Acetylcholinesterase Inhibitory and Antioxidant Activity of the Compounds Isolated from Vanda roxburghii.

Vanda roxburghii has been used in traditional medicine to treat nervous system disorders including Alzheimer's disease (AD). We reported earlier a high acetylcholinesterase inhibitory and antioxidant activity in the chloroform fraction of this plant. Therefore, this study was designed to explore the compounds with acetylcholinesterase inhibitory and antioxidant activities from the chloroform fraction of Vanda roxburghii. Phytochemical investigation led to the isolation for the first time of a fatty acid ester: methyl linoleate (1), and three phenolics: syringaldehyde (2), vanillin (3), and dih

Biological Activity and Chemical Constituents of Essential Oil and Extracts of Murraya microphylla.

Murraya microphylla is the most closely related species to M. koenigii (Curry tree). Inspired by the traditional effects of M koenigii, the antioxidant, anti- inflammatory, and cytotoxic activities of the essential oil and extracts of M. microphylla were evaluated for the first time. The light petroleum and chloroform extracts were found to be able to scavenge DPPH free radicals, inhibit linoleic acid peroxidation, and nitric oxide production, as well as to present cytotoxicity to the human cancer cell lines HepG2, Bel7402, Bel7403, and Hela, but the essential oil only showed moderate activiti

Light exposure at night disrupts host/cancer circadian regulatory dynamics: impact on the Warburg effect, lipid signaling and tumor growth prevention.

The central circadian clock within the suprachiasmatic nucleus (SCN) plays an important role in temporally organizing and coordinating many of the processes governing cancer cell proliferation and tumor growth in synchrony with the daily light/dark cycle which may contribute to endogenous cancer prevention. Bioenergetic substrates and molecular intermediates required for building tumor biomass each day are derived from both aerobic glycolysis (Warburg effect) and lipid metabolism. Using tissue-isolated human breast cancer xenografts grown in nude rats, we determined that circulating systemic f

Ultrasound-responsive and size-transformable self-carrier nanodroplets for augmented deep vein thrombosis therapy.

Deep vein thrombosis (DVT) treatment remains challenging due to the limited efficacy of conventional thrombolytic agents and the short half-lives of anticoagulants. In this study, we develop a low-molecular-weight heparin (LMWH)-functionalized, ultrasound-responsive nanodroplet with tunable size, designate as LMWH-LA@PFP. Our approach incorporates LMWH itself as an intrinsic carrier scaffold, linked to linoleic acid (LA) for self-assembly and perfluoropentane (PFP) encapsulation. This design integrates three synergistic therapeutic functions including targeted delivery, ultrasound-triggered th

[Study of nutritional factors related to breast cancer prevention. Importance of animal models approaches].

Breast cancer is the most common worldwide neoplasia in women. The totality of etiology factors of breast cancer is unknown and thus an effective preventive strategy has not been developed. Risk factors associated to breast cancer can be grouped into three broad categories: a) family history (hereditary) factors, b) endocrine and reproductive factors and c) environmental and life-style factors including diet. Animal models of chemical induced mammary carcinogenesis with 7, 12-dimethylbenz[a]anthracene and N-methyl-N-nitrosourea have been useful in the study of biology, treatment and prevention

Lysophospholipids Contribute to Oxaliplatin-Induced Acute Peripheral Pain.

Oxaliplatin, a platinum-based chemotherapeutic drug, which is used as first-line treatment for some types of colorectal carcinoma, causes peripheral neuropathic pain in patients. In addition, an acute peripheral pain syndrome develop in almost 90% of patients immediately after oxaliplatin treatment, which is poorly understood mechanistically but correlates with incidence and severity of the later-occurring neuropathy. Here we investigated the effects of acute oxaliplatin treatment in a murine model, showing that male and female mice develop mechanical hypersensitivity 24 h after oxaliplatin tr

Integrative analysis of pancreatic microbiota and metabolome in patients with pancreatic ductal adenocarcinoma.

The importance of microbiota-associated metabolites in pancreatic ductal adenocarcinoma (PDAC) tumorigenesis highlights the need to investigate the microbiota and metabolome along the normal-adjacent-carcinoma sequence to explore their mechanistic implications in PDAC tumorigenesis. We characterized the pancreatic microbiota and metabolome in four distinct tissue groups using 16S rRNA amplicon sequencing and untargeted metabolomics: tumor tissues from 23 PDAC patients (PDAC T), their matched para-carcinoma tissues (PCT), tumor tissues from 13 pancreatic benign tumor patients (Control T), and t

Chronic salicylate administration induces transcriptomic and metabolomic remodeling in the rat cochlear nucleus and hippocampus.

