The impact of nutritional, environmental, and lifestyle factors on neurological disorders: therapeutic implications and mechanistic insights.

Neurological disorders like Alzheimer's, Parkinson's, multiple sclerosis, and primary psychiatric conditions are complex, arising from a mix of genetic and modifiable risks. Growing evidence indicates that nutrition, environment, and lifestyle significantly influence disease development, progression, and treatment response. Nutrients such as vitamins, minerals, omega-3 fatty acids, and polyphenols affect neuroinflammation, oxidative stress, mitochondrial health, and neurotransmitter function. Dietary patterns like the Mediterranean and ketogenic diets offer protective benefits in clinical and

PHD3-Mediated Inhibition of Retinal Neovascularization in Retinopathy of Prematurity.

Retinopathy of prematurity is characterised by abnormal retinal neovascularization in response to hypoxia stress. Prolyl 4-hydroxylase domain protein 3 (PHD3) is a well-known molecular oxygen sensor. However, the role that PHD3 plays in retinopathy of prematurity remains unclear. In this work, a mouse model of oxygen-induced retinopathy (OIR) was used for in vivo studies. Compared with the mice in room air, OIR mice showed sprouting of retinal neovascularization and increased level of PHD3. It was further found that PHD3 overexpression weakened OIR-induced retinal neovascularization and promot

Unique immune and other responses of human nasal epithelial cells infected with H5N1 avian influenza virus compared to seasonal human influenza A and B viruses.

Highly pathogenic avian influenza (HPAI) virus (e.g. H5N1) infects the lower airway to cause severe infections, and constitute a prime candidate for the emergence of disease X. The nasal epithelium is the primary portal of entry for respiratory pathogens, serving as the airway's physical and immune barrier. While HPAI virus predominantly infects the lower airway, not much is known about its interactions with the nasal epithelium. Hence, we sought to elucidate and compare the differential responses of the nasal epithelium against HPAI infection that may contribute to its pathology, and to ident

Allogeneic fibroblast sheets prevent pulmonary air leaks caused by rat pleural defects without adhesion to the thoracic wall.

INTRODUCTION: Pulmonary air leak (PAL) is one of a complication of thoracic surgery and an unavoidable problem after lung resection or pleural adhesion detachment. Conventional procedures to close PAL by applying polyglycolic acid (PGA) sheets are prone to pleural adhesion. This study evaluated the ability of allogeneic fibroblast sheet transplantation to prevent PALs. METHODS: Rat skin fibroblasts were prepared from transgenic rats expressing green fluorescent protein (GFP) and cultured on temperature-responsive culture dishes to harvest fibroblast sheets. Allogeneic fibroblast sheets or PGA

Pentraxin-related protein PTX3 alleviates blast-induced acute lung injury by promoting efferocytosis to eliminate ferroptotic alveolar epithelial cells in vitro.

Blast injuries are common in both peacetime and war, and one of the main types is lung injury, characterised by internal severity, rapid changes in condition, high mortality, and complex injury patterns. The primary pathological manifestation is pulmonary haemorrhage caused by damage to alveolar epithelial cells (AECs) and capillary endothelial cells. Treatment is complex, making it essential to fully understand the characteristics and pathogenesis of blast-induced acute lung injury (bALI) to improve treatment efficacy. In this study, a rat blast-injury model was established through a fuel-air

Effects of elevated atmospheric carbon dioxide to Microcystis aeruginosa under different forms of phosphorus sources.

Human activities have led to an increase in atmospheric carbon dioxide (CO2) concentration, which can enhance the flux of CO2 from air to water, thus impacting algal growth. Phosphorus (P) is a key factor influencing the formation of cyanobacteria blooms. Nutrient utilization is closely related to carbon (C) metabolism, but the effects of elevated CO2 on microalgae under different P sources are rarely studied. In this study, we investigated the growth and physiological and biochemical responses of Microcystis aeruginosa (M. aeruginosa) under ambient (400 ppm) and elevated (550 ppm) CO2 levels

Associated transcription factors of allergic rhinitis: SP1 aggravates nasal epithelial barrier damage.

Allergic rhinitis (AR) is a respiratory airway disorder characterized by inflammation and barrier dysfunction. The transcription factor specificity protein 1 (SP1), a member of the Sp/Krüppel-like factor family, is known to be associated with inflammation. This study aimed to explore the impacts of SP1 on AR. An AR murine model was developed through multiple intranasal challenges with ovalbumin (OVA). Human nasal epithelial cells (HNEpCs) were cultured at the air-liquid interface (ALI) and treated with IL-13 to create a cell model with barrier damage. Bioinformation samples revealed that SP1 e

Auranofin Ameliorates Gouty Inflammation by Suppressing NLRP3 Activation and Neutrophil Migration via the IL-33/ST2-CXCL1 Axis.

