Discovery of Novel Peptidomimetics as Irreversible CHIKV NsP2 Protease Inhibitors Using Quantum Mechanical-Based Ligand Descriptors.

Chikungunya virus (CHIKV) is a mosquito-borne alphavirus. Recent outbreaks of CHIKV infections have been reported in Asia, Africa, and Europe. The symptoms of CHIKV infection include fever, headache, nausea, vomiting, myalgia, rash, and chronic persistent arthralgia. To date, no vaccines or selective antiviral drugs against this important emerging virus have been reported. In this study, the design, synthesis, and antiviral activity screening of new topographical peptidomimetics revealed three potential prototype agents 3a, 4b, and 5d showing 93-100% maximum inhibition of CHIKV replication in

Identification of thioredoxin glutathione reductase inhibitors that kill cestode and trematode parasites.

Parasitic flatworms are responsible for serious infectious diseases that affect humans as well as livestock animals in vast regions of the world. Yet, the drug armamentarium available for treatment of these infections is limited: praziquantel is the single drug currently available for 200 million people infected with Schistosoma spp. and there is justified concern about emergence of drug resistance. Thioredoxin glutathione reductase (TGR) is an essential core enzyme for redox homeostasis in flatworm parasites. In this work, we searched for flatworm TGR inhibitors testing compounds belonging to

Dispersive Optical Activity of (S)-2-Methylaziridine: Intrinsic Properties and Extrinsic Solvation Effects.

The dispersive optical activity of conformationally flexible S-2-methylaziridine (S-2-MeAz) has been investigated experimentally under isolated and solvated conditions to elucidate the impacts of large-amplitude (conformational) and small-amplitude (vibrational) nuclear degrees of freedom on intrinsic chiroptical response and the extrinsic perturbations caused by seven distinct solvents. All solution-phase measurements of optical rotatory dispersion (ORD) revealed plain-looking curves of uniformly negative polarity in the near-ultraviolet/visible regions, while their solvent-free counterpart s

State-Averaged Density Matrix Embedding Theory for Local Excitations.

Density matrix embedding theory (DMET) provides an elegant framework in quantum chemistry to describe local properties of chemical systems, which allows a high-level method to solve an embedded subsystem constructed based on a low-level treatment of the whole system and therefore achieves a balance between efficiency and accuracy. However, because the embedded subspace in DMET is typically constructed from the mean-field ground-state Slater determinant, the resulting bath orbitals inherently favor the ground state, leading to unbalanced descriptions of ground and excited states for local excit

Molecular insights into the selective interaction of endocrine-disrupting chemicals with poly(mannuronate) in aqueous systems: a multiscale computational study.

Estrogens and other steroid hormones, also known as endocrine-disrupting chemicals (EDCs), are emerging contaminants in wastewater effluent due to human activities. Although these compounds play essential roles in medical treatments and modern agriculture, their release into the environment has raised concerns because of their potential ecological and health impacts. The selective removal of EDCs from aqueous environments is a challenge due to competition with water molecules for binding sites. This work presents a systematic computational investigation of the intermolecular interactions and p

Stabilization of the Ferryl═Oxoheme Form of Staphylococcus aureus IsdG by Electron Transfer from a Second-Sphere Tryptophan.

The ferryl heme forms of Staphylococcus aureus IsdG and IsdI have novel UV/vis absorption spectra that are distinct from those of the three forms of ferryl heme typically found in biological systems: compound I, compound II, and compound ES. In this work, the ferryl heme form of IsdG was characterized because it is an analogue for the immediate product of enzyme-catalyzed heme hydroxylation. The ferryl heme form of IsdG generated following the addition of meta-chloroperoxybenzoic acid to the ferric heme form of IsdG has a half-life of 4.0 ± 0.2 min, which is more than 100 times longer than the

Optical Properties and Excited-State Dynamics of Safranin O Monomers and Dimers in Aqueous Solution.

