Potential genetic markers for nonsyndromic oral clefts in the Brazilian population: A systematic review and meta-analysis.

BACKGROUND: Although various genes and genomic regions were described as of susceptibility for nonsyndromic oral clefts (NOC), recent reports have demonstrated significant interethnic variations in the genetic predisposition, a situation that affects the Brazilian population, one of the most admixed populations in the world. Therefore, the purpose of this review was to describe the available information on genetic risk markers for NOC in the Brazilian population. METHODS: A systematic search of the literature was performed using LILACS, LIVIVO, PubMed, Scopus, and Web of Science databases, an

The genetics of cholesteatoma. A systematic review using narrative synthesis.

OBJECTIVE: A cholesteatoma is a mass of keratinising epithelium in the middle ear. It is a rare disorder that is associated with significant morbidity, and its causative risk factors are poorly understood; on a global scale, up to a million people are affected by this each year. We have conducted a systematic literature review to identify reports about the heritability of cholesteatoma or any constitutional genetic factors that may be associated with its aetiology. DATA SOURCES: A systematic search of MEDLINE (EBSCO) and two databases of curated genetic research (OMIM and Phenopedia) was cond

The etiology of congenital nephrotic syndrome: current status and challenges.

BACKGROUND: Congenital nephrotic syndrome (CNS), defined as heavy proteinuria, hypoalbuminemia, hyperlipidemia and edema presenting in the first 0-3 months of life, may be caused by congenital syphilis, toxoplasmosis, or congenital viral infections (such as cytomegalovirus). However, the majority of CNS cases are caused by monogenic defects of structural proteins that form the glomerular filtration barrier in the kidneys. Since 1998, an increasing number of genetic defects have been identified for their involvements in the pathogenesis of CNS, including NPHS1, NPHS2, WT1, PLCE1, and LAMB2. DA

Common TLR1 genetic variation is not associated with death from melioidosis, a common cause of sepsis in rural Thailand.

Melioidosis, infection caused by the Gram-negative bacterium Burkholderia pseudomallei, is a common cause of sepsis in northeast Thailand. In white North Americans, common functional genetic variation in TLR1 is associated with organ failure and death from sepsis. We hypothesized that TLR1 variants would be associated with outcomes in Thais with melioidosis. We collated the global frequencies of three TLR1 variants that are common in white North American populations: rs5743551 (-7202A/G), rs4833095 (742A/G), and rs5743618 (1804G/T). We noted a reversal of the minor allele from white North Amer

Detailed analysis of gene polymorphisms associated with ischemic stroke in South Asians.

The burden of stroke is disproportionately high in the South Asian subcontinent with South Asian ethnicity conferring a greater risk of ischemic stroke than European ancestry regardless of country inhabited. While genes associated with stroke in European populations have been investigated, they remain largely unknown in South Asians. We conducted a comprehensive meta-analysis of known genetic polymorphisms associated with South Asian ischemic stroke, and compared effect size of the MTHFR C677T-stroke association with effect sizes predicted from homocysteine-stroke association. Integrative analysis using Mendelian randomization and network toxicology reveals genetic support and potential mechanisms of atrazine in breast cancer.

OBJECTIVE: Breast cancer (BC) is the leading cause of cancer-related mortality in women. Atrazine, a prevalent herbicide, is a persistent environmental pollutant with bioaccumulative potential. This study investigated the potential mechanisms underlying the association between atrazine exposure and BC. METHODS: We integrated potential common targets of atrazine in BC through multiple databases (e.g., PubChem, CTD, GeneCards, OMIM). Genetically supported target-level associations were assessed using Mendelian randomization (MR). The protein-protein interaction (PPI) network was constructed via

No Causal Relationship Between Helicobacter Pylori Infection and Rosacea: A 2-Sample Mendelian Randomization Study.

BACKGROUND: Previous studies have shown that patients with rosacea have a higher prevalence of Helicobacter pylori (H. pylori) infection. However, whether H. pylori infection contributes to the development of rosacea remains unclear, and no genetic association studies between the two have been conducted to date. Genome-wide association studies (GWAS) database is a public resource that stores and shares data that aims to identify genetic links to complex diseases, physiological traits, or drug responses. Mendelian randomization (MR) is an epidemiological approach to investigate the effect of th

Comprehensive genomic study of Plasmodium falciparum in Sierra leone: genetic variation and resistance patterns.

BACKGROUND: Malaria remains a significant global health concern, with Plasmodium falciparum being the most dangerous of the malaria-causing parasites. Sierra Leone, with year-round transmission of malaria, continues to experience high infection rates, largely due to P. falciparum. Although genomic databases have provided valuable insights into malaria transmission patterns, drug resistance, and population structure, data from Sierra Leone has been limited. This study aims to build on our previous report by incorporating new samples and providing a more comprehensive genomic analysis of P. falc

Correlation between choroid plexus carcinoma and Li-Fraumeni syndrome: implications of TP53 mutations and management strategies-a case-based narrative review.

BACKGROUND: Choroid plexus carcinomas (CPCs) are rare, aggressive grade 3 tumors of the central nervous system associated with Li-Fraumeni syndrome (LFS) in a notable percentage of cases due to TP53 germline mutations. Understanding the correlation between CPCs and LFS is crucial for tailored management strategies. However, distinguishing CPCs from benign choroid plexus papillomas (CPPs) remains challenging, relying largely on histologic features. This study aimed to explore the association between CPCs and LFS, emphasizing the impact of TP53 mutations on diagnosis, treatment, and clinical out

Genetic Aspects of Keratoconus: A Literature Review Exploring Potential Genetic Contributions and Possible Genetic Relationships with Comorbidities.

INTRODUCTION: Keratoconus (KC) is a complex, genetically heterogeneous, multifactorial degenerative disorder that is accompanied by corneal ectasia which usually progresses asymmetrically. With an incidence of approximately 1 per 2000 and 2 cases per 100,000 population presenting annually, KC follows an autosomal recessive or dominant pattern of inheritance and is, apparently, associated with genes that interact with environmental, genetic, and/or other factors. This is an important consideration in refractive surgery in the case of familial KC, given the association of KC with other genetic d

Genetic Burden in Congenital Anomalies of the Mitral and Tricuspid Valves: A Case-Control Study.

Congenital heart disease (CHD) is the most common congenital malformation, with most cases exhibiting a multifactorial etiology involving genetic and environmental factors. Congenital anomalies of the atrioventricular valve or septum (CAAVAS) and functionally univentricular heart (FUH) are complex subtypes of CHD, where disruptions in key molecular pathways are implicated. This study investigates the genetic burden contributing to these anomalies. This case-control study included 48 participants: 24 patients diagnosed with CAAVAS or FUH and 24 healthy controls. Whole-exome sequencing (WES) was
