Structural anomalies of the peripheral olfactory system in psychosis high-risk subjects.

BACKGROUND: Olfactory impairments are prominent in both schizophrenia and the preceding at-risk state. Their presence prior to illness predicts poor functional outcome. In schizophrenia, these impairments reflect peripheral olfactory structural abnormalities, which are hypothesized to arise during early embryonic development. If this is correct, then similar structural anomalies should be apparent among clinical high-risk subjects. METHODS: Thirty-nine clinical high-risk (CR) subjects (24M/15F) were compared to 36 low-risk (LR) subjects (19M/17F). Olfactory measures derived from 3T MRI scans

Methylxanthines induce structural and functional alterations of the cardiac system in zebrafish embryos.

BACKGROUND: Zebrafish embryos are emerging as a model for pharmacological and toxicological studies. We used zebrafish embryos to study the general toxicity and cardiovascular effects of eight methylxanthines: aminophylline, caffeine, diprophylline, doxofylline, etophylline, 3-isobutyl-1-methylxanthine (IBMX), pentoxifylline and theophylline. METHODS: Microinjections of the eight methylxanthines were performed in 1-2 cell stage zebrafish embryos and the general toxicity and cardiovascular effects were analyzed at different time points. Embryotoxicity and teratogenicity were evaluated to under

Isolation, characterization, and mapping of the mouse Fgd3 gene, a new Faciogenital Dysplasia (FGD1; Aarskog Syndrome) gene homologue.

FGD1 gene mutations result in faciogenital dysplasia (FGDY, Aarskog syndrome), an X-linked developmental disorder that adversely affects the formation of multiple skeletal structures. FGD1 encodes a guanine nucleotide exchange factor (GEF) that specifically activates the Rho GTPase Cdc42. By way of Cdc42, FGD1 regulates the actin cytoskeleton and activates the c-Jun N-terminal kinase signaling cascade to regulate cell growth and differentiation. Previous work shows that FGD1 is the founding member of a family of related genes including the mouse Fgd2 gene and the rat Frabin gene. Here, we repo

Total auricular reconstruction with a three-dimensional costal cartilage framework.

Microtia (hypoplasia of the auricle) is known to result from incomplete development of the auricle during embryonic development so that the size and shape of the deformity vary from case to case. Total auricular reconstruction for microtia is dependent on the degree of hypoplasia in comparison to that of the normal auricle on which the specific reconstruction method is based. I have classified microtia into 3 major types: 1) The lobule type. 2) The concha type, and 3) The small concha type which has been previously described [32]. Regardless of the size and shape of the deformity, the ultimate

Toxicomorphomics and toxicokinetics of quinalphos on embryonic development of zebrafish (Danio rerio) and its binding affinity towards hatching enzyme, ZHE1.

This study outlines the toxic effects of Quinalphos (QP), an organophosphrous insecticide on the development of zebrafish (Danio rerio) embryos, with special emphasis on toxicomorphomics and toxicokinetics of target enzyme, AChE. A range of concentrations was used to elucidate the median lethal concentration (LC50) of Quinalphos. Furthermore, embryos were exposed to two sub-lethal concentrations LC10 (0.66mg/L) and LC20 (1.12mg/L) along with a median lethal concentration (3.0mg/L) for 96h. Several morphological aberrations like lordosis, kyphosis, scoliosis, heart edema, breaks in the neuronal

Left-right asymmetry in vertebrate embryogenesis.

Embryonic development results in animals whose body plans exhibit a variety of symmetry types. While significant progress has been made in understanding the molecular events underlying the early specification of the antero-posterior and dorso-ventral axes, little information has been available regarding the basis for left-right (LR) differences in animal morphogenesis. Recently however, important advances have been made in uncovering the molecular mechanisms responsible for LR patterning. A number of genes (including well-known signaling molecules such as Sonic hedgehog and activin) are asymme

Developmental toxicity study in rats and rabbits administered an emulsion containing medium chain triglycerides as an alternative caloric source.

Triglyceride-containing lipid emulsions have been designed as caloric sources that can be administered intravenously to patients that cannot meet their nutritional needs by conventional parenteral therapies. In their study, we evaluate the developmental toxicity of a 20% lipid emulsion that contains a 3:1 ratio of medium chain triglyceride (MCT) to one long chain containing lipid emulsion (LCT). This emulsion was administered by intravenous infusion to rats and rabbits at dosages of 1 and 4.28 g lipid/kg body weight (g lipid/kg) at dose volumes of 5 and 21.4 mL/kg, respectively, once daily dur

Mechanisms of action of the congenital diaphragmatic hernia-inducing teratogen nitrofen.

Congenital diaphragmatic hernia (CDH) is a developmental anomaly that results in significant mortality and morbidity. The underlying etiology is poorly understood. Insights will arise from an understanding of the mechanisms by which the teratogen nitrofen induces CDH in rodent models. In this study, we use in vitro cell assays in conjunction with whole animal rodent studies to test hypotheses regarding nitrofen's mechanism of action. The first component examined the interaction of nitrofen with various aspects of the retinoid signaling pathway including uptake proteins, binding proteins, recep

Identification of small-molecule inhibitors of the immune system repressor Regnase-1.

Regnase-1 is an essential RNase that suppresses immune cell activation by targeting the mRNA of pro-inflammatory cytokines such as interleukin 6 (IL-6). Although genetic inhibition of Regnase-1 activity in CD8+ T cells has been shown to enhance antitumor immunity, no molecular inhibitors of Regnase-1 have been reported. By exploiting a library of nucleic acid analogs in conjunction with an RNA displacement assay, we identified multiple candidate competitors of Regnase-1 RNA substrates. Among these, suramin-an approved drug for treating African sleeping sickness-and several of its derivatives d

Declining ribosome function limits oocyte longevity.