Phenazines, phenoxazines, and phenothiazines are a structurally similar set of compounds that exhibit antibiotic and photocatalytic activity and that have been used as histological stains and optical sensors. Safranin O (SO) is a phenazine that has been found useful to quantify polysaccharides in cartilage and that has been shown to have a unique ability to inhibit ice growth. In both of these applications, the high concentration of SO can lead to aggregation, which can result in drastic changes in optical properties and excited-state dynamics. In this work, we characterize the optical propert

Iodine Clusters in the Atmosphere I: Computational Benchmark and Dimer Formation of Oxyacids and Oxides.

The contribution of iodine-containing compounds to atmospheric new particle formation is still not fully understood, but iodic acid and iodous acid are thought to be significant contributors. While several quantum chemical studies have been carried out on clusters containing iodine, there is no comprehensive benchmark study quantifying the accuracy of the applied methods. Here, we present the first study in a series that investigate the role of iodine species in atmospheric cluster formation. In this work, we have studied the iodic acid, iodous acid, iodine tetroxide, and iodine pentoxide mono

Origins of Electromagnetic Radiation from Spintronic Terahertz Emitters: A Time-Dependent Density Functional Theory plus Jefimenko Equations Approach.

Microscopic origins of charge currents and electromagnetic (EM) radiation generated by them in spintronic THz emitters-such as, femtosecond laser pulse-driven single magnetic layer or its heterostructures with a nonmagnetic layer hosting strong spin-orbit coupling (SOC)-remain poorly understood despite nearly three decades since the discovery of ultrafast demagnetization. We introduce a first-principles method to compute these quantities, where the dynamics of charge and current densities is obtained from real-time time-dependent density functional theory, which are then fed into the Jefimenko

Synergistic Effects of Unconventional Hydrogen Bonds and π-Stacking Interaction and Their Excited-State Dependence: The Origin of Unusual Photophysical Properties of Aromatic Thioketones in Acetonitrile and Hydrocarbons.

It has been established experimentally that aromatic thioketones possess several inherently unique photophysical properties, some of which are highly sensitive even to common hydrocarbon solvents. However, the deeper reasons and the underlying mechanisms remain unclear up to date. In this study, the multistate complete active space second-order perturbation theory (MS-CASPT2) has been utilized to investigate the five lowest-lying electronic states (S0, T1, S1, T2, and S2) of 4H-1-benzopyran-4-thione (BPT) in acetonitrile and hydrocarbons. The results show that the S1, T1, and T2 states of BPT

Insight into the interaction between amino acids and SO2: Detailed bonding modes.

CONTEXT: Amino acids are a highly effective and environmentally friendly adsorbent for SO2. However, there has been no comprehensive study of the binding modes between amino acids and SO2 at the molecular level. In this paper, the binding modes of three amino acids (Asp, Lys, and Val) with SO2 are studied comprehensively and in detail using quantum chemical calculations. The results indicate that each amino acid has multiple binding modes: 22 for Asp, 49 for Lys, and 10 for Val. Both the amino and carboxyl groups in amino acids, as well as those in side chains, can serve as binding sites for c

Room-Temperature Phosphorescence from Pd(II) and Pt(II) Complexes as Supramolecular Luminophores: The Role of Self-Assembly, Metal-Metal Interactions, Spin-Orbit Coupling, and Ligand-Field Splitting.

The synthesis as well as the structural and photophysical characterization of two isoleptic bis-cyclometalated Pt(II) and Pd(II) complexes, namely [PtL] and [PdL], bearing a tailored dianionic tetradentate ligand (L2-) are reported. The isostructural character and intermolecular interactions of [PtL] and [PdL] were assessed by NMR spectroscopy and X-ray diffraction analysis. Both complexes show fully ligand-controlled aggregation, demonstrating that a judicious molecular design can tune the photophysical properties. In fact, by introduction of fluorine atoms on defined positions and methoxy gr
