Cardiovascular Safety of Ozanimod in Patients With Ulcerative Colitis: True North and Open-Label Extension Analyses.

BACKGROUND & AIMS: Evaluating cardiovascular safety of sphingosine 1-phosphate (S1P) receptor modulators is warranted due to S1P receptor expression on cardiomyocytes and vascular endothelial cells. This analysis reports the cardiovascular safety of ozanimod, an S1P receptor modulator, in patients with moderately to severely active ulcerative colitis from the phase 3 True North (TN) and open-label extension (OLE). METHODS: All patients who received ozanimod in TN (n = 796) and all eligible TN patients who entered the OLE (n = 823) were included. Cardiovascular-related adverse events were eval…

Long non-protein coding RNA DANCR functions as a competing endogenous RNA to regulate osteoarthritis progression via miR-577/SphK2 axis.

Long noncoding RNAs (lncRNAs) have been known to be involved in multiple diverse diseases, including osteoarthritis (OA). This study aimed to explore the role of differentiation antagonizing non-protein coding RNA (DANCR) in OA and identify the potential molecular mechanisms. The expression of DANCR in cartilage samples from patients with OA was detected using quantitative reverse transcription-polymerase chain reaction. The effects of DANCR on the viability of OA chondrocytes and apoptosis were explored using cell counting kit 8 assay and flow cytometry assay, respectively. Additionally, the

Functional and structural alterations in the endoplasmic reticulum and mitochondria during apoptosis triggered by C2-ceramide and CD95/APO-1/FAS receptor stimulation.

C(2)-ceramide (C(2)-cer) and binding of the CD95/APO-1/FAS (aCD95) receptor are acknowledged inducers of apoptosis. In spite of that, their effects on the endoplasmic reticulum (ER) and mitochondria during early phases of apoptotic onset are poorly characterized. Here, by employing various approaches, we followed structural and functional modifications of these organelles at the beginning of cellular demise. In detail, we observed that C(2)-cer, but not CD95 activation, markedly modifies the morphology of the ER and promotes Ca(2+) release. Accordingly, mitochondria of C(2)-cer-treated, but no

Alteration of the 4-sphingenine scaffolds of ceramides in keratinocyte-specific Arnt-deficient mice affects skin barrier function.

Aryl hydrocarbon receptor nuclear translocator (ARNT), a transcription factor of the Per/AHR/ARNT/Sim family, regulates gene expression in response to environmental stimuli including xenobiotics and hypoxia. To examine its role in the epidermis, the Cre-loxP system was used to disrupt the Arnt gene in a keratinocyte-specific manner. Gene-targeted, newborn mice with almost normal appearance died neonatally of severe dehydration caused by water loss. Histology showed small changes in the architecture of cornified layers, with apparently preserved intercorneocyte lamellar structures responsible f

Radio-sensitive murine thymoma cell line 3SB: characterization of its apoptosis-resistant variants induced by repeated X-irradiation.

3SB, a mouse thymoma cell line, is one of the most radio-sensitive cells (D0 = 0.3 Gy), and its rapid apoptosis (4 h after 5 Gy irradiation, 90% apoptosis) seems to play a decisive role in enhancing the radiosensitivity. To understand the molecular mechanisms underlying extremely high radiosensitivity and rapid apoptosis, we attempted to isolate X-ray-resistant (XR) variants from 3SBH5, a stable subclone of 3SB, by repeating exposure of the cells to 2-5 Gy X-rays. Four independent stable XR variants, R111, R223, R316 and R429, were isolated by the repeated irradiation protocols. All XR cells p

Endothelial cell-specific TGM2 deficiency promotes progression of temporal lobe epilepsy by disrupting S1P metabolism and intercellular communication.

BACKGROUND: Temporal lobe epilepsy (TLE) is a common and medically refractory neurological disorder with a complex pathogenesis. Blood-brain … reduced the expression of tight junction protein CLDN5. Mechanistically, TGM2 positively regulated sphingosine-1-phosphate (S1P) synthesis. Reduced S1P production abolished the inhibition of RelA nuclear translocation, thereby activating downstream inflammatory genes and triggering endothelial inflammation. Molecular docking confirmed the direct binding betw …

Longitudinal humoral and cellular immune responses to COVID-19 vaccination in persons with multiple sclerosis across disease-modifying therapy classes.

BACKGROUND: Humoral responses to COVID-19 vaccination are attenuated in some people with multiple sclerosis (PwMS), particularly those receiving B-cell-depleting therapies or sphingosine-1-phosphate receptor modulators (S1PR). Less is known about how disease-modifying therapies (DMTs) shape cellular responses over time. OBJECTIVE: Evaluate longitudinal SARS-CoV-2 Spike IgG and antigen-specific T-cell responses in vaccinated PwMS across DMT classes in a decentralized study. METHODS: COVER-MS participants underwent decentralized biospecimen collection through Quest Diagnostics Patient Service …

The interplay between bile acid metabolism and gut microbiome in biliary tract cancers.

The gut microbiota and bile acids (BAs) exist in a tightly regulated, bidirectional relationship that influences host metabolism, immune function, and disease. Primary BAs synthesized in the liver are chemically transformed by intestinal microbes into a diverse pool of secondary BAs, which exert antimicrobial effects and activate host signaling pathways including Farnesoid X Receptor (FXR), Takeda G protein-coupled receptor 5 (TGR5), and sphingosine-1-phosphate receptor 2 (S1PR2). These pathways regulate BA homeostasis, epithelial barrier integrity, inflammation, and carcinogenesis. Disruption…

How Biologics Are Changing the Global Burden of Inflammatory Bowel Disease: A Review.

Inflammatory bowel disease (IBD), comprising Crohn's disease and ulcerative colitis, has evolved from a condition concentrated in high-income Western nations into a global disease. The therapeutic landscape has been transformed by biologic agents, including anti-tumor necrosis factor (anti-TNF) antibodies, anti-integrin therapy (vedolizumab), interleukin (IL)-12/23 blockade (ustekinumab), and selective IL-23 (p19) inhibitors (risankizumab and mirikizumab); more recently, oral small-molecule advanced therapies (Janus kinase (JAK) inhibitors and sphingosine-1-phosphate (S1P) receptor modulators)…
