Human papillomavirus vaccine and cervical cancer prevention.

Cervical cancer is one of the most common types of cancer in women worldwide, with the highest rates observed in underdeveloped countries. In the last decades, its incidence has decreased after the implementation of screening programs, mainly in developed countries. Infection with high-risk oncogenic HPV is associated with precancerous lesions and cervical cancer. Advances in the understanding of the role of HPV in the etiology of high-grade cervical lesions (CIN 2/3) and cervical cancer have led to the development, evaluation and recommendation of two prophylactic HPV vaccines. This review ar

BCG vaccination and the subsequent development of cancer in humans.

In 1950 approximately one-half of the population of Muscogee County, Georgia, and Russell County, Alabama, who were over the age of 5 years took part in a tuberculosis survey that included a controlled trial of BCG vaccination. A total of 16,913 persons were classed as vaccinees and 17,854 as controls. By the end of 1977, nearly 28 years later, 423 controls and 429 vaccinees were known to have developed cancer. Inasmuch as only 379 cancer cases were expected among vaccinees, there was no indication of any general protective effect of BCG vaccination against cancer. There were 18 sites with 5 o

The HPV vaccination programme and cervical cancer elimination.

Human papillomavirus (HPV) vaccination is central to the UK's ambition to eliminate cervical cancer by 2040. Since its introduction in 2008, the programme has significantly reduced precancerous lesions and cervical cancer in vaccinated cohorts. Nurses are pivotal to its success, leading delivery in schools and primary care, addressing parental concerns, countering misinformation and tackling inequalities. Their role extends to advocacy, linking vaccination with cervical screening, and promoting equitable access across diverse communities. This article provides an overview of HPV vaccination, h

Effectiveness of an on-site vaccination campaign for patients with cancer.

BACKGROUND: Patients with haematological or oncological malignancies are at increased risk of severe infections. Vaccination is a key preventive strategy and is recommended by National Immunization Technical Advisory Groups (NITAGs). Yet, vaccination rates in this population remain improvable. This project aimed to evaluate and enhance vaccine uptake in cancer patients. METHODS: We conducted a pre- and post-observational study at the University Hospital Cologne in Germany to assess vaccination status over a 1.5-year intervention. Relevant infections included COVID-19, influenza, herpes zoster

Vaccine-Based Immunotherapy for Oropharyngeal and Nasopharyngeal Cancers.

Viral infections such as human papillomavirus (HPV) and Epstein-Barr virus (EBV) play a critical role in the onset of oropharyngeal (OPC) and nasopharyngeal cancer (NPC), respectively. Despite advancements in targeted therapies and immunotherapies, in the recurrent/metastatic setting, these tumors remain incurable diseases with poor prognosis. The development of therapeutic tumor vaccines, utilizing either neoantigens or oncoviral antigens, represents a promising addition to the cancer immunotherapy arsenal. Research on vaccine-based immunotherapy for OPC and NPC focuses on targeting viral ant

Nucleic acid vaccination strategies for ovarian cancer.

High grade serous carcinoma (HGSC) is one of the most lethal ovarian cancers that is characterised by asymptomatic tumour growth, insufficient knowledge of malignant cell origin and sub-optimal detection. HGSC has been recently shown to originate in the fallopian tube and not in the ovaries. Conventional treatments such as chemotherapy and surgery depend upon the stage of the disease and have resulted in higher rates of relapse. Hence, there is a need for alternative treatments. Differential antigen expression levels have been utilised for early detection of the cancer and could be employed in

Bilateral Iritis after Vaccine for Bladder Cancer.

PURPOSE: Bacille Calmette-Guérin (BCG) is a vaccine that can be instilled into the urinary bladder as immunotherapy against superficial bladder cancer. Several case reports have implicated intravesical BCG in the development of uveitis. Patients treated with BCG therapy may present with systemic symptoms resembling reactive arthritis and, less frequently, have ocular adverse effects including bilateral panuveitis or chorioretinitis. In all but three previously reported cases of uveitis associated with BCG treatment, HLA-B27 has been positive. No patients have been reported to be positive for r

Delivery of DNA-based cancer vaccine with polyethylenimine.

