Mouse strain differences in opiate reward learning are explained by differences in anxiety, not reward or learning.

Gene-targeting techniques to produce null mutations provide a powerful method for evaluating the contribution of particular candidate genes involved in motivation. The embryonic stem cell lines in which homologous recombination is undertaken are derived from 129 mice, but because of the impoverished performance of 129 mice on a number of behavioral tasks, mice chimeric for the mutation are often bred with a C57BL/6 mouse strain. Thus, an examination of both parental strains is important in the study of the knock-out mice. Although the C57BL/6 behavioral phenotype is well documented, details of

Sunlight Modulates Fruit Metabolic Profile and Shapes the Spatial Pattern of Compound Accumulation within the Grape Cluster.

Vineyards are characterized by their large spatial variability of solar irradiance (SI) and temperature, known to effectively modulate grape metabolism. To explore the role of sunlight in shaping fruit composition and cluster uniformity, we studied the spatial pattern of incoming irradiance, fruit temperature and metabolic profile within individual grape clusters under three levels of sunlight exposure. The experiment was conducted in a vineyard of Cabernet Sauvignon cv. located in the Negev Highlands, Israel, where excess SI and midday temperatures are known to degrade grape quality. Filterin

Biologically-constrained spiking neural network for neuromodulation in locomotor recovery after spinal cord injury.

Presynaptic inhibition after spinal cord injury (SCI) has been hypothesised to disproportionately affect flexion reflex loops in locomotor spinal circuitry. Reducing gamma-aminobutyric acid (GABA) inhibitory activity increases the excitation of flexion circuits, restoring muscle activation and stepping ability. Conversely, nociceptive sensitisation and muscular spasticity can emerge from insufficient GABAergic inhibition. To investigate the effects of neuromodulation and proprioceptive sensory afferents in the spinal cord, a biologically constrained spiking neural network (SNN) was developed.

Olfactory bulb circuits drive ketamine-enhanced high-frequency oscillations via kainate and GABAergic mechanisms.

Ketamine produces rapid antidepressant effects accompanied by marked changes in fast brain rhythms. High-frequency oscillations (HFO; 130-180 Hz) are an increasingly recognized signature of ketamine across species. However, their relationship to classical gamma activity and their circuit-level organization remain unclear. Using simultaneous multiregion recordings in freely moving male Wistar rats, we show that ketamine differentially reorganizes fast oscillations across cortical networks: neocortical gamma power increased broadly, whereas the olfactory bulb (OB) showed suppressed gamma alongsi

Polysaccharides from Baihe Dihuang decoction ameliorate depression-like behaviors in rats subjected to chronic unpredictable mild stress through modulation of metabolic homeostasis.

Baihe Dihuang Decoction (BDD) is a traditional Chinese medicinal formula with well-documented antidepressant effects. However, the role of its polysaccharide fraction (BDDP) remains poorly understood. This study aimed to investigate the structural characteristics and antidepressant mechanisms of BDDP through chemical analysis and in vivo validation. BDDP was obtained by ethanol precipitation and analyzed by high-performance anion-exchange chromatography (HPAEC), revealing a heteropolysaccharide composed of rhamnose, arabinose, galactose, glucose, mannose, and fructose, with an average molecula

Regulation Progresses of Selenium Improving Intestinal and Extra-Intestinal Tissues Health Through Regulating Gut Microbiota.

Selenium (Se) is an essential trace element that exerts pleiotropic effects on host physiology, yet the mechanisms by which it coordinates systemic health remain incompletely understood. Emerging evidence regards the gut microbiota as a key mediator of Se biological functions, giving rise to the Se-gut-tissue axis. This review synthesizes the current research progresses on how dietary Se may shape gut microbial composition and metabolism, and how these microbial shifts are associated with protective effects in both intestinal and extra-intestinal tissues. Se sources (particularly organic or ne

Major Depressive Disorder Signatures: A Review of Artificial Intelligence (AI)-Powered Insights Into Gut Dysbiosis.

BACKGROUND AND AIMS: Major Depressive Disorder (MDD) is a highly common neuropsychiatric disorder globally. A variety of factors contribute to the neuropathology of MDD. Microbiome research in neuropsychiatric disorders such as MDD has recently attracted attention. Indeed, the gut-brain axis could influence the course of MDD through metabolites such as Gamma-Aminobutyric Acid (GABA), Quinolinate, and other factors. Such metabolites may modulate the balance of excitatory and inhibitory signals. Moreover, MDD features abundant hyperinflammatory bacteria, whereas anti-inflammatory butyrate-synthe

Emerging Roles of Postbiotics in Gut-Brain-Microbiome Axis Modulation and Neurobiological Pathways of Chronic Stress-Related Brain Dysfunction.

Chronic psychological stress disrupts the gut-brain-microbiome axis (GBMA) through gut dysbiosis, intestinal and blood-brain barrier disruption, hypothalamic-pituitary-adrenal (HPA) axis dysregulation, and neuroinflammation, collectively impairing neurotransmitter signaling and neuroplasticity. Addressing these interconnected pathological processes requires therapeutic strategies capable of acting across multiple nodes of the GBMA simultaneously. Postbiotics, defined as preparations of inanimate microorganisms and/or their components that confer a health benefit on the host, have emerged as pr

A novel fluorescence-based method for the determination of Zuranolone using Tinopal CBS-X in the pharmaceutical formulation and spiked human plasma.

Zuranolone is a neuroactive steroid modulating GABA-A receptors, providing fast and effective treatment for depressive disorders. Despite its therapeutic promise, Zuranolone lacks strong chromophores or fluorophores, complicating its quantitative analysis using traditional optical methods. This study presents a validated spectrofluorimetric method for Zuranolone determination, utilizing Tinopal CBS-X, a fluorescent dye, as a probe. Under optimized conditions, a stable ion-pair complex forms between Tinopal CBS-X and Zuranolone, with characteristic florescence emission signal at 520 nm. The met

Resolving native GABAA receptor structures from the human brain.

Type A GABA (γ-aminobutyric acid) receptors (GABAA receptors) mediate most fast inhibitory signalling in the brain and are targets for drugs that treat epilepsy, anxiety, depression and insomnia and for anaesthetics1,2. These receptors comprise a complex array of 19 related subunits, which form pentameric ligand-gated ion channels. The composition and structure of native GABAA receptors in the human brain have been inferred from subunit localization in tissue1,3, functional measurements and structural analysis from recombinant expression4-7 and in mice8. However, the arrangements of subunits t

Intracellular mechanisms underlying modulation of inhibitory synaptic transmission by appetite-related peptides in the insular cortex.

BACKGROUND: The insular cortex (IC) integrates multimodal sensory inputs, including gustatory, visceral, nociceptive, and oral somatosensory information, and is widely regarded as the primary gustatory cortex. These functions are closely associated with feeding behaviors such as mastication, swallowing, and digestion, which are affected by appetite- and energy balance-related peptides. HIGHLIGHT: Orexin, leptin, and insulin have received particular attention because of their effects on IC circuits. Orexin, produced in the hypothalamus, activates postsynaptic phospholipase C, thereby inducing
