Vesicle-like nanoparticles extracted from Pueraria lobata decoction alleviate colitis by modulating the intestinal microbiota.

Aim: This study aimed to investigate whether vesicle-like nanoparticles derived from the GeGen decoction (GGD-PDVLNs) represent a key bioactive component responsible for its anti-colitis effects and to elucidate their underlying mechanisms, particularly focusing on gut microbiota modulation. Methods: The GeGen decoction (GGD) was subjected to differential centrifugation following boiling, yielding vesicle-like nanoparticles. Structural analysis confirmed that these nanoparticles have a lipid bilayer and can resist digestion by simulated gastrointestinal fluids. These nanoparticles were adminis

Multi-wavelength photo-activation of transmembrane chloride transport in distinct vesicle populations using photocaged indolocarbazole anionophores.

Synthetic ionophores enable the study of transmembrane ion transport and show promise as therapeutics or for engineering responsive synthetic cells. Incorporation of photolabile protecting groups (photocages) allows for OFF-ON control over transport to spatiotemporally regulate activity; however, systems incorporating two different photocages such that two wavelengths of irradiation are required for activation are unknown. Here, we report a family of photocaged indolocarbazole-based anionophores that enable multi-wavelength photo-activation of chloride transport in lipid bilayer vesicles. Mono

Interactions of Glucagon-Like Peptide-1 Receptor Agonists With GLP-1R-Gs Complex: Insights From Gaussian-Accelerated Molecular Dynamics.

The glucagon-like peptide-1 receptor (GLP-1R) is a critical therapeutic target for metabolic diseases. GLP-1 receptor agonists (GLP-1RAs) have been developed to treat type 2 diabetes and promote weight loss. In this study, Gaussian accelerated molecular dynamics (GaMD) simulations, MM-PBSA calculations, and conformational dynamic analyses were systematically employed to elucidate the binding mechanisms and dynamic stability of three representative agonists-native GLP-1, semaglutide, and tirzepatide-bound to the GLP-1R-Gs membrane protein complex. Molecular docking and trajectory analyses revea

Controlling membrane tension by dynamic DNA nanorings.

Membrane tension, defined as the energy cost to expand the membrane surface area, is a fundamental mechanical property of the lipid bilayer and an important modulator of various cellular processes. Liposomes provide model systems to study protein-mediated membrane dynamics, where membrane tension can be controlled by osmotic pressure and micropipette aspiration. However, it is challenging to individually control membrane tension of nanometer-sized liposomes, such as small unilamellar vesicles (SUVs), that are widely used for modeling subcellular membrane structures and drug delivery. To bridge

Lauric Acid Microemulsions Inhibit Staphylococcus aureus Through Cell Membrane Disruption and Potential Interference with Peptidoglycan Biosynthesis.

Staphylococcus aureus (S. aureus) is a prominent foodborne pathogen that poses a continuous threat to global public health and food safety due to its possession of a variety of toxins and its multidrug resistance. Medium-chain fatty acids (MCFAs), notably lauric acid (LA), exhibit strong antimicrobial properties, but their application is heavily constrained by poor water solubility. In this study, optimized LA emulsions stabilized by chitosan (CS) and polyvinyl alcohol (PVA) were evaluated for their antibacterial activity and detailed mode of action against S. aureus ATCC 6538. The antibacteri

Natural Product-Derived Vesicles in Nanotherapeutics.

Natural product-derived vesicles (NPVs) are lipid bilayer-enclosed nanovesicles isolated from diverse sources, including plants, fungi, and marine organisms, and are increasingly recognized as an emerging class of extracellular vesicles with potential therapeutic relevance. Their abundance, renewability, and capacity to transport endogenous metabolites and exogenous cargoes highlight their potential as scalable biological delivery vehicles. NPVs can interact with mammalian cells, participate in intercellular communication, and influence conserved biological pathways that modulate disease, supp

Self-protected circular DNAzyme for integrated enrichment and quantification of small extracellular vesicles.