Protein synthesis declines with age across many tissues, but whether this decline actively contributes to physiological aging remains unclear. Oocytes may be particularly vulnerable because they rely on ribosomes synthesized during growth and maintained for prolonged periods without continual replacement. Here we show that declining translational capacity is a causal component of oocyte aging. An unbiased genetic screen identified ribosome dosage as a major determinant of oocyte longevity, and experimentally reducing ribosome abundance was sufficient to accelerate the loss of developmental com

Acute and Chronic Toxicity of Propylparaben in the Freshwater Snail Biomphalaria glabrata: Effects on Survival, Growth, Reproduction, and Histopathology.

Propylparaben (PP) is a widely used preservative in cosmetics, pharmaceuticals, and food products, and its potential toxicity to non-target aquatic invertebrates remains a concern. This study used the freshwater snail Biomphalaria glabrata as a model organism to evaluate the toxic effects of PP through acute and chronic exposures at embryonic, newly hatched, and adult stages. Acute exposure experiments showed concentration-dependent mortality and developmental inhibition, with LC50 values of 36.69 mg/L (embryos, 168 h), 33.48 mg/L (newly hatched snails, 96 h), and 57.05 mg/L (adults, 72 h). Microglial Maturation and Functional Heterogeneity: Mechanistic Links to Neurodevelopmental Disorders.

As the brain's resident macrophages, microglia on the one side are increasingly recognized as essential players in discrete developmental stages, where immune, metabolic, and activity-derived signals are coordinately integrated to guide brain development. On the other side, the precise temporal and molecular coordination of microglial maturation is imperative for the structural and functional integrity of the developing central nervous system (CNS). In this review, we synthesize recent data that reposition microglia from a uniform population of immune sentinels to temporally programmed and reg

scDIAGRAM: detecting chromatin compartments from individual single-cell Hi-C matrix without imputation or reference features.

Single-cell Hi-C (scHi-C) provides unprecedented insight into 3D genome organization, but its sparse and noisy data pose challenges in accurately detecting A/B compartments, which are crucial for understanding chromatin structure and gene regulation. We presented scDIAGRAM, a data-driven method for annotating A/B compartments in single cells using direct statistical modeling and graph community detection. Unlike existing approaches, scDIAGRAM infers chromatin compartments directly from individual scHi-C matrix without imputation or external reference features, and subsequently assigns A/B labe

Post traumatic response of coelomocytes in the common sea star Echinastersepositus.

Regenerative dynamics stem from the coordination of pathways shared with processes of reproduction, post-embryonic development and any tissue mechanisms triggered to reconstitute damaged or lost cells and tissues. In living organisms, following a wound, immune cells are recruited to the injured area to defend against pathogens and promote the structural and functional restoration of the damaged tissue. This repair process is thoroughly investigated in sea stars throughout their development, from the initial repair stages to the actual regenerative phase. In this context, circulating coelomocyt

Standardized Isolation, Culture, and Imaging of Contracting Cardiomyocytes from Neonatal Mouse Hearts.

Cardiomyocyte maturation represents the final stage of functional development in the heart. During this critical process, fetal cardiomyocytes gradually acquire adult phenotypes and become capable of sustaining billions of contraction-relaxation cycles throughout an organism's lifespan. This transition is driven by gene expression changes that remodel cardiomyocyte structure, metabolism, and functions. In mice, cardiomyocyte maturation takes place during embryonic day (E)18.5 to postnatal two weeks, during which the heart loses its regenerative capacity in response to injury. Hence, characteri

Brain implantation of soft bioelectronics via embryonic development.

Developing bioelectronics capable of stably tracking brain-wide, single-cell, millisecond-resolved neural activity in the developing brain is critical for advancing neuroscience and understanding neurodevelopmental disorders. During development, the three-dimensional structure of the vertebrate brain arises from a two-dimensional neural plate1,2. These large morphological changes have previously posed a challenge for implantable bioelectronics to reliably track neural activity throughout brain development3-9. Here we introduce a tissue-level-soft, submicrometre-thick mesh microelectrode array

Spinal Cord-Like Scaffold with Rapid Tissue Integration Enhanced Spinal Cord Nerve Repair.

Spinal cord injury (SCI) repair represents an immense challenge globally. Despite various beneficial strategies having been attempted for SCI repair, the ideal therapy has not yet been achieved. In this study, a multidimensional biomimetic spinal cord-like scaffold (MBSLS) is designed with a hierarchical structure and embryonic neurogenetic microenvironment and neural regeneration in SCI repair. MBSLS successfully regulated the neural differentiation of loaded neural stem cells (NSCs) and regenerated spinal cord-like tissue in vitro by promoting neurogenesis, gliogenesis, axonogenesis, axon en

Vesicle-associated membrane protein 5 is an intrinsic defense factor for embryonic stem cells against coronaviruses.

Embryonic stem cells (ESCs) display a distinctive resistance against various viruses, irrespective of any interferon response. Nevertheless, the underlying mechanism of this resistance remains unclear. In this study, we identify vesicle-associated membrane protein 5 (VAMP5) as a potent cell-autonomous defense factor against coronaviruses, including SARS-CoV-2, with high expression levels observed in ESCs and mesoderm. VAMP5 not only exhibits functional conservation in restricting the replication of SARS-CoV-2 and its variants, as well as other highly pathogenic coronaviruses, but also shows ef
