In silico design of a multi-epitope vaccine against Cryptosporidium parvum using structural and immunoinformatics approaches.

BACKGROUND: Cryptosporidium parvum is a waterborne protozoan parasite responsible for diarrheal illness in humans and animals. The lack of effective vaccines and the emergence of antimicrobial resistance underscore the urgent need for novel prophylactic strategies. METHODS: A structure-based immunoinformatics approach was used to design a multi-epitope subunit vaccine (MESV) targeting immunogenic regions of C. parvum. Three proteins, Cp15, Cp23, and CpP2, were selected based on antigenicity, non-allergenicity, non-homology with host proteins, and absence of transmembrane domains. B-cell, CD4+

In vivo immunogenicity assessment and vaccine efficacy evaluation of a chimeric tandem repeat of epitopic region of OMP31 antigen fused to interleukin 2 (IL-2) against Brucella melitensis in BALB/c mice.

BACKGROUND: Designing a potent recombinant vaccine, using the appropriate subunits with the greatest effect on stimulating the immune system, especially in the case of intracellular pathogens such as gram negative Brucella Melitensis bacteria, is of great importance. In this study, three repeats of 27 amino acids of the immunogenic epitope derived from OMP31 antigen (3E) from the Brucella melitensis, in a protective manner against Brucellosis have been used. To fortify the delivery system of recombinant antigens, IL-2 cytokine as a molecular adjuvant was fused to recombinant constructs. Recomb

Lassa virus-like particles displaying all major immunological determinants as a vaccine candidate for Lassa hemorrhagic fever.

BACKGROUND: Lassa fever is a neglected tropical disease with significant impact on the health care system, society, and economy of Western and Central African nations where it is endemic. Treatment of acute Lassa fever infections has successfully utilized intravenous administration of ribavirin, a nucleotide analogue drug, but this is not an approved use; efficacy of oral administration has not been demonstrated. To date, several potential new vaccine platforms have been explored, but none have progressed toward clinical trials and commercialization. Therefore, the development of a robust vacc

Role for the αV Integrin Subunit in Varicella-Zoster Virus-Mediated Fusion and Infection.

UNLABELLED: Varicella-zoster virus (VZV) is an alphaherpesvirus that causes varicella and herpes zoster. Membrane fusion is essential for VZV entry and the distinctive syncytium formation in VZV-infected skin and neuronal tissue. Herpesvirus fusion is mediated by a complex of glycoproteins gB and gH-gL, which are necessary and sufficient for VZV to induce membrane fusion. However, the cellular requirements of fusion are poorly understood. Integrins have been implicated to facilitate entry of several human herpesviruses, but their role in VZV entry has not yet been explored. To determine the in

B subunit of cholera toxin fused with VP7 from GCRV (grass carp reovirus) was expressed in E. coli and folds into an active protein.

Grass carp reovirus (GCRV) is one of the most serious pathogens threatening grass carp (Ctenopharyngodon idella) production and results in high mortality in China. To obtain a genetically engineered oral vaccine against GCRV, the cholera toxin B subunit (CTB) of Vibrio cholerae was fused to VP7 (CTB-VP7) and transformed into BL21(DE3) for expression. SDS-PAGE and Western blotting showed that the purified rCTB-VP7 fusion protein (rCTB-VP7) was approximately 49.0 kDa. The monomeric nature of rCTB-VP7 through multistage purification showed a binding affinity for GM1, a receptor for biologically a

Immunotargeting of Antigen xCT Attenuates Stem-like Cell Behavior and Metastatic Progression in Breast Cancer.

Resistance to therapy and lack of curative treatments for metastatic breast cancer suggest that current therapies may be missing the subpopulation of chemoresistant and radioresistant cancer stem cells (CSC). The ultimate success of any treatment may well rest on CSC eradication, but specific anti-CSC therapies are still limited. A comparison of the transcriptional profiles of murine Her2(+) breast tumor TUBO cells and their derived CSC-enriched tumorspheres has identified xCT, the functional subunit of the cystine/glutamate antiporter system xc(-), as a surface protein that is upregulated spe

A one dose experimental cholera vaccine.

The number of cholera vaccine doses required for immunity is a constraint during epidemic cholera. Protective immunity following one dose of multiple Vibrio cholerae (Vc) colonization factors (Inaba LPS El Tor, TcpA, TcpF, and CBP-A) has not been directly tested even though individual Vc colonization factors are the protective antigens. Inaba LPS consistently induced vibriocidal and protective antibodies at low doses. A LPS booster, regardless of dose, induced highly protective secondary sera. Vc protein immunogens emulsified in adjuvant were variably immunogenic. CBP-A was proficient at induc

The Shiga toxin B-subunit targets antigen in vivo to dendritic cells and elicits anti-tumor immunity.

The non-toxic B-subunit of Shiga toxin (STxB) interacts with the glycolipid Gb3, which is preferentially expressed on dendritic cells (DC) and B cells. After administration of STxB chemically coupled to OVA (STxB-OVA) in mice, we showed that the immunodominant OVA(257-264) peptide restricted by K(b) molecules is specifically presented by CD11c+ CD8alpha- DC, some of them displaying a mature phenotype. Using mice carrying a transgene encoding a diphtheria toxin receptor (DTR) under the control of the murine CD11c promoter, which allows inducible ablation of DC, we showed that DC are required fo

Contriving multiepitope vaccine for breast cancer by targeting the estrogen receptor using in-silico method.

