Airway infection in stable lung transplant patients is associated with decreased intracellular T-helper type 1 pro-inflammatory cytokines in bronchoalveolar lavage T-cell subsets.

Current immunosuppression protocols to prevent lung transplant rejection reduce pro-inflammatory and T-helper type 1 (Th1) cytokines. However, Th1 T-cell pro-inflammatory cytokine production is important in host defense against bacterial infection in the lungs. Excessive immunosuppression of Th1 T-cell pro-inflammatory cytokines leaves patients susceptible to infection. To investigate whether pulmonary infection in lung transplant recipients is associated with reduced Th1 T-cell pro-inflammatory cytokines, whole blood and bronchoalveolar lavage (BAL) fluid from 13 stable lung transplant patien

Antagonism of endogenous putative P2Y receptors reduces the growth of MDCK-derived cysts cultured in vitro.

P2Y receptors couple to G proteins and either mobilize intracellular Ca(2+) or alter cAMP levels to modulate the activity of Ca(2+)- and cAMP-sensitive ion channels. We hypothesize that increased ion transport into the lumen of MDCK cysts can osmotically drive fluid movement and increase cyst size. Furthermore, activation of the adenylate cyclase/cAMP pathway may trigger cell proliferation via an extracellular signal-related kinase cascade. To test this hypothesis, several P2Y receptor inhibitors were used on the MDCK in vitro model of renal cyst formation. The nonspecific P2 receptor inhibito

Background potassium concentrations and epileptiform discharges. I. Electrophysiological characteristics of neuronal activity.

Intra- and extracellular recording techniques were used to study the epileptiform activity generated by guinea pig hippocampal slices perfused with free-magnesium artificial cerebrospinal fluid in the presence of physiologic (4 mM), reduced (2 mM) or elevated (8 mM) extracellular potassium concentrations ([K(+)](o)). Extracellular field potentials along with intracellular recordings were recorded in CA1 or CA3 region. Reduction of [K(+)](o) significantly increased the latency of epileptiform field potential (EFP) appearance as well as burst discharge duration and decreased EFP repetition rate.

Increased antiviral activity of 1-O-hexadecyloxypropyl-[2-(14)C]cidofovir in MRC-5 human lung fibroblasts is explained by unique cellular uptake and metabolism.

Recently, there has been renewed interest in finding orally active drugs against smallpox. Cidofovir (CDV) given by parenteral injection has been shown to protect against lethal poxvirus infection. We have been interested in the synthesis and evaluation of orally active derivatives of CDV. Previous studies showed that the CDV and cyclic cidofovir (cCDV) analogs 1-O-hexa-decyloxypropyl-CDV (HDP-CDV) and 1-O-hexadecyloxypropyl-cCDV (HDP-cCDV), show >100-fold increases in antiviral activity versus the unmodified nucleosides against cells infected with orthopoxviruses, cowpox, and vaccinia virus.

Cooling abolishes neuronal network synchronization in rat hippocampal slices.

PURPOSE: We sought to determine whether cooling brain tissue from 34 to 21 degrees C could abolish tetany-induced neuronal network synchronization (gamma oscillations) without blocking normal synaptic transmission. METHODS: Intracellular and extracellular electrodes recorded activity in transverse hippocampal slices (450-500 microm) from Sprague-Dawley male rats, maintained in an air-fluid interface chamber. Gamma oscillations were evoked by afferent stimulation at 100 Hz for 200 ms. Baseline temperature in the recording chamber was 34 degrees C, reduced to 21 degrees C within 20 min. RESULT

Infection of mice with the helminth Strongyloides stercoralis suppresses pulmonary allergic responses to ovalbumin.

Asthma and helminth infections induce similar immune responses characterized by the presence of peripheral blood eosinophilia and elevated serum IgE levels. Epidemiological surveys have reported either increases or decreases in the development of atopic diseases and asthma based on the prevalence of helminth infections in the population. The aim of this study was to determine if a pre-existing helminth infection would increase or decrease subsequent allergic responses to an unrelated allergen in the lungs. BALB/cByJ mice were infected with the nematode parasite Strongyloides stercoralis prior

Heterogeneity of chloride/base exchange in rabbit superficial and juxtamedullary proximal convoluted tubules.

