In vivo molecular imaging analysis of a nasal vaccine that induces protective immunity against botulism in nonhuman primates.

Nasal administration is an effective route for a needle-free vaccine. However, nasally administered Ags have the potential to reach the CNS … ioactivity or the traditional [(111)In]-radiolabel method in dissected tissues of mice and nonhuman primates. We found no deposition of BoHc/A in the cerebrum or olfactory bulb after nasal administration of (18)F-labeled BoHc/A in both animals. We also established a real-time quantitative profile of elimination of this nasal vaccine candidate and demonstra …

Continuous neuromonitoring using transcranial Doppler reflects blood flow during carbon dioxide challenge in primates with global cerebral ischemia.

OBJECTIVE: At present, there is no consensus on the optimal monitoring method for cerebral blood flow (CBF) in neurointensive care patients. The aim of the present study was to investigate whether continuous transcranial Doppler (TCD) monitoring with modulation of partial pressure of CO2 reflects CBF changes. This hypothesis was tested in 2 pathological settings in which cerebral ischemia can be imminent: after an episode of cerebral ischemia and during vasospasm after subarachnoid hemorrhage. METHODS: Sixteen cynomolgus monkeys were divided into 3 groups: 1) chemoregulation in control animal

Efficacy and safety of live attenuated persistent and rapidly cleared Mycobacterium tuberculosis vaccine candidates in non-human primates.

Tuberculosis (TB) remains a global health burden for which safe vaccines are needed. BCG has limitations as a TB vaccine so we have focused on live attenuated Mycobacterium tuberculosis mutants as vaccine candidates. Prior to human studies, however, it is necessary to demonstrate safety in non-human primates (NHP). In this study, we evaluate the safety and efficacy of two live attenuated M. tuberculosis double deletion vaccine strains mc(2)6020 (DeltalysA DeltapanCD) and mc(2)6030 (DeltaRD1 DeltapanCD) in cynomolgus macaques. In murine models, mc(2)6020 is rapidly cleared while mc(2)6030 persi…

Virulence and pathophysiology of the Congo Basin and West African strains of monkeypox virus in non-human primates.

Monkeypox virus is divided into Congo Basin and West African strains. The virulence and pathophysiology of two strains, Zr-599 (a Congo Basin monkeypox virus) and Liberia (a West African monkeypox virus), were evaluated in non-human primates. Four monkeys were infected by the subcutaneous (SC) and two by the intranasal (IN) inoculation routes for Zr-599 and Liberia at a dose of 10(6) p.f.u. One monkey in the Liberia/SC group was demonstrated to be co-infected with Gram-positive cocci and was excluded from analyses. Infections in three of the four Zr-599/SC monkeys and in one of the three Liber…

Causing and curing infantile esotropia in primates: the role of decorrelated binocular input (an American Ophthalmological Society thesis).

PURPOSE: Human infants at greatest risk for esotropia are those who suffer cerebral insults that could decorrelate signals from the 2 eyes during an early critical period of binocular, visuomotor development. The author reared normal infant monkeys, under conditions of binocular decorrelation, to determine if this alone was sufficient to cause esotropia and associated behavioral as well as neuroanatomic deficits. METHODS: Binocular decorrelation was imposed using prism-goggles for durations of 3 to 24 weeks (in 6 experimental, 2 control monkeys). Behavioral recordings were obtained, followed

The effects of selective amygdala, orbital frontal cortex or hippocampal formation lesions on reward assessment in nonhuman primates.

We examined the effects of bilateral amygdaloid, hippocampal or orbital frontal cortex lesions on reward assessment in rhesus monkeys (Macaca mulatta). In Experiment 1, basic preferences for foods and inedible nonfoods were measured pre- and postsurgery. None of the lesions produced changes in animals' preferences for palatable foods or raw meat relative to presurgery, although amygdaloid or hippocampal lesions yielded increased preference for inedible nonfoods postsurgery. When the reinforcement value of each animal's highest-preferred food was decreased by selective satiation, only animals w

Assessing the mammary gland of nonhuman primates: effects of endogenous hormones and exogenous hormonal agents and growth factors.

This review provides a summary of the normal biology, development, and morphology of the breast in nonhuman primates (macaques), and of the major published work addressing hormonally-induced changes in the breast of these animals. The mammary glands of macaques are anatomically, developmentally, and physiologically similar to the human breast, with similar expression of sex steroid receptors (estrogen receptors alpha and beta, progesterone receptor A and B, androgen receptors), estrogen dependent markers, and steroid metabolizing enzymes. Genetic similarity between human beings and macaques is…

Chronic oral nicotine normalizes dopaminergic function and synaptic plasticity in 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine-lesioned primates.

Our recent studies show that chronic oral nicotine partially protects against striatal damage in 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-treated nonhuman primates. To identify the cellular changes associated with this protective action, we investigated the effects of nicotine treatment on stimulus-evoked dopamine release, dopamine turnover, and synaptic plasticity in striatum from lesioned and unlesioned animals. Monkeys were chronically (6 months) treated with nicotine in the drinking water and subsequently lesioned with the dopaminergic neurotoxin MPTP (6 months) while nicotine w…

Long-term effects of early-life environmental manipulations in rodents and primates: Potential animal models in depression research.

