Humoral immune responses against particulate bacterial antigens are dependent on marginal metallophilic macrophages in the spleen.

Lymphoid organs are populated by different macrophage subpopulations. In the spleen, four subpopulations can be characterized using differences in, for example, morphology, localization, cell markers and repopulation kinetics after liposome-mediated depletion. The involvement of the different macrophage subpopulations in the immune response to particulate antigens was studied in vivo by intravenous injection of clodronate containing liposomes to eliminate the splenic macrophages, followed by immunization with trinitrophenylated Brucella abortus (TNP-BA; thymus-independent (TI)) or TNP-Lactobac…

Some effects of silical treatment on Marek's disease.

Treatment of newly hatched chicks with silica by the intraperitoneal route delayed the onset of mortalities due to the JM strain of Marek's … t hemagglutination test but not if measured by the agar gel diffusion test, whereas the response to Brucella abortus was enhanced significantly in N-line (MD-resistant) chicks but not significantly in P-line (MD-susceptible) chicks. Five days after infection, silica-treated chicks had significantly less fluorescing antigen in thymus and bursa than did untreated ch …

Bovine brucellosis seropositivity in Mpumalanga Province, South Africa, 2021-2024: Temporal, and spatial trends.

INTRODUCTION: Bovine brucellosis, caused primarily by Brucella abortus, remains a major constraint to livestock productivity and a persistent zoonotic threat. Although brucellosis is a controlled disease in South Africa, detailed subnational epidemiological evidence is limited, particularly for Mpumalanga Province. Understanding temporal, seasonal, and spatial patterns is essential for improving risk-based surveillance and control. MATERIALS AND METHODS: A retrospective cross-sectional analysis was conducted using routine diagnostic records from the Mpumalanga Provincial Veterinary Laboratory…

Ruminant brucellosis in Upper Egypt (2005-2008).

Brucellosis is endemic among humans and ruminant in Egypt and recent reports suggest that its incidence may be increasing. In this study we describe the frequency of brucellosis among different ruminant species in Upper Egypt and its spatial distribution using the data generated by a large-scale control campaign undertaken between 2005 and 2008. A total of 120,090 individual animals of different ruminant species were tested during the campaign. The true proportions of brucellosis were estimated as 0.79% (CI: 0.71%-0.87%), 0.13% (CI: 0.08%-0.18%), 1.16% (1.05%-1.27%) and 0.44% (0.34%-0.54%) amo

Glucose and electrolyte supplementation of drinking water improve the immune responses of poults with inanition.

Enteric disorders predispose poultry to malnutrition. The objectives of this paper were 1) to simulate the inanition of poult enteritis mortality syndrome by restricting feed intake and 2) to develop a drinking water supplement that supports the immune functions of poults with inanition. Poults were restricted to 14 g of feed/d for 7 d beginning at 14 d of age then fed ad libitum until 36 d (recovery). The control was fed ad libitum. During the feed-restriction period, duplicate groups of 6 poults received 1 of 5 drinking water treatments: 1) restricted feed, unsupplemented water; 2) restricte

A study of delayed hypersensitivity and desensitization in guinea pigs after Brucella infection.

We made use of the guinea pig model to demonstrate delayed hypersensitive manifestations in the knee as well as the liver rather than the skin. Experiments revealed that several factors took part in this immune response, i.e. nature of sensitizing agents, route and dosage of allergen procured and functional involvement of local tissue or organ. For instance, the knee swelling was triggered by intraarticular introduction of allergen while the distended liver gave rise to a pathogenic fatal outcome after large dose intravenously. Animal models were also established to abolish partially all these

A TLR4 agonist liposome formulation effectively stimulates innate immunity and enhances protection from bacterial infection.

Stimulation of innate immunity can protect against infectious insult and could be used in combination with other therapies. Since antibiotic resistance is an increasing concern, strategies to reduce the dose or eliminate the need for these drugs are warranted. Lipo-CRX is a formulation in which the TLR4 agonist CRX-527 is incorporated into lipid membranes in liposomes. Lipo-CRX is less inflammatory than either CRX-527 or LPS, but retains unique capacity to enhance host defense responses. We compared lipo-CRX to other agonists in vitro using mammalian cells and in vivo in mice, and assessed ind

Risk factors for new bovine brucellosis infections in Colombian herds.

BACKGROUND: Bovine brucellosis is a zoonotic disease that causes substantial economic losses and has a strong impact on public health. The main objective of this paper is to determine the risk factors for new infections of Brucella abortus on Colombian cattle farms previously certified as being free of brucellosis. A case-control study was conducted by comparing 98 cases (farms certified as brucellosis-free for three or more years but became infected) with 93 controls (farms that remained brucellosis-free during at least the previous three years). The farms were matched by herd size and geogra…

Winter feeding of elk in the Greater Yellowstone Ecosystem and its effects on disease dynamics.

Providing food to wildlife during periods when natural food is limited results in aggregations that may facilitate disease transmission. This is exemplified in western Wyoming where institutional feeding over the past century has aimed to mitigate wildlife-livestock conflict and minimize winter mortality of elk (Cervus canadensis). Here we review research across 23 winter feedgrounds where the most studied disease is brucellosis, caused by the bacterium Brucella abortus Traditional veterinary practices (vaccination, test-and-slaughter) have thus far been unable to control this disease in elk, …

Neurobrucellosis: A report of two cases.

Neurobrucellosis is a rare complication of brucellosis, a common zoonosis with multisystem involvement. Its clinical presentation is quite heterogeneous and diagnosis requires a high index of suspicion in patients from endemic areas. We present two cases of neurobrucellosis with widely varying clinical involvement from a tertiary center in Pakistan. Our case report emphasizes that neurobrucellosis should be considered in evaluation of patients with unexplained neurological symptoms..

Initial presentation of Brucella endocarditis in a previously healthy eighteen-year-old young man.

Brucellosis is a reported endemic in the Middle East, Arabian Peninsula, South Asia, and Central and South America, with a high fatality rate of 80%. We present a case report of Brucella endocarditis in a young 18-year-old male who developed aortic and mitral vegetation, subaortic abscess and mitral leaflet perforation. He was started on antibiotics but his cardiac status rapidly deteriorated and eventually needed a double valve replacement. We would like to share our experience and the challenges we faced in managing this case, which is uncommon, especially in young adults.

The LsrB Protein Is Required for Agrobacterium tumefaciens Interaction with Host Plants.

Agrobacterium tumefaciens infects and causes crown galls in dicot plants by transferring T-DNA from the Ti plasmid to the host plant via a t … etoxification enzymes for oxidative stress. Moreover, expression of the lsrB gene from S. meliloti, Brucella abortus, or A. tumefaciens rescued the defects observed in the S. meliloti or A. tumefaciens lsrB deletion mutant. Our findings suggest that a conserved mechanism of LsrB function exists in symbiotic and pathogenic bacteria of the family Rhizobiaceae.

Effects of management and climate on elk brucellosis in the Greater Yellowstone Ecosystem.

Every winter, government agencies feed approximately 6000 metric tons (6 x 10(6) kg) of hay to elk in the southern Greater Yellowstone Ecosystem (GYE) to limit transmission of Brucella abortus, the causative agent of brucellosis, from elk to cattle. Supplemental feeding, however, is likely to increase the transmission of brucellosis in elk, and may be affected by climatic factors, such as snowpack. We assessed these possibilities using snowpack and feeding data from 1952 to 2006 and disease testing data from 1993 to 2006. Brucellosis seroprevalence was strongly correlated with the timing of th…
