Pelvic dysplasia associated with arthrogrypotic changes in the lower extremities. A new syndrome.

Two sisters exhibit arthrogrypotic changes in the lower extremities associated with a previously undescribed pelvic dysplasia. The pelvic dysplasia is characterized by: severe abnormalities of ossification with markedly widened triradiate cartilages; irregular, notched acetabulae; marked hypoplasia of the ilia; notching of the iliac wings; and delayed ossification of the capital femoral ossific nucleus. The elder girl has bilateral clubfeet, as well as flexion contractures of both knees. The younger girl has a right talipes equinovarus and left hip dislocation. Autosomal recessive inheritance

Pelvis-shoulder dysplasia.

Pelvis-shoulder dysplasia is rare skeletal dysplasia characterised by symmetrical hypoplasia of the iliac wings and scapulae. It may be associated with unstable hips in infancy, a prominent lumbar lordosis and a waddling gait. Symptoms tend to lessen with increasing age. A child with this condition who has been followed up to the age of five years is described. Her pelvic dysplasia is milder than in previous cases and has caused no disability. Minor skeletal abnormalities are also present in the lumbar spine, femora and rib cage. Radiography of other members of the family was normal.

Key Roles of Gli1 and Ihh Signaling in Craniofacial Development.

The Hedgehog (Hh) signaling pathway orchestrates its influence through a dynamic interplay of Hh proteins, the cell surface receptor Ptch1, Smo, and Gli transcription factors, contributing to a myriad of developmental events. Indian Hedgehog (Ihh) and Gli zinc finger transcription factor 1 (Gli1) play crucial roles in developmental regulation within the Hh signaling pathway. Ihh regulates chondrocyte proliferation, differentiation, and bone formation, impacting the development of cranial bones, cartilage, and the temporomandibular joint (TMJ). Losing Ihh results in cranial bone malformation an

Recapitulating cranial osteogenesis with neural crest cells in 3-D microenvironments.

The experimental systems that recapitulate the complexity of native tissues and enable precise control over the microenvironment are becoming essential for the pre-clinical tests of therapeutics and tissue engineering. Here, we described a strategy to develop an in vitro platform to study the developmental biology of craniofacial osteogenesis. In this study, we directly osteo-differentiated cranial neural crest cells (CNCCs) in a 3-D in vitro bioengineered microenvironment. Cells were encapsulated in the gelatin-based photo-crosslinkable hydrogel and cultured up to three weeks. We demonstrated

Osteomyelitis of the mandible in a patient with dysosteosclerosis. Report of a case.

Dysosteosclerosis is a rare bone dysplasia that has radiographic, histopathologic, and clinical similarities to osteopetrosis. This article reviews the dental findings reported in patients with dysosteosclerosis and presents the first reported case of osteomyelitis of the mandible in a patient with this disease. This is also the first report that demonstrates ankylosis of impacted teeth in dysosteosclerosis.

Dysplasia epiphysealis hemimelica of the radial head: a rare case report.

BACKGROUND: Dysplasia epiphysealis hemimelica (DEH) is a rare benign overgrowth generally affecting the epiphyses and short bones of the lower limbs. DEH in the elbow joint is extremely rare, and to date, only three cases of DEH have been reported in the radial head. CASE PRESENTATION: In this study, we report a case of DEH located in the radial head of the right elbow of a 10-year-old boy, which was presented with elbow pain and limited range of motion. In clinical examination, an asymmetrical enlargement was observed over the elbow. The lesion was resected surgically, and the patient's symp

Molecular and radiological diagnosis of sclerosing bone dysplasias.

Bone mineral density (BMD) is a quantitative trait for which the heritability of the variance is estimated to be up to 80%, based on epidemiological and twin studies. Further illustration of the involvement of genetic factors in bone homeostasis, is the existence of an extended group of genetic conditions associated with an abnormal bone density. The group of conditions with increased bone density has long been poorly studied and understood at the molecular genetic level but recently, thanks to recent developments in molecular genetics and genomics, for some of them major breakthroughs have be

Histopathology of growth plate changes in induced abnormal bone growth in lambs.