Salicylate reliably induces tinnitus, yet its systemic effects on the central auditory and limbic systems remain incompletely characterized. Through integrated transcriptomic and metabolomic profiling of the rat cochlear nucleus and hippocampus, we observed pronounced region-specific remodeling following chronic tinnitus-inducing salicylate treatment. All differential and enrichment analyses were filtered using a nominal p-value cutoff (p < 0.05) without multiple-testing correction; thus, the findings should be interpreted as exploratory. We identified 150 differentially expressed genes (DEGs)

Soybean oil supplementation of a high-concentrate diet does not affect site and extent of organic matter, starch, neutral detergent fiber, or nitrogen digestion, but influences both ruminal metabolism and intestinal flow of fatty acids in limit-fed lambs.

Our objective was to measure ruminal fermentation characteristics and site and extent of nutrient digestion in sheep limit-fed an 81.6% (DM basis) concentrate diet supplemented with increasing levels of soybean oil. Eight white-faced wether lambs (39.9+/-3.0 kg BW) fitted with ruminal, duodenal, and ileal cannulas were used in a replicated 4 x 4 Latin square experiment. Diets were formulated to contain 15.0% CP (DM basis) and included bromegrass hay (18.4%), cracked corn, soybean oil, corn gluten meal, urea, and limestone. Soybean oil was added to diets at 0, 3.2, 6.3, and 9.4% of dietary DM.

Combined Analysis of Transcriptome and Metabolome of Large-Size Triploid Rainbow Trout in Different Seasons.

Seasonal temperature variation poses a significant physiological challenge for cold-water aquaculture species; however, the molecular mechanisms underlying seasonal adaptation in large-sized triploid rainbow trout (Oncorhynchus mykiss) remain largely unexplored. In this study, we integrated liver transcriptomics and LC-MS-based untargeted metabolomics across four seasons (February, May, August, November) to investigate this phenomenon. Seasonal comparisons revealed pronounced molecular reprogramming, with the comparison between May and August showing the largest transcriptomic shift (13,359 di

Untargeted and targeted metabolomics profiling reveals the underlying pathogenesis and abnormal arachidonic acid metabolism in laying hens with fatty liver hemorrhagic syndrome.

As a metabolic disease, fatty liver hemorrhagic syndrome (FLHS) has become the major factor responsible for the noninfectious cause of mortality in laying hens, which lead to huge economic losses to poultry industry. However, the pathogenesis of FLHS remains unclear. The aim of present study was to identify novel liver metabolites associated with FLHS. Twenty healthy Chinese commercial Jing Fen laying hens aged 90 d were used in present study. After acclimatization for 2 wk, the hens were divided into 2 treatments (n = 10): control group (normal diet) and FLHS group (high-energy low-protein di

Effects of an oil enriched in gamma linolenic acid on locomotor activity and behaviour in the Morris Maze, following in utero ethanol exposure in rats.

Ethanol is known to decrease the quantity of polyunsaturated fatty acids in brain membranes possibly as the repercussion of an inhibition of delta-5- and delta-6-desaturases. Consequently behavioural changes may occur. Evening Primrose Oil (EPO) which is rich in gamma linolenic acid (10% 8:3 (n-6)) was proposed to try to circumvent these deleterious effects of ethanol. An animal model of the foetal alcohol syndrome was used in which ethanol was administered throughout gestation and into the weaning period, in a milk diet. EPO was administered concurrently with ethanol to establish the effect o

In depth study on the mechanism of Xiao-Chai-Hu-Tang in ameliorating irinotecan-induced intestinal toxicity and enhancing antitumor immunity via intestinal microbiota and fecal metabolomics.

Irinotecan (CPT-11) is a cornerstone chemotherapeutic for advanced colorectal cancer (CRC), but its clinical use is limited by dose-dependent gastrointestinal (GI) toxicity. Xiao-Chai-Hu-Tang (XCHT), a classical traditional Chinese medicine formula, alleviates CPT-11-induced adverse effects, yet its mechanism remains unclear. This study established a CPT-11-induced intestinal toxicity model in CRC rats, using multi-omics analyses (16S rRNA sequencing, fecal metabolomics, SCFA quantification) and an antibiotic depletion model to explore XCHT's mechanism via the gut microbiota-metabolite axis.