Breast cancer is an invasive cancer that arises either in ductal epithelial cells or lobules of the glandular tissues of the breast. The main culprit behind this misery in most cases is Estrogen receptor which allows binding of estrogen protein that helps in the proliferation, differentiation, and metastasis of breast cells. The present study designed multi-epitope B-cell stimulating vaccine by using estrogen receptor alpha as a template for epitope prediction. The predicted epitopes were checked for antigenicity, and toxicity using Vaxijen, Aller-Pred and Toxin-Pred. The final vaccine constru

Safety and immunogenicity of intramuscularly administered CS6 subunit vaccine with a modified heat-labile enterotoxin from enterotoxigenic Escherichia coli.

INTRODUCTION: Enterotoxigenic Escherichia coli (ETEC) is a common cause of infectious diarrhoea and a leading cause of morbidity and mortality in children living in resource-limited settings. It is also the leading cause of travellers' diarrhoea among civilian and military travellers. Its dual importance in global public health and travel medicine highlights the need for an effective vaccine. ETEC express colonization factors (CFs) that mediate adherence to the small intestine. An epidemiologically prevalent CF is coli surface antigen 6 (CS6). We assessed the safety and immunogenicity of a CS6

Interventions against West Nile virus, Rift Valley fever virus, and Crimean-Congo hemorrhagic fever virus: where are we?

ARBO-ZOONET is an international network financed by the European Commission's seventh framework program. The major goal of this initiative is capacity building for the control of emerging viral vector-borne zoonotic diseases, with a clear focus on West Nile virus, Rift Valley fever virus, and Crimean-Congo hemorrhagic fever virus. To evaluate the status quo of control measures against these viruses, an ARBO-ZOONET meeting was held in Istanbul, Turkey, from 19 to 20 November 2009. The symposium consisted of three themes: (1) vaccines: new and existing ones; (2) antivirals: existing and new deve

Immunogenicity, Efficacy, and Safety of a Novel Synthetic Microparticle Pre-Erythrocytic Malaria Vaccine in Multiple Host Species.

We previously reported a protective antibody response in mice immunized with synthetic microparticle vaccines made using layer-by-layer fabrication (LbL-MP) and containing the conserved T1BT* epitopes from the P. falciparum circumsporozoite protein. To further optimize the vaccine candidate, a benchtop tangential flow filtration method (LbL-by-TFF) was developed and utilized to produce vaccine candidates that differed in the status of base layer crosslinking, inclusion of a TLR2 ligand in the antigenic peptide, and substitution of serine or alanine for an unpaired cysteine residue in the T* ep

A phase 1 randomized controlled trial to evaluate the safety and immunogenicity of a HIV monomeric gp120 protein B-cell lineage targeting HIV vaccine in healthy adults.

BACKGROUND: The isolation of many HIV broadly neutralizing antibodies (bnAbs) from people living with HIV (PLWH) and rigorous characterization of their ontogeny has promoted the goal of reverse engineering their natural development as a strategy for achieving an effective preventive HIV vaccine. We previously described the developmental process of CH103, a CD4-binding site (CD4bs)-specific monoclonal antibody, and the associated evolution of HIV Envelopes (Envs) within the person (CH505) from whom it was isolated. A series of monomeric gp120 protein subunit immunogens representing the transmit

Non-Coding DNA-Derived Mimotopes of Aβ₄₂ as Novel Candidates for Alzheimer's Peptide Vaccine Design.

Alzheimer's disease (AD), the leading cause of dementia, is a progressive neurodegenerative disorder marked by memory loss, cognitive decline, and characteristic neuropathology involving amyloid-β (Aβ) plaques and tau tangles. Among emerging therapeutic strategies, Aβ-targeted immunotherapy using monoclonal antibodies or peptide vaccines offers the most promising disease-modifying potential. Mimotopes, short peptides that mimic antigenic epitopes of Aβ, have recently gained attention as safe and effective candidates for vaccine development. In this study, we employed a computational immunoinfo

Bioinformatics and immunoinformatic approaches for structural modeling and vaccine design of the CCHFV whole genome to develop a novel broad-spectrum multiepitope vaccine candidate.

Crimean-Congo hemorrhagic fever virus (CCHFV), the causative agent of Crimean-Congo hemorrhagic fever (CCHF), represents the most widely distributed tick-borne viral disease in humans and is currently managed only through supportive care. Consequently, considerable efforts have been directed toward the development of an effective vaccine to control this pathogen. Here, we report the first comprehensive in silico prediction of immunogenic CD8⁺ T cells, CD4⁺ T cells, and B-cell epitopes across the S, M, and L segments of CCHFV. Candidate epitopes were evaluated for conservation in mouse and huma

Phage-Mediated Presentation of a Conserved HA2 Epitope From Influenza A Virus Elicits Significant IgY Antibody Responses in Broiler Chickens.

Peptides alone often exhibit limited immunogenicity, necessitating the development of effective antigen delivery systems to facilitate recognition and presentation, ultimately eliciting robust immune responses and activating T and B lymphocytes. Filamentous bacteriophages, such as M13, are recognized as efficient platforms for peptide expression and presentation via their capsid surfaces. The conserved amino acid sequence HA2 1-9 (GLFGAIAGF) from the haemagglutinin (HA) transmembrane protein of Influenza A virus (IAV) has been identified as a promising immunogen for eliciting broad-spectrum im

Oral immunization of Nile tilapia with Chlorella-bioencapsulated multi-epitope fusion antigen effectively protects against Streptococcus agalactiae infection.

Streptococcus agalactiae is an important bacterial pathogen of farmed tilapia; however, practical oral vaccines with stable protective efficacy remain limited because of poor antigen stability and low intestinal delivery efficiency. In this study, three immunodominant epitopes derived from the S. agalactiae surface immunogenic protein Sip, C5a peptidase ScpB, and leucine-rich repeat protein LrrG, as well as a multi-epitope fusion antigen (MEFA) constructed from these epitopes, were individually expressed in Chlorella sorokiniana, thereby establishing a microalgae-based oral vaccine system.
