C terminal half fragment (50 kDa) of heavy chain components of Clostridium botulinum type C and D neurotoxins can be used as an effective vaccine.

Recombinant whole heavy chains (H, 100 kDa) and their N-terminal (Hn, 50 kDa) and C-terminal (Hc, 50 kDa) half fragments of Clostridium botulinum type C and D neurotoxins were expressed as glutathione S-transferase (GST) fusion proteins in Escherichia coli. GST eliminated-preparations of H (10 microg), Hn (5 microg), Hc (5 microg), or a mixture of Hn (5 microg) and Hc (5 microg) of types C and D were mixed with an equal volume of adjuvant, and then were twice injected into mice subcutaneously. After immunization, the mice were challenged with up to 10(6) the minimum lethal doses (MLD)/0.5 ml o

A Poly(lactic Acid)-Porphyrin-Cobalt Vaccine Adjuvant System Elicits Protein- or Peptide-Specific Humoral and Cellular Immunity.

There is growing interest in vaccine adjuvant systems that are scalable, adaptable, and capable of stimulating humoral and cellular immune responses with recombinant protein or synthetic peptide immunogens. In this study, we introduced a "plug-and-play" nanoadjuvant system consisting of poly(lactic acid)-porphyrin-cobalt ion co-formulated with immunostimulatory saponin QS-21 (PPCoQ). PPCoQ facilitated subunit vaccine assembly by coordinating antigens' polyhistidine tags with immobilized cobalt (Co2+) within a porphyrin-modified poly(lactic acid). Using the receptor-binding domain (RBD) of seve

Alcaligenes lipid A as a sublingual adjuvant to augment protective immune responses in the respiratory and gastrointestinal tracts.

We previously identified Alcaligenes as symbiotic bacteria residing within Peyer's patches and demonstrated that their primary components, lipopolysaccharides, and their active center, lipid A, are excellent adjuvants for mucosal vaccination. Here, we evaluated the effectiveness of Alcaligenes-derived lipid A as an adjuvant for sublingual immunization, a novel vaccination route. Mice sublingually immunized with Alcaligenes lipid A and ovalbumin (OVA) showed enhanced production of OVA-specific IgA in both the respiratory and gastrointestinal tracts. In addition, increased serum levels of OVA-sp

Generation and Protective Ability of Influenza Virus-Specific Antibody-Dependent Cellular Cytotoxicity in Humans Elicited by Vaccination, Natural Infection, and Experimental Challenge.

BACKGROUND: Nonneutralizing antibodies (Abs) involved in antibody-dependent cellular cytotoxicity (ADCC) may provide some protection from influenza virus infection. The ability of influenza vaccines to induce ADCC-mediating Abs (ADCC-Abs) in adults and children is unclear. METHODS: We quantified ADCC-Abs in serum samples from adults who received a dose of inactivated subunit vaccine (ISV) targeting monovalent 2009 pandemic influenza A(H1N1) virus or live-attenuated influenza vaccine (LAIV) or who had laboratory-confirmed influenza A(H1N1) virus infection. We also measured ADCC-Abs in children

Development of capsular polysaccharide-based glycoconjugates for immunization against melioidosis and glanders.

Burkholderia pseudomallei and Burkholderia mallei, the etiologic agents of melioidosis and glanders, respectively, cause severe disease in humans and animals and are considered potential agents of biological warfare and terrorism. Diagnosis and treatment of infections caused by these pathogens can be challenging and, in the absence of chemotherapeutic intervention, acute disease is frequently fatal. At present, there are no human or veterinary vaccines available for immunization against these emerging/re-emerging infectious diseases. One of the long term objectives of our research, therefore,

TLR4-mediated activation of dendritic cells by the heat shock protein DnaK from Francisella tularensis.

