Biliary excretion of acetaminophen-glutathione as an index of toxic activation of acetaminophen: effect of chemicals that alter acetaminophen hepatotoxicity.

Acetaminophen (AA) is converted, presumably by cytochrome P-450, to an electrophile which is conjugated with glutathione (GS). AA-GS is excreted into bile, therefore the biliary excretion rate of AA-GS may reflect the rate of activation of AA in vivo. In order to test this hypothesis, the effect of agents capable of altering the activation of AA including cytochrome P-450 inducers and inhibitors, cobaltous chloride which decreases the amount of P-450, prostaglandin synthetase inhibitors (indomethacin and naproxen), antioxidants (butylated hydroxyanisole, alpha-tocopherol, ascorbic acid and asc

Hepatoprotective action of Dendrobium officinale polysaccharide against MAFLD: modulation of gut microbiota-metabolite cascade controls TLR4/NF-κB inflammatory response.

INTRODUCTION: Metabolic dysfunction-associated fatty liver disease (MAFLD) threatens public health worldwide, and Dendrobium officinale polysaccharide (DOP) exhibits promising hepatoprotective potential, yet its regulatory mechanism targeting the gut microbiota-metabolite-TLR4/NF-κB inflammatory axis remains poorly defined. METHODS: In this study, in-vivo rat and in-vitro HepG2 cell MAFLD models were established. Male Sprague-Dawley (SD) rats received 12-week high-fat diet (HFD) feeding to induce MAFLD; HepG2 cells were exposed to a 1 mmol/L free fatty acid (FFA) mixture (oleic acid/palmitic

Salvianolic acid B ameliorates metabolic dysfunction-associated steatohepatitis in mice by inhibiting hepatocyte ferroptosis via regulation of the SIRT1/p53 axis.

BACKGROUND: MASH has become the most prevalent chronic liver disease worldwide. Hepatocyte ferroptosis is a key driver in the progression of MASH. Salvianolic acid B (Sal B), a natural antioxidant active ingredient, has an anti-MASH effect. However, the underlying mechanism by which Sal B inhibits hepatocyte ferroptosis and improves MASH remains unclear. METHODS: An in vivo MASH model was established by feeding C57BL/6J mice a HFHC diet for 30 weeks. In the 24th week, mice were treated with Sal B, obeticholic acid (OCA) or SRT1720. In vitro, over-expression and knockdown of SIRT1 in hepatocyt

Metabolic associated fatty liver disease induced hepatocyte apoptosis to eliminate hepatitis B virus.

BACKGROUD AND AIM: With the rising prevalence of chronic hepatitis B (CHB) co-occurring with metabolic dysfunction-associated fatty liver disease (MAFLD), an in-depth investigation into the impact and mechanisms of MAFLD on CHB is urgently needed. Prior studies have reported that patients with concomitant CHB and MAFLD exhibit lower levels of hepatitis B virus (HBV) replication; however, the underlying mechanistic basis remains unclear. Accumulating evidence indicates that induction of apoptosis is a critical mechanism for HBV clearance. Therefore, it remains to be determined whether lipotoxic

Evaluation of phytochemicals and essential oils of Cupressus semprevirens in controlling cattle tick Rhipicephalus annulatus (Acari: Ixodidae).

BACKGROUND: Cupressus sempervirens is one of the conifer plants, that is used as an antimicrobial, antioxidant, anthelminthic, and many other health purposes. Rhipicephalus annulatus is one of the hard tick genera affecting the production and health of domestic animals in Egypt. Extensive use of chemical acaricides in the management of ticks caused acaricide resistance, environmental contamination, residues in meat and milk, and harmful effects on non-target species. For these reasons, there is an urgent need to create efficient, environmentally friendly acaricides. This work aimed to assay th

Effects of 1-oleate-2-palmitate-3-linoleate glycerol supplementation on the small intestinal development and gut microbial composition of neonatal mice.

Recent studies have shown that 1-oleo-2-palmito-3-linoleyl glycerol (OPL) is the most abundant triacylglycerol in human breast milk in China. Epidemiologic studies have shown that sn-2 palmitate improves the absorption of fatty acids and calcium in infants. However, there have been few studies of the specific mechanism by which OPL affects intestinal function. In the present study, we have characterized the effects of various levels of OPL supplementation on the development of the intestinal epithelium and the intestinal microbiota of neonatal mice. OPL supplementation increased the body masse

Inhibitory Effect of Styrylpyrone Extract of Phellinus linteus on Hepatic Steatosis in HepG2 Cells.

The prevalence of nonalcoholic fatty liver disease (NAFLD) is estimated to be approximately about 25.24% of the population worldwide. NAFLD is a complex syndrome and is characterized by a simple benign hepatocyte steatosis to more severe steatohepatitis in the liver pathology. Phellinus linteus (PL) is traditionally used as a hepatoprotective supplement. Styrylpyrone-enriched extract (SPEE) obtained from the PL mycelia has been shown to have potential inhibition effects on high-fat- and high-fructose-diet-induced NAFLD. In the continuous study, we aimed to explore the inhibitory effects of SPE

Metabolomics reveals that CAF-derived lipids promote colorectal cancer peritoneal metastasis by enhancing membrane fluidity.

