Clonal Diversity of Extraintestinal Pathogenic Escherichia coli Strains Isolated from Canine Urinary Tract Infections in Brazil.

BACKGROUND/OBJECTIVES: Extraintestinal pathogenic Escherichia coli (ExPEC) strains, particularly those belonging to phylogenetic group B2, are clinically significant due to their frequent involvement in urinary tract infections (UTIs) and display antimicrobial resistance profiles. While the association of phylogroup B2 E. coli with human urinary tract infections is well established, the growing number of reports of ExPEC strains in canine UTIs highlights their clinical relevance in small animal medicine and raises concerns about their potential role in zoonotic transmission. This study investi

Emergence and traceability of Salmonella enterica serotype Mbandaka harboring blaOXA-10 from chickens in China.

Salmonella enterica serotype Mbandaka (S. Mbandaka), a multi-host adapted non-typhoidal Salmonella, has emerged as a significant public health concern in recent years. In this study, we isolated S. Mbandaka strains carrying a multidrug-resistant IncHI2A/IncHI2 plasmid from deceased chickens in China and performed whole-genome sequencing and comparative genomic analyses to investigate their global dissemination and evolutionary adaptation. The multidrug-resistant IncHI2A/IncHI2 plasmid in isolate YK35 harbored multiple antibiotic resistance genes (ARGs) including blaOXA-10, which was firstly ob

Mycobacteria that cause tuberculosis have retained ancestrally acquired genes for the biosynthesis of chemically diverse terpene nucleosides.

Mycobacterium tuberculosis (Mtb) releases the unusual terpene nucleoside 1-tuberculosinyladenosine (1-TbAd) to block lysosomal function and promote survival in human macrophages. Using conventional approaches, we found that genes Rv3377c and Rv3378c, but not Rv3376, were necessary for 1-TbAd biosynthesis. Here, we introduce linear models for mass spectrometry (limms) software as a next-generation lipidomics tool to study the essential functions of lipid biosynthetic enzymes on a whole-cell basis. Using limms, whole-cell lipid profiles deepened the phenotypic landscape of comparative mass spect

Complete Genome Sequencing of Leptospira interrogans Isolates from Malaysia Reveals Massive Genome Rearrangement but High Conservation of Virulence-Associated Genes.

The ability of Leptospirae to persist in environments and animal hosts but to cause clinically highly variable disease in humans has made leptospirosis the most common zoonotic disease. Considering the paucity of data on variation in complete genomes of human pathogenic Leptospirae, we have used a combination of Single Molecule Real-Time (SMRT) and Illumina sequencing to obtain complete genome sequences of six human clinical L. interrogans isolates from Malaysia. All six contained the larger (4.28-4.56 Mb) and smaller (0.34-0.395 Mb) chromosome typical of human pathogenic Leptospirae and 0-7 p

Resident or transient? Whole-genome approach to tracking colistin-resistant Escherichia coli in the broiler chicken processing chain.

This study presents a genome-informed surveillance model to investigate the persistence and spread of colistin-resistant Escherichia coli in broiler chicken processing. The study targeted a high-throughput poultry facility-previously linked to retail meat contamination by colistin-resistant E. coli and Salmonella-where 200 carcasses were sampled across ten production batches to assess the prevalence and genomic characteristics of antimicrobial-resistant strains within the processing line. We analyzed one E. coli isolate per carcass to characterize antimicrobial resistance (AMR), and utilized w

Antimicrobial resistance in foodborne Escherichia coli and Salmonella spp. from animal-origin foods: Transmission pathways, global surveillance gaps, and alternative therapeutic strategies.

Antimicrobial resistance (AMR) in enteric pathogens such as Escherichia coli and Salmonella spp. has emerged as a critical global health challenge affecting both human and animal populations. The widespread use of antibiotics in food-producing animals for therapeutic, prophylactic, and growth-promoting purposes has accelerated the selection and dissemination of resistant bacteria and resistance genes throughout the food chain. Animal-origin foods, including meat, milk, eggs, and fish, serve as important vehicles for the transmission of multidrug-resistant organisms and AMR genes to humans, rep

The cytosolic and glycosomal isoenzymes of glyceraldehyde-3-phosphate dehydrogenase in Trypanosoma brucei have a distant evolutionary relationship.

Trypanosoma brucei contains two isoenzymes for glyceraldehyde-3-phosphate dehydrogenase: one enzyme resides in a microbody-like organelle, the glycosome; the other is found in the cytosol. Previously we have reported the characterization of the gene for the glycosomal enzyme [Michels, P. A. M., Poliszczak, A., Osinga, K. A., Misset, O., Van Beeumen, J., Wierenga, R. K., Borst, P. & Opperdoes, F. R. (1986) EMBO J. 5, 1049-1056]. Here we describe the cloning and analysis of the gene that codes for the cytosolic isoenzyme. The gene encodes a polypeptide of 330 amino acids, with a calculated molec

Genetic diversity of the outer surface protein C gene of southern Borrelia isolates and its possible epidemiological, clinical, and pathogenetic implications.

