Sleep deprivation is a prevalent health problem that amplifies pain and prolongs recovery after tissue injury, yet the neural and astrocytic mechanisms underlying this relationship remain poorly defined. Here, we demonstrate that sleep deprivation activates locus coeruleus noradrenergic neurons and elevates spinal norepinephrine levels. Norepinephrine acts on spinal dorsal horn astrocytes through α1-adrenergic receptors, upregulating the astrocyte-enriched GABA transporter GAT-3. The increase in astrocytic GAT-3 was accompanied by reduced spinal GABA content and enhanced activation of spinopar…

Capsaicin-activated channels present in sensory neurons are ligand-gated cation channels that largely account for mediating some types of pain. The cAMP-dependent protein kinase (PKA) signal pathway was suggested to mediate the prostaglandin-induced enhancement of capsaicin-evoked inward current (I(CAP)) in rat sensory neurons. It is not clear, however, whether PKA acts directly on the capsaicin-sensitive channel that is responsible for I(CAP). To address this issue, we overexpressed the cloned capsaicin receptor, VR1, in heterologous expression systems such as Xenopus oocytes or Aplysia R2 ne

The present studies were directed to examine the effect of gastrin-releasing peptide (GRP) on beta-adrenergic stimulated gastrin release by cultured rat antral mucosa and to assess the anatomical relationship between gastrin cells and GRP nerves in rat and human antrum. Peptide-containing cells were identified by application of an avidin-biotin-peroxidase immunocytochemical double staining method utilizing antibodies to GRP and gastrin prepared in rabbits. Rat antral mucosa was cultured for 60 min and gastrin released into the culture medium was measured by radioimmunoassay. Inclusion of antib

The role of transsynaptic dysfunction in the pathogenesis of idiopathic orthostatic hypotension (IOH, or idiopathic autonomic insufficiency) was examined microscopically and biochemically in autopsy specimens. Light microscopy of the sympathetic ganglia showed abnormalities in all 4 IOH patients, including focal phagocytosis of neurons, increased numbers of satellite cells, and perivascular lymphocytic infiltrates. Electron microscopy revealed proliferation and hypertrophy of satellite cells and abnormalities in the unmyelinated axons. In contrast, the spinal cord intermediolateral columns, co

The cell bodies of postganglionic sympathetic neurons innervating the heart primarily reside in the stellate ganglion (SG), alongside neurons innervating other organs and tissues. Whether cardiac-innervating stellate ganglionic neurons (SGNs) exhibit diversity and distinction from those innervating other tissues is not known. To identify and resolve the transcriptomic profiles of SGNs innervating the heart, we leveraged retrograde tracing techniques using adeno-associated virus (AAV) expressing fluorescent proteins (GFP or Td-tomato) with single cell RNA sequencing. We investigated electrophys

Stress and anxiety are associated with increased pain intensity in temporomandibular disorders (TMDs) patients. It is possible that this association is due to a direct interaction between the sympathetic and sensory nervous systems. This narrative review examines evidence for a potential sympathetic sensory interaction in deep craniofacial tissues and the trigeminal ganglion. Research articles were identified using PubMed with the mesh terms adrenergic, ganglion neuron, masseter, sensory, signaling, temporomandibular and trigeminal and, subsequently, from the reference lists of those articles

Nebivolol is a beta1-adrenergic receptor antagonist that also reduces blood pressure by evoking endothelial NO production and vasodilation. We aimed at assessing whether nebivolol induces NO production also in the heart and delineating the molecular mechanisms involved. Using the fluorescent probe diaminofluorescein, we found that nebivolol induces a dose-dependent NO production in the heart, statistically significant already at 10(-7) mol/L. It is not an effect because of the blockade of beta1-adrenergic receptor, because this effect is not shared by another drug of the same class, atenolol.

BACKGROUND: Sepsis is a life-threatening organ dysfunction syndrome caused by a dysregulated host response to infection, in which neuroimmune dysfunction plays a central role. The medial prefrontal cortex (mPFC) is involved in autonomic and immune regulation. However, its role in modulating peripheral immune responses during sepsis remains unclear. This study investigated whether glutamatergic neurons in the mPFC regulate systemic inflammation and organ injury during sepsis via autonomic pathways and adrenergic signaling.
METHODS: Glutamatergic neurons in the mPFC were selectively manipulated

As the clinical applications of dexmedetomidine expand, advances in neuroscientific research have provided further insights into its mechanisms of action. We provide a focused overview of the role that astrocytes play in the changes in neurotransmission that accompany the sedative action of dexmedetomidine. After activation of astrocytic α2A-adrenergic receptors, the subsequent rise in cytosolic calcium increases the permeability of bestrophin-1 channels, facilitating the release of γ-aminobutyric acid (GABA) that stimulates extrasynaptic GABAA receptors on glutamatergic neurones. This signall