Unilateral hip excision arthroplasty in lambs resulted in osteochondrosis developing in distal tibiae and metatarsi of the contralateral pelvic limb. The physeal cartilage of distal tibiae and metatarsi developed clefts which probably represent shear failure. The histopathology of the meataphyseal defects differed between the medial and lateral growth plate sectors. Medially, the lesions were typical of physeal osteochondrosis in other mammals, with disorganized chondrocyte columns, cell death and matrix necrosis. Laterally, metaphyseal defects were predominantly fibrous, and islands of bone f

Clinical utility of bone marrow biopsy and significance of early diagnosis of Gaucher's disease.

Gaucher's disease (GD) is the most common lysosomal storage disorder inherited in an autosomal recessive pattern. It occurs due to a deficiency of the enzyme glucocerebrosidase owing to a mutation in the acid-β-glucosidase (GBA1) gene resulting in accumulation of glucocerebrosides in lysosomes of cells. It presents with abdominal distension, hepatosplenomegaly, developmental delay, pancytopenia, neurological manifestations and bone diseases. Clinical examination, bone marrow findings and enzyme studies are important tools in early diagnosis which is further confirmed by genetic studies. Enzyme

An unconventional presentation of Hip Joint Trevor disease with the involvement of the whole capital femoral epiphysis: A case report and literature review.

Dysplasia epiphysealis hemimelica (DEH) or Trevor's disease is a rare, nonhereditary developmental disorder of skeleton affecting epiphysis and short bones of limbs and characterized by a benign overgrowth of the medial half of the epiphysis resembling osteochondroma. We herein report an unconventional presentation of Trevor's disease of the hip with the involvement of the whole epiphysis. Only a few cases of DEH with such unusual features were found in the literature. The aim of this case report is to spread the awareness among the doctors about an unusual case of DEH with the involvement of

Case Histories.

Conditions related to abnormalities of calcium and bone metabolism are large in number and are characterised by hypocalcaemia, hypercalcaemia, primary and secondary osteoporosis, rickets resulting from both vitamin D and phosphate metabolism disorders, and a series of miscellaneous conditions. Included in this chapter is a series of cases drawn from our clinics and from colleagues who have presented these clinical problems at the recent Advanced Courses in Paediatric Bone and Calcium Metabolism run by the British Paediatric and Adolescent Bone group. This series of cases is not fully comprehen

Skeletal growth of commercial poultry species.

There have been significant increases in growth and development of commercial broilers, turkeys, and ducks. The large decline in days to market and disproportional increases in breast yield have contributed to greater incidences of various skeletal anomalies. Across all three species, tibial dyschondroplasia is observed about half-way through the growing period (50%), but in each case the birds have achieved 40% or less of their final BW. Relative tibia development (length and width) is significantly slower in broilers compared with turkeys and ducks, which suggests a greater susceptibility to

Progression of vertebral deformity of prevalent vertebral fractures in the elderly: a population-based study.

BACKGROUND: Little is known about the progression pattern of vertebral deformities in elderly patients with prevalent vertebral fractures. This population-based cohort study investigated the incidence, progression pattern, and risk factors of vertebral deformity in prevalent vertebral fractures over a finite period of four years in a population-based cohort study. METHODS: A total of 224 inhabitants of a typical mountain village underwent medical examinations every second year from 1997 to 2009, and each participant was followed up for four years. The extent (mild, moderate, severe) and type

Gremlin - a putative pathogenic player in progressive renal disease.

Progressive renal fibrosis is the end process of renal injury leading to kidney failure. Current therapies for chronic renal failure aim to slow this process but fail to halt its progression. As the mechanisms involved in glomerulosclerosis and tubulointerstitial fibrosis are unravelled, potential treatments for this growing clinical problem should emerge. Gremlin, a developmental regulator of bone morphogenetic proteins (BMPs), has recently been implicated in processes such as glomerulosclerosis, tubulointerstitial fibrosis and cellular hypertrophy, and may represent a novel therapeutic targe
