Abatement of acrylonitrile-induced gastric injury by 2-Methoxyestradiol through inhibition of TLR4/MYD88/phospho-p38 MAPK axis.

Acrylonitrile (ACN) is a widely used industrial chemical. It has a wide range of toxicities, including gastric mucosal damage. 2-Methoxyestradiol (2ME) is one of the estrogen metabolites with a plethora of beneficial pharmacological activities. This study aimed to evaluate the possible preventive effects of 2ME against ACN-induced gastric injury in rats. 2ME was injected intraperitoneally into rats at two dose levels (1 and 5 mg/kg) prior to a single oral dose of ACN (30 mg/kg). 2ME obviously prevented histopathological alteration in gastric tissues. 2ME prevented ACN-induced oxidative damage

An ERRα-ZEB1 transcriptional signature predicts survival in triple-negative breast cancers.

BACKGROUND: Transcription factors (TFs) act together with co-regulators to modulate the expression of their target genes, which eventually dictates their pathophysiological effects. Depending on the co-regulator, TFs can exert different activities. The Estrogen Related Receptor α (ERRα) acts as a transcription factor that regulates several pathophysiological phenomena. In particular, interactions with PGC-1 co-activators are responsible for the metabolic activities of ERRα. In breast cancers however, ERRα exerts several tumor-promoting, metabolism-unrelated activities that do not depend on PGC

The role of probiotics in modulating RANKL in osteoporosis: A systematic review of animal interventional studies and RCTs.

BACKGROUND: Osteoporosis is a major global health concern characterized by reduced bone mass and structural deterioration. The RANK/RANKL/OPG signaling pathway plays a pivotal role in bone remodeling, particularly in osteoclastogenesis. Emerging evidence suggests that probiotics may modulate this pathway through the gut-bone axis, offering potential therapeutic benefits for bone loss. OBJECTIVE: This systematic review aims to evaluate the effects of probiotic supplementation on Receptor activator of nuclear factor kappa-Β ligand (RANKL) in animal and human interventional studies related to os

Phenolic-derived compounds in osteoporosis-Mechanisms, clinical evidence, and drug delivery: A review.

Osteoporosis is a degenerative skeletal disorder characterized by reduced bone mass and the deterioration of bone microarchitecture, resulting in an increased risk of fractures. Its development is driven by an imbalance in bone remodeling, where osteoclastic bone resorption surpasses osteoblastic bone formation. Factors such as oxidative stress, chronic inflammation, ferroptosis, and hormonal changes, particularly estrogen deficiency in postmenopausal women, contribute to this imbalance. Metabolites derived from phenolic compounds have emerged as promising natural agents for osteoporosis preve

Raloxifene prevents stress granule dissolution, impairs translational control and promotes cell death during hypoxia in glioblastoma cells.

Glioblastoma (GBM) is the most common primary malignant brain tumor, and it has a uniformly poor prognosis. Hypoxia is a feature of the GBM microenvironment, and previous work has shown that cancer cells residing in hypoxic regions resist treatment. Hypoxia can trigger the formation of stress granules (SGs), sites of mRNA triage that promote cell survival. A screen of 1120 FDA-approved drugs identified 129 candidates that delayed the dissolution of hypoxia-induced SGs following a return to normoxia. Amongst these candidates, the selective estrogen receptor modulator (SERM) raloxifene delayed S

An efficient inducible RPE-Selective cre transgenic mouse line.

Cre-mediated modulation of gene function in the murine retinal pigment epithelium (RPE) has been widely used, but current postnatal RPE-selective Cre driver lines suffer from limited recombination efficiency and/or ectopic or mosaic expression. We sought to generate a transgenic mouse line with consistently efficient RPE-selective Cre activity that could be temporally regulated. We used ϕC31 integrase to insert a DNA construct encoding a human BEST1 promoter fragment driving a Cre recombinase estrogen receptor fusion (BEST1-CreERT2) at the Rosa26 locus of C57BL/6J mice. Rosa26BEST1-CreERT2 mic

Testosterone and Vaginal Function.

INTRODUCTION: Androgens have been shown to exert beneficial effects on vaginal physiology, at least partially independent of their aromatization to estrogens. Androgen deficiency in the vagina and in the other genitourinary tissues contributes to the development of vulvovaginal atrophy and genitourinary syndrome of menopause, resulting in impaired arousal and lubrication and dyspareunia. OBJECTIVES: To summarize the role of testosterone in modulating vaginal structure and function. METHODS: A qualitative review of the relevant literature on the topic was performed using the PubMed database.

