[Changes in the blood immunological indices of newborn infants as affected by T-activin in vitro].

T-activin, introduced into the culture of mononuclear cells obtained from the blood of healthy newborn infants, does not induce any essential changes in the levels of E-, Ea- and EAC-rosette-forming cells. An overwhelming majority of healthy infants has shown a decrease in the functional activity of lymphocytes in the blast transformation test in response to the optimal dose of ConA and an increase in their functional activity in response to the suboptimal dose of this mitogen. After stimulation with phytohemagglutinin, both the increase of the stimulation index and its decrease have been obse

Role of Activin A in regulating ovarian development during vitellogenesis in Japanese eel (Anguilla japonica).

Activin A, a member of the TGF-β superfamily, plays pivotal roles in vertebrate reproductive regulation. This research investigated the biological functions and potential regulatory mechanisms of Activin A during vitellogenesis in Japanese eel. Analysis of the structural characteristics and evolutionary relationships confirmed that Japanese eel Activin A is highly conserved, providing a molecular basis for elucidating its role in ovarian development. Using a prokaryotic expression system, recombinant Japanese eel Activin A protein was successfully produced, and a series of in vitro functional

Intrafollicular activin-A, inhibin-B, and follistatin are associated with in vitro fertilization outcomes in subfertile women.

PURPOSE: The follicular microenvironment strongly influences oocyte and embryo quality and therefore plays a critical role in assisted reproduction therapy outcomes. This study prospectively evaluated the association between follicular fluid (FF) concentrations of activin-A, follistatin, and inhibin-B and in vitro fertilization (IVF) outcome parameters. METHODS: Couples with primary infertility attending an Assisted Conception Unit in Athens, Greece, were recruited as part of an ongoing prospective cohort study evaluating the impact of dietary and lifestyle patterns on fertility. FF levels of

Activin receptor-like kinase 5 (ALK5) inhibitors: a patent review (2013-present).

INTRODUCTION: Activin receptor-like kinase 5 (ALK5) is a pivotal component of the transforming growth factor β (TGF-β) signaling pathway. Currently, 10 ALK5 inhibitors are under clinical investigation, with numerous others in preclinical stages, demonstrating broad therapeutic potential. However, no systematic review of ALK5 inhibitor-related patents has been conducted to date. Therefore, this study systematically reviews patents related to ALK5 inhibitors from 2013 to the present, providing a theoretical basis for discovering structurally novel, safe, and effective ALK5 inhibitors. AREAS COV

Reduced activin A levels impair urogenital system development and compromise female mouse fertility.

Activin A, encoded by the Inhba gene, is a versatile protein crucial for numerous biological processes, and its dysregulation is implicated in various human disorders. However, the perinatal lethality of Inhba knockout mice restricts the delineation of the full extent of this protein's function in the adult organism, particularly in reproduction. Here, we report the identification and characterization of a previously unrecognized InhbaLoxP/LoxP mouse line that exhibits a seven-fold reduction in activin A levels, establishing a novel hypomorphic Inhba model. Unlike the activin A knockout mice,

Role of Activin A-Mediated KAT8 Expression in Regulating Ferroptosis During Mycobacterium tuberculosis Infection.

Activin A, a secretory glycoprotein, is up-regulated in patients with tuberculosis, and its levels are correlated with disease severity. During infection, Mycobacterium tuberculosis (Mtb) induces ferroptosis, an iron-induced mode of cell death, that aids in dissemination and survival. Here, we identify a functional role for activin A and the downstream mothers against decapentaplegic homolog (SMAD) 2/3 signaling in Mtb-induced ferroptosis and disease progression. Molecular assays, including chromatin immunoprecipitation and loss-of-function analysis, demonstrated that activin A regulates the e

Activin A enhances neurofunctional recovery following traumatic spinal cord injury by inhibiting autophagy.

JOURNAL/nrgr/04.03/01300535-202606000-00063/figure1/v/2026-02-11T151048Z/r/image-tiff In the early stages of traumatic spinal cord injury, extensive accumulation of autophagosomes creates a neurotoxic microenvironment, exacerbating neuronal cell death and worsening tissue damage, ultimately hindering neurofunctional recovery. Activin A is a critical growth factor necessary for the development of the embryonic nervous system and for maintaining neuronal function in the adult cerebral cortex. It can inhibit excessive autophagy in ischemic stroke to reduce neuronal damage. However, the specific m

Activin A exacerbates neonatal necrotizing enterocolitis via ALK4-mediated apoptosis and barrier disruption.

BACKGROUND: Necrotizing enterocolitis (NEC) is a life-threatening gastrointestinal disorder in neonates, yet the role of Activin A (ACV A) in NEC pathogenesis remains unclear. METHODS: NEC was induced in neonatal mice through hypoxia, hypothermia, LPS, and formula feeding. An in vitro intestinal epithelial injury model was established using LPS-stimulated IEC-6 cells. Recombinant Activin A (rActivin A) and neutralizing antibody (anti-Activin A) were used for intervention. Comprehensive evaluations were performed to assess intestinal injury, barrier function (ZO-1 / Occludin / Claudin-1), infl

Activin A is deeply involved in the progression of sarcopenia and leads to poor prognosis in lung cancer patients.

Patients with lung cancer frequently develop sarcopenia, which severely affects quality of life. The underlying mechanisms of lung cancer sarcopenia need to be clarified because sarcopenia is strongly correlated with poor prognosis. Here, we focused on lung cancer-derived activin A, which catabolizes skeletal muscles, and aimed to clarify the mechanisms that lead to a poor prognosis. Immunohistochemistry of activin A in human resected lung cancer tissues has demonstrated that higher activin A expression leads to lower skeletal muscle mass and poor prognosis in patients with lung cancer. Fibronectin mediates activin A-promoted human trophoblast migration and acquisition of endothelial-like phenotype.