DNA-based vaccine directed to tumor-specific antigens is an attractive strategy in cancer prevention and therapy. In view of the poor immunogenicity of most tumor-associated antigens, studies were carried out here to examine the adjuvant effect of polyethylenimine (PEI), a cationic polymer widely used in non-viral gene delivery, on the efficacy of cancer vaccine strategy. Treatment of animals with the DNA/PEI complexes resulted in antigen-specific cell lysis and activation of B3Z cells, an ovalbumin/K(b)-specific cytotoxic clone that recognizes the target cells through the class I major histoc

Photodynamic therapy-generated vaccine for cancer therapy.

A target tumor-derived whole cancer cell therapeutic vaccine was developed based on an in vitro pre-treatment by photodynamic therapy (PDT) and was investigated using a poorly immunogenic tumor model. The vaccine was produced by incubating in vitro expanded mouse squamous cell carcinoma SCCVII cells for 1 h with photosensitizer benzoporphyrin derivative (BPD), then exposing to light (690 nm, 1 J/cm2) and finally to a lethal X-ray dose. Treatment of established subcutaneous SCCVII tumors growing in syngeneic C3H/HeN mice with 2 x 10(7) PDT-vaccine cells per mouse by a peritumoral injection prod

Vaccination strategies for the treatment and prevention of cervical cancer.

PURPOSE OF REVIEW: Immunotherapy of HPV-induced premalignant anogenital lesions and cervical cancer has made impressive progress. HPV as causative agent is targeted by prophylactic and therapeutic vaccination strategies. Preclinical and clinical studies have shown induction of natural and/or vaccine-induced immune responses. This review will summarize the status of vaccine development and clinical testing published since March 2003. RECENT FINDINGS: For prophylactic vaccines there is first clinical evidence of effectivity (ie, 100% protection from HPV infection and dysplasia by virus-like par

Vaccines for control of fertility and hormone dependent cancers.

Two vaccines, namely one inducing antibodies against hCG and the other against GnRH, are now in clinical trials. The hCG vaccine has entered Phase II clinical trials in three centres in India after successfully completing Phase I clinical studies in several centres in India and in four countries abroad. The vaccine was found to be devoid of side-effects; its effect was reversible. The available data on 179 cycles indicate that the vaccine prevents pregnancy at antibody titres above 50 ng/ml. A genetically engineered version of the vaccine has also been approved for trials in human lung cancer

mRNA vaccines in cancer immunotherapy: current progress and perspectives in solid tumors and hematologic malignancies.

The unprecedented success of mRNA vaccines during the COVID-19 pandemic has accelerated the development of nucleic acid-based therapeutics, particularly in oncology. Decades of foundational research on mRNA design, delivery, and immunogenicity have laid the groundwork for the application of mRNA vaccines in cancer treatment. Herein, we summarize the key principles of synthetic mRNA engineering, including the optimization of structural elements, nucleoside modification, and codon usage to improve stability, enhance translation efficiency, and modulate immune responses. We highlight diverse anti

Personalized mRNA Neoantigen Vaccines in Cancer: Linking Precision Antigen Selection, Immune Remodeling, and Checkpoint Blockade.

Personalized mRNA neoantigen vaccines represent an emerging form of precision cancer immunotherapy designed to generate de novo T-cell responses against patient-specific tumor mutations. Their therapeutic activity, however, depends not only on neoantigen selection and vaccine-induced immune priming but also on whether these responses can remain functional within an immunosuppressive tumor microenvironment. Immune checkpoint blockade provides a mechanistically complementary strategy by relieving inhibitory signaling that constrains vaccine-expanded tumor-reactive lymphocytes. This review examin

Dual prophylactic and therapeutic potential of iPSC-based vaccines and neoantigen discovery in colorectal cancer.

Rationale: Induced pluripotent stem cells (iPSCs) share transcriptomic similarities with cancer cells and express tumor-specific and tumor-associated antigens, highlighting their potential as cancer vaccines. Our previous study demonstrated that an iPSC-based vaccine effectively prevented tumor growth in various mouse models, including melanoma, breast, lung, and pancreatic cancers. However, the underlying mechanisms and the therapeutic efficacy of the iPSC-based vaccine remain unclear. Colorectal cancer (CRC), the third most common cancer with a rising incidence worldwide, presents an urgent …
