Characterization of cross-resistance gene optrA-carrying Campylobacter coli.

Campylobacter is a significant foodborne pathogen causing human campylobacteriosis. The optrA gene, which encodes an ABC-F protein, confers cross-resistance to oxazolidinones and phenicols, and recent studies have identified the emergence of optrA in Campylobacter. However, detailed information of optrA- carrying C. coli from food-producing animals and various other sources globally, as analyzed by Whole Genome Sequencing (WGS), remains unknown. In this study, our objective was to explore the distribution and genotypic characteristics of optrA-positive C. coli isolates while also analyzing the

Antibiotic-resistant bacteria in marine productive zones of the eastern Arabian Sea: Implications for human and environmental health.

The increasing threat of antibiotic resistance is a major global concern affecting human and environmental health. Marine environments, though underexplored, are emerging as significant reservoirs for antibiotic resistance genes (ARGs). This study provides genome-resolved shotgun metagenomic insights into the seasonal and spatial dynamics of ARGs in the chlorophyll maximum zones of the eastern Arabian Sea, focusing on bacterial communities from coastal (30 m) and offshore (600 m) depths. Using a shotgun metagenomic approach, 31 potential ARGs were identified across both non-monsoon and monsoon

Zoonotic potential of ESBL-producing coliforms in pastorally managed ruminants with subclinical mastitis in Plateau State, Nigeria.

BACKGROUND: Environmental coliform bacteria are frequently the cause of subclinical mastitis (SCM), a serious health issue in the dairy industry. Extended-spectrum β-lactamase (ESBL)-producing coliforms in livestock are a serious public health concern, particularly in environments where people and animals coexist. With an emphasis on their zoonotic and One Health implications, this study sought to evaluate the incidence of SCM and the occurrence of ESBL-producing coliforms in ruminants in Plateau State, Nigeria. METHODS: The California Mastitis Test (CMT) was used to screen 287 milk samples t

Potential of ZnO nanoparticles for multi-drug resistant Escherichia coli having CRISPR-Cas from poultry market in Lahore.

BACKGROUND AND OBJECTIVES: Apart from known factors such as irrational use of antibiotics and horizontal gene transfer, it is now reported that clustered regularly interspaced short palindromic repeats (CRISPR) are also associated with increased antimicrobial resistance. Hence, it is critical to explore alternatives to antibiotics to control economic losses. Therefore, the present study aimed to determine not only the association of CRISPR-Cas system with antibiotic resistance but also the potential of Zinc Oxide nanoparticles (ZnO-NPs) for avian pathogenic Escherichia coli (APEC) isolated fro

Diffusion and enrichment of high-risk antibiotic resistance genes (ARGs) via the transmission chain (mulberry leave, guts and feces of silkworm, and soil) in an ecological restoration area of manganese mining, China: Role of heavy metals.

This study investigated the diffusion and enrichment of antibiotic resistance genes (ARGs) and pathogens via the transmission chain (mulberry leaves - silkworm guts - silkworm feces - soil) near a manganese mine restoration area (RA) and control area (CA, away from RA). Horizontal gene transfer (HGT) of ARGs was testified by an IncP a-type broad host range plasmid RP4 harboring ARGs (tetA) and conjugative genes (e.g., korB, trbA, and trbB) as an indicator. Compared to leaves, the abundances of ARGs and pathogens in feces after silkworms ingested leaves from RA increased by 10.8% and 52.3%, res

Genome data vs MLST for exploring intraspecific evolutionary history in bacteria: Much is not always better.

Genome-based phylogeny has been proposed to be more accurate than phylogeny based in a few genes as MLST-based phylogeny. However, much is not always better. Here we analyzed 368 complete genomes corresponding to 9 bacterial species in order to address intraspecific phylogeny. The studied species were: Burkholderia pseudomallei, Campylobacter jejuni, Chlamydia trachomatis, Helicobacter pylori, Klebsiella pneumoniae, Listeria monocytogenes, Salmonella enterica, Staphylococcus aureus and Streptococcus pyogenes. The intra-specific phylogenies were inferred using the complete genome sequences of d

Comparative genomics and transduction potential of Enterococcus faecalis temperate bacteriophages.

To determine the relative importance of temperate bacteriophage in the horizontal gene transfer of fitness and virulence determinants of Enterococcus faecalis, a panel of 47 bacteremia isolates were treated with the inducing agents mitomycin C, norfloxacin, and UV radiation. Thirty-four phages were purified from culture supernatants and discriminated using pulsed-field gel electrophoresis (PFGE) and restriction mapping. From these analyses the genomes of eight representative phages were pyrosequenced, revealing four distinct groups of phages. Three groups of phages, PhiFL1 to 3, were found to

Mobile Colistin Resistance (mcr) Gene-Containing Organisms in Poultry Sector in Low- and Middle-Income Countries: Epidemiology, Characteristics, and One Health Control Strategies.