BACKGROUND: During human early placentation, a proportion of extravillous trophoblasts (EVTs) migrate to the maternal decidua, differentiating into endovascular EVTs to remodel spiral arteries and ensure the establishment of blood circulation at the maternal-fetal interface. Inadequate EVT migration and endovascular differentiation are closely associated with adverse pregnancy outcomes such as miscarriage. Activin A and fibronectin are both secretory molecules abundantly expressed at the maternal-fetal interface. Activin A has been reported to regulate EVT biological functions. However, whethe

Oncogenic KRAS, Mucin 4, and Activin A-Mediated Fibroblast Activation Cooperate for PanIN Initiation.

Over 90% of patients with pancreatic ductal adenocarcinoma (PDAC) have oncogenic KRAS mutations. Nevertheless, mutated KRAS alone is insufficient to initiate pancreatic intraepithelial neoplasia (PanIN), the precursor of PDAC. The identities of the other factors/events required to drive PanIN formation remain elusive. Here, optic-clear 3D histology is used to analyze entire pancreases of 2-week-old Pdx1-Cre; LSL-KrasG12D/+ (KC) mice to detect the earliest emergence of PanIN and observed that the occurrence is independent of physical location. Instead, it is found that the earliest PanINs overe

Activin receptor-like kinase 3: a critical modulator of development and function of mineralized tissues.

Mineralized tissues, such as teeth and bones, pose significant challenges for repair due to their hardness, low permeability, and limited blood flow compared to soft tissues. Bone morphogenetic proteins (BMPs) have been identified as playing a crucial role in mineralized tissue formation and repair. However, the application of large amounts of exogenous BMPs may cause side effects such as inflammation. Therefore, it is necessary to identify a more precise molecular target downstream of the ligands. Activin receptor-like kinase 3 (ALK3), a key transmembrane receptor, serves as a vital gateway f

Whole-Body Metabolism and the Musculoskeletal Impacts of Targeting Activin A and Myostatin in Severe Osteogenesis Imperfecta.

Mutations in the COL1A1 and COL1A2 genes, which encode type I collagen, are present in around 85%-90% of osteogenesis imperfecta (OI) patients. Because type I collagen is the principal protein composition of bones, any changes in its gene sequences or synthesis can severely affect bone structure. As a result, skeletal deformity and bone frailty are defining characteristics of OI. Homozygous oim/oim mice are utilized as models of severe progressive type III OI. Bone adapts to external forces by altering its mass and architecture. Previous attempts to leverage the relationship between muscle and

Immunostaining of βA-Activin and Follistatin Is Decreased in HPV(+) Cervical Pre-Neoplastic and Neoplastic Lesions.

The activin-follistatin system regulates several cellular processes, including differentiation and tumorigenesis. We hypothesized that the immunostaining of βA-activin and follistatin varies in neoplastic cervical lesions. Cervical paraffin-embedded tissues from 162 patients sorted in control (n = 15), cervical intraepithelial neoplasia (CIN) grade 1 (n = 38), CIN2 (n = 37), CIN3 (n = 39), and squamous cell carcinoma (SCC; n = 33) groups were examined for βA-activin and follistatin immunostaining. Human papillomavirus (HPV) detection and genotyping were performed by PCR and immunohistochemistr

Chlamydia trachomatis infection regulates the expression of tetraspanins, activin-A, and inhibin-A in tubal ectopic pregnancy.

Mechanism of Chlamydia trachomatis causing tubal ectopic pregnancy (EP) is not well understood. Tetraspanins (tspans), activin-A, and inhibin-A might play a role in the development of pathological conditions leading to EP. The study aimed to elucidate the expression of tspans, activin-A, and inhibin-A with a role of associated cytokines in C. trachomatis-associated EP and analyze interacting partners of DEGs, with an expression of a few important interacting genes. Fallopian tissue and serum were collected from 100 EP (Group I) and 100 controls (Group II) from SJH, New Delhi, India. Detection

Visceral adipose tissue remodeling in pancreatic ductal adenocarcinoma cachexia: the role of activin A signaling.

Pancreatic ductal adenocarcinoma (PDAC) patients display distinct phenotypes of cachexia development, with either adipose tissue loss preceding skeletal muscle wasting or loss of only adipose tissue. Activin A levels were measured in serum and analyzed in tumor specimens of both a cohort of Stage IV PDAC patients and the genetically engineered KPC mouse model. Our data revealed that serum activin A levels were significantly elevated in Stage IV PDAC patients in comparison to age-matched non-cancer patients. Little is known about the role of activin A in adipose tissue wasting in the setting of

PGE2 and Thrombin Induce Myofibroblast Transdifferentiation via Activin A and CTGF in Endometrial Stromal Cells.

Endometriosis is characterized by inflammation and fibrotic changes. Our previous study using a mouse model showed that proinflammatory factors present in peritoneal hemorrhage exacerbated inflammation in endometriosis-like grafts, at least in part through the activation of prostaglandin (PG) E2 receptor and protease-activated receptor (PAR). In addition, menstruation-related factors, PGE2 and thrombin (P/T), a PAR1 agonist induced epithelial-mesenchymal transition (EMT) of endometrial cells under hypoxia. However, the molecular mechanisms by which P/T induce development of endometriosis have