Active transcellular NaCl transport in the proximal convoluted tubule (PCT) is via apical parallel Na/H and Cl/base exchange. The mechanism of Cl/base exchange remains unclear. The present in vitro microperfusion study examined the mechanism of Cl/base exchange in superficial and juxtamedullary PCT by examining the rate of change in intracellular pH in response to luminal Cl removal. In superficial PCT the rate of Cl/base exchange was 24.0 +/- 2.3 without formate, 36.4 +/- 6.6 with 10 microM formate (P < 0.05), and 43.6 +/- 2.8 pmol.mm-1.min-1 (P < 0.001) with 1 mM luminal formate. Bicarbonate transport under nominally bicarbonate-free conditions in bovine corneal endothelium.

Fluid transport by the corneal endothelium is supported in air equilibrated, nominally HCO3- free Ringer's, yet it is inhibited by carbonic anhydrase inhibitors indicating dependence on the presence of CO2. It is possible that the CO2/HCO3- contained in air and/or the CO2 derived from oxidative metabolism is sufficient to support HCO3- transport. This study investigates whether endothelial HCO3- transport mechanisms, Na+:HCO3- cotransport and Cl-/HCO3- exchange, are active in the nominal absence of HCO3-. In cultured bovine corneal endothelial cells the rate of intracellular pH (pHi) recovery

Role of G proteins in stimulation of Na-H exchange by cell shrinkage.

Many cells respond to shrinkage by stimulating specific ion transport processes (e.g., Na-H exchange). However, it is not known how the cell senses this volume change, nor how this signal is transduced to an ion transporter. We have studied the activation of Na-H exchange in internally dialyzed barnacle muscle fibers, measuring intracellular pH (pHi) with glass microelectrodes. When cells are dialyzed to a pHi of approximately 7.2, Na-H exchange is active only in shrunken cells. We found that the shrinkage-induced stimulation of Na-H exchange, elicited by increasing medium osmolality from 975

Tubuloglomerular feedback responses in the rat during calmodulin inhibition.

Previously, we have suggested that a cytosolic calcium system participates in the transmission of signals between distal tubular fluid and the glomerular vascular elements. Since the Ca-binding protein calmodulin has been implicated in the intracellular actions of cytosolic Ca, we evaluated the effects of the calmodulin antagonists trifluoperazine (TFP) and calmidazolium (R24571) on tubuloglomerular feedback responses. In the rat, stop-flow pressure (SFP) was measured during retrograde perfusion into the distal tubule at 15 nl/min for 5 min. Perfusion with an isotonic Ringer solution resulted

A high-concentrate diet induces inflammatory injury via regulating Ca2+/CaMKKβ-mediated autophagy in mammary gland tissue of dairy cows.

INTRODUCTION: Calmodulin-dependent protein kinase β (CaMKKβ) is closely related to Ca2+ concentration. An increase in Ca2+ concentration in the cytoplasm activates CaMKKβ, and activated CaMKKβ affects the activities of AMPK and mTOR and induces autophagy. A high-concentrate diet leads to Ca2+ disorder in mammary gland tissue. OBJECTIVES: Therefore, this study mainly investigated the induction of mammary gland tissue autophagy by a high-concentrate diet and the specific mechanism of lipopolysaccharide (LPS)-induced autophagy in bovine mammary epithelial cells (BMECs). Extracellular Vesicle Communication at the Neural Graft-Host Interface: From Bidirectional Paracrine Signaling to Graft Fate and Neural Circuit Integration.

Neural cell transplantation is progressing from nonspecific trophic support toward regionally defined cell replacement and functional circuit reconstruction. Yet transplanted cells do not develop in isolation; they remain embedded within a dynamic host environment composed of neurons, glia, vascular-associated cells, cerebrospinal fluid, and systemic signals. Extracellular vesicles (EVs) may provide a membrane-enclosed route for this reciprocal communication by transferring nucleic acids, proteins, lipids, and organelle-associated cargo across the graft-host interface. Moving beyond EVs as cel

Evaluation of sNfL as a Biomarker for Paclitaxel-Induced Peripheral Neurotoxicity Through an Integrated PKPD Model.

