Opportunistic Pathogens in Cystic Fibrosis: Epidemiology and Pathogenesis of Lung Infection.

Cystic fibrosis (CF) is one of the most common life-shortening genetic diseases in Caucasians. Due to abnormal accumulation of mucus, respiratory failure caused by chronic infections is the leading cause of mortality in this patient population. The microbiology of these respiratory infections includes a distinct set of opportunistic pathogens, including Pseudomonas aeruginosa, Burkholderia spp., Achromobacter spp., Stenotrophomonas maltophilia, anaerobes, nontuberculous mycobacteria, and fungi. In recent years, culture-independent methods have shown the polymicrobial nature of lung infections,

Atypical microbial keratitis.

Atypical microbial keratitis refers to corneal infections caused by micro-organisms not commonly encountered in clinical practice. Unlike infections caused by common bacteria, cases of atypical microbial keratitis are often associated with worse clinical outcomes and visual prognosis. This is due to the challenges in the identification of causative organisms with standard diagnostic techniques, resulting in delays in the initiation of appropriate therapies. Furthermore, due to the comparatively lower incidence of atypical microbial keratitis, there is limited literature on effective management

Structural Insights into TMB-1 and the Role of Residues 119 and 228 in Substrate and Inhibitor Binding.

Metallo-β-lactamases (MBLs) threaten the effectiveness of β-lactam antibiotics, including carbapenems, and are a concern for global public health. β-Lactam/β-lactamase inhibitor combinations active against class A and class D carbapenemases are used, but no clinically useful MBL inhibitor is currently available. Tripoli metallo-β-lactamase-1 (TMB-1) and TMB-2 are members of MBL subclass B1a, where TMB-2 is an S228P variant of TMB-1. The role of S228P was studied by comparisons of TMB-1 and TMB-2, and E119 was investigated through the construction of site-directed mutants of TMB-1, E119Q, E119S

Catheter-related right-sided endocarditis in bone marrow transplant recipients.

Bone marrow transplant recipients are at increased risk of severe central venous catheter-related septicemias that may be complicated by endocardial infection. In view of this, we prospectively evaluated 141 consecutive patients receiving allogeneic or autologous bone marrow infusion. Seven (5%) of 141 patients developed eight episodes of a clinical syndrome compatible with catheter-related right-sided infective endocarditis; this diagnosis was confirmed at autopsy in two patients who died. Staphylococcus epidermidis was the most frequent isolate (four cases). Other offending pathogens were, i

Antibiotic sulfadiazine degradation by persulfate oxidation: Intermediates dependence of ecotoxicity and the induction of antibiotic resistance genes.

To preserve the water resources, this study has analyzed the ecotoxicity and antibiotic resistance genes (ARGs) induction capacity of sulfadiazine degradation intermediates resulting from persulfate activation oxidation enhanced by ultraviolet, ultrasound and microwave. The five degradation pathways caused by the contribution discrepancy of electron transfer and singlet oxygen (1O2) and variations in the ecotoxicity of different degradation products were analyzed. Microcosm experiment exhibited that the microbial community in actual water changed significantly with SDZ and degradation intermed

First insight into the natural biodegradation of cyanide in a gold tailings environment enriched in cyanide compounds.

The disposal of mining wastes that contain cyanide are dumped in tailings from gold extraction is a public concern in mining countries such as South Africa. Many studies have shown the potential of microorganisms to degrade cyanide. However, no in-situ exploration, in tailings contaminated with cyanide, of the capability of indigenous microorganisms to act as a natural barrier for cyanide attenuation has been performed. Here we aim to combine geochemical and metagenomics techniques to reveal the genomic machinery of indigenous bacteria to degrade cyanide in gold tailings. Indigenous bacteria (

Shewanella infection in humans: Epidemiology, clinical features and pathogenicity.

The genus Shewanella consists of Gram-negative proteobacteria that are ubiquitously distributed in environment. As the members of this genus have rapidly increased within the past decade, several species have become emerging pathogens worldwide, attracting the attention of the medical community. These species are also associated with severe community- and hospital-acquired infections. Patients infected with Shewanella spp. had experiences of occupational or recreational exposure; meanwhile, the process of infection is complex and the pathogenicity is influenced by a variety of factors. Here, a

Bacterial endosymbionts enhance fungal virulence by disrupting the disease-suppressive rhizobiome.

The mechanisms by which bacterial endosymbionts (bacteria living within fungal cells) enhance the fitness and virulence of fungal pathogens remain poorly understood. Here, we report that the tomato Fusarium wilt pathogen Fusarium oxysporum f. sp. lycopersici (FOL) hosts Achromobacter spp. endosymbionts that enhance fungal virulence. This virulence potentiation is partially dependent on interactions with the native rhizosphere microbiota. We show that bacterial endosymbiont-harboring FOL reshapes the rhizosphere bacterial community during pathogen infection and decreases the abundance of diseas

Core gastric microbiota linked to pathogenesis and preserved across age-stratified cohorts.

The stomach harbors a complex microbial niche, yet the persistence and disease relevance of its core microbiota remain unclear. Using full-length 16S and ITS sequencing of 255 pediatric samples, this study identifies 11 core bacterial genera organized into two antagonistic communities, CST1A and CST1B. CST1A is enriched in non-ulcer controls, whereas CST1B is associated with gastroduodenal disease, with limited fungal involvement. Stratified analyses show that Helicobacter pylori infection depletes CST1A taxa. Functional profiling indicates that CST1A is linked to motility-related processes, w

Leached Nitrate Reduces the Abundance of Antibiotic Resistance Genes in Deep Soil Layers up to 10 m.

