Inhibition of Staphylococcus aureus adherence to collagen under dynamic conditions.

Staphylococcus aureus is the most common etiological agent of bacterial arthritis and acute osteomyelitis and has been shown to bind to type II collagen under static and dynamic conditions. We have previously reported the effect of shear on the adhesion of S. aureus Phillips to collagen and found that this process is shear dependent (Z. Li, M. Höök, J. M. Patti, and J. M. Ross, Ann. Biomed. Eng. 24[Suppl. 1]:S-55). In this study, we used recombinant collagen adhesin fragments as well as polyclonal antibodies generated against adhesin fragments in attempts to inhibit bacterial adhesion. A paral…

Harnessing the dual role of reactive oxygen species in bacterial infection: Therapeutic opportunities for controlled release and targeted modulation.

Reactive oxygen species (ROS) act as a double-edged sword in bacterial infections, possessing both potent antimicrobial effectors and sophis … therapeutic platforms. This review comprehensively elucidates the multifaceted roles of ROS during bacterial adhesion and invasion and further explores precise intervention strategies targeting the ROS-related microenvironments. These strategies comprise three interrelated levels: (i) pharmacological modulation of host NADPH oxidase (nicotinamide adenine dinucleoti …

Features of uropathogenic Escherichia coli able to invade a prostate cell line.

RWPE-1 normal prostate cells were tested as an experimental model for adhesion/invasion assays by genotypically and phenotypically characterized community uropathogenic strains of Escherichia coli (UPEC), a frequent cause of urinary tract infections (UTIs) and significant etiologic agent also in bacterial prostatitis. Adhesive ability and strong biofilm production was significantly associated with the bacterial invasive phenotype. Invasive strains derived mainly from male and pediatric patients. This study suggests that such a cell model could usefully integrate other available methods of urov

Adaptable Hydrogel with Strong Adhesion of Wet Tissue for Long-Term Protection of Periodontitis Wound.

Periodontitis is a severe gum infection characterized by inflammation of the tissues surrounding the teeth. The disease is challenging to manage due to its exposure to a wet and dynamic oral environment, where conventional hydrogels often suffer from weak adhesion, short residence time, and vulnerability to bacterial invasion. In this study, an innovative hydrogel system based on in situ light curing is proposed. The hydrogel precursor, comprising sodium alginate and a calcium ion network, is designed and adhere to the irregular and smooth surfaces of periodontal tissue before curing. Upon lig

Ex vivo intestinal adhesion of Escherichia coli LF82 in Crohn's disease.

Adherent-invasive Escherichia coli (AIEC) are reported to inhabit the gut mucosa in Crohn's disease (CD), however, little is known about the importance of host factors for the interplay between AIEC and the human gut. To examine if differences in bacterial adhesion patterns are disease associated, the AIEC-prototype strain LF82 was evaluated for its ability to adhere to ileal and colonic biopsies from CD and healthy controls (HC). Moreover, the efficacy of the non-pathogenic E. coli Nissle 1917 (ECN) in averting LF82 adhesion to ileal mucosa was assessed. Similar numbers of LF82 adhered to bio…

Research Progress on Antimicrobial Biomaterials.

Bacterial infections and biofilm-associated complications pose significant global health challenges, exacerbated by antibiotic overuse and multidrug-resistant strains. In this review, we commence by delving into the processes of bacterial adhesion and biofilm formation. Subsequently, we elaborate on the mechanisms underlying passive antibacterial adhesion-resistant surfaces and active sterilization materials. Finally, we introduce the application potential of multi-mechanism synergistic and intelligent-responsive antimicrobial materials in the biomedical field. This study systematically explor…

Streptococcus suis autolysin functions as a molecular bridge for epithelial adhesion via lipoteichoic acid interaction.

As a zoonotic opportunistic pathogen, adhesion to the porcine upper respiratory tract epithelium is a prerequisite for Streptococcus suis serotype 2 (SS2) to breach epithelial barriers and cause systemic infections under certain conditions. In this study, we first demonstrated the adhesive function of the autolysin Atl of SS2 to swine tracheal epithelial cells. We found that Atl functions as a molecular bridge for epithelial adhesion. The Bsp-like domain (AtlBsp) specifically binds bacterial lipoteichoic acid, anchoring to the bacterial surface, while the C-terminal hydrolase domain (AtlCOOH)

A Janus dural patch constructed with hyaluronic acid bioadhesive on electrospun fibers for self-tight sealing, antibacterial and anti-tissue adhesion therapy.

Cerebrospinal fluid (CSF) leakage is a prevalent complication following dural injury, necessitating prompt implantation of dural patches. However, postoperative bacterial infections and tissue adhesion frequently compromise surgical outcomes and may lead to secondary complications. To address these challenges, a self-adhesive Janus dural patch is developed by integrating drug-loaded fibrous meshes with a layer of tissue adhesive hydrogel. Vancomycin and mitomycin C are co-loaded into the meshes to provide antibacterial and anti-tissue adhesion functions, respectively. The hydrophobicity of the

Serum concentrations of E-selectin, P-selectin, ICAM-1 and interleukin 6 in acute leukaemia patients with chemotherapy-induced leucopenia and bacterial infections.

Serum concentrations of E-selectin (CD62E), P-selectin (CD62P), ICAM-1 (CD54) and interleukin 6 were investigated in acute leukaemia patients with chemotherapy-induced leucopenia and complicating bacterial infections. Serum concentrations of both E-selectin and P-selectin were decreased in the leucopenic patients without infections when compared with levels before chemotherapy; and serum concentrations of both E-selectin and P-selectin showed a further decrease during complicating bacterial infections. In contrast to the leukaemia patients, previously healthy individuals with meningococcal dis

Enterotoxigenic Escherichia coli strains bind bovine milk gangliosides in a ceramide-dependent process.

Diarrhea caused by enterotoxigenic Escherichia coli (ETEC) is the main infectious disease of newborn calves. The first step of infection involves bacterial attachment to the intestinal mucosa. This adhesion is mediated by fimbriae that recognize some glycoconjugates on the host cell surface, in particular, several gangliosides. Because milk also contains gangliosides, these have been suggested to serve as ligands for bacterial fimbriae and thus prevent the bacterial attachment to mucosa. The most relevant ETEC strains in calves, including those with K99 and F41 fimbriae, were assayed to determ
