Uukuniemi Virus as a Tick-Borne Virus Model.

UNLABELLED: In the last decade, novel tick-borne pathogenic phleboviruses in the family Bunyaviridae, all closely related to Uukuniemi virus (UUKV), have emerged on different continents. To reproduce the tick-mammal switch in vitro, we first established a reverse genetics system to rescue UUKV with a genome close to that of the authentic virus isolated from the Ixodes ricinus tick reservoir. The IRE/CTVM19 and IRE/CTVM20 cell lines, both derived from I. ricinus, were susceptible to the virus rescued from plasmid DNAs and supported production of the virus over many weeks, indicating that infect

Borrelia sp. phylogenetically different from Lyme disease- and relapsing fever-related Borrelia spp. in Amblyomma varanense from Python reticulatus.

BACKGROUND: Species of the genus Borrelia are causative agents of Lyme disease and relapsing fever. Lyme disease is the most commonly reported vector-borne disease in the northern hemisphere. However, in some parts of the world Lyme borreliosis and relapsing fever may be caused by novel Borrelia genotypes. Herein, we report the presence of a Borrelia sp. in an Amblyomma varanense collected from Python reticulatus. METHODS: Ticks were collected from snakes, identified to species level and examined by PCR for the presence of Borrelia spp. flaB and 16S rRNA genes. Phylogenetic trees were constru

Rhipicephalus (Boophilus) microplus resistant to acaricides and ivermectin in cattle farms of Mexico.

Ticks and the diseases they transmit cause great economic losses to livestock in tropical countries. Non-chemical control alternatives include the use of resistant cattle breeds, biological control and vaccines. However, the most widely used method is the application of different chemical classes of acaricides and macrocyclic lactones. Populations of the cattle tick, Rhipicephalus (Boophilus) microplus, resistant to organophosphates (OP), synthetic pyrethroids (SP), amitraz and fipronil have been reported in Mexico. Macrocyclic lactones are the most sold antiparasitic drug in the Mexican veter

Acaricidal action of destruxins produced by a marine-derived Beauveria felina on the bovine tick Rhipicephalus (Boophilus) microplus.

The increasing resistance of Rhipicephalus (Boophilus) microplus tick to commercial insecticides requires alternative methods for the control of this cattle plague. The enthomopathogenic fungus Beauveria felina produces destruxins in culture media, cyclic depsipeptides which display an array of biological activities. The present investigation aimed to evaluate the acaricide action of destruxins isolated from B. felina culture media on R. (B.) microplus engorged females. B. felina was grown in MF medium under 19 different growth conditions. HPLC-PDA analysis of chromatographic fractions obtaine

The fecal odor of sick hedgehogs (Erinaceus europaeus) mediates olfactory attraction of the tick Ixodes hexagonus.

Parasite loads of animals vary among individuals, but the underlying mechanisms have not been fully identified. Here, we investigated whether health status of hedgehogs (Erinaceus europaeus) is correlated with tick burden, and whether chemical cues linked to the health status of the host mediate attraction of the tick Ixodes hexagonus. An ecological survey conducted over 10 years, involving 226 wild hedgehogs, revealed a strong association between health status and tick burden of hedgehogs, with healthy animals being less likely to carry ticks than unhealthy ones. Behavioral choice tests demon

Hosts of the exotic ornate kangaroo tick, Amblyomma triguttatum triguttatum Koch, on southern Yorke Peninsula, South Australia.

Amblyomma triguttatum triguttatum is assumed to be endemic to south-western Western Australia (including Barrow Island), Queensland (excluding Cape York Peninsula), and New South Wales, south to Dubbo and Barham. The species has been recorded on a range of mammalian hosts including macropods and domestic animals. In Queensland, A. triguttatum triguttatum is implicated in the epidemiology of Q fever. In 2000, the species was detected on southern Yorke Peninsula in South Australia. We aimed to identify A. triguttatum triguttatum's hosts through trapping, sampling of carcasses, and opportunistic

Effectiveness of Metarhizium anisopliae (Deuteromycetes) against Ixodes scapularis (Acari: Ixodidae) engorging on Peromnyscus leucopus.