Patients with peritoneal metastasis (PM) of colorectal cancer (CRC) have poorer overall survival outcomes than those without PM. Cancer-associated fibroblasts (CAFs) are a major component of the tumor microenvironment and mediate CRC progression and PM. It is imperative to identify and develop novel therapeutic targets for PM-CRC driven by CAFs. Using lipidomics, we reveal that the abundance of phosphatidylcholine (PC) with unsaturated acyl chains was increased in clinical PM-CRC specimens. Additionally, we found that CAFs were present at a higher relative abundance in primary PM-CRC tumors an

A fatty acid-induced decrease in pyruvate dehydrogenase activity is an important determinant of beta-cell dysfunction in the obese diabetic db/db mouse.

We studied the effects of fatty acid oxidation on insulin secretion of db/db mice and underlying molecular mechanisms of these effects. At 2-3 months of age, db/db mice were markedly obese, hyperglycemic, and hyperinsulinemic. Serum free fatty acid (FFA) levels were increased in 2-month-old (1.5 +/- 0.1 vs. 1.1 +/- 0.1 mmol/l, P < 0.05) and 3-month-old (1.9 +/- 0.1 vs. 1.2 +/- 0.1 mmol/l, P < 0.01) mice compared with the age and sex-matched db/+ mice serving as controls. Glucose-induced insulin release from db/db islets was markedly decreased compared with that from db/+ islets and was specifi

Circulating endocannabinoid levels in SARS-CoV-2 infection and their potential role in the inflammatory response.

Plasma levels of endocannabinoids (eCBs) are very dynamic and variable in different circumstances and pathologies. The aim of the study was to determine the levels of the main eCBs and N-acylethanolamines (NAEs) in COVID-19 patients during the acute and post-acute phase of SARS-CoV-2 infection. Samples collected before December 31, 2020 were used for the determination of circulating eCB levels by liquid chromatography-tandem mass spectrometry (LC-MS/MS). The association between plasma eCB measurements and biochemical and hematological parameters, as well as serum IL-6 levels, was evaluated. Sa

Clinical applications of 3-hydroxy fatty acid analysis by gas chromatography-mass spectrometry.

BACKGROUND: L-3-Hydroxy fatty acids are unusual metabolites and rarely occur in significant quantities in normal human physiology. Genetic defects of both long-chain and medium-/short-chain mitochondrial L-3 hydroxyacyl coenzyme A dehydrogenases (LCHAD, M/SCHAD) have been identified as significant metabolic diseases in humans often with severe clinical phenotypes and pathophysiology that appears to differ from other defects of straight chain fatty acid oxidation. It is felt that accumulation of these atypical fatty acid species may play a role in this pathology. We have therefore developed an

Possible Anandamide and Palmitoylethanolamide involvement in human stroke.

BACKGROUND: Endocannabinoids (eCBs) are ubiquitous lipid mediators that act on specific (CB1, CB2) and non-specific (TRPV1, PPAR) receptors. Despite many experimental animal studies proved eCB involvement in the pathogenesis of stroke, such evidence is still lacking in human patients. Our aim was to determine eCB peripheral levels in acute stroke patients and evaluate their relationship with clinical disability and stroke volume. METHODS: A cohort of ten patients with a first acute (within six hours since symptoms onset) ischemic stroke and a group of eight age- and sex-matched normal subject

LGR6 couples maresin 1 signaling to glycolytic flux and protein lactylation for the rescue of diabetic cardiomyopathy.

BACKGROUND: Diabetic cardiomyopathy (DbCM) is driven by metabolic inflexibility and impaired mitochondrial clearance. While the pro-resolving lipid mediator Maresin 1 (MaR1) shows therapeutic promise, its capacity to correct the underlying metabolic-epigenetic disturbances in DbCM remains undefined. MATERIALS: Db/db mice and palmitic acid (PA)-exposed H9c2 cardiomyocytes were treated with MaR1. Cardiac function and structure were assessed by echocardiography and histological analysis. Myocardial oxidative stress, inflammation, and key molecular pathways were evaluated via immunoblotting, qPCR

CPT1A alters granulosa cell proliferation and steroidogenic activity in association with fatty acid oxidation during goat follicular development.

Follicular development in goats relies on the extensive proliferation and hormone synthesis of granulosa cells (GCs), processes that require an adequate energy supply. Fatty acid oxidation (FAO) represents a primary energy source for granulosa cells. However, how its rate-limiting enzyme, carnitine palmitoyltransferase 1A (CPT1A), modulates granulosa cell function remains unclear. This study examined CPT1A expression in healthy and atretic follicles and investigated its association with FAO-associated gene expression, ATP generation, proliferation, steroidogenesis, and palmitic acid (PA)-induc

Carnitine deficiency alters fuel metabolism and voluntary wheel running in mice.

Carnitine plays an obligatory role in energetics owing to its role in the translocation of long-chain fatty acids into the mitochondrion for oxidation. Here, we determined the metabolic and behavioral consequences of systemic carnitine deficiency (SCD) in mice. Female C57BL/6J mice were randomized to receive normal drinking water (control, n = 8) or drinking water supplemented with mildronate 4g.L-1 (mildronate, n = 8) for 21 days. Body composition was assessed at baseline and post-treatment. Metabolic and behavioral phenotyping was performed continuously over 72 h following 14 days of control