Mobile colistin resistance (mcr) genes (mcr-1 to mcr-10) are plasmid-encoded genes that threaten the clinical utility of colistin (COL), one of the highest-priority critically important antibiotics (HP-CIAs) used to treat infections caused by multidrug-resistant and extensively drug-resistant bacteria in humans and animals. For more than six decades, COL has been used largely unregulated in the poultry sector in low- and middle-income countries (LMICs), and this has led to the development/spread of mcr gene-containing bacteria (MGCB). The prevalence rates of mcr-positive organisms from the pou

Enrofloxacin metabolism and antibiotic resistance in Monopterus albus differs between pond cage and greenhouse micro-flow aquaculture systems.

Enrofloxacin (ENR) is a fluoroquinolone antibiotic widely used in aquaculture, yet its metabolic fate and resistance selection dynamics remain poorly characterized. Here, we investigated ENR metabolism, tissue distribution, withdrawal periods, gut microbial communities, and antibiotic resistance genes (ARGs) in Monopterus albus reared in pond net cage (PNC) and greenhouse micro-flow (GMF) systems following a 5-day medicated feed treatment (20 mg ENR/kg body weight). Eight ENR metabolites were tentatively identified, with ciprofloxacin (CIP) as the predominant metabolite. The GMF system signifi

Mesorhizobium bavaricum sp. nov. and Mesorhizobium monacense sp. nov., two novel Lotus-associated species harbouring symbiotic plasmids.

Legumes establish a mutualistic interaction with nitrogen-fixing rhizobia. Lotus japonicus is a model for studying this symbiosis; however, only a limited number of rhizobial species nodulating this host have been taxonomically described. Here, we characterise four Mesorhizobium strains (DC-1.1T, Qj1B1, DC-1.5T, and Qj2B2) isolated from root nodules of Lotus japonicus and Lotus burttii. Multi-locus phylogeny and phylogenomic analyses resolved these isolates into two well-supported monophyletic clades. Genome-based comparisons supported their classification as distinct taxa, with strains DC-1.1

Environmental occurrence, ecotoxicity, and management perspectives of β-lactam antibiotics.

β-Lactams are the most widely prescribed antibiotic class globally, comprising penicillins, cephalosporins, carbapenems, and monobactams. Their continual release to the environment raises concerns about both direct ecotoxicity and potential indirect effects on ecosystem function. This review highlights three key findings; 1) environmental concentrations can exceed predicted no-effect concentrations for resistance selection; for example ceftriaxone had been reported 6160 µg/L in wastewater influent and 4150 µg/L in effluent in India, indicating that many β-lactams have removal efficiency below

Interactions of antiphage defense systems in the ESKAPE pathogen plasmids.

BACKGROUND: The global rise of multidrug resistant (MDR) ESKAPE pathogens represents a serious threat to antimicrobial therapy. While phage therapy has re-emerged as a promising alternative, its effectiveness may be compromised by bacterial defense systems, particularly those encoded on plasmids. Comprehensive surveillance of the distribution, diversity, and mobilome context of plasmid-encoded defense systems in ESKAPE pathogens remains key to the design of effective phage therapies. RESULTS: We analyzed 7,330 dereplicated plasmids from ESKAPE pathogens to characterize the prevalence, diversi

Phylogenetic analysis and characterization of hybrid enteroaggregative/uropathogenic Escherichia coli strains isolated from urinary tract infection.

BACKGROUND: Hybrid EAEC/UPEC Escherichia coli strains are emerging uropathogens that combine intestinal and extraintestinal virulence traits through horizontal gene transfer. Although relatively uncommon, these hybrids exhibit strong biofilm formation, enhanced epithelial adherence, and notable antimicrobial resistance, underscoring the need for detailed molecular characterization and epidemiological assessment. The present study aimed to determine the frequency of hybrid EAEC/UPEC isolates among E. coli recovered from patients with urinary tract infections and to characterize their lineage us

Characterization of the novel transposon Tn7722 harboring blaNDM-1: Insights into the evolutionary dynamics of resistance in Klebsiella pneumoniae.

BACKGROUND: Klebsiella pneumoniae is an opportunistic pathogen responsible for invasive infections. The rise of carbapenem-resistant K. pneumoniae, largely driven by acquisition of blaNDM genes, represents a global health threat. In French Polynesia, sporadic cases of NDM-producing Enterobacteriales have been reported. This study characterizes the genomic features of NDM-producing K. pneumoniae isolates from French Polynesia and evaluates the roles of clonal expansion and mobile genetic element (MGE)-mediated horizontal gene transfer in blaNDM dissemination. Role of Mobilome in Carbapenem Resistance.

Growing resistance to carbapenem antibiotics is a major public health problem as these antibiotics are considered the last line of therapy for infections caused by multidrug-resistant (MDR) Gram-negative bacteria. The rapid emergence and dissemination of carbapenem-resistant bacterial strains are mainly due to horizontal gene transfer (HGT) within or between bacterial cells via the mobilome. The aim of this article is to discuss the role of mobile genetic elements (MGEs) that capture and disseminate resistance determinants of carbapenem antibiotics, as a comprehensive review integrating the co