BACKGROUND: Serum neurofilament light chain (sNfL), a biomarker of axonal damage, has shown promise in clinical studies for monitoring paclitaxel-induced peripheral neurotoxicity (PIPN). The latter involves pathological changes in PIPN sites such as the dorsal root ganglia, peripheral nerves, and brain. However, the mechanistic link between paclitaxel and NfL concentrations in these tissues remains poorly understood, necessitating preclinical investigation. METHODS: We developed a semi-mechanistic pharmacokinetic-pharmacodynamic model to characterize: (i) total and unbound paclitaxel concentr

Cell-specific MicroRNA networks orchestrate the pathogenesis of Alzheimer's disease.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder marked by extracellular amyloid-β (Aβ) plaques, intracellular neurofibrillary tangles of hyperphosphorylated tau, synaptic dysfunction, and chronic neuroinflammation. AD pathogenesis involves multiple central nervous system (CNS) cell types-including neurons, astrocytes, microglia, and oligodendrocytes, and, less prominently, neural stem cells (NSCs), ependymal cells, and endothelial cells-which undergo coordinated but cell-type-specific pathological changes. These include neuronal loss, reactive gliosis, impaired myelin main

Biomarker-informed PBPK modelling of meropenem in paediatric severe pneumonia: implications for target-site PK/PD.

OBJECTIVES: Optimal antimicrobial exposure in epithelial lining fluid (ELF) is critical for meropenem efficacy in pneumonia, yet ELF pharmacokinetic data remain scarce, particularly in children. To address this, we aimed to develop a model capable of predicting meropenem concentrations in both plasma and ELF for evaluating pharmacodynamic target attainment under clinical dosing strategies. METHODS: A physiologically based pharmacokinetic (PBPK) model was developed to simulate unbound meropenem concentrations in plasma and ELF. An empirical penetration coefficient (ρ) was incorporated to link

Impact of in ovo myo-inositol administration on immune modulation in broiler chickens.

Myo-inositol (MI) plays an important role in intracellular signaling, cell proliferation, and cell growth, and has been associated with positive effects on the immune system and health in fish. However, its potential immunological effects in poultry have not been investigated yet. This study aimed to evaluate the effects of in ovo MI administration on immune parameters in broiler chickens after hatching. A total of 480 fertilized Ross 308 eggs were randomly assigned as hatched to four groups and injected on embryonic day 17 with either 12 µmol/mL MI (MI-12), 24 µmol/mL MI (MI-24), 0.9% saline

C-reactive protein in bronchoalveolar lavage fluid as a potential biomarker of bacterial bronchopneumonia in dogs.

Diagnosis of canine bacterial bronchopneumonia (bBP) is challenging and mostly based on bronchoalveolar lavage fluid (BALF) cytology and bacterial culture (BC) results. However, bacterial contamination of BALF is frequent, intracellular bacteria are not always visualized on BALF cytology, and BC results may not be related to antimicrobial requirements. Based on local expression of C-reactive protein (CRP) by alveolar macrophages, we aimed to evaluate whether CRP was detectable in canine BALF, and to determine whether BALF-CRP detection could assist in differentiating bBP from non-bacterial bro

Mechanisms, Mediators, and Pharmacological Approaches Targeting Brain Cholesterol Transport in Alzheimer's Disease.

Cholesterol transport within the brain represents a highly regulated process essential for maintaining neuronal function and central nervous system (CNS) homeostasis. Unlike peripheral tissues, the brain relies on in situ cholesterol synthesis, primarily by astrocytes and other glial cells, which supply neurons via high-density lipoprotein (HDL)-like particles, identified in the human cerebrospinal fluid (CSF). The major component of HDL-like lipoproteins is the apolipoprotein E (ApoE), whose E4 isoform represents the strongest genetic risk factor for late-onset Alzheimer's disease (AD). Growi

Neurofilament light chain (NfL) as a biomarker for neurodegeneration: Protein biology, detection methods, and clinical applications.

Neurofilaments are vital structural components of neurons and play a central role in preserving axonal architecture and function. Among these, neurofilament light chain (NfL) has emerged as a sensitive biomarker of neuroaxonal injury. This chapter delineates the molecular organisation of neurofilaments, tracing their hierarchical assembly from coiled-coil dimers to mature filaments, and emphasises their contribution to axonal stability, calibre regulation, and intracellular transport. It further examines how perturbations in neurofilament homeostasis reflect neuronal damage across a broad spec

Distinct fibrosis-associated macrophage subsets coordinate iron metabolism in pulmonary fibrosis.

Pulmonary fibrosis has a poor prognosis because of challenges in early diagnosis and therapeutic intervention. Current treatments remain largely ineffective due to an incomplete understanding of the complex pathology, including the interactions between fibroblasts and profibrotic immune cells within fibrotic lungs. To elucidate the dynamics of fibrosis, we performed single-cell RNA sequencing on bronchoalveolar lavage fluid obtained from patients with interstitial lung disease (ILD). We identified the SPP1- and APOE-expressing macrophage population that is commonly present across ILDs. Histolo