Small extracellular vesicles (sEVs) hold immense potential for liquid biopsy given the wealth of biological information they carry. Currently, the clinical application of these methods is limited due to their low abundance and the complexities associated with traditional isolation techniques. To address this, we developed a strategy integrating cholesterol-mediated capture with a Self-Protected DNAzyme Walker for the rapid and simultaneous specific isolation and quantification of small extracellular vesicles (sEVs). Upon specific binding to CD63, the blocker strand is released, which activates

Design and evaluation of a novel peptide-EV complex for targeted Alzheimer's disease therapy.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder that involves the formation of amyloid-β (Aβ) aggregates, and the development of targeted therapeutic strategies is needed. In the current work, we report the rational design of H102-CP05, a 22-residue chimeric peptide that integrates the β-sheet breaker peptide H102 with the CD63-targeting anchor CP05 to enable extracellular vesicle (EV)-mediated delivery of an Aβ-inhibitory payload. Computational analysis confirmed favourable physicochemical properties and a non-allergenic profile. In silico immunogenicity assessment and C-

In vitro reconstitution implicates multiple OMP assembly factors in CdiAEC93 toxin delivery.

Contact-dependent inhibition is a mechanism of interbacterial competition in gram-negative bacteria. A large cell surface protein called CdiA is the key component of CDI systems; it inhibits the growth of neighboring, nonsibling bacteria by delivering a protein toxin through their cell envelopes. Here, we describe an in vitro reconstitution of the initial steps of the toxin delivery process, which normally occur in the outer membrane of a target cell. We show that a large, purified fragment of a CdiA protein from Escherichia coli EC93 spontaneously assembles into proteoliposomes containing the

Structural basis of active state coupling of metabotropic glutamate receptor 8 to beta-arrestins.

Metabotropic glutamate receptors (mGluRs) are prototypical, dimeric family C G protein-coupled receptors (GPCR) that perform crucial modulatory roles throughout the nervous system. While mGluR activation and signaling through G proteins has been studied extensively, how these receptors interact with and are desensitized by β-arrestins (β-arrs) is not well understood. Here, we use an integrative biophysical and structural approach to probe the coupling of mGluR8 and β-arrs. Using negative stain electron microscopy (EM), we identify tail- and core-bound orientations and stoichiometries of mGluR8

Exosomal lncRNAs in Cerebrovascular Diseases: Biomarkers, Pathological Mechanisms, and Therapeutic Potential.

Background: Cerebrovascular diseases have complex pathogenesis and pose a serious threat to human health; thus, novel diagnostic and therapeutic strategies are needed. Small extracellular vesicles (sEVs), commonly referred to as exosomes, are 30-150 nm lipid-bilayer vesicles that shield long noncoding RNAs (lncRNAs) from degradation. Because exosomal lncRNAs are more stable than free lncRNAs in blood and cerebrospinal fluid and can cross the blood-brain barrier, they are promising as biomarkers and therapeutic vectors. This review summarizes the roles and mechanisms of exosomal lncRNAs in cere

Neurovascular Impairment in Type 2 Diabetes Mellitus: The Role of Adipocyte-Derived Exosomes.

Type 2 diabetes mellitus (T2DM) is a major metabolic disorder characterized by chronic hyperglycemia with far-reaching morbidities. Among these, diabetes-related cerebrovascular complications such as ischemic and hemorrhagic stroke, cerebral blood vessel disease, and vascular dementia are significant contributors to morbidity and mortality. Adipose tissue is a metabolically active endocrine organ that becomes dysfunctional in T2DM and communicates with distant tissues via secreted factors, including extracellular vesicles such as exosomes (EXs), phospholipid bilayer-enclosed nanosized particle

Knifing Action of WS2 Nanosheets Toward the Inner Membrane of Escherichia coli Compared to Its Mild and Conditional Detrimental Effect on the Bacterial Membrane of S. aureus.