The ospC genes of 20 southern Borrelia strains were sequenced. The strains consisted of B. burgdorferi sensu stricto, B. andersonii, B. bissettii, one undescribed genospecies, MI-8, and one probably new Borrelia species, TXW-1. A high degree of similarity exists between B. burgdorferi sensu stricto and B. bissettii and between B. bissettii and B. andersonii. Lateral transfers of the ospC gene probably occurred between B. burgdorferi sensu stricto and B. bissettii and between B. bissettii and B. andersonii. Internal gene recombination appears to occur among them. The highest degree of genetic d

Predicted and inducible prophages display contrasting virulence gene profiles within the prophage-SaPI mobilome of Staphylococcus aureus.

Prophages play a significant role in bacterial evolution by shaping genomic diversity, virulence, and host adaptation. This study investigated the prophage composition of 109 clinical Staphylococcus aureus isolates obtained from four tertiary care hospitals in Mexico City and compared these results with data from 993 global genomes. Prophages were present in 97% of local isolates. Consistently, analysis of the global genome collection revealed a 99% prevalence, supporting the near ubiquity of prophages in S. aureus. Analysis identified 216 genomic regions corresponding to the predicted prophag

Dissemination of antimicrobial resistance in surface waters: surveillance of enterococci and coliform bacteria in Slovakia.

Antimicrobial-resistant faecal bacteria represent an increasing environmental concern in surface waters impacted by anthropogenic activities. Surveillance of enterococci and coliform bacteria, particularly Escherichia coli, is widely used to assess the dissemination of antimicrobial resistance in aquatic environments. These ecosystems may act as reservoirs of resistance determinants, where subinhibitory antibiotic concentrations promote their persistence and horizontal transfer, posing a potential public health risk. This study aimed to survey antibiotic‑resistant coliform bacteria and enteroco

Emerging zoonotic pathogen Staphylococcus borealis mediates longitudinal increase and environmental contamination of tigecycline resistance throughout the pig production period.

The widespread use of tetracyclines in swine production has accelerated the emergence of tigecycline-resistant Staphylococcus, posing a threat to public health. This study investigated the prevalence and mechanisms of tigecycline resistance in pigs, where tigecycline is unapproved yet resistance is present. The emerging zoonotic pathogen Staphylococcus borealis played an important role. Longitudinal surveillance revealed that tigecycline-resistant S. borealis (TRSB) ST64 colonized newborn piglets and persisted throughout the entire production period, exhibiting environmental contamination from

The pet café is a neglected site for transmission of antimicrobial-resistant Escherichia coli in urban life.

The process of urbanization has brought with it several novel lifestyles, but it remains to be seen whether such lifestyles are the potential driver behind the spread of antimicrobial resistance (AMR) in modern society. Hence, this study employs the pet café as a proof of concept to observe how one pathway of AMR transmission occurs within a megacity. A total of 111 samples were collected from consumers, workers, animals and the surrounding environment from three pet cafés in Guangzhou, and 163 bacterial strains were isolated, with Escherichia coli (n=60) being the most dominant species. Pathogen-encoded Rum DNA polymerase drives rapid bacterial drug resistance.

The acquisition of multidrug resistance by pathogenic bacteria is a potentially incipient pandemic. Horizontal transfer of DNA from mobile integrative conjugative elements (ICEs) provides an important way to introduce genes that confer antibiotic (Ab)-resistance in recipient cells. Sizable numbers of SXT/R391 ICEs encode a hypermutagenic Rum DNA polymerase (Rum pol), which has significant homology with Escherichia coli pol V. Here, we show that even under tight transcriptional and post-transcriptional regulation imposed by host bacteria and the R391 ICE itself, Rum pol rapidly accelerates deve

Nothing in Evolution Makes Sense Except in the Light of Genomics: Read-Write Genome Evolution as an Active Biological Process.

The 21st century genomics-based analysis of evolutionary variation reveals a number of novel features impossible to predict when Dobzhansky and other evolutionary biologists formulated the neo-Darwinian Modern Synthesis in the middle of the last century. These include three distinct realms of cell evolution; symbiogenetic fusions forming eukaryotic cells with multiple genome compartments; horizontal organelle, virus and DNA transfers; functional organization of proteins as systems of interacting domains subject to rapid evolution by exon shuffling and exonization; distributed genome networks i
