrillator (ICD) therapy in ischemic cardiomyopathy (ICM) and nonischemic cardiomyopathy (NICM) patients and to evaluate 4 LGE border-zone algorithms. We used routinely collected data to develop and assess machine learning (ML) algorithms to predict unplanned ACU within 90 days of ICI treatment initiation. METHODS: We used aggregated electronic health record data from 7,960 patients receiving ICI treatments to train and assess eight ML algorithms.
