RNA amplification with transcript sequencing (RAWTS) is a rapid and sensitive method of direct sequencing that involves complementary DNA synthesis, polymerase chain reaction (PCR) with a primer or primers containing a phage promoter, transcription from the phage promoter, and reverse transcriptase-mediated sequencing. By means of RAWTS, it was possible to sequence each of four tissue-specific human messenger RNAs (blue pigment, factor IX, phenylalanine hydroxylase, and tyrosine hydroxylase) in four cell types examined (white blood cells, liver, K562 erythroleukemia cells, and chorionic villus

The development of the vasculature of the pectoral fin in the Australian lungfish, Neoceratodus forsteri, was studied by the dye-injection method. Only a single primitive subclavian artery appears from the dorsal aorta for the fin anlage, and it passes laterally through the postaxial region of the structure. The venous channel draining into the posterior cardinal vein is located in the preaxial region medially. As development proceeds, the arteriovenous arrangement in the pectoral fin anlage changes as follows: 1) one artery and one venous plexus, 2) two arteries and one vein, 3) three arterie

Acute cohesin loss causes widespread reorganization of three-dimensional (3D) chromatin architecture but has relatively minor effects on steady-state transcription. It remains unclear whether its role in gene regulation becomes more critical during mitotic exit, when 3D chromatin architecture and transcription are globally re-established. To address this, we acutely depleted RAD21 in mouse embryonic stem cells during mitotic exit under self-renewal or differentiation conditions. Here we show that, although most loops failed to reform without cohesin, the few cohesin-independent loops were link

Nephrotoxicity remains a critical concern in drug development, with limited preclinical models that balance physiological relevance, ethical considerations, and scalability. Here, we evaluate the chicken embryo (CE) as an in vivo platform for nephrotoxicity assessment, using gentamicin-induced acute kidney injury as a proof-of-principle. Using an ex ovo culture system, we assessed renal responses in the developing CE kidney. By embryonic day 12, the CE exhibited advanced organogenesis, including a functional metanephric kidney. Immunohistochemical analyses demonstrated conservation of key stru

During morphogenesis, multicellular patterns emerge from coordinated processes at the single-cell level. This is characterised in flat epithelial layers, where average neighbour distributions follow conserved arrangements. However, less is known about how curvature defines tissue architecture. Epithelial shells constitute a critical structural stage in early mammalian embryonic development. Our analysis reveals how the closed, curved geometry of epithelial shells limits their 3D organisation. Here, we designed computational models that predict shell surface topology across different cell numbe

Environmental exposure to microplastics, especially polystyrene microplastics (PS-MPs), poses growing concerns for male reproductive health. In this study, we demonstrate that PS-MPs internalize into Sertoli cells, triggering CEBPB-mediated lysosomal hyperactivation and the degradation of tight junction proteins, thereby impairing the structural integrity of the blood-testis barrier (BTB). Concurrently, PS-MPs accumulate in mature human sperm, damaging mitochondrial ultrastructure and inducing ROS-driven lipid peroxidation and ferroptosis-like injury. These alterations result in reductions in

The study aimed to optimize and develop novel PEGylated co-loaded nanoliposome entrapped with paclitaxel (PTX) and 6-gingerol (Gn) (PEG-Lipo-PTX-Gn) by response surface methodology (RSM) approach to prevent the diffusion and resistant-related issues of PTX in oncotherapy. Physiochemical characterization studies results revealed that the prepared PEG-Lipo-PTX-Gn attained optimum particle size, shape, charge, polydispersity index (PDI) as well as showed synergistic entrapment of PTX and Gn, sustained drug release, better colloidal stability and structural integrity. PEG-Lipo-PTX-Gn exhibited a n

Neural tube defects (NTDs) are complex multigenic disorders and are the most prevalent and severe congenital malformations that affect the central nervous system. PCSK9 is identified as a molecular marker for the prenatal diagnosis of NTDs during its early stages in fetuses; however, its role in NTD neurulation and pathogenesis remains unclear. This study introduces PCSK9 knockout embryonic stem cells (ESCs) into neural organoid (NO) and neural progenitor cell (NPC) models and finds that PCSK9 loss leads to an incomplete neural tube structure in NOs and microfilament network disorder in NPCs.

Analysis of retinas from mice with a targeted disruption of the gene encoding the forkhead transcription factor Foxn3 revealed additional displaced amacrine interneurons and retinal astrocytes in the inner plexiform and ganglion cell layers, as well as ectopic primary cilia on bipolar and amacrine interneurons. Foxn3 is a transcriptional repressor and numerous genes linked to cilia structure or assembly were upregulated in embryonic retinas without Foxn3 , including Foxj1, a forkhead transcription factor required for motile cilia. CUTCRUN analysis revealed that many of the upregulated retinal

Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease characterized by the loss of motor neurons in the spinal cord and brain, resulting in motor deficits and muscle atrophy. Approximately 5-10% of ALS patients are familial (fALS), while the rest are sporadic (sALS). Currently, early diagnosis of ALS cannot be achieved based on clinical manifestations and electromyography due to the lack of effective and easily available biomarkers. The skin and central nervous system (CNS) share the same embryonic origin. Several skin biomarkers have been found in many neurodegenerative dis

Epigenetic modifications can influence the phenotypes of subsequent generations through intergenerational and transgenerational effects. The aim of the research was to assess the impact of epigenetic factors acting during embryonic development on the structure and cellular composition of lymphoid organs in three generations of chickens. Two groups of eggs were injected once (in the F1 generation) with a synbiotic (SYNs) or a synbiotic + choline (SYNCHs), while two other groups were injected in each successive generation (F2, F3-SYNr, SYNCHr). Synbiotic administration resulted in an increased c

Throughout my research career, I have focused on understanding the extracellular matrix (ECM) and its roles in disease pathogenesis and the aging process. My initial interest in muscular dystrophies gradually expanded to include various organs and systems, including the kidneys, brain, cartilage, skin, and eyes. During my tenure at the National Institutes of Health (NIH), I made substantial contributions to generating and analyzing conditional knockout mouse models for key ECM molecules, including laminin α1 and perlecan. These studies elucidated the roles of ECM components in hereditary disea

Neurotrophins (NTs) are four small proteins produced by both neuronal and non-neuronal cells; they include nerve growth factor (NGF), brain-derived neurotrophic factor (BDNF), neurotrophin-3 (NT-3), and neurotrophin-4 (NT-4). NTs can exert their action through both genomic and non-genomic mechanisms by interacting with specific receptors. Initial studies on NTs have identified them only as functional molecules of the nervous system. However, recent research have shown that some tissues and organs (such as the lungs, skin, and skeletal and smooth muscle) as well as some structural cells can sec

Oxygen is a fundamental requirement for cellular metabolism. Hypoxia is a state of oxygen deprivation of the tissues. Cells develop numerous adaptive mechanisms to survive hypoxic insult. Alveolar bone is a unique structure that encases and protects the tooth. Literature reports that hypoxia, in all forms, impacts alveolar bone health. The hypoxia-inducible pathway appears to play a key role in mediating changes in alveolar bone metabolism. Embryonic hypoxia plays a vital role in craniofacial skeletal development. Further, hypoxia has been anticipated in the repair of extraction sockets. Alveo

A major barrier to the impact of genomic diagnosis in patients with congenital malformations is the lack of understanding regarding how sequence variants contribute to disease pathogenesis and whether this information could be used to generate patient-specific therapies. Congenital diaphragmatic hernia (CDH) is among the most common and severe of all structural malformations; however, its underlying mechanisms are unclear. We identified loss-of-function sequence variants in the epigenomic regulator gene SIN3A in two patients with complex CDH. Tissue-specific deletion of Sin3a in mice resulted

Despite their distinct embryonic origins, the skull and brain are highly integrated. Understanding the covariation between the skull and brain can shed light on anatomical, cognitive, and behavioral traits in extant and extinct species. Domestic dogs offer a unique opportunity to investigate skull-brain covariation due to their diverse skull morphologies and neural anatomy. To assess this question, we examined T2-weighted MRI studies of 62 dogs from 33 breeds, plus an additional 17 dogs of mixed or unknown breeds. Scans were opportunistically collected from a veterinary teaching hospital of do

Low-density lipoprotein receptor-related protein 6 (LRP6) is a co-receptor of the Wnt signaling pathway, which plays an essential role in various biological activities during embryonic and postnatal development. LRP6 is exceptionally associated with rare diseases and always with autosomal dominant inheritance. Here we report a familial phenotype of high bone mass associated with skeletal anomalies and oligodontia but also persistent left superior vena cava, inguinal hernia, hepatic cysts, abnormal posterior fossa and genital malformations. Molecular analysis revealed a novel heterozygous varia

Rhyzopertha dominica Prip (RdPrip) cDNA was cloned (GenBank accession no. OK318454), and the encoded 276-amino-acid protein indicated the typical aquaporin structure, including six transmembrane regions and two NPA motifs. The developmental and tissue profiles of RdPrip transcription were determined. RdPrip was highly transcribed in female adults, followed by larvae, pupae, and male adults. The transcriptional expression levels of RdPrip were significantly high in the ovary and hindgut (including cryptonephridial systems) compared with the foregut, testis, midgut, and Malpighian tubules. Dominant missense mutations in DNAJB6, a co-chaperone of HSP70, cause limb girdle muscular dystrophy (LGMD) D1. No treatments are currently available. Two isoforms exist, DNAJB6a and DNAJB6b, each with distinct localizations in muscle. Mutations reside in both isoforms, yet evidence suggests that DNAJB6b is primarily responsible for disease pathogenesis. Knockdown treatment strategies involving both isoforms carry risk, as DNAJB6 knockout is embryonic lethal. We therefore developed an isoform-specific knockdown approach using morpholinos. Selective reduction of each isoform was achieved in vit
