Pyrethroid pediculicide resistance of head lice in Canada evaluated by serial invasive signal amplification reaction.

BACKGROUND: Most people in the United States and Canada with pediculosis will be treated with neurotoxic pediculicides containing pyrethrins or pyrethroids. Their widespread use led to significant resistance reported from various countries. Although treatment failures are frequently observed in Canada, the resistance frequency to pyrethroid pediculicide of human head lice (Pediculus humanus capitis) has not been determined. OBJECTIVE: To determine the knockdown resistance (kdr) allele frequency in human head louse populations in Canada. METHODS: Patients infested with Pediculus humanus capit

Field evaluation of a novel tick control strategy aiming a reduced selection for drug resistance: A case for "host-level chemical mosaic".

Widespread multiple acaricide resistance in Rhipicephalus microplus has challenged conventional strategic tick-control schemes using acaricides that prioritize short-term efficacy while largely overlooking how treatment structure affects resistance selection. Here, we propose and validate a novel protocol for chemical control designed to reduce pressure selection for acaricide resistance: host-level chemical mosaic (HLCM). This approach combines two initial uniform treatments with subsequent alternation of drugs between subgroups of animals within the same paddock. The first and second applica

Integration of publicly available datasets reveals pesticide-associated genetic variation in a wild non-target animal.

Pesticide use is ubiquitous. While pesticide resistance in target species such as insects is a well-known phenomenon, it is less known how pesticide exposures affect the genetics of non-target species. Here, we demonstrate the research value of integrating existing datasets to study the relationship between chemicals and biodiversity from the genetics perspective by analyzing spatial pesticide concentration data and genetic data from Europe that were not initially generated for toxicological purposes. Specifically, we found associations between environmental pesticide concentrations and geneti

Carvone-Rich Essential Oils and Their Agrobiological Interactions: A Review.

Carvone-rich essential oils (EOs), and carvone specifically, exhibit a broad spectrum of protective effects against major agricultural threats. They display strong antifungal and moderate antibacterial effects, effectively inhibiting numerous phytopathogenic fungi. EOs exhibit significant insecticidal, acaricidal, and repellent activity against various insects and mites, and some EOs are highly effective against agricultural nematodes, suppressing mobility and egg hatching. Crucially, the EOs demonstrate a strong capacity to suppress the germination and initial growth of different weed species

Functional Analysis of PxylPBP2 Responding to Repellent Activity of Natural Pyrazine Against Diamondback Moth.

2,3-dimethyl-6-(1-hydroxy)-pyrazine showed significant repellent activity to diamondback moth (Plutella xylostella), the PxylPBP2 was considered as the potential molecular target, though this has not been validated. In this study, the repellent rate was significantly decreased by about 20% when the PxylPBP2 was silenced using RNAi. There was obvious interaction between 2,3-dimethyl-6-(1-hydroxy)-pyrazine and PxylPBP2, with a Kd (Dissociation constant) value of 7.78 μmol/L via microscale thermophoresis, while the value was changed to 4.45 μmol/L when the PxylOR31 (Pxyl, Plutella xylostella; OR3

How to successfully express insect microsomal P450 using a baculovirus system.

Cytochrome P450 enzymes (P450s) play a key role in the metabolism of a diverse array of exogenous compounds. In insects they are often implicated in the phase I metabolism of many insecticides, which can lead to the emergence of resistance. Studies of such P450s have frequently employed heterologous expression in insect cell lines for in vitro functional characterisation. However, identifying optimal conditions for insect P450s expression is challenging. Here, we address this challenge by developing an improved protocol for insect P450s expression in insect cell lines employing two new concept

From the Eye of the Storm to Epidemiological Footprints After the Floods: Viral, Vector-Borne, and One Health Risks Post-Hurricane Melissa in Jamaica.

Hurricanes cause severe impacts on lives, livelihoods, and essential systems. Hurricane Melissa impacted Jamaica as a Category 5 cyclone, resulting in estimated losses of approximately 41% of national GDP (US$8.8 billion) and eliciting widespread damage to housing, healthcare, agriculture, and urban infrastructure. Agriculture sustained heavy losses, with 41,000 hectares of damaged farmland and the loss of more than 1 million livestock animals. These impacts resulted in exposed animal closures with biological hazards. Using systems thinking, the PESTHEEL framework, and a One Health lens, we ar

Anopheles mosquito survival and pharmacokinetic modeling show the mosquitocidal activity of nitisinone.