Impact of 27-hydroxylase (CYP27A1) and 27-hydroxycholesterol in breast cancer.

The impact of systemic 27-hydroxycholesterol (27HC) and intratumoral CYP27A1 expression on pathobiology and clinical response to statins in breast cancer needs clarification. 27HC is an oxysterol produced from cholesterol by the monooxygenase CYP27A1, which regulates intracellular cholesterol homeostasis. 27HC also acts as an endogenous selective estrogen receptor (ER) modulator capable of increasing breast cancer growth and metastasis. 27HC levels can be modulated by statins or direct inhibition of CYP27A1, thereby attenuating its pro-tumorigenic activities. Herein, the effect of statins on s

Hepatic toxicological and multi-omics alterations associated with short- and long-term nonylphenol exposure in mice.

Nonylphenol (NP) is an alkylphenol environmental contaminant with endocrine-disrupting activity and potential hepatotoxicity. However, the molecular responses associated with different NP exposure durations remain insufficiently characterized. In this study, male Kunming mice were exposed to NP through drinking water at 500 μg/L for 10 days (NP-S) or 90 days (NP-L) (n = 8/group), and hepatic toxicological alterations were evaluated using biochemical, histopathological, transcriptomic, metabolomic, and integrative analyses. Network toxicology analysis was first applied as a hypothesis-generatin

Association between breastfeeding, mammographic density, and breast cancer risk: a review.

BACKGROUND: Mammographic density has been associated with breast cancer risk, and is modulated by established breast cancer risk factors, such as reproductive and hormonal history, as well as lifestyle. Recent epidemiological and biological findings underscore the recognized benefits of breastfeeding in reducing breast cancer risk, especially for aggressive subtypes. Current research exploring the association among mammographic density, breastfeeding, and breast cancer is sparse. MAIN FINDINGS: Changes occur in the breasts during pregnancy in preparation for lactation, characterized by the pr

[Endocrine adjuvant treatment specific features for young breast cancer women].

For young patients with hormone receptor-positive early breast cancer, tamoxifen for at least 5 years is the standard endocrine treatment. Prolonged endocrine treatment over 5 years is recommended for patients with high risk of late relapse. When adjuvant chemotherapy is indicated, prolonged iatrogenic amenorrhea is a strong pronostic factor. WIthout persistant amenorrhea after chemotherapy, it is indicated to associate ovarian suppression to the endocrine treatment. Optimal duration of this induced amenorrhea is unknown.

Bazedoxifene protects cerebral autoregulation after traumatic brain injury and attenuates impairments in blood-brain barrier damage: involvement of anti-inflammatory pathways by blocking MAPK signaling.

OBJECTIVE: Traumatic brain injury (TBI) is a significant cause of death and long-term deficits in motor and cognitive functions for which there are currently no effective chemotherapeutic drugs. Bazedoxifene (BZA) is a third-generation selective estrogen receptor modulator (SERM) and has been investigated as a treatment for postmenopausal osteoporosis. It is generally safe and well tolerated, with favorable endometrial and breast safety profiles. Recent findings have shown that SERMs may have therapeutic benefits; however, the role of BZA in the treatment of TBI and its molecular and cellular

Female hormones enhance skin T cell expansion and suppress IFN-γ production.

INTRODUCTION: Estrogen receptors (ERs) regulate immune function, but their expression across immune cell subsets and hormone responsiveness in human skin are not fully understood. METHODS: We used 10-color flow cytometry to quantify ER (ERα, ERβ, and GPER1) expression in peripheral blood mononuclear cells (PBMCs) from healthy donors. We analyzed sex hormone gene expression using public single-cell RNA datasets. Skin immune responses to sodium lauryl sulfate (SLS) were assessed in pre- and post-menopausal women, with or without hormone replacement therapy (HRT). Furthermore, we measured the pr

ERβ mediates sex-specific protection in the App-NL-G-F mouse model of Alzheimer's disease.

BACKGROUND: Menopausal loss of neuroprotective estrogen is thought to contribute to the sex differences in Alzheimer's disease (AD). Activation of estrogen receptor beta (ERβ) can be clinically relevant since it avoids the adverse systemic effects of ERα activation. However, very few studies have explored ERβ-mediated neuroprotection in AD, and no information on its contribution to the sex differences in AD exists. In the present study, we specifically explored the role of ERβ in mediating sex-specific protection against AD pathology in the AppNL-G-F knock-in mouse model of amyloidosis, and if