Francisella tularensis is the causative agent of tularemia, a severe, debilitating disease of humans and other mammals. As this microorganism is also classified as a "category-A pathogen" and a potential biowarfare agent, there is a need for an effective vaccine. Several antigens of F. tularensis, including the heat shock protein DnaK, have been proposed for use in a potential subunit vaccine. In this study, we characterized the innate immune response of murine bone marrow-derived dendritic cells (DC) to F. tularensis DnaK. Recombinant DnaK was produced using a bacterial expression system and

Oral Food-Derived Proanthocyanidins as Immunostimulants: Enhanced Vaccine Immunogenicity via Host Immune Modulation in a Murine CSFV Model.

Traditional injectable vaccine adjuvants are limited by local granulomatous reactions, chronic inflammation, and reduced compliance, creating an urgent need for safe, host-directed strategies to enhance vaccine immunogenicity. Proanthocyanidins (PACs)-natural polyphenols abundant in grapes, cocoa, and tea-are food-derived bioactive compounds with antioxidant, anti-inflammatory, and immunomodulatory activities, positioning them as attractive candidates for oral immunostimulants. Identification and immunological characterization of three potential vaccinogens against Cryptosporidium species.

Cryptosporidiosis is a ubiquitous infectious disease, caused by the protozoan parasites Cryptosporidium hominis and Cryptosporidium parvum, leading to acute, persistent, and chronic diarrhea with life-threatening consequences in immunocompromised individuals. In developing countries, cryptosporidiosis in early childhood has been associated with subsequent significant impairment in growth, physical fitness, and intellectual abilities. Currently, vaccines are unavailable and chemotherapeutics are toxic and impractical, and agents for immunoprophylaxis or treatment of cryptosporidiosis are a high

Association between perfluoroalkyl and polyfluoroalkyl substances and response to the COVID-19 vaccine in a paediatric prospective observational cohort in Arizona.

BACKGROUND: Previous research has shown a significant association between increased serum perfluoroalkyl and polyfluoroalkyl substances (PFAS) concentration and reduced antibody response to vaccinations in children, but this association has not yet been studied for COVID-19 vaccinations. We explored this relationship using data from the Arizona Healthcare, Emergency Response and Other Essential Workers Surveillance (AZ-HEROES) Kids study, a prospective cohort that began recruiting participants in July 2021 and followed through April 2023. METHODS: AZ-HEROES Kids participants aged 6 months to

Immunoinformatic design of a multi-epitope vaccine candidate targeting multidrug-resistant Pseudomonas aeruginosa.

BACKGROUND: Pseudomonas aeruginosa, a World Health Organization (WHO) priority pathogen, causes severe multidrug-resistant (MDR) infections in immunocompromised and hospitalized patients, highlighting the critical need for effective vaccines. To this end, we designed a novel multi-epitope vaccine by integrating immunodominant domains from three key virulence proteins (PcrV, OprE, and CupC2) with the cholera toxin B subunit (CTB) as a mucosal adjuvant. METHODS: Immunoinformatics was used to predict epitopes and construct a vaccine with EAAAKEAAAK linkers. In silico assessments included antigen

Comparing adjuvanted H28 and modified vaccinia virus ankara expressingH28 in a mouse and a non-human primate tuberculosis model.

Here we report for the first time on the immunogenicity and protective efficacy of a vaccine strategy involving the adjuvanted fusion protein "H28" (consisting of Ag85B-TB10.4-Rv2660c) and Modified Vaccinia Virus Ankara expressing H28. We show that a heterologous prime-boost regimen involving priming with H28 in a Th1 adjuvant followed by boosting with H28 expressed by MVA (H28/MVA28) induced the highest percentage of IFN-γ expressing T cells, the highest production of IFN-γ per single cell and the highest induction of CD8 T cells compared to either of the vaccines given alone. In contrast, in

An oral rotavirus-vectored vaccine confers protection against Clostridium perfringens and rotavirus.