The global rise in mortality rates due to drug-resistant infections necessitates the development of medicinal alternatives, with membrane-targeting antibacterial nanomaterials representing a promising strategy due to their inherently low propensity for resistance development. Notably, two-dimensional transition-metal dichalcogenides (TMDs), such as tungsten disulfide (WS2), have emerged as promising broad-spectrum, membrane-mediated antibacterial agents with favorable biocompatibility. Herein, we investigate the antibacterial mode of action of WS2 nanosheets against two popular pathogenic bact

Primary amine-functionalized radially amphiphilic polypeptides target bacterial phospholipids in polyanionic matrices for biofilm therapy.

Bacterial biofilm infections, a key contributor to antibiotic resistance, pose a critical global health challenge. Although antimicrobial peptides are promising candidates, their cationic amphipathic structures often lead to nonspecific sequestration by polyanionic biofilm matrix components. Here, we report a class of primary amine-functionalized radially amphiphilic antimicrobial polypeptides (paRAPs) that achieve potent antibiofilm activity by selectively targeting bacterial phosphatidylglycerol (PG) in polyanionic biofilm matrices. Simulation studies support a mechanism of PG-responsive str

Extracellular vesicles and the lung: from disease pathogenesis to biomarkers and treatments.

Nanosized extracellular vesicles (EVs) are released by all cells to convey cell-to-cell communication. EVs, including exosomes and microvesicles, carry an array of bioactive molecules, such as proteins and RNAs, encapsulated by a membrane lipid bilayer. Epithelial cells, endothelial cells, and various immune cells in the lung contribute to the pool of EVs in the lung microenvironment and carry molecules reflecting their cellular origin. EVs can maintain lung health by regulating immune responses, inducing tissue repair, and maintaining lung homeostasis. They can be detected in lung tissues and

Machine Learning-Driven Discovery of Structurally Related Natural Products as Activators of the Cardiac Calcium Pump SERCA2a.

A key molecular dysfunction in heart failure is the reduced activity of the cardiac sarcoplasmic reticulum Ca2+-ATPase (SERCA2a) in cardiac muscle cells. Reactivating SERCA2a improves cardiac function in heart failure models, making it a validated target and an attractive therapeutic approach for heart failure therapy. However, finding small-molecule SERCA2a activators is challenging. In this study, we used a machine learning-based virtual screening to identify SERCA2a activators among 57,423 natural products. The machine learning model identified ten structurally related natural products from

Extracellular Vesicles in Periodontitis: Pathogenic Mechanisms and Therapeutic Potential.

Periodontitis is a prevalent yet frequently overlooked oral disease that is linked to a range of systemic conditions. Although basic treatment and periodontal surgery can alleviate the symptoms of periodontitis to a certain extent, the treatment of severe tissue defects or refractory cases is not effective. Extracellular vesicles (EVs) are subcellular lipid bilayer particles that come from a variety of sources and are prevalent in the biological fluids of vertebrates. They play a key role in intercellular communication by transporting multiple signaling molecules. Recent research has indicated

Design, synthesis, and evaluation of phosphorylated rhein derivatives with enhanced antiproliferative activity.

Rhein, a natural anthraquinone, possesses anticancer properties but is hampered by modest potency and poor bioavailability. To overcome these limitations, we synthesized a series of novel phosphonate derivatives of rhein and evaluated their antiproliferative activity across several cancer cell lines. The derivative C9 emerged as the most potent compound against human hepatocellular carcinoma (HepG2) cells, with an IC50 value of 11.88 ± 0.93 μM, representing a 6.5-fold enhancement over the parent compound while maintaining a favorable selectivity index (SI = 8.0) against normal hepatocytes. pH-Sensitive Multiliposomal Containers for Encapsulation and Rapid Release of Bioactive Substances.

A new method of the design of stimuli-sensitive multiliposomal containers for encapsulation and controlled drug release is described. Despite quite a wide choice of pH-sensitive containers, there is still a considerable challenge to synthesize those that respond quickly to small variations in pH and release most of the encapsulated drug in a short time. The suggested AMS-containing multiliposomal complexes demonstrated an excellent rate of encapsulated substance release under altering the pH of the outer solution. To improve the efficiency of the delivery of bioactive compounds to target cells