Depression is one of the most common human illnesses and is of immense clinical and economic significance. Knowledge of the neuro-psychology … bnormalities, across the life span. Studies of postnatal environmental manipulations in rodents and primates can potentially yield evidence that abnormal early-life experience leading to dysfunction of the neurobiology, physiology and behaviour of emotion is a general mammalian characteristic, and therefore, that this approach can be used to develop animal …

Attenuated, replication-competent herpes simplex virus type 1 mutant G207: safety evaluation of intracerebral injection in nonhuman primates.

This study examined the safety of intracerebral inoculation of G207, an attenuated, replication-competent herpes simplex virus type 1 (HSV-1) recombinant, in nonhuman primates. Sixteen New World owl monkeys (Aotus nancymae [karyotype 1, formerly believed to be A. trivirgatus]), known for their exquisite susceptibility to HSV-1 infection, were evaluated. Thirteen underwent intracerebral inoculation with G207 at doses of 10(7) or 10(9) PFU, two were vehicle inoculated, and one served as an infected wild-type control and received 10(3) PFU of HSV-1 strain F. HSV-1 strain F caused rapid mortality …

Material of tongues of Tupaia glis, Microcebus murin … illae filiformes and fungiformes at the apex linguae; the same seems to be true for other genera of primates. It is self-explanatory that the accumulation of papillae makes the tip of the tongue a particularly sensitive area; this has been found to be true even in man. Intestinal absorption, liver uptake, and excretion of 3H-folic acid in folic acid-deficient, alcohol-consuming nonhuman primates.

Nonhuman primates fed folic acid-deficient diets +/- 30% kcal ethanol were used to determine alcohol effects on megaloblastic anemia development and folate bioavailability. Lower hemoglobin (Hb) and red blood cell (RBC) counts and higher mean corpuscular volume (MCV) occurred after 13 wk in alcohol-fed monkeys, later in controls. Plasma, RBC, and liver folate declined and urinary formiminoglutamic acid (FIGLU) was elevated in both groups with FIGLU increasing more among alcohol-fed monkeys at 38 wk. After 40 wk, the bioavailability of oral 3H-folic acid was investigated and showed increased fe…

Representations of the body surface in cortical areas 3b and 1 of squirrel monkeys: comparisons with other primates.

Microelectrode multiunit mapping techniques were used to determine the somatotopic organization of postcentral parietal cortex in the squirrel monkey Saimiri sciurus. Recordings were largeLy confined to architectonic areas 3b and 1. Results were compared to those from similar studies of owl (Merzenich et al., '78) and macaque (Nelson et al., '80) monkeys. As in these previous investigations, separate representations of the body surface were found in areas 3b and 1 of squirrel monkeys. These representations were organized in parallel, so that both proceeded from the tail on the medial wall of t

A comparative study of the toxicity of Fusarium verticillioides (= F. moniliforme) to horses, primates, pigs, sheep and rats.

An isolate of Fusarium verticillioides (MRC826) that induced experimental leukoencephalomalacia, also caused acute toxicity when fed to pigs and administered per rumen fistula to sheep. Pigs developed severe pulmonary oedema while sheep manifested severe nephrosis and hepatosis. A less toxic isolate (F. verticillioides MRC602), fed to baboons, resulted in acute congestive heart failure or hepatic cirrhosis, depending on the dose. Both isolates were toxic to rats and caused similar lesions, namely, hepatic cirrhosis and intraventricular cardiac thrombosis.

Influenza A virus infection induces immune dysregulation in the placenta and fetus without vertical transmission in nonhuman primates.

Influenza A virus (IAV) infection during pregnancy is associated with stillbirth and preterm birth. We hypothesized that maternal IAV infection disrupts placental and fetal immune networks in ways that correlate with disease severity. Pregnant pigtail macaques were inoculated with IAV [A/California/07/2009 (H1N1)] and underwent delivery and fetal necropsy 5 days postinfection (N = 11), with comparison to uninfected controls (N = 16). Stillbirth occurred in 18% of infected pregnancies and in no controls. Although vertical transmission was not observed, low levels of viral RNA were detected in t

Glymphatic dysfunction contributes to thalamic iron retention and secondary thalamic injury after stroke: evidence from primates and rodents.

BACKGROUND: Secondary neurodegeneration, characterized by neuronal loss and neuroinflammation, in the remote thalamus is associated with post-stroke cognitive impairment (PSCI). This study aimed to elucidate common pathological mechanism of the secondary neurodegeneration in both primates and rodents. METHODS: Thalamic amyloid-β (Aβ), iron deposition and glymphatic dysfunction was assessed across primate and rodent stroke models in different time points, using histopathological, and magnetic resonance imaging methods. Proteomic analysis was performed to explore the molecular mechanisms underly…

Dose-dependent power and connectivity modulation of low frequency oscillations through transcranial magnetic stimulation in non-human primates.

Transcranial magnetic stimulation (TMS) is a powerful non-invasive tool for safely modulating neural activity in humans. In particular, the … , we investigate the effects of single-pulse TMS applied to the left prefrontal cortex in non-human primates on LFPs recorded through intracranial EEG. Compared to sham TMS, the intensity of active TMS pulses scaled with LFP power changes in a 1-13 Hz range at contacts close to the stimulation site in the prefrontal cortex (e.g., caudate nucleus, anterior …