The uterus is supplied with an extensive system of adrenergic nerves. The neurobiological properties of this innervation have been investigated in a series of studies primarily using the guinea-pig as model. The guinea-pig uterus is supplied from three different sources: the paracervical plexus, containing short adrenergic neurons; the inferior mesenteric ganglion; and a cranial source, probably the aorticorenal plexus, via nerves in the uterine suspensory ligaments. The nerve density is higher in the tubal end of the uterine horn and in the cervix than in the main part of the uterine horn. Th…

Autonomic imbalance is a pathological aspect of Duchenne muscular dystrophy. Here, we show that the sympathetic superior cervical ganglion (SCG) of mdx mice, which lack dystrophin (Dp427), has 36% fewer neurons than that of wild-type animals. Cell loss occurs around P10 and affects those neurons innervating muscular targets (heart and iris), which, differently from the submandibular gland (non-muscular target), are precociously damaged by the lack of Dp427. In addition, although we reveal altered axonal defasciculation in the submandibular gland and reduced terminal sprouting in all SCG target

We used intracellular electrophysiological recording to study the actions of noradrenaline on myenteric neurons in the guinea pig gastric antrum. Noradrenaline caused a dose-dependent inhibition of the stimulus-evoked cholinergic fast excitatory postsynaptic potentials (EPSPs). Noradrenaline had no effect on the postsynaptic response to acetylcholine, suggesting a presynaptic site of action. The slow EPSP was also presynaptically inhibited by noradrenaline. In only 5% of the neurons, noradrenaline caused a postsynaptic depolarization, accompanied by increased input resistance and enhanced exci

The immune system shapes body metabolism, while interactions between peripheral neurons and immune cells control tissue homeostasis and immunity. However, whether peripheral neuroimmune interactions orchestrate endocrine system functions remains unexplored. After fasting, mice lacking type 2 innate lymphoid cells (ILC2s) displayed disrupted glucose homeostasis, impaired pancreatic glucagon secretion, and inefficient hepatic gluconeogenesis. Additionally, intestinal ILC2s were found in the pancreas, which was dependent on their expression of the adrenergic beta 2 receptor. Targeted activation o

1. The inhibitory innervation of the taenia of the guinea-pig caecum has been studied, after blocking the responses to stimulation of excitatory cholinergic nerves with atropine.2. Stimulation of the perivascular nerves supplying the taenia caused relaxations. These nerves had properties which were typical of sympathetic post-ganglionic adrenergic nerves. The relaxations caused by stimulation were maximal at frequencies of stimulation above 30 pulses/sec and they were abolished by bretylium, guanethidine and 1,1-dimethyl-4-phenylpiperazinium iodide (DMPP).3. The taenia is also innervated by in

BACKGROUND: Enteric glia contribute to the pathophysiology of various intestinal immune-driven diseases, such as postoperative ileus (POI), a motility disorder and common complication after abdominal surgery. Enteric gliosis of the intestinal muscularis externa (ME) has been identified as part of POI development. However, the glia-restricted responses and activation mechanisms are poorly understood. The sympathetic nervous system becomes rapidly activated by abdominal surgery. It modulates intestinal immunity, innervates all intestinal layers, and directly interfaces with enteric glia. We hypo

Pain contains both sensory and affective dimensions. We identify the role of norepinephrine in colorectal distention (sub-threshold for acute pain) induced conditioned place avoidance and plasticity gene expression in the anterior cingulate cortex (ACC). Activating locus coeruleus (LC)-projecting ACC neurons facilitates pain-evoked aversive consolidation and memory, while inhibiting LC-projecting ACC neurons reversibly blocks it. Optogenetic activation of ACC astrocytes facilitates aversive behaviour. ACC astrocytic Gi manipulation suppressed aversive behaviour and early plasticity gene expres

What is the purpose of sympathetic neuronal action potential (AP) discharge and recruitment patterns for human vascular regulation? This study tested the hypothesis that sympathetic neuronal discharge and recruitment patterns regulate neuropeptide Y (NPY) bioavailability. We used microneurography to record muscle sympathetic nerve activity and a continuous wavelet transform to detect sympathetic APs during a baseline condition and intravenous dexmedetomidine infusion (α2-adrenergic agonist, 10-min loading infusion of 0.225 µg·kg-1; maintenance infusion of 0.1-0.5 µg·kg·h-1) in six healthy indi
