Studies on metabolic activation of vinyl chloride in Drosophila melanogaster after pretreatment with phenobarbital and polychlorinated biphenyls.

It is known that vinyl chloride is metabolized by the mixed function oxygenase system in the liver to reactive mutagenic and carcinogenic metabolites. This metabolic activation was studied in Drosophila melanogaster by measuring the uptake of 14C from labelled vinyl chloride in different strains and with different pretreatments with phenobarbital and polychlorinated biphenyl (PCB) Clophen A50), well known inducers of cytochrome P-450. In accordance with previously obtained data on vinyl chloride induced sex linked recessive lethals, it was shown that pretreatment with inducers increased the up

Molecular surveillance of the Plasmodium falciparum Pfmdr1 N86Y mutation linked to antimalarial drug resistance in Mbouda, Cameroon.

BACKGROUND: Malaria, mainly caused by Plasmodium falciparum, remains a major health concern in Cameroon due to the emergence of drug-resistant parasite strains. The Plasmodium falciparum multidrug resistance 1 (Pfmdr1) gene, particularly the N86Y mutation, is a key marker associated with reduced susceptibility to several antimalarial drugs. This study investigated the prevalence of the Pfmdr1-N86Y mutation in isolates from patients attending three health facilities in the Mbouda Health District to support improved malaria control strategies in the area. METHODS: The study was conducted among p

Antimalarial Activity of Solvent Fractions of the Stem Bark of Periploca linearifolia Quart.-Dill. & A.Rich. (Asclepiadaceae) Against Plasmodium berghei in Mice.

BACKGROUND: Currently, malaria elimination is intimidated by the emergence of antimalarial drug and insecticide resistance. This scenario urges scientists to develop new antimalarial drugs, where plant-based compounds are the major candidates. OBJECTIVE: The objective of this experiment was to examine the antimalarial activities of the solvent fractions of the stem bark of P. linearifolia in mice. METHODS: The dried hydromethanolic extract was treated with n-hexane, chloroform, and ethyl acetate solvents based on increasing order of polarity. For the suppressive test, after three hours post-…

Genetic differentiation in the African malaria vector, Anopheles gambiae s.s., and the problem of taxonomic status.

Of the seven recognized species of the Anopheles gambiae complex, A. gambiae s.s. is the most widespread and most important vector of malari … ct discontinuity was observed for this species. Autosomal inversions in one chromosome arm (2R), an insecticide resistance gene on 2L, and this single X-linked region indicate at least two genetically differentiated subpopulations of A. gambiae. Yet, rather extensive studies of other regions of the genome have failed to reveal genetic discontinuity. Evidently, incomplet …

The paper discusses important gaps in our understanding of the ecology of larval habitats, insecticide resistance monitoring, and thorough studies of vector incrimination beyond primary sites. We suggest that the biogeography of the sand fly vectors that transmit leishmaniasis in Pakistan is driven by their dynamic and climate-sensitive nature and, as such, sho …

Plasmodium falciparum Pfs47 haplotype compatibility to Anopheles gambiae in Kisumu, a malaria-endemic region of Kenya.

Insecticide resistance and outdoor transmission have reduced the effectiveness of existing malaria transmission prevention strategies. As a result, targeted approaches to support continuing malaria control, such as transmission-blocking vaccines, are required. Cross-sectional mass blood screening in children between 5 and 15 years was conducted in Chulaimbo, Kisumu, during the dry and wet seasons in 2018 and 2019. Plasmodium falciparum gametocyte carriers were identified by Microscopy. Subsequently, carriers were used to feed colony bred Anopheles gambiae females in serum replacement and whole…

Effects of regional crop rotations on autumn insect pests in winter oilseed rape.

BACKGROUND: Chemical control of insect pests in oilseed rape (OSR) is becoming increasingly difficult due to the development of resistance and restrictive insecticide approvals in Europe. At the same time, there is a lack of preventive and alternative control measures. Crop rotation mostly fails to control insects due to their mobility; however, changing regional cropping densities can dilute or concentrate pest pressure. In this study, we investigated whether the local occurrence of Psylliodes chrysocephala and Delia radicum, serious insect pests in winter OSR, is influenced by distance from

Effects of ivermectin treatment of backyard chickens on mosquito dynamics and West Nile virus transmission.

BACKGROUND: Vector control strategies typically rely on pesticides to target mosquitoes involved in enzootic and zoonotic transmission of West Nile virus (WNV). Nevertheless, increasing insecticide resistance and a desire to reduce pesticide usage provide the impetus for developing alternative strategies. Ivermectin (IVM), an antiparasitic drug which is widely used in human and veterinary medicine, is a potential alternative for targeted control because Culex mosquitoes experience increased mortality following ingestion of IVM in bloodmeals. METHODOLOGY/PRINCIPAL FINDINGS: We conducted a rand…

Microbiome Differences Between Human Head and Body Lice Ecotypes Revealed by 16S RRNA Gene Amplicon Sequencing.

Human head lice and body lice (Pediculus humanus) are neglected ectoparasites. Head lice continue to be prevalent in children worldwide, and insecticide resistance in these insects has complicated their treatment. Meanwhile, body lice, which are most common in the developing world, are resurging among marginalized populations in developed nations. Today, the microbiome is being increasingly recognized as a key mediator of insect physiology. However, the microbial communities that inhabit human lice have remained unknown beyond only a few species of bacteria. Knowledge of the microbiomes of hea…

Experimental hut evaluation of linalool spatial repellent agar gel against Anopheles gambiae sensu stricto mosquitoes in a semi-field system in Bagamoyo, Tanzania.

BACKGROUND: Malaria vector control is in need of new tools to face its current challenges such as the spread of pyrethroid-resistance and the increase of outdoor feeding mosquitoes. New strategies such as spatial repellents need to be evaluated as supplemental tools to existing control measures such as insecticide treated bed nets and indoor residual spraying. Linalool is a naturally occurring terpene alcohol commonly found in flowers and spices with reportedly repellent properties. METHODS: Four experimental huts fitted with exit traps and enclosed inside a large screened semi-field system w

Impact of plant growth-promoting rhizobacteria and natural enemies on Myzus persicae (Hemiptera: Aphididae) infestations in pepper.

Management of green peach aphid, Myzus persicae (Sulzer) (Hemiptera: Aphididae), in bell pepper, Capsicum annuum L., was explored through a combination of plant growth-promoting rhizobacteria (PGPR) and endemic biological control in New York in 2006 and 2007. We hypothesized that by using PGPR-treated peppers 1) M. persicae infestations would be reduced via induced resistance, 2) natural enemies would be lured to plants through the elicitation of volatile organic compounds, and 3) yield amount and quality would be improved. Pepper seed was planted in soil containing the PGPR formulation BioYie

Resistance to Cry14A family Bacillus thuringiensis crystal proteins in Caenornabditis elegans operates via the nhr-31 transcription factor and vacuolar-type ATPase pathway.

Bacillus thuringiensis (Bt) has been successfully used commercially for more than 60 years for biocontrol of insect pests. Since 1996, transgenic plants expressing Bt crystal (Cry) proteins have been used commercially to provide protection against insects that predate on corn and cotton. More recently, Bt Cry proteins that target nematodes have been discovered. One of these, Cry14Ab, has been expressed in transgenic soybean plants and found to provide significant protection against the soybean cyst nematode, Heterodera glycines. However, to date there has been no description of high-level resi