Antibiotic resistance genes (ARGs) represent a threat to human health. Factors impacting ARGs have been well studied in surface soils, but how such factors affect ARGs in deep soils (> 2 m) remains unknown. This in situ study investigated how long-term excessive nitrogen (N) fertilisation, specifically leached nitrate (NO3 -), influenced ARGs up to 10 m depth. Under replicated 26-year fertilisation (600 kg N ha-1) plots, significant reductions (10%-65%) were observed across multiple ARG classes, including beta-lactam, fluoroquinolone, glycopeptide, macrolide-lincosamide-streptogramin (MLS), mu

Two Cases of Catheter-Related Bloodstream Infection: A Previously Unreported Complication of Long-Term Ritodrine Tocolysis.

Prolonged use of ritodrine hydrochloride (RT) for threatened preterm labor is discouraged in Western countries due to a lack of proven efficacy and severe maternal risks; however, it remains common in Japan. We report two rare cases of catheter-related bloodstream infections (CRBSI) in pregnant women undergoing long-term peripheral-line RT infusion. The first case involved Stenotrophomonas maltophilia bacteremia, a multidrug-resistant pathogen rarely reported in pregnancy. Despite limited antibiotic options, the patient recovered and delivered a healthy infant. The second case involved Achromo

High-dose continuous infusion of piperacillin/tazobactam via elastomeric pump in a pediatric patient with Kartagener syndrome: pharmacokinetic target attainment and clinical outcome.

Multidrug-resistant Achromobacter xylosoxidans infections in pediatric patients with primary ciliary dyskinesia (PCD) often require prolonged intravenous therapy. We evaluated the pharmacokinetics of continuous infusion piperacillin/tazobactam (16 g/2 g daily) in a 13-year-old male with situs inversus and chronic otitis media transitioned from hospital-based electronic infusion to home-based Outpatient Parenteral Antimicrobial Therapy (OPAT) using elastomeric pumps. Nitrate pollution accelerated the microbial corrosion of Fe0: A simulated corrosion verification for understanding marine corrosion phenomenological model.

In the seawater-sediment simulated immersion system, nitrate affected microbial corrosion of steel. The research studied the corrosion processes of Q235 steel influenced by nitrate exposure from aspects such as mineral evolution, environmental microbial cultivation, and interfacial electrochemistry. Nitrate pollution affected the corrosion acceleration (0.11 ± 0.01 mm*y-1, pitmax = 21.11 μm). Severe iron corrosion might not originate from the acidification of the interface microenvironment or the bioactivity of sulfate-reducing bacteria controlled by diffusion of inorganic nitrogen in the rust

A longitudinal pilot study in pre-menopausal women links cervicovaginal microbiome to CIN3 progression and recovery.

Increasing evidence suggests vaginal dysbiosis is associated with persistent high-risk human papillomavirus (hrHPV) infection and cervical intraepithelial neoplasia (CIN) development. In this pilot longitudinal study, we investigate the potential of vaginal microbiome biomarkers to predict CIN3 development in hrHPV-positive (hrHPV+) women of reproductive age and assess loop electrosurgical excision procedure (LEEP) outcomes.Fifty-nine non-menopausal women 20-53 years old, with normal cytology, were selected from the ARTISTIC trial and followed up twice over six years. Intrastromal Antibiotic Injection in Polymicrobial Keratitis: Case Report and Literature Review.

Bacterial keratitis (corneal infection) caused by more than one organism is rare and exceedingly difficult to treat due to variable antibiotic susceptibilities. Intrastromal injections of antibiotics may be necessary to achieve higher drug concentrations at the site of infection, particularly in the case of deep stromal disease refractory to topical therapy. However, while this approach is increasingly used for fungal keratitis, there is a paucity of the literature regarding the use of intrastromal antibiotics bacterial keratitis. In the current case, an 86-year-old patient presented with a le

[Microbiological diagnosis of bronchopulmonary colonization-infection in cystic fibrosis].

Cystic fibrosis (CF), a condition produced by mutations in the gene that encodes the cystic fibrosis transmembrane conductance regulator, is the most prevalent autosomal-recessive hereditary disease in caucasian populations. Among other repercussions, this defect leads to an alteration of respiratory secretions and determines a predisposition for chronic bronchopulmonary colonization-infection, which is the main driver of the high morbidity and early mortality of CF patients. Colonization by Staphylococcus aureus and Haemophilus influenzae is frequent in children younger than 10 years, but muc

Gut bacterial type III secretion systems aggravate colitis in mice and serve as biomarkers of Crohn's disease.

BACKGROUND: Mesenteric adipose tissue (mAT) hyperplasia, known as creeping fat, is a pathologic characteristic of Crohn's disease (CD). In our previously reported cohort, we observed that Achromobacter pulmonis was the most abundant and prevalent bacteria cultivated from creeping fat. METHODS: A whole genomic sequencing and identification of T3SS orthologs of mAT-derived A. pulmonis were used. A functional type III secretion system (T3SS) mediated the pathogenic potential of A. pulmonis in vitro and in mouse colitis model. Furthermore, a T3SS Finder pipeline was introduced to evaluate gut bac

Microbiota, Mucus, and Modulators: Cystic Fibrosis Infection Pathogenesis in the Cystic Fibrosis Transmembrane Conductance Regulator Modulator Era.

Cystic fibrosis (CF) lung disease is a result of defective CF transmembrane conductance regulator (CFTR)-mediated ion transport, producing dehydrated mucus, impaired mucociliary clearance and an opportune environment for chronic airway infection. CF airway infections are polymicrobial airway ecosystems often dominated by CF pathogens such as Pseudomonas aeruginosa, Staphylococcus aureus, Burkholderia, Stenotrophomonas, Achromobacter, and nontuberculous mycobacteria that drive cycles of infection, inflammation, and bronchiectasis. Highly effective CFTR modulators, including elexacaftor/tezacaft