With the incidence of Lyme disease increasing throughout the United States, reducing risk of exposure to the disease is of the utmost concern. In the northeastern U.S., the blacklegged tick, Ixodes scapularis, is the primary vector and the white-footed mouse, (Peromyscus leucopus), the primary reservoir for Borrelia burgdorteri, the bacterium causing Lyme disease. Targeting I. scapularis engorging on white-footed mice with an effective biological control agent, such as the fungus Metarhizium anisopliae, could be an effective and relatively safe control technique. In 2002-2003, we performed lab

Interaction and transmission of two Borrelia burgdorferi sensu stricto strains in a tick-rodent maintenance system.

In the northeastern United States, the Lyme disease agent, Borrelia burgdorferi sensu stricto, is maintained by enzoonotic transmission, cycling between white-footed mice (Peromyscus leucopus) and black-legged ticks (Ixodes scapularis). B. burgdorferi sensu stricto is genetically variable and has been divided into three major genotypes based on 16S-23S ribosomal DNA spacer (RST) analysis. To better understand how genetic differences in B. burgdorferi sensu stricto may influence transmission dynamics in nature, we investigated the interaction between an RST1 and an RST3 strain in a laboratory s

Skin test and tick immune status in susceptible and resistant cattle in Brazil.

A cutaneous hypersensitivity test (CHT) was used to correlate host resistance to ticks and type of reaction elicited to unfed larval extract-ULE of the cattle tick Boophilus microplus in European and Indian cattle. Twenty calves were separated into four groups of five animals each: naïve or preinfested Indian or European cattle. CHT was induced by intradermal inoculation of 0.1 ml of ULE cattle tick B. microplus (50 micrograms protein) in the calf ear. Ear thickness was measured using calipers before and 10 min, 1, 2, 6, 18, 24, 48, 72, 96, and 144 h postinoculation (PI). Preinfested European

Arthropod parasites of some wild animals in South Africa and Namibia.

Earlier research on the parasites of wild animals in South Africa is reviewed and the findings of more recent research discussed. The life cycles of various gasterophilid and oestrid fly species are described and the seasonal abundance of their larvae in their zebra and antelope hosts is considered. The seasonal abundance of fleas, lice and ixodid ticks on their hosts is given and the role played by both small and large mammals and some birds in the maintenance of tick populations is described. Factors contributing to severe parasitism of wild animals are listed and the chemical and biological

Co-Infection Patterns in Individual Ixodes scapularis Ticks Reveal Associations between Viral, Eukaryotic and Bacterial Microorganisms.

Ixodes scapularis ticks harbor a variety of microorganisms, including eukaryotes, bacteria and viruses. Some of these can be transmitted to and cause disease in humans and other vertebrates. Others are not pathogenic, but may impact the ability of the tick to harbor and transmit pathogens. A growing number of studies have examined the influence of bacteria on tick vector competence but the influence of the tick virome remains less clear, despite a surge in the discovery of tick-associated viruses. In this study, we performed shotgun RNA sequencing on 112 individual adult I. scapularis collecte

Tick toxicosis in a dog bitten by Ornithodoros brasiliensis.

Ticks are hematophagous parasites of people and animals and are a public health hazard in several countries. They are vectors of infectious diseases; in addition, the bite of some ticks, mainly from the Ornithodoros genus, may lead to local lesions and systemic illness, referred to as tick toxicosis. In this report, we describe a dog bitten by Ornithodoros brasiliensis, popularly known as the mouro tick. The main clinical findings were disseminated skin rash, pruritus, mucosal hyperemia, lethargy, and fever. Laboratory abnormalities 48 hours after the bites occurred included mild nonregenerati

Dog tick-borne diseases in Sicily.