The segmented genome of live-attenuated rotavirus (RV) vaccines provides an ideal platform for developing multivalent vaccines against enteric pathogens. Here, we engineered the commercial human RV vaccine strain LLR as a vector to deliver Clostridium perfringens alpha-toxin (CPA), a critical virulence factor associated with gastroenteritis. A screen of four 2A peptide sequences identified that porcine teschovirus-1 2A (P2A) preceded by an N-terminal GSG spacer enabled optimal cleavage and antigen expression. In mice, the resulting recombinant virus elicited potent systemic antibody responses

In silico design of a multi-epitope vaccine targeting DENV-1 and DENV-3.

The co-infection of DENV-1 and DENV-3 during endemic outbreaks can be potentially fatal and complicate the diagnostic process. In our study, we have focused on the development of a multiepitope vaccine against DENV-1 and DENV-3 co-infection, utilizing non-structural protein 1 (NS1) and envelope protein (E) as key antigens. B cell and T cell epitopes were predicted for their immunogenicity, antigenicity, and ability to elicit an IFN-γ response. The final construct showed predicted stability (Instability Index: 30.63), antigenicity (0.5509), non-allergenicity, and hydrophilic character (GRAVY: -

Trimeric Class I Viral Fusion Protein Vaccine Immunogens Using a Trimeric Autotransporter in a Killed Whole-Cell Bacteria Vaccine Platform: Applications to HIV MPER.

BACKGROUND: Trimeric envelope-proximal domains in viral class I fusion proteins are conserved targets of broadly neutralizing antibodies (bNAbs), but it has proven difficult to develop vaccines against those targets. The HIV-1 gp41 membrane-proximal external region (MPER) is one such target. Induction of a neutralizing response likely depends on the immunogen having a close-to-native structure. METHODS: Native sequence MPER was displayed on genome-reduced bacteria as a coiled-coil homotrimer using a Haemophilus influenzae Hia trimeric autotransporter. Vaccine designs incorporated additional f

Optimization and production of a GLP-grade recombinant Sm29 vaccine against schistosomiasis: From expression to preclinical assessment.

Schistosomiasis remains a major neglected tropical disease, and the development of a safe, effective vaccine is a global priority. Sm29, a Schistosoma mansoni surface antigen associated with naturally acquired resistance, has emerged as a promising vaccine candidate; however, its translational advancement requires both high-quality antigen production and evaluation with clinically acceptable adjuvants. Here, we report a Good Laboratory Practice-aligned process for generating tag-free recombinant Sm29 in Escherichia coli, including multi-step anion-exchange chromatography followed by subsequent

Diagnostic potential of recombinant Mycobacterium tuberculosis PcaA antigen and its enhancement of protective efficacy as a subunit vaccine booster following BCG priming.

Bacillus Calmette-Guérin (BCG) is the only vaccine currently used in clinical practice to prevent tuberculosis (TB), however, it remains limited in preventing latent infection, TB reactivation, and providing comprehensive protection. In this study, the pcaA gene, an antigen associated with persistent infection, was selected, and the recombinant plasmid pET28a-PcaA was successfully constructed for protein expression and purification. The specificity of the antigen was further verified using chemiluminescence, enzyme-linked immunosorbent assay (ELISA), flow cytometry, and mycobacterial growth in

Designing a broad-spectrum multi-epitope vaccine against influenza A and Mycoplasma pneumoniae: an immunoinformatics approach.

INTRODUCTION: Influenza A and Mycoplasma pneumoniae (M. pneumoniae) are common respiratory pathogens, causing severe co-infections in clinical diseases. Current vaccines have failed to provide comprehensive protection against both of the pathogens, highlighting the requirement of integrated solution. METHODS: In this study, a novel dual-target multi-epitope vaccine was developed based on the immunoinformatics method. Based on the consistent sequences, we used the HA and NA proteins of influenza A virus, and the p1, p65, HMW1-3 proteins of M. pneumoniae to determine the immunodominant T- and B