Gout is a form of sterile inflammatory arthritis in which monosodium urate (MSU) crystals deposit and provoke a neutrophil-predominant response, primarily driven by activation of the NACHT, leucine-rich repeat, and pyrin domain-containing protein 3 (NLRP3) inflammasome. Here, we show that auranofin, a Food and Drug Administration (FDA)-approved anti-rheumatic agent, exerts anti-inflammatory effects in both in vitro and in vivo models of gout. Auranofin inhibited NLRP3 inflammasome activation in human THP-1 cells and murine macrophages, leading to reduced cleavage of caspase-1, interleukin-1β (

Evaluation of the protective efficacy of six major immunogenic proteins of Mycoplasma Synoviae.

Mycoplasma synoviae (MS) is a primary avian pathogen prevalent worldwide that causes airsacculitis and synovitis in birds. Vaccination is recommended as the most cost-effective strategy in the control of MS infection. Novel alternative vaccines are needed for eradicating and controlling MS infection in flocks. DnaK, enolase, elongation factor Tu (EF-Tu), MSPB, NADH oxidase and LP78 are the major immunogenic antigens of MS and are promising targets for subunit vaccine candidates. In the present study, genes encoding DnaK, enolase, EF-Tu, MSPB, LP78, and NADH oxidase were cloned and expressed in

Deafness due to loss of a TRPV channel eliminates mating behavior in Aedes aegypti males.

Attraction and mating between male and female animals depend on effective communication between conspecifics. However, in mosquitoes, we have only a rudimentary understanding of the sensory cues and receptors critical for the communication that is essential for reproductive behavior. While it is known that male Aedes aegypti use sound to help them identify females, it is not unclear whether sound detection is absolutely required since other cues such as vision may also participate in mating behavior. To determine the effect of eliminating hearing on mating success, we knocked out the Ae. aegyp

Muscle and Muscle-like Autoantigen Expression in Myasthenia Gravis Thymus: Possible Molecular Hint for Autosensitization.

The thymus is widely recognized as an immunological niche where autoimmunity against the acetylcholine receptor (AChR) develops in myasthenia gravis (MG) patients, who mostly present thymic hyperplasia and thymoma. Thymoma-associated MG is frequently characterized by autoantibodies to the muscular ryanodine receptor 1 (RYR1) and titin (TTN), along with anti-AChR antibodies. By real-time PCR, we analyzed muscle-CHRNA1, RYR1, and TTN-and muscle-like-NEFM, RYR3 and HSP60-autoantigen gene expression in MG thymuses with hyperplasia and thymoma, normal thymuses and non-MG thymomas, to check for mole

Club cell protein (CC16) in serum as an effect marker for small airway epithelial damage caused by diesel exhaust and blasting fumes in potash mining.

OBJECTIVE: The effect marker club cell protein (CC16) is secreted by the epithelium of the small respiratory tract into its lumen and passes into the blood. Increased amounts of CC16 in serum are observed during acute epithelial lung injury due to air pollutants. CC16 in serum was determined as part of this cross-sectional study in underground potash miners on acute and chronic health effects from exposures to diesel exhaust and blasting fumes. METHODS: Nitrogen oxides, carbon monoxide, and diesel particulate matter were measured in 672 workers at a German potash mining site on a person-by-pe

Chylous ascites and pancreatic pseudocyst on a child following blunt abdominal trauma; a case report.

INTRODUCTION: Chylous ascites is the collection of milky-like fluid rich in triglycerides within the peritoneal cavity. It results from disruption of normal intestinal lymphatic flow. It is caused mainly by congenital anomalies, trauma, and malignancy. Chylous ascites following blunt abdominal injury is uncommon in pediatrics. Chyle duct and pancreatic injuries present a rare clinical sequela yet to be reported in the literature. Conservative management is the mainstay of treating chyle duct injuries, reserving invasive measures for unique circumstances. CASE PRESENTATION: A case of an eleven

Nitrated fatty acid, 10-nitrooleate protects against hyperoxia-induced acute lung injury in mice.

The antioxidant and anti-inflammatory effects of electrophilic nitrated fatty acid (NFA); 10-nitrooleate, have been reported. The present study investigated whether 10-nitrooleate has a protective role against hyperoxic-induced acute lung injury (HALI). Using a C57BL/6 mice model of HALI, we investigated the protective effect of 10-nitrooleate. C57BL/6 mice were administered with NFA intratracheally, exposed to hyperoxia for 48 h to induce HALI, and kept at room air for 24 h. Bronchoalveolar lavage (BAL) fluid and lung samples were collected after 24 h of post hyperoxia to analyze markers asso

Melatonin alleviates PM2.5 -induced glucose metabolism disorder and lipidome alteration by regulating endoplasmic reticulum stress.

Exposure to fine particulate matter (PM2.5 ) was associated with an increased incidence of liver metabolic disease. Melatonin has been shown to prevent liver glucolipid metabolism disorders. However, whether melatonin could rescue PM2.5 -induced liver metabolic abnormalities remains uncertain. This study was to evaluate the mitigating effect of melatonin on PM2.5 -accelerated hepatic glucose metabolism imbalance in vivo and in vitro. Schiff periodic acid shiff staining and other results showed that PM2.5 led to a decrease in hepatic glycogen reserve and an increase in glucose content, which wa