In Sicily many tick borne diseases are endemic, in particular way those that see like main carrier ticks that prefer, for their vital cycle, climatic conditions characterized by high temperatures and a warmth-humid atmosphere. The more important pathologies transmitted by ticks causing diseases in dogs are babesiosis and ehrlichiosis. Borrelia burgdorferi, Anaplasma phagocytophilum, Rickettsia conorii, Coxiella burnetii and tick transmitted encephalitis virus assume particular relevance because they are agents of zoonosis. Our centre, C.R.A.Ba.R.T, have conducted many researches and carried ou

Geographic distribution of ticks (Acari: Ixodidae) in Iowa with emphasis on Ixodes scapularis and their infection with Borrelia burgdorferi.

In Iowa, public concern regarding Lyme disease has increased markedly over the last decade. In response to these concerns, a statewide surveillance program was initiated in 1990 based on ticks received by the Department of Entomology at Iowa State University. Ticks were received from health care professionals, state government agencies, and the general public. A total of 5,343 ticks from all 99 Iowa counties were identified during the 12 years of this study. Dermacentor variabilis was the most numerous species, followed by Amblyomma americanum, and Ixodes scapularis. Dermacentor variabilis wer

Laboratory and field investigations into the Theileria parva carrier-state in cattle in Zimbabwe.

The Theileria parva carrier-state in cattle on commercial farms on Zimbabwe was investigated using parasitological and serological methods. The proportion of cattle showing Theileria piroplasms on two farms, which had recent histories of disease outbreaks, were 64% (n = 106, total of heifers and weaned calves examined) and 71.5% (n = 60) while the proportion of T. parva antibodies for the same animals were 59% and 98.5%, respectively. On four farms where no cases of the disease occurred for over 10 years, the average proportion of animals showing piroplasms and antibodies were 55.4% (range 32-

A protective "concealed" antigen from Boophilus microplus. Purification, localization, and possible function.

A membrane protein that can be used successfully to vaccinate cattle against the tick Boophilus microplus has been purified and characterized. The mature protein, which is referred to as Bm91, has an apparent m.w. of 86,000, an isoelectric point of 4.8 to 5.2, and is glycosylated, with an affinity for lentil lectin. Bm91 is of relatively low abundance, with approximately 300 to 400 micrograms being recovered from 1 kg of semiengorged adult female ticks. The protein is located largely in the salivary gland and gut of these ticks. Partial amino acid sequence data for the protein show striking si

Successful vaccination against Boophilus microplus and Babesia bovis using recombinant antigens.

Current methods for the control of the cattle tick Boophilus microplus and the agent of bovine babesiosis, Babesia bovis are unsatisfactory. Effective immunological control of both parasites would have great advantages. However, naturally acquired immunity to the tick is generally unable to prevent serious production losses. A vaccine against the tick, based on a novel form of immunization, is being developed. A protective antigen has been isolated from the tick, characterized and produced as an effective, recombinant protein. A vaccine incorporating this antigen is currently undergoing field

Epidemiology of theileriosis in calves in an endemic area of Kenya.

Thirty-one calves born into five Maasai zebu cattle herds over a period of 1 month in the Trans-Mara Division of Kenya, endemic for theileriosis, were recruited for an intensive study of theileriosis. No calves up to 6 months of age died but all developed Theileria infections as judged by slide examination and serology. Parasitosis by T. mutans schizonts in lymph node smears was usually higher than that of T. parva. The T. mutans schizonts usually occurred at an earlier age but persisted at a patent level for a shorter time than those of T. parva. Serological findings using the indirect fluore

Morphological and molecular characterization of ixodid tick vectors in Algeria, North Africa.

Ticks are hematophagous ectoparasites of veterinary and medical importance, serving as vectors for a wide array of pathogens that affect livestock, wildlife, and humans. Given their role in pathogen dissemination, this study aimed to provide an updated molecular characterization and phylogenetic analysis of hard tick species in Algeria using two mitochondrial markers: Cytochrome c oxidase subunit 1 (Cox1) and the 12S rRNA gene. We analyzed fourteen tick species from the archival collection of the Pasteur Institute of Algeria, originally collected from various provinces across the country. Thes