One approach to interrupting the transmission of insect-borne diseases that is successfully used in veterinary medicine is exploiting the ability of antiparasitic drugs to make vertebrate blood toxic for blood-feeding insects. Recent studies have identified 4-hydroxyphenylpyruvate dioxygenase (HPPD), an enzyme of the tyrosine detoxification pathway, as essential for hematophagous arthropods to digest their blood meals. Such blood-feeding insects include anopheline mosquitoes, which transmit malaria-causing Plasmodium parasites. A US Food and Drug Administration-approved HPPD enzyme inhibitor c

Evaluating Beauveria bassiana Strains for Insect Pest Control and Endophytic Colonization in Wheat.

Certain entomopathogenic fungi, such as Beauveria bassiana, are highly pathogenic to arthropod pests and are able to colonize plant tissues, thereby enhancing both plant growth and disease resistance. This study assessed three B. bassiana strains (CBM1, CBM2, and CBM3) for their pathogenicity toward insect larvae and colonization potential in wheat. The insecticidal activity of the fungi against the larvae of the major lepidopteran pests Helicoverpa armigera, Spodoptera frugiperda, Mythimna separata, and Plutella xylostella was determined. The fungi were then applied to wheat plants using seed

Global Health Priority Box─Proactive Pandemic Preparedness.

The coronavirus pandemic outbreak of 2019 highlighted the critical importance of preparedness for current and future public health threats (https://www.mmv.org/mmv-open/global-health-priority-box/about-global-health-priority-box). While the main attention for the past few years has been on COVID-19 research, this focus has reduced global resources on research in other areas, including malaria and neglected tropical diseases (NTDs). Such a shift in focus puts at risk the hard-earned progress in global health achieved over the past two decades (https://www.who.int/news-room/spotlight/10-global-h

Review: Presence, distribution and current pesticides used in Spanish agricultural practices.

To guarantee an adequate food supply for the world's growing population, intensive agriculture is necessary to ensure efficient food production. The use of pesticides helps maintain maximum productivity in intensive agriculture by minimizing crop losses due to pests. However, pesticide contamination of surface waters constitutes a major problem as they are resistant to degradation and soluble enough to be transported in water. In recent years, all groups of pesticides defined by the World Health Organization have increased their use and, therefore, their prevalence in the different environment

Antimicrobial, Insecticidal and Cytotoxic Activity of Linear Venom Peptides from the Pseudoscorpion Chelifer cancroides.

Linear cationic venom peptides are antimicrobial peptides (AMPs) that exert their effects by damaging cell membranes. These peptides can be highly specific, and for some, a significant therapeutic value was proposed, in particular for treatment of bacterial infections. A prolific source of novel AMPs are arthropod venoms, especially those of hitherto neglected groups such as pseudoscorpions. In this study, we describe for the first time pharmacological effects of AMPs discovered in pseudoscorpion venom. We examined the antimicrobial, cytotoxic, and insecticidal activity of full-length Checacin

Mosquitoes Larvicidal Activity of Ocimum kilimandscharicum Oil Formulation under Laboratory and Field-Simulated Conditions.

Mosquitoes are vectors of many severe diseases, including malaria, yellow as well as dengue fever, and lymphatic filariasis. The use of synthetic chemical insecticides for mosquito control has been associated with resistance development and detrimental human, and ecological effects. For a safer alternative, the emulsified Ocimum kilimandscharicum oil formulation was evaluated for its larvicidal activity. The oil was analyzed by GC and GC/MS. The formulations were evaluated against third instar mosquito larvae in the laboratory and later compared with Bacillus thuringiensis subsp. israelensis a

Antioxidant enzymes and their contributions to biological control potential of fungal insect pathogens.

Filamentous fungal insect pathogens represent a source of biological insecticides and acaricides formulated using intact cells, such as conidia or other spores. These mycoinsecticides infect arthropod pests through cuticular penetration. In field application, formulated fungal cells are exposed to environmental stresses, including solar UV irradiation, high temperature, and applied chemical herbicides and fungicides, as well as stress from host immune defenses. These stresses often result in accumulation of toxic reactive oxygen species (ROS), generating oxidative stress to the fungal cells an

A paratransgenic strategy to block transmission of Xylella fastidiosa from the glassy-winged sharpshooter Homalodisca vitripennis.

BACKGROUND: Arthropod-borne diseases remain a leading cause of human morbidity and mortality and exact an enormous toll on global agriculture. The practice of insecticide-based control is fraught with issues of excessive cost, human and environmental toxicity, unwanted impact on beneficial insects and selection of resistant insects. Efforts to modulate insects to eliminate pathogen transmission have gained some traction and remain future options for disease control. RESULTS: Here, we report a paratransgenic strategy that targets transmission of Xylella fastidiosa, a leading bacterial pathogen
